Four new chiral 1,2,3,4-tetrahydroisoquinoline-derived β-amino alcohols were synthesized from commercially available L-DOPA.These ligands were evaluated in the asymmetric addition of diethylzinc to benzaldehydes and showed different catalytic activities (up to 86% ee).The solvent played an important role in the enantioselective process.The transition state models were proposed to explain the reversion of the product configuration.  相似文献

A series of chiral C1- and C2-symmetric ferrocenyl Schiff bases (1a-c), ferrocenyl aminoalcohols (2a), ferrocenylphosphinamides (2b-c), 1,1′-ferrocenyl-diol (3), and 1,1′-ferrocenyl-disulfonamide (4) were prepared and employed as base catalysts or as ligand for titanium(IV) complexes in the asymmetric addition of diethylzinc to aromatic aldehydes. High enantioselectivity up to almost 100% ee was achieved for the alkylation of benzaldehyde and p-methoxybenzaldehyde with 1 or 3. In contrast, however, the β-aminoalcohol (2a) and phosphinamides (2b and c) that are ubiquitous classes of base catalysts for this reaction proved inefficient in our hands, regardless of the types of substrates or reaction conditions. Comparative studies show that there exist various reaction parameters governing not only chemical yields but also optical yields. These include steric and electronic environment of the substrate, the solvent, the reaction temperature, and the nature of the ferrocene moieties.  相似文献

A series of readily known enantiomerically pure 3-aminoquinazolinones 1ad were synthesised from easily accessible chiral pool α-hydroxy acids and α-amino acids in only four steps without any requirement of chromatography. These quinazolinones were examined as chiral ligands for catalytic enantioselective diethylzinc and phenylacetylene additions to aldehydes. For enantioselective alkylations, the effects of temperature, solvent, diethylzinc and ligand criteria were analysed, and the desired chiral alcohols were obtained in up to 86% ee. 3-Aminoquinazolinones 1ad were also shown to be very useful ligands in enantioselective alkynylations of aldehydes. Based upon the optimised conditions, the corresponding propargylic alcohols were obtained in up to 94% ee.  相似文献

Chiral BINOL was covalently anchored on two different pore sized mesoporous silica (SBA-15 (7.5 nm) and MCF (14 nm)). These heterogenized ligands were used in Ti-catalyzed asymmetric addition of diethylzinc to aldehydes. High catalytic activity with excellent enantioselectivity (up to 94% ee) for secondary alcohols was achieved using MCF supported chiral BINOL under heterogeneous reaction conditions. Good to excellent enantioselectivity (ee, 68–91%) was also achieved with various small to bulkier aldehydes. The MCF supported catalyst was reused in multiple catalytic runs without loss of enantioselectivity.  相似文献

The synthesis and resolution of new tridentate isoquinoline-derived ligands has been developed. The key steps in the synthetic sequence include successive, chemo-selective Suzuki-Miyaura cross-couplings of 1,3-dichloroisoquinoline with suitable arylboronic acids. The new ligands prepared in this manner were resolved either via molecular complexation with N-benzylcinchonidinium chloride as with 1-[3-(2-hydroxyphenyl)isoquinolin-1-yl]naphthalen-2-ol or via chromatographic separation of its epimeric camphorsulfonates as for 1,3-bis-(2-hydroxynaphthalen-1-yl)isoquinoline. 4-tert-Butyl-2-chloro-6-[1-(2-hydroxymethylnaphthalen-1-yl)isoquinolin-3-yl]phenol was resolved by chiral semi-preparative HPLC. The application of these ligands in the diethylzinc addition to aldehydes was investigated. In certain cases, the desired secondary alcohols were obtained in high yield with excellent enantiomeric excess (ee?>?99%) at low catalyst loading (1?mol%).  相似文献

A series of oxadiazines derived from l-phenylalanine bearing phenolic substituents have been synthesized in a multistep, one pot process. This process involves the reaction of a mixed anhydride with a β-hydrazino alcohol, methanesulfonylation of the alcohol moiety, and base induced cyclization. Enantioselective addition of diethylzinc to a series of aromatic aldehydes was developed using a modular amino acid amide chiral ligand (2S)‐3‐phenyl‐N‐((R)‐1‐phenyl‐ethyl)‐2‐(tosylamino)propanamide without using titanium complex. The catalytic system employing 10 mol% of 1g was found to promote the addition of diethylzinc (ZnEt2) to a wide range of aromatic aldehydes with electron‐donating and electron‐withdrawing substituents, giving up to 82% ee of the corresponding secondary alcohol under mild conditions. The readily available ferrocenyl substituted aziridinylmethanol 1 (FAM-1) was used as a chiral catalyst in the diethylzinc addition reaction to aromatic and aliphatic aldehydes to give secondary alcohols in high yields and up to 99% enantiomeric excess at room temperature. Purpose of the research was to determine the activity of chiral bis(sulfonamide) ligands derived from camphor in the addition of diethylzinc to benzaldehyde. Chiral bis(ketosulfonamides) and bis(hydroxysulfonamides), have been synthesized in a reaction of diamines with camphorsulfonic acid chloride. Their activity in a reaction of asymmetric addition of dialkylzinc to benzaldehyde in a presence of titanium(IV) tetraisopropoxide was determined. The bis(ketosulfonamide) ligands reveal low enantioselectivity, with the ee% not exceeding 12%. The bis(hydroxysulfonamides) reveal much higher asymmetric induction in the investigated ZnEt2 addition. The best enantiomeric excess (62%) has been observed for bis(hydroxysulfonamide) obtained from 1,3-diaminepropane. The yields of the reaction obtained after 18 h are 92–96%. Crystal structures have been solved for bis(ketosulfonamide) ligands obtained from diamines based on C2 to C4 chain. The (2R) configuration in the rings systems of bis(hydroxysulfonamide) containing the C3 bridge was also determined by the crystal structure analysis. The sulfonamides have been characterized by IR, 1H and 13C NMR.  相似文献

Enantiopure C2‐symmetric diol bidentate ligands have been synthesized in a straightforward manner through a three‐step reaction with good yields. The synthesized C2‐symmetric diol bidentate ligands were used in the addition of diethylzinc to various aromatic aldehydes, a general catalytic benchmark reaction, in order to assess their enantioselective induction properties. The enantioselective addition of diethylzinc to 1‐naphthaldehyde and 3‐chlorobenzaldehyde was achieved with an enantiomeric excess (ee) of up to 98%. All synthesized ligands were also evaluated in the addition of diethyzinc to aromatic aldehydes including an extra metal such as Ti(IV) (up to 99% ee). Several novel ligands that are based on the camphor skeleton or contain the O‐sulfonyl group were synthesized and used in the asymmetric addition of phenylacetylene to aldehydes.
