9-羟基-9-咕吨羧酸甲酯是合成咕吨类抗胆碱能药物的一种重要的中间体.本文以9-咕吨羧酸为原料,通过微波催化先合成了中间体9-咕吨羧酸甲酯,然后很简便地得到了9-羟基-9-咕吨羧酸甲酯.在中间体的合成中,利用正交设计,探讨了微波辐射时间、功率以及催化剂用量对反应的影响,实验结果表明微波辐射时间的影响最为显著.  相似文献

以醋酸酐为溶剂, 将9, 9-二(4-羟基苯基)呫吨与浓硝酸在15 ℃左右发生硝化反应1.5 h, 形成中间体-9, 9-二(3-硝基-4-羟基苯基)呫吨, 产率为88.5%;继而以Pd/C 为催化剂, 80%的水合肼为还原剂, 在乙醇和1, 4-二氧六环混合溶剂中, 于70 ℃将中间体进行还原反应10 h, 得到目标产物9, 9-二(3-氨基-4-羟基苯基)呫吨, 产率为90.8%, 两步反应总收率为80.4%。 中间体与目标化合物经NMR、FTIR和元素分析等确定了它们的结构。  相似文献

以呫吨酮为原料,经二氯亚砜氯化得到9,9-二氯呫吨中间体,无需分离纯化,将其分别与邻甲酚和2,6-二甲基苯酚进行取代反应,制得2个含呫吨结构的双酚化合物9,9-二(4-羟基-3-甲基苯基)呫吨和9,9-二(4-羟基-3,5-二甲基苯基) 呫吨,产率分别为81.0%和80.5%。 用FTIR、1H NMR、13C NMR和元素分析对其组成和结构进行了表征。 该法具有操作简单、反应条件温和及收率高等优点。  相似文献

以水杨酸、间苯三酚为原料合成了1,3-二羟基呫吨酮, 经醚化、环化反应得到1-羟基呋喃并呫吨酮3a和3b, 再经Mannich反应合成了10个呋喃并呫吨酮衍生物4和5, 接着通过季铵化反应得到相应的10个季铵盐6和7. 运用IR、一维和二维NMR、MS、元素分析等对化合物进行了结构表征, 考察了化合物4~7对乙酰胆碱酯酶的抑制作用及化合物6, 7的抗癌活性. 结果表明: 化合物4~7对乙酰胆碱酯酶具有较好的抑制活性, IC50=2.0~12.4 μmol/L; 化合物6, 7对肝癌(HepG2)、肺癌(SPC-A)、口腔上皮癌(KB)、乳腺癌(MCF-7)这四种癌细胞株的增殖均有抑制作用, 其中化合物6c对癌细胞株HepG2、化合物7d对癌细胞株MCF-7的抑制作用最强, IC50分别为0.82和0.77 μmol/L.  相似文献

以间苯三酚为起始原料,在伊顿试剂的催化作用下,与不同取代水杨酸反应合成1,3-二羟基呫吨酮类化合物3a~3l 12个,收率63%~87%,并探讨了反应机理及结构-反应活性关系.利用紫外分光光度法测定了所合成化合物对亚硝酸盐的清除作用,结果表明,在37℃、pH 3及反应时间为30 min条件下,1,3-呫吨酮类化合物对亚硝酸根均有较好的清除作用,其中化合物3f和3b的清除作用最强,清除率达34%.同时对1,3-二羟基呫吨酮类化合物清除亚硝酸盐的作用机制及构效关系进行了探讨.  相似文献

在碘化亚铜、四丁基溴化铵和磷酸钾存在下,9,9-二(4-羟基苯基)呫吨(1)和4-甲基碘苯(2)于N,N-二甲基甲酰胺溶剂中发生Ullmann偶联反应,加热回流反应24 h,以95%的产率合成了中间体--9,9-二[4-(4-甲基苯氧基)苯基]呫吨(3),继而加入催化量的N-溴代丁二酰亚胺并在光照条件下,将中间体3氧化得到一种新型芳香族二羧酸--9,9-二[4-(4-羧基苯氧基)苯基]呫吨(4),其产率为84%,二步反应总收率为79.8%。 目标化合物4经1H NMR、13C NMR、IR和元素分析测试技术确定了其结构。 该法具有原料易得,操作简单,反应条件温和,收率高等优点。  相似文献

以呫吨酮为起始原料,经二氯亚砜作用得到9,9-二氯呫吨中间体,继而与苯酚发生取代反应,探索了"一锅、二步"法制备含呫吨结构的双酚单体———9,9-二(4-羟基苯基)呫吨(BHPX)的新方法.在氮气保护下,在K2CO3存在下,以环丁砜为溶剂,将其与1,3-二(4-氟苯甲酰基)苯进行亲核缩聚反应,制得了一种高分子量的(数均分子量为45000,多分散性指数为1.9)新型含呫吨结构的Cardo型聚醚醚酮酮(PEEKK-X),并用FTIR,DSC,TG及WAXD等方法对其进行了分析表征.结果表明,所制得的聚合物(PEEKK-X)为无定形结构,其玻璃化转变温度(Tg)为215℃;在氮气气氛中5%的热失重温度(Td)为558℃,700℃时的残炭率为67%;在常温下易溶于非质子极性溶剂(如NMP、DMF和DMAc)和极性较弱的溶剂如CHCl3中.该聚合物可通过溶液浇铸成膜,所得到的薄膜韧性好,透明且耐折,其拉伸强度为81.5 MPa,杨氏膜量为2.48 GPa,断裂伸长率为16.8%.  相似文献

系统地研究了9-(邻羧基苯基)-6-羟基-3H-呫吨酮-3(FL)与Fe~(2 )、Tween-80褪色反应测定铁的光度条件。试验表明,在HOAc-NaOAc缓冲溶液中和非离子表面活性剂Tween-80存在的条件下,体系吸光度明显降低,利用这一现象可直接测定铁,其表观摩尔吸光系数为8.4×10~4,线性范围0.5~9.0μg/25ml,方法选择性较高,一定量的常见离子不干扰测定,可直接用于矿泉水中铁的测定。  相似文献

以(9Z)-9-去氧-9-羟基亚胺基红霉素A为原料,甲醇/水为反应体系,对甲苯磺酰氯为催化剂,硼氢化钠为还原剂,首次用一步法合成了9-去氧-8a-氮杂-8a-同型红霉素A,其结构经1H NMR和FAB-MS表征。在pH8~9,于-3℃反应9 h的较佳反应条件下,收率94.1%,纯度93.0%。  相似文献

A new approach to the synthesis of novel medicinally-important mono- and bis-9-anilinoacridine (9-AnA) peptidyl derivatives is described. The method generates efficiently 9-AnAs with variable spacer lengths and charged, polar or hydrophobic residues at desired positions, which can increase binding affinity, conformational stability, intracellular transport and/or biological activity. The synthetic routes reported in this work are both general and applicable, and significantly expand the scope of potential 9-aminoacridine (9-AA) anticancer candidates.  相似文献

C-Functionalization of pyrrolo[3,2-d]pyrimidine scaffold in positions 2, 4, and 7 using cross-coupling reactions was performed. Thus, 2-(5-(benzyloxymethyl)-2,4-dichloro-5H-pyrrolo[3,2-d]pyrimidin-7-yl)ethanol, a versatile synthetic precursor for 9-deazapurines and 4,6-diazaindoles, was prepared by vinylation of the corresponding iodide followed by hydroboration of the double bond. A synthesis of 9-(1,2-dihydroxyethyl)-9-deazaadenine, a 9-deaza-1′-nor congener to antiviral DHPA, was developed. In addition, an abnormal regioselectivity in methylalumination of the terminal triple bond in position 7 of the pyrrolo[3,2-d]pyrimidine scaffold leading to a transformation into (Z)-prop-1-enyl was observed.  相似文献

A protocol has been developed to purify 9-(trifluoroacetyl)anthracene from anthracene, the major impurity associated in its synthesis. The protocol utilizes a chemoselective thermal Diels–Alder reaction, employing maleic anhydride as the dienophile. The resulting Diels–Alder adduct can then be removed by extractive workup followed by recrystallization to give 9-(trifluoroacetyl)anthracene with ≥99% chemical purity.  相似文献

An expeditious and convenient method to synthesize 9-allenylpurines via cesium carbonate catalyzed isomerization of 9-alkynylpurines has been successfully developed. The reactions proceeded rapidly under the base conditions and formed the desired products in good to excellent yields. The method was suitable with a broad substrate scope and proceeded well even on a multgram-scale. The obtained 9-allenylpurines were successfully applied to prepare various potential bioactive 9-acyclic nucleosides with high regioselectivity promoted by AgNO3.  相似文献

The first direct synthesis of 3-N-methyl-9-formylcytisine via electrophylic formylation is described. It is established, that Vilsmeier-Haack and Gatterman variants of this reaction are unsuccessful in the case with 3-substituted (-)-cytisine derivatives, but Duff procedure (with hexamethylenetetramine in trifluoroacetic acid) gives a possibility to obtain the target pseudo aromatic aldehyde with the 69% yield. Convenient precursors for [4 + 2]- or [3 + 2]-cycloaddition reactions are obtained by means of condensation of synthesized 3-N-methyl-9-formylcytisine with acetone, nitromethane and phosphorous ylides with yields from 70 to 87%. Alternative aprroach to alkenyl products and to 9-alkynyl-3-methylcytisine is realized using the Heck and Sonogashira cross-coupling reactions of methyl vinyl ketone, cyclohexenone or trimethylsilylacetylene with 9-bromo-3-methylcytisine (55, 70 and 60% accordingly). It is shown, that interaction of 3-N-methyl-9-formylcytisine with hydroxylamines leads to corresponding nitrone (93%) and oxime (70%). All individual compounds are isolated by column chromatography and completely characterized on the basis of NMR spectroscopy data.  相似文献

The estimation of the binding of a set of molecules against BRD9 protein was carried out through an in silico molecular dynamics-driven exhaustive analysis to guide the identification of potential novel ligands. Starting from eight crystal structures of this protein co-complexed with known binders and one apo form, we conducted an exhaustive molecular docking/molecular dynamics (MD) investigation. To balance accuracy and an affordable calculation time, the systems were simulated for 100 ns in explicit solvent. Moreover, one complex was simulated for 1 µs to assess the influence of simulation time on the results. A set of MD-derived parameters was computed and compared with molecular docking-derived and experimental data. MM-GBSA and the per-residue interaction energy emerged as the main indicators for the good interaction between the specific binder and the protein counterpart. To assess the performance of the proposed analysis workflow, we tested six molecules featuring different binding affinities for BRD9, obtaining promising outcomes. Further insights were reported to highlight the influence of the starting structure on the molecular dynamics simulations evolution. The data confirmed that a ranking of BRD9 binders using key parameters arising from molecular dynamics is advisable to discard poor ligands before moving on with the synthesis and the biological tests.  相似文献

Palladium catalyzed domino C-H functionalization reaction of arylaldehyde with dihaloarene has been developed to access 9-flourenone molecules. Bidentate ligand assisted strategy, single step reaction, high yield and excellent functional group tolerance make this method concise and effective for the synthesis of 9-flourenone.
