Comparison of reaction products afforded by phenylguanidine derivatives with β-ketoesters or propiolic esters, respectively (synthesis of pyrimidones). 2-Anilino-imidazolines, when treated with either β-ketoesters or propiolic esters, yield two isomeric groups of pyrimidones. The isomerism is based on different positions of the carbonyl group in the pyrimidone ring. The mass spectra permit an unequivocal assignment of constitution I to the product formed with β-ketoesters and of constitution V to that formed with propiolic esters. Additionally, 2-(2-amino-anilino)-2-imidazoline (XII) when treated with methyl phenylpropiolate yields IX; while treatment with ethyl benzoylacetate yields XIII as an intermediate, which eliminates spontaneously one molecule of water to give the benzimidazole derivative XIV. Phenylguanidines (XV) add propiolic esters in the same way as do imidazoline derivatives. Photochemical cleavage of two carbon atoms with their adherent hydrogen atoms from the imidazole ring of the pyrimidones (V) leads to aminopyrimidine derivatives, e.g. XVI.  相似文献

An infrared spectral investigation of a number of pyrimidine and pyrimidone hexachloroantimonates is reported. Typical aromatic absorption bands found in pyrimidinium salts were absent in the pyrimidone cations. 2-And 4-Pyrimidone hexachloroantimonates, as well as their N-methyl- and N,N'-dimethyl derivatives, exhibit intense carbonyl absorption bands between 1720–1729 cm?1 (solid state) which corroborates N-protonation of pyrimidones.  相似文献

Abstract

The reaction of 2-(prop-2-ynylsulfanyl)pyrimidone 1 with various iodobenzenes in the presence of a palladium catalyst leads to regioselective cyclization to 3-benzylthiazolo [3,2-a] pyrimidones 3. The reaction of 4-(prop-2-ynylsulfanyl) pyrimidone 5 with various iodobenzenes in the same condition give the corresponding 3-benzylthiazolo[3,2-c]pyrimidones 6.  相似文献

Aryl pyrimidones are pharmacologically relevant compounds whose optical properties have only been partially explored. We report the synthesis and optical characterization of a series of aryl- and diaryl-2(1H)-pyrimidones. The electronic transitions of these chromophores are modulated by the extent of conjugation between the pendant phenyl ring and the pyrimidone core as well as the presence of electron-donating auxochromes. Monoprotonation of the pyrimidone ring results in large hyperchromic and bathochromic shifts as well as switching of fluorescence making these phenyl pyrimidones of interest as sensory materials.  相似文献

An efficient synthesis of functionalized pyrimidones via microwave-accelerated rearrangement reaction of amidoxime DMAD adducts is described. In most cases, the pyrimidone formation was furnished in reasonable yield after 2 min of microwave irradiation.  相似文献

α-Pyrones and pyrimidones are common structural motifs in natural products and bioactive compounds. They also display photochemistry that generates high-energy intermediates that may be capable of protein reactivity. A library of pyrones and pyrimidones was synthesized, and their potential to act as photoaffinity probes for nondirected affinity-based protein profiling in several crude cell lysates was evaluated. Further "proof-of-principle" experiments demonstrate that a pyrimidone tag on an appropriate scaffold is equally capable of proteome labeling as a benzophenone.  相似文献

Soluble poly(arylene thiophenes) containing amide bridge bonds and free nitrile groups have been synthesized through polycondensation of new arylene-bis(2-aminothiophene-3-carbonitrile)s with arylene- dicarboxylic acid dichlorides, and their physical and photochemical properties have been studied. Thermostable and acid-resistant poly(thiene pyrimidones) co ntaining main-chain thiophene, arylene, and pyrimidone fragments have been obtained via the isomerization polycyclomerization of the poly(arylene thiophenes).  相似文献

Methyl 2-[bis(acetyl)ethenyl]aminopropenoate ( 4 ) was prepared in 3 steps from acetylacetone ( 1 ) via 4-(N,N-dimethylamino)-3-acetylbut-3-en-2-one ( 2 ) and methyl N-[2,2-bis(acetyl)ethenyl]glycinate ( 3 ). Compound 4 reacts with N- and C-nucleophiles to give fused heterocyclic systems. Derivatives of pyrido[1,2-a]pyrimidones 14–16 and thiazolo[3,2-a]pyrimidones 17 and 18 were prepared from 2-aminopyridines and 2-aminothiazoles, respectively. With C-nucleophiles derivatives of pyrido[1,2-a]-pyridinone 19 and 2H-1-benzopyran-2-one 20–22 were prepared.  相似文献

Direct construction of fused aromatic ring systems by "zipper-mode" double C-H bond activation is described. Treatment of (Z)-3-bromo-N-(2-bromo-3-phenylprop-2-enyl)aniline derivatives with a catalytic amount of Pd(OAc)2 and PCy3.HBF4 in the presence of Cs2CO3 in dioxane affords 4,5-naphtho[3,2,1-cd]indole derivatives in good yields. Introduction of heterocycles such as benzofuran, benzothiophene, or indole moieties into the substrates leads to the efficient construction of highly fused heterocyclic aromatic ring systems via C-H bond activation of the heteroaromatic rings.  相似文献

In this personal account, our recent studies of novel synthetic methods of 3,4‐fused tricyclic indole derivatives using 3‐alkylidene indoline derivatives as versatile precursors are discussed. Two types of cascade reactions producing 3,4‐fused tricyclic 3‐alkylidene indolines were developed based on a palladium‐catalyzed intramolecular Heck insertion to an allene‐allylic amination cascade and a platinum‐catalyzed intramolecular Friedel‐Crafts type C?H coupling‐allylic amination cascade. Furthermore, three types of 3,4‐fused tricyclic indoles were accessible from a single 3‐alkylidene indoline precursor via acid‐promoted olefin isomerization or oxidative treatments. The application of the developed methods to the synthesis of natural products bearing a 3,4‐fused tricyclic indole skeleton, (?)‐aurantioclavine, fargesine, and synthetic studies of dragmacidin E are also highlighted.  相似文献

Benzothiophene, benzofuran, indole, and indene derivatives are privileged heterocyclic motifs. These are present in a wide range of bioactive natural products and pharmaceutical drugs and are the subject of materials science research. However, the construction of benzothiophene, benzofuran, indole, and indene frameworks have been long‐standing challenges to organic chemists. In this review, we classify the derivatives of four structures synthesized from 2‐benzylidene‐1‐benzofuran‐3‐one and their analogues in terms of their ring size (from three‐ to ten‐membered) and type (fused or spiro), as well as summarizing the developments of this field. Finally, we discuss the ring opening and 1,4‐addition reactions.  相似文献

The fused heterocycle 4-(phenylsulfonyl)-4H-furo[3,4-b]indole, which is an indole-2,3-quinodimethane synthetic analogue, is prepared in five steps from indole in 46% yield. A similar sequence is used to synthesize C-3 derivatives (3-methyl, 3-phenyl, and 3-heptyl). Thus, indole-3-carbaldehyde (1) is protected as the N-phenylsulfonyl derivative 2 and converted to the ethylene acetal 6. Lithiation at C-2 followed by treatment with an aldehyde affords the expected hydroxy acetals 7 and 8. Exposure to acid effects cyclization to the furoindoles 5 and 9. Furthermore, C-1 lithiation of furo[3,4-b]indole 9c followed by treatment with methyl iodide affords disubstituted furo[3,4-b]indole 10.  相似文献

Bis(2-lithioallyl)amines derived from bis(2-bromoallyl)amines undergo intramolecular carbometallation of a lithiated double bond, giving dilithiated dihydropyrroles. The cyclizations are promoted by N,N,N',N'-tetramethylethylenediamine (TMEDA). Reaction of these intermediates with electrophiles allows the preparation of some new fused and nonfused five-membered functionalized heterocycles. Although 2-lithioallylamines do not suffer intermolecular carbometallation, dimerization products are obtained with their copper or zirconium derivatives. Finally, the application of this new reaction to 2-lithio-N-(2-lithioallyl)anilines leads to 3-lithiomethylindole derivatives, which are transformed to functionalized indole derivatives by reaction with electrophiles.  相似文献

Tubulin inhibitors can interfere with normal cell mitosis and inhibit cell proliferation through interfering with the normal structure and function of microtubules, forming spindle filaments. Indole, as a privileged pharmacological skeleton, has been widely used in anti-cancer inhibitors. A variety of alkaloids containing an indole core obtained from natural sources have been proven to inhibit tubulin polymerization, and an ever-increasing number of synthetic indole-based tubulin inhibitors have been reported. Among these, several kinds of indole-based derivatives, such as TMP analogues, aroylindoles, arylthioindoles, fused indole, carbazoles, azacarbolines, alkaloid nortopsentin analogues and bis-indole derivatives, have shown good inhibition activities towards tubulin polymerization. The binding modes and SARs investigations of synthetic indole derivatives, along with a brief mechanism on their anti-tubulin activity, are presented in this review.  相似文献

Palladium‐catalyzed cascade C? H alkenylation and arylation provides convenient access to polycyclic aromatic compounds. Treatment of 3‐bromoaniline derivatives bearing a bromocinnamyl group on the nitrogen atom with a catalytic amount of [Pd(OAc)2] and PCy3?HBF4 in the presence of Cs2CO3 in dioxane affords naphthalene‐fused indole derivatives in good yields. This double cyclization reaction is also applicable to heterocyclic substrates, giving fused indoles containing a heteroaromatic ring such as dibenzofuran, dibenzothiophene, carbazole, indole, or benzofuran through heterocyclic C? H arylation. When using a 2,6‐unsubstituted aniline derivative, the first C? H arylation preferentially proceeds at the more hindered position of the aniline ring.  相似文献

[STRUCTURE: SEE TEXT] Polycyclic ring-fused 2-pyridones (5a-e and 9a-e) have been prepared via a microwave-assisted acyl-ketene imine cyclocondensation. Starting from 3,4-dihydroisoquinolines (4a-b) or 3,4-dihydroharman (8), fused 2-pyridones could be prepared in a one-step procedure. By using either Meldrum's acid derivatives (1a-d) or 1,3-dioxine-4-ones (7a-b) as acyl-ketene sources, mono- or disubstitution of the fused 2-pyridone ring could be accomplished. As an application of the method, a formal synthesis of the indole alkaloid sempervilam was performed.  相似文献
