A novel, powerful analytical technique, preparative temperature rising elution fractionation (prep TREF)/high-temperature (HT)-HPLC/Fourier transform infrared spectroscopy (FTIR)/high-performance differential scanning calorimetry (HPer DSC)), has been introduced to study the correlation between the polymer chain microstructure and the thermal behaviour of various components in a complex impact polypropylene copolymer (IPC). For the comprehensive analysis of this complex material, in a first step, prep TREF is used to produce less complex but still heterogeneous fractions. These chemically heterogeneous fractions are completely separated by using a highly selective chromatographic separation method—high-temperature solvent gradient HPLC. The detailed structural and thermal analysis of the HPLC fractions was conducted by offline coupling of HT-HPLC with FTIR spectroscopy and a novel DSC method—HPer DSC. Three chemically different components were identified in the mid-elution temperature TREF fractions. For the first component, identified as isotactic polypropylene homopolymer by FTIR, the macromolecular chain length is found to be an important factor affecting the melting and crystallisation behaviour. The second component relates to ethylene–propylene copolymer molecules with varying ethylene monomer distributions and propylene tacticity distributions. For the polyethylene component (last eluting component in all semi-crystalline TREF fractions), it was found that branching produced defects in the long crystallisable ethylene sequences that affected the thermal properties. The different species exhibit distinctively different melting and crystallisation behaviour, as documented by HPer DSC. Using this novel approach of hyphenated techniques, the chain structure and melting and crystallisation behaviour of different components in a complex copolymer were investigated systematically.

The aim of this study was to learn to what extent the selected instrumental techniques, differential scanning calorimetry (DSC), as well as Fourier-transform infrared (FTIR) and Raman spectroscopies, can be used to detect both organic or inorganic magnesium compounds in the dietary supplements and medicinal products. Besides magnesium compounds as the active pharmaceutical ingredients (APIs), the preparations contain also other organic and inorganic APIs and several excipients. The study will be extended over the analysis of the products manufactured by various firms but containing the same API at different levels. In this way, it will be possible to assess the impact of excipients on the DSC scans and the FTIR and Raman spectra of a dominant constituent present in a studied preparation. The study on thirty commercially available dietary supplements and medicinal products has shown that in the majority of cases the DSC, FTIR and Raman techniques could be used for the detection of APIs in these commercial products. This was possible with the aid of the endothermic DSC peaks and the so-called matching factors of the FTIR and Raman spectra to those of substances used as standards. Both the complex composition and low levels of API in the studied preparations have been identified as the factors which have a strong impact on the usefulness of the three techniques for the detection of APIs in the dietary and medicinal products.  相似文献

The applicability of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) to the qualitative and quantitative analysis of most mammalian phospholipid (PL) classes was demonstrated in a crude extract of porcine lens membranes. When 2,5-dihydroxybenzoic acid (DHB) was used as the matrix, positive-ion spectra allowed the accurate quantification of phosphatidylcholines (PCs) and sphingomyelins (SMs). Other PLs such as phosphatidylethanolamines (PEs), phosphatidylethanolamine plasmalogens (PEps), phosphatidylethanolamine ethers (PEes) and phosphatidylserines (PSs), could also be detected, but their lower ionization efficiency led to negative errors in their quantification. Despite this limitation, it was possible to determine relative changes among PLs extracted from cortical and nuclear regions. Negative-ion spectra were acquired with the use of p-nitroaniline (PNA) as the matrix. Because neither PCs nor SMs produce negative ions, other PL classes can be analyzed selectively. The absolute quantification of the various PL classes detectable in negative-ion spectra was also affected by differences in ionization efficiencies. However, the trends in compositional changes between cortical and nuclear-fiber PLs were in agreement with those obtained by (31)P NMR spectroscopy. MALDI-TOFMS also offers the possibility of studying variations in the acyl-chain distribution of the various species comprising each PL class. For porcine lenses, PCs, PEs and phosphatidylinositols (PIs) exhibited the greatest depletions in going from cortical to nuclear membranes. Among their individual species, those with two or more sites of unsaturation suffered the most significant reduction.  相似文献

Solid-state 13C nuclear magnetic resonance (13C NMR) with magic-angle spinning (MAS) and with cross-polarization and magic-angle spinning (CP/MAS) spectra, and differential scanning calorimetry (DSC) techniques were used to obtain structural data from a sample of acetyl aleuritolic acid (AAA) extracted from the stem bark of Croton cajucara Benth. (Euphorbiaceae) and recrystallized from acetone. Since solid-state 13C NMR results suggested the presence of more than one molecule in the unitary cell for the AAA, DSC analysis and molecular modeling calculations were used to access this possibility. The absence of phase transition peaks in the DSC spectra and the dimeric models of AAA simulated using the semi-empirical PM3 method are in agreement with that proposal.  相似文献

Thermogravimetry (TG), differential thermal analysis (DTA), differential scanning calorimetry (DSC) as well as X-ray diffraction powder (DRX) patterns and Fourier transformed infrared spectroscopy (FTIR) were used to study ketoprofen, ibuprofen, and naproxen. The chemical or physical properties of the studied compounds were established and when possible by X-ray powder diffractometry and/or infrared spectroscopy were used. In this investigation, quantum chemical approach was used to determine the molecular structures using Becke three-parameter hybrid method and the Lee–Yang–Par (LYP) correlation functional. The performed molecular calculations in this work were done using the Gaussian 03 routine. Theoretical calculations help in interpretations of FTIR spectra supplying structural and physicochemical parameters.  相似文献

In certain fire situations, a structural or load‐bearing polymer matrix composite (PMC) may be exposed to excessive thermal loads that degrade the matrix. In this paper, we report the results of a study to assess the utility of ultrasonic spectroscopy as a means of assessing the residual physical and mechanical characteristics of PMCs exposed to excessive thermal loads. We show that the measured power spectra of ultrasonic energy correlates with performance of graphite fiber epoxy matrix composites exposed to thermal degradation. Unidirectional composites were exposed to short term–high intensity thermal loads at one end of the specimen. Thus, inducing a thermal gradient along the length of the specimen. Simultaneous thermogravimetric analysis–differential scanning calorimetry (TGA/DSC) and Fourier transform infrared spectroscopy (FTIR) analysis of the aged specimens revealed a gradient in thermal degradation. The thermal loads induced substantial degradation of the composite. However, the amplitude of the power spectra is observed to increase gradually then sharply prior to its complete attenuation due to delaminations. Mode I fracture toughness tests correlate with the observed changes in the ultrasonic spectra. FTIR, TGA/DSC, fracture toughness, and ultrasonic spectral analysis all indicate the same critical temperature at which thermally induced damage sharply increased. © 1999 John Wiley & Sons, Inc. Characterization of PVDF membranes by vibrational spectroscopy   总被引:3,自引:0,他引:3  
In order to investigate the effectiveness of vibrational spectroscopy in the characterization of polymeric membranes, several poly(vinylidene fluoride) (PVDF) membranes with different porous structures were prepared by the phase inversion process using different casting solvents. An accurate analysis of the Fourier transform Raman (FT-Raman) and the Fourier transform infrared (FTIR) spectra was performed for each sample and the scanning electron microscopy (SEM) results were noted. To highlight the specific problems related to porosity and surface roughness in the acquisition of spectra by different sampling techniques, the attenuated total reflection (ATR) and photoacoustic spectroscopy (PAS) spectra were compared with corresponding spectra obtained from dense films. A detailed analysis of these spectra highlighted their ability in determining the differences in the polymer structure between the two membrane sides. This indicates that (considering the results given by all the different techniques) thorough qualitative membrane characterization can generally be achieved. Moreover, the good quality spectra of the PVDF membrane provide information on a portion of material which depends on its structure, highlighting the usefulness of FTIR-PAS in studying porous materials which, as a rule, give low quality infrared spectra when other sampling techniques are used. However, the complex and inhomogeneous structure of these materials can make quantitative analysis more, or less, difficult.  相似文献

In this article we investigated the compositional and structural characteristics of the principal biomolecules such as carbohydrates, proteins, lipids, nucleic acids and chlorophyll pigments present in biofilm cultures of Ostreopsis spp. and in batch cultures of Ostreopsis ovata. Our approach based on the use of infrared (FTIR) and near infrared (FTNIR) spectroscopy showed the marked differences existing between biofilm cultures and batch cultures. FTIR spectroscopy showed the higher contents of polysaccharides and chlorophyll pigments in O. ovata from batch cultures with respect to Ostreopsis spp. Second derivative FTIR spectroscopy showed different features concerning the secondary structure of proteins because in O. ovata samples the beta sheet and beta turn structures were observed whereas in Ostreopsis spp. samples the alpha helix structure was the most evident. FTNIR spectroscopy showed other structural differences observed existing between O. ovata and Ostreopsis spp. mainly related to hydrogen bond interactions determining more packed structures in the nucleus of O. ovata. In addition, the interpretation of FTIR and FTNIR spectral information was also supported by the application of two statistical methods, the independent component analysis (ICA) and the spectral cross correlation analysis (SCCA). ICA was used as spectral deconvolution technique to separate the effects of the interference bicarbonate ion from algal FTIR spectra so to verify the high similar qualitative composition of the three biofilm samples of Ostreopsis spp. At last, SCCA applied to FTIR and FTNIR spectra was useful to evidence some structural differences involving -CH and CH(2) groups of aliphatic chains in O. ovata and Ostreopsis spp. samples. Though preliminary, these results agree with some previous studies suggesting that the presence of different ecophysiological characteristics in O. ovata and Ostreopsis spp. depending on the parameters related to the condition growth.  相似文献

First-line drugs (rifampicin, RIF; isoniazid, INH; ethambutol, ETA; and pyrazinamide, PZA) recommended in conventional treatment of tuberculosis were analyzed in 1:1 w/w binary mixtures with microcrystalline cellulose MC 101 (CEL) and lactose supertab® (LAC) by differential scanning calorimetry (DSC), thermogravimetry (TG), differential thermal analysis (DTA), and Fourier transformed infrared analysis (FTIR) as part of development of fixed dose combination (FDC) tablets. Evidence of interaction between drug and pharmaceutical excipients was supposed when peaks disappearance or shifting were observed on DTA and DSC curves, as well as decreasing of decomposition temperature onset and TG profiles, comparing to pure species data submitted to the same conditions. LAC was showed to interact with RIF (absence of drug fusion and recrystallization events on DSC/DTA curves); INH (thermal events of the mixtures different from those observed for drug and excipient pure in DSC/DTA curves); PZA (decrease on drug fusion peak in DSC/DTA curves), and ETA (shift on drug onset fusion and absence of pure LAC events on DSC/DTA curves). In all cases, an important decrease on the temperature of drug decomposition was verified for the mixtures (TG analysis). However, FTIR analysis showed good correlation between theoretical and experimental drug-LAC spectra except for INH–LAC mixture, evidencing high incompatibility between these two species and suggesting that those interactions with PZA and RIF were thermally induced. No evidence of incompatibilities in CEL mixtures was observed to any of the four-studied drugs.  相似文献

Previously Fourier transform infrared(FTIR) spectroscopy has been applied to detecting thyroid cancer during operations and to discriminating cervical metastatic ones from non-metastatic lymph nodes. This study explored the possibility of establishing a sensitive, accurate and noninvasive screen or diagnosis by preoperative FTIR spectroscopy. 111 patients undergone a thyroid operation and 50 healthy volunteers were enrolled in the study. The FTIR spectra were obtained by two mid-infrared optical fibers with an attenuated total reflectance(ATR) probe closely contacting the subjects' skin on the thyroid nodules. The FTIR spectra obtained from normal thyroid, nodular goiter(NG) and papillary thyroid carcinoma(PTC) patients were compared. A Fisher's discriminant analysis was created based on these data. There were 41 PTC patients and 70 NG patients according to their histopathological examinations. A total of 23(of 39) parameters were statistically different among the three groups(P<0.05). The F1300 and F1080 parameters were significantly different between the three groups. In total, 9 out of 39 FTIR parameters were selected as independent factors by the Wilks' lambda stepwise discriminant analysis. The discrimination accuracy of papillary thyroid carcinoma in the three groups was 88.8%. Surface detection of PTC by FTIR spectroscopy is feasible. FTIR spectroscopy can be used for rapid and noninvasive PTC screen and auxiliary diagnosis.  相似文献

Various polyethylenes (PEs) and the effects of high-energy radiation on their structures were widely studied in the past using conventional Differential Scanning Calorimetry (DSC) measurements. In this work, we used the Temperature Modulated Differential Scanning Calorimetry (TMDSC) technique in order to obtain more information about the influence of the initial structural differences and gamma radiation on the evolution in structure and thermal properties of different polyethylenes. For this reason, low density polyethylene (LDPE), linear low density polyethylene (LLDPE) and high density polyethylene (HDPE) samples were exposed to gamma radiation, in air, to a wide range of absorbed doses (up to 2400 kGy). The separation of the total heat flow TMDSC signal into a reversing and non-reversing part enabled us to observe the low-temperature enthalpy relaxation (related to the existence of the “rigid amorphous phase”) and recrystallisation processes, as well as to follow their radiation-induced evolution and/or that of melting in a more revealing manner compared to the case of the conventional DSC. Consequently, our results indicate that TMDSC could improve the understanding of radiation-induced effects in polymers.  相似文献

Four new cross-linked polymers poly(vinyl m-nitrobenzene)-polyglycidylazides (PVMNB-GAPs) were successfully synthesized using toluene diisocyanate as the cross-linking agent. Their structure was confirmed by their FTIR, UV–Vis, 1H and 13C NMR spectroscopy. Moreover, the thermal properties of cross-linked polymers were evaluated by DTA, TGA and DSC techniques, which confirmed that synthesized polymers exhibited good resistance to thermal decomposition up to 200°C. In addition, their compatibility with the main energetic components of 2,4,6-trinitrotoluene-based melt-cast explosives were also evaluated by the non-isothermal differential thermal analysis method.  相似文献

For a better understanding of the molecular and organizational changes in human dermis, biophysical methods were tested. The aim of this study was to find suitable and reproducible biomarkers for further clinical studies on intrinsic and extrinsic aging of dermis. Thermoporometry, hydric organization and thermal transitions of fresh and frozen skins were determined by differential scanning calorimetry (DSC). Fourier transform infrared spectroscopy (FTIR) was used to identify the absorption bands of the dermis especially in the 1800–1000 cm?1 zone and to discriminate between the different secondary structures of proteins. A widening of the pore size distribution is evidenced with freezing, but there is no significant difference between the hydric organization and the endothermic collagen denaturation of fresh and frozen skins. The global FTIR spectra and the second derivative spectra in the scanned zone are also identical in fresh and frozen dermis, validating the storage protocol. DSC and FTIR are well-suited techniques to characterize human skin, giving accurate results with high reproducibility. The acquisition of thermal and vibrational biomarkers of the skin at the mesoscale and nanoscale contributes to its better knowledge and is promising for further studies on skin aging.  相似文献

通过粘度、紫外光谱、红外光谱和DSC谱的测定,探讨了PVP/HEC分子间的缔合机理.结果表明,PvP/HEC复合体系相对于单一体系产生了粘度的负协同效应,NaCl的加入使单一体系和复合体系的粘度均下降;在紫外可见吸收光谱中,PVP/HEC复合体系的最大吸收波长λmax相对于PVP、HEC各自的λmax都发生红移;PVP/HEC复合物的红外光谱在2370 cm-1出现新的吸收峰;DSC谱中复合物的峰温高于简单混合物,而其焓变低于简单混合物,这一切均证明HEC与PVP之间未发生疏水缔合,而是HEC的羟基与PVP的羰基之间通过氢键缔合成复合物,复合物的分子链排列较为紧密.  相似文献

This paper reports the thermal stability and thermal degradation of six derivatives of indole by means of TG-DSC (in air) and TG-FTIR (in nitrogen) techniques. The compounds were also characterized by infrared spectroscopy. In addition, IR spectra were calculated and compared with the experimental data. In particular, the potential energy distribution analysis was performed to assign IR signals. The studied compounds are characterized by good thermal stability in oxidizing and inert atmospheres which is important for potential medical application. Thermogravimetric measurements in air atmosphere showed that the decomposition of compounds proceeds in two or three main stages. The thermal degradation of compounds is preceded by the melting process. The pyrolysis of samples is a one-step process. Together with the analyses performed in nitrogen, the FTIR spectra of the evolved gas phase products were recorded. On the FTIR spectra of gaseous products, only the bands of water, carbon dioxide and carbon oxide molecules are present. In the case of indole derivatives containing the p-chlorobenzyl substituent in position 1, the bands of anisole, p-chlorotoluene and chlorobenzene also appear.  相似文献

Differential scanning calorimetry (DSC), isothermal stress testing–Fourier transform infrared spectroscopy (IST–FTIR), isothermal stress testing–high-performance liquid chromatography, and powder X-ray diffraction (PDRX) were used as screening techniques for assessing the compatibility of tobramycin with some currently employed ophthalmic excipients. In the first phase of the study, DSC was used as a tool to detect any interaction. The absolute value of the difference between the enthalpy of the pure tobramycin melting peak and that of its melting peak in the different analyzed mixtures was chosen as a parameter of the drug–excipient interaction degree. DSC results demonstrated that benzalkonium chloride, monobasic sodium phosphate, boric acid, edetate disodium, sodium metabisulfite, thimerosal, and potassium sorbate interact with tobramycin. Taking into account these results, it could be suggested that some of the changes observed in the IST–FTIR spectra of binary blends of tobramycin and some of the excipients would account for a possible interaction between the mixture component. In this study, PDRX did not provide much information, since only tobramycin–thimerosal interactions could be detected. DSC and IST–FTIR are suitable and simple methods for the detection of potential incompatibilities between active pharmaceutical ingredient (API) and excipients.

The purpose of this research study was evaluation of the utility of two common multivariate techniques, agglomerative cluster analysis (CA) and principal component analysis (PCA), as supplementary means of detecting incompatibilities, which can occur between active pharmaceutical ingredients and excipients at the preformulation stage of a solid dosage form. For the detection of incompatibilities between atenolol (beta blocker) and selected excipients (mannitol, lactose, starch, methylcellulose, β-cyclodextrin, meglumine, chitosan, polyvinylpyrrolidone and magnesium stearate), the thermogravimetry (TG), differential scanning calorimetry (DSC) and Fourier transform infrared spectroscopy (FTIR) were chosen. The results have shown that compatibility between atenolol and an excipient can be identified in a CA dendrogram by two large clusters, from which one groups an excipient and physical mixtures with a high concentration of the excipient. Another cluster encompasses atenolol and mixtures with a high content of the drug. In the PCA plot, all samples are located along the first principal component axis (PC1), beginning from a single component located with the most negative PC1 value, through mixtures with gradually varying concentration of both ingredients, till the second component located close to the most positive PC1 values. The results have shown that CA and PCA fulfil their role as supporting techniques in the interpretation of the data acquired from the TG curves, and the obtained data are compatible with the results of DSC and FTIR analyses.  相似文献

FTIR spectra of polycrystalline samples of adenosine and uridine, pure and containing small (<10%) quantity of N(O)H or N(O)D groups, were measured in KBr pellets from 4000 to 400 cm(-1) at temperatures from 300 to 20 K. For the first time, the bands of narrow isotopically decoupled proton stretching vibration nu1 mode of NH- and OH- groups were found and assigned to ordered hydrogen bonds according to crystal structural data for both nucleosides. The FTIR adenosine spectra in the out-of-plane bending proton nu4 mode range (lower than 1000 cm(-1)) of N(O)H groups revealed at low temperature at least twice more bands, than in the nu1 range, which are influenced by isotopic exchange and (or) cooling. Almost all of them have their counterparts in the N(O)D substance spectrum with an isotopic frequency ratio of 1.30-1.40. These bands were assigned to the differently H-bound disordered NH and OH protons, which could not be seen with crystal structural methods. The energy and length of different H-bonds were estimated from peak positions of both mode bands (as the red shift of nu1 or blue shift of nu4 relatively free molecules) with well-established empirical correlations between spectral, thermodynamic and structural parameters of hydrogen bonds. The detection of phospholipids (PLs) by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was demonstrated nearly a decade ago. However, its use as a conventional tool for PL analysis has been hindered by ambiguities in peak assignments caused by spectral overlaps and difficulties in the detection of some PL classes when analytes with positively charged head groups, such as sphingomyelins (SMs) and phosphatidylcholines (PCs) are present. In this work, either a strong cation-exchange resin or CsCl crystals were added directly to the PL samples to reduce spectral complexity and enhance sensitivity. The quantitative exchange resulted in virtually only protonated or Cs+ adducts. To alleviate difficulties in the detection and identification of PL classes with ionization efficiencies lower than those of SMs and PCs, improvements in the sensitivity of negative-ion mass spectra were sought. For this purpose, several neutral and basic matrices were tried. Among them, p-nitroaniline (PNA) proved to be an advantageous alternative to the use of 2,5-dihydroxybenzoic acid (DHB), the most commonly used matrix in PL analysis. Because of its lower acidity, PNA increased the relative amount of deprotonated species and improved the sensitivity of negative-ion mass spectra. It was possible to confirm peak assignments for PL classes that normally give weak signals when DHB is used. Noteworthy is the detection (in both positive and negative modes) and conclusive identification of species in natural mixtures of phosphatidylethanolamines (PEs) and PE plasmalogens (PEps). PNA allowed the identification of PEs and PEps even in mixtures containing SMs and PCs. Although some cations related to PCs and PEs overlapped in positive-ion spectra, these interferences were eliminated in the negative mode as only the deprotonated forms of PEs and PEps were detectable and those of SMs and PCs were absent owing to their neutrality.  相似文献
