Design, synthesis, and biological evaluation of several domains of the thiopeptide antibiotic thiostrepton led to the discovery of a biologically active fragment. The biological properties of this novel small organic molecule include antibiotic activity against methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant Enterococcus faecalis (VREF) bacterial strains, as well as cytotoxic action against a number of cancer cell lines.  相似文献

A 15-step and fully stereocontrolled total synthesis of the title alkaloid 1 has been accomplished using the enantiomerically pure and enzymatically-derived cis-1,2-dihydrocatechol 2 as starting material. The final and pivotal step involved the intramolecular hetero-Michael addition of secondary amine 16 to a tethered enone moiety followed by trapping of the resulting enolate through its reaction with an adjacent ester residue.  相似文献

Eudistomidin B, isolated from a marine tunicate, was originally assigned a tetrahydro-β-carboline structure with a 2-p-tolylethanamine residue. Asymmetric total synthesis of the reported structure of natural product, which features an intramolecular diastereoselective Pictet-Spengler cyclization, suggests that the reported structure of the natural product needs to be revised.  相似文献

An efficient and low-cost protocol for the manual synthesis of Peptide Nucleic Acids is reported here. The protocol relies on coupling reactions carried out with 2.5 equiv of PNA monomers activated with HOBT/HBTU, in the presence of pyridine/NMM. The protocol has been tested on four PNA oligomers with a length ranging from 9 to 12 bases and a purine content up to 67%.  相似文献

Summary The aim of this investigation was the synthesis of -aryl propionic acids which might be expected to have antiinflammatory activity. The total synthesis of -(4-benzoylphenyl)--methyl propionic acid was monitored by thin-layer chromatography. The adsorption of the compounds of interest was found to be dependent upon the polarity of the eluting solvents. The observed retentions were mathematicaly analysed, and the system that showed the best separation of reactants evaluated. A least squares method was used to determine the best geometric curve expressing the relationship between values for Rf and eluent composition.Presented at the 14th International Symposium on Chromatography London, September, 1982  相似文献

Herein, we disclose an approach for the asymmetric synthesis of both enantiomers of an anti-inflammatory furanone. The approach is based on the utilization of a Morita-Baylis-Hillman adduct as starting material and has as key step a selective epoxide-opening/benzylic oxidation mediated by Palladium (II). This sequence afforded an advanced intermediate, which was used to accomplish the total synthesis. Experimental evidences allowed us to suggest a mechanistic proposal for the oxidation Palladium(II)-mediated.  相似文献

Efficient access: the 204-residue covalent-dimer vascular endothelial growth factor with full mitogenic activity was prepared from three unprotected peptide segments by one-pot native chemical ligations. An indole alkaloid bearing an oxazolone ring, christened almazolone, has been isolated from a new collection of Haraldiophyllum sp. from Dakar (Senegal), as an 88:12 mixture of (Z)/(E) stereoisomers. The relative ratio could be modified under controlled photochemical and thermal processes. The product (Z)-3-indolyl-2-(phenyl-propionylamino)-acrylic acid obtained by oxazolone ring opening has also been observed. Its formation has been confirmed by alkaline hydrolysis of (Z)-almazolone available from synthesis, where condensation of indole-3-carboxaldehyde with (3-phenylpropionylamino)-acetic acid represents the key step.  相似文献

Texaline, an antimycobacterial oxazole-containing alkaloid previously isolated from Amyris texana and A. elemifera, and related compounds have been synthesized in order to explore aspects of the structure-antituberculosis activity relationship. While texaline was found to be inactive in our assays, simpler diaryloxazoles were more active whilst also exhibiting modest toxic selectivity, leading to their identification as potential lead compounds.  相似文献

A strategy for a total synthesis of the structurally novel Lycopodium alkaloid lycopladine H has been investigated. Key steps that have been tested include: 1. a regioselective Diels-Alder cycloaddition of nitroethylene with an o-quinone ketal to produce the bicyclo[2.2.2]octane moiety of the alkaloid; 2. a stereoselective Henry reaction to generate the requisite functionality and configuration at C-5; 3. a stereoselective catalytic hydrogenation of a trisubstituted alkene to set the C-15 methyl configuration.  相似文献

A concise and step-economical total synthesis of biologically active frutinones has been achieved. Tributylphosphine (PBu3) efficiently induced the tandem acyl transfer-cyclization of carbonates 5 to afford 3-methoxycarbonylflavone derivatives 4 in excellent yields. A monoprotective approach, involving acetyl ester as unique protective group in oligosaccharides synthesis, has been developed. Starting from peracetylated monosaccharides and glycals, by using an efficient and selective chemo-enzymatic ‘one-pot’ strategy (a regioselective hydrolysis catalyzed by immobilized lipases followed by a chemical acyl migration), different carbohydrate acceptors, only protected with acetyl ester, can be achieved. If combined with the use of an acetylated glycosyl donor, the glycosylation reaction with these glycosyl acceptors leads to peracetylated oligosaccharides. These compounds can be directly used as intermediates for the synthesis of glycopeptides used as antitumoral vaccines and, at the end of the process, can be easily fully deprotected in only one step. Thus, these key building blocks have been successfully used in glycosylation reactions for an efficient construction of peracetylated disaccharides, such as the biological relevant lactosamine, in multigram scale. Subsequently, glycosylation with the 3OH-tetraacetyl-α-d-galactose, used as carbohydrate acceptor, allowed the synthesis of a peracetylated N-trisaccharidic precursor of the lacto-N-neo-tetraose antigen. Extending this strategy to a 3OH-di-acetyl galactal, one peracetylated precursor of the T tumor-associated carbohydrate antigen has been synthesized.This efficient approach, characterized by the use of the acetyl ester as only protecting group during all the synthetical steps expected, represents an easy and efficient alternative to the classical synthetic methods in carbohydrate chemistry that involve several protecting group manipulation.  相似文献
