Cleavage and some modifications of the 7-amide group of the cephamycins

The cleavage and some modifications of the 7-amide group of cephamycins are described. Cephamycin derivatives 16b, c which were synthesized from the naturally occurring cephamycin C (16a) were converted to the corresponding oxamic acid derivatives 17a, e respectively by the reaction with oxalyl chloride and successive treatment with water. The reaction of the oxamic acid 17a with diphenylcarbodiimide gave 7-aminocephamycinoic acid (7-ACMA) benzhydryl ester (21a) which was further converted to cefoxitin (21c). These compounds 17a, b, c, d, e, f thus obtained from cephamycin C appear to be favorable intermediates for the syntheses of cephamycin analogues such as cefmetazole (28c).

A large body of appropriate calculations now exists and its main conclusions are summarized in this critical review. The present paper completes the recent reviews by Pyykk? (2004, 2005) (529 references).

Investigations of the author in thiophene chemistry developed during the last four decades are reviewed. Stereochemical studies, metallation and halogen-metal exchange, spectral studies (mainly those of NMR in thiophene and related series), studies of tautomerism in hydroxythiophenes and atropisomerism in bithienyl derivatives, ring opening of 3-thienyllithium derivatives and thiophene-1,1-dioxides, as well as studies of thieno fused heteroaromatic boron compounds are considered in detail.     

The chemistry of chromyl fluoride III. Reactions with inorganic systems

Reactions of CrO₂F₂ with MF or MF₂ gave the corresponding M₂CrO₂F₄ and MCrO₂F₄ fluorochromates. With the Lewis Acids (SO₃, TaF₅, SbF₅) and (CF₃CO)₂O known and new chromyl compounds [CrO₂(CF₃COO)₂, CrO₂(SO₃F)₂, CrO₂FTaF₆, CrO₂FSbF₆, CrO₂FSb₂F₁₁] were produced. Chromyl fluoride and inorganic salts (CF₃COONa and NaNO₃) produced the following complexes - Na₂CrO₂F₂(CF₃COO)₂ and Na₂CrO₂F₂(NO₃)₂. Unusual solid products were obtained with CrO₂F₂ and NO, NO₂, SO₂.A new method of preparing CrO₂F₂ is also presented.

The synthesis of substituted benzimidazole, benzothiazole, oxadiazole and triazine systems from dipotassium N-cyanodithioimidocarbonate are described.     

The tert-butyl group in chemistry and biology

The unique reactivity pattern elicited by the crowded tert-butyl group is highlighted by summarising characteristic applications. Starting from the use of this simple hydrocarbon moiety in chemical transformations, via its relevance in Nature and its implication in biosynthetic and biodegradation pathways, the way through to its possible application in biocatalytic processes is described.     

Studies on the analytical chemistry of iodonium compounds. Part III

Summary A procedure is described for the determination of the diphenyliodonium cation in the range 1.0 to 4.0 mg, by extraction from solution buffered at p_H 9.5 into chloroform as the diethyldithiocarbamate followed by conversion into the copper(II) complex. Absorbance measurements are made on the chloroform solution of the copper(II) diethyldithio-carbamate at 435 nm, and a linear relation exists between absorbance and cation content within the range specified. The optimum conditions for the extraction are discussed.

Zusammenfassung Ein Verfahren zur Bestimmung von Diphenyliodoniumkationen in der Größenordnung von 1,0 bis 4,0 mg wurde beschrieben. Es beruht auf deren Extraktion aus gepufferter Lösung (p_H=9,5) mit Chloroform als Diäthyldithiocarbamat und dessen Überführung in einen Kupfer(II)-komplex. Die Absorption des Kupfer(II)-diäthyldithiocarbamats in Chloroformlösung wurde bei 435 nm gemessen. Sie steht in linearem Verhältnis zur Kationenkonzentration. Die optimalen Bedingungen für die Extraktion werden erörtert.

Résumé On décrit un procédé pour le dosage du cation diphényliodonium dans le domaine de 1,0 à 4,0 mg, en l'extrayant d'une solution tamponnée à p_H 9,5 par le chloroforme, à l'état de diethyldithiocarbamate puis en le transformant en complexe du cuivre-II. On effectue les mesures de densité optique sur la solution dans le chloroforme du diéthyldithiocarbamate de cuivre-II à 435 nm; il existe une relation linéaire entre la densité optique et la teneur en cation dans le domaine spécifié. On discute les conditions optimales pour effectuer l'extraction.
