A simple and highly selective molecularly imprinted solid-phase extraction (MISPE) combined with ultrasound-assisted dispersive liquid-liquid microextraction (DLLME) was developed for the determination of four Sudan dye (I, II, III, and IV) residues in sausage products. The novel molecularly imprinted microspheres (MIMs) synthesized by aqueous suspension polymerization using phenylamine-naphthol as the dummy template show high affinity to the four Sudan dyes and were applied as selective sorbents of MISPE-DLLME to overcome the drawbacks of template leakage in quantitative analysis. Good linearity was obtained in a range of 0.005-2.0 μg g(-1) and the average recoveries of the four Sudan dyes at three spiked levels ranged from 86.3 to 107.5%. The MISPE-DLLME-HPLC protocol significantly improved the purification and enrichment of the analytes and eliminated the template leakage of the conventional MISPE on quantitative analysis.  相似文献

A new kind of aniline-naphthol molecularly imprinted microsphere (MIM) synthesized by aqueous suspension polymerization was applied as a selective sorbent of miniaturized matrix solid-phase dispersion combining with dispersive liquid-liquid microextraction (MSPD-DLLME) for the simultaneous determination of four Sudans in egg yolk samples. The solid sample was directly blended with MIM in MSPD procedure and the eluent of MSPD was used as the dispersive solvent of the followed DLLME for further purification and enrichment of the analytes before HPLC analysis. Good linearity for all the Sudan dyes was ranged from 0.02 μg g(-1) to 2.0 μg g(-1) (r(2)≥0.9990) and their recoveries at three spiked levels were ranged from 87.2% to 103.5% with RSD less than 6.1% (n=3). The presented MIM-MSPD-DLLME method combined the advantages of MIM, MSPD and DLLME, and could be applied for the determination of Sudans in complicated food samples.  相似文献

In this work, we developed a novel molecularly imprinted solid‐phase microextraction with capillary electrophoresis method for the selective extraction and determination of protoberberines in complicated samples. The imprinted monolith was prepared in a micropipette tip‐based device by using acrylamide as the functional monomer, ethyleneglyoldimethacrylate as the cross‐linker and dimethylsulfoxide as the porogen, and exhibited an imprinting factor of 2.41 to berberine, 2.36 to palmatine and 2.38 to jatrorrhizine. Good capillary electrophoresis separation was achieved by using 20 mM phosphate buffer at pH 7 as running buffer with the addition of organic modifier of 10% methanol. Parameters such as sample pH value, sample flow rate and sample volume were investigated for imprinted monolith‐based solid‐phase microextraction. An imprinted solid‐phase microextraction with capillary electrophoresis method was developed, the method showed a wide linear range (0.3–50 μg/mL), good linearity (R2 ≥ 0.9947) and good reproducibility (relative standard deviations ≤ 0.73%), the limit of detection was as low as 0.1 μg/mL, which was lower than some reported methods based on capillary electrophoresis for protoberberines. The method has been applied for determination of three common protoberberines in Cortex Phellodendri Chinensis, by using a molecularly imprinted monolith as the selective sorbent, most of the matrices in the Cortex Phellodendri Chinensis sample were removed and three protoberberines were selectively enriched and well determined.  相似文献

Molecularly imprinted polymers (MIPs) are synthetic polymers having a predetermined selectivity for a given analyte, or group of structurally related compounds, that make them ideal materials to be used in separation processes. In this sense, during past years a huge amount of papers have been published dealing with the use of MIPs as sorbents in solid-phase extraction, namely molecularly imprinted solid-phase extraction (MISPE). Although the majority of these papers were restricted to describe the use of different templates for different applications, several attempts proposing new alternatives to minimize the inherent drawbacks of the preparation and use of MIPs (i.e. template bleeding, tedious synthesis procedure, etc.) have been reported. Thus, this paper does not pretend to be a collection of MISPE-related papers but to give an overview on the significant attempts carried out during recent years to improve the performance of MIPs in solid-phase extraction. In addition, the use of MIPs packed in high performance liquid chromatography (HPLC) columns for the direct injection of crude sample extracts and the preparation of imprinted fibres for solid-phase microextraction will be also discussed.  相似文献

Solid-phase microextraction (SPME) and capillary electrophoresis (CE) are two of the main inventions that shaped 20th Century analytical chemistry. SPME is an effective microscale sampling and sample preparation technique, and CE is a high-efficiency microanalytical method. Online coupling of SPME with CE can be a powerful combination because of the significant advantages of the two techniques. The progress in the development of online SPME-CE coupling is surveyed in this review. Problems encountered and solutions reported are highlighted.  相似文献

Organophosphorus insecticides are widely employed in agriculture, and residues of them can remain after harvesting or storage. Pesticide residue control is an important task for ensuring food safety. Common chromatographic methods used in the determination of pesticide residues in food require clean-up and concentration steps prior to quantitation. While solid-phase extraction has been widely employed for this purpose, there is a need to improve selectivity. Due to their inherent biomimetic recognition systems, molecularly imprinted polymers (MIP) allow selectivity to be enhanced while keeping the costs of analysis low. In this work, a MIP that was designed to enable the selective extraction of fenitrothion (FNT) from tomatoes was synthesized using a noncovalent imprinting approach. The polymer was prepared using methacrylic acid as functional monomer and ethyleneglycol dimethacrylate as crosslinking monomer in dichloromethane (a porogenic solvent). The polymer was characterized by Fourier transform infrared spectroscopy, solid-state nuclear magnetic resonance (NMR), scanning electron microscopy (SEM), and nitrogen sorption porosimetry. The pore structure and the surface area were evaluated using the BET adsorption method. To characterize the batch rebinding behavior of the MIP, the adsorption isotherm was measured, allowing the total number of binding sites, the average binding affinity and the heterogeneity index to be established. A voltammetric method of quantifying FNT during the molecularly imprinted solid-phase extraction (MISPE) studies was developed. The polymer was placed in extraction cartridges which were then used to clean up and concentrate FNT in tomato samples prior to high-performance liquid chromatographic quantitation. The material presented a medium extraction efficiency of 59% (for analyses performed with three different cartridges on three days and a fortification level of 5.0 μg g−1) and selectivity when used in the preparation of tomato samples, and presented the advantage that the polymer could be reused several times after regeneration. A method was developed to sensitively determine safranine T in wolfberry by molecularly imprinted solid-phase extraction (MISPE) coupled with high-performance liquid chromatography and laser-induced fluorescence detection (HPLC-LIF). The MISPE capillary monolithic column was prepared by water-bath in situ polymerization, using safranine T, methacrylic acid (MAA), and ethylene dimethacrylate (EDMA) as template, functional monomer, and cross-linker, respectively. The properties of the homemade MISPE capillary monolithic column, including capacity and specificity, were investigated under optimized conditions and the morphologies of inner polymers were characterized by scanning electron microscopy (SEM). The mean recoveries of safranine T in wolfberry ranged from 91.2 % to 92.9 % and the intraday and interday relative standard deviation (RSD) values all ranged from 3.4 % to 4.2 %. Good linearity was obtained over 0.001–1.0 μg mL–1 (r?=?0.9999) with a detection limit (S/N?=?3) of 0.4 ng g–1. Under the selected conditions, enrichment factors of over 90-fold were obtained and the extraction on the monolithic column effectively cleaned up the wolfberry matrix. The results demonstrated that the proposed MISPE-HPLC-LIF method could be applied to sensitively determine safranine T in wolfberry.

A simple solid-phase microextraction device was fabricated for use in on-line immunoaffinity capillary electrophoresis (CE). The device, designed in the form of a four-part cross-shaped or cruciform configuration, includes a large-bore tube to transport samples and washing buffers and a small-bore fused-silica capillary for separation of analytes. At the intersection of the transport and separation tubes, a small cavity was fabricated, termed the analyte concentrator-microreactor, which contains four porous walls or semipermeable membranes (one for each inlet and outlet of the tubes) permitting the confinement of beads or suitable microstructures. The surface of the beads in the analyte concentrator carried a molecular recognition adsorbing chemical or affinity ligand material. The improved cruciform configuration of the analyte concentrator-microreactor device, designed for use in on-line immunoaffinity CE, enables it to specifically trap, enrich, and elute an analyte from any biological fluid or tissue sample extract without any sample pretreatment except filtration, centrifugation, and/or dilution allowing the separation and characterization of target analyte(s) with improved speed, sensitivity, and lower cost than existing techniques. As a model system, Fab' fragments derived from a purified immunoglobulin G (IgG) antibody were covalently bound to controlled-porosity glass and used as constituents of the analyte-microreactor device. The high-specificity polyclonal antibodies employed in these experiments were individually raised against the acidic nonsteroidal anti-inflammatory drugs ibuprofen and naproxen, and the neuropeptides angiotensin II, and neurotensin. These compounds, which were present in simple and complex matrices were captured by and eluted from the analyte concentrator-microreactor using a 50 mM sodium tetraborate buffer solution, pH 9.0, followed by a 100 nL plug of 300 mM glycine buffer, pH 3.4. Two analyte concentrators were tested independently: one containing Fab' fragments derived from antibodies raised against ibuprofen and naproxen; the other containing Fab' fragments derived from antibodies raised against angiotensin II and neurotensin. Each resulting electropherogram demonstrated the presence of two eluted materials in less than 20 min. Immunoaffinity CE performed in a cruciform structure was simpler and faster than previously reported in the literature using on-line microextraction devices designed in a linear format. The new concentration-separation system operated consistently for many runs, maintaining reproducible migration times and peak areas for every analyte studied.  相似文献

Ultrasound-assisted emulsification microextraction and dispersive liquid-liquid microextraction were compared for extraction of ephedrine, norephedrine, and pseudoephedrine from human urine samples prior to their determination by capillary electrophoresis. Formation of a microemulsion of the organic extract with an aqueous solution (at pH 3.2) containing 10% methanol facilitated the direct injection of the final extract into the capillary. Influential parameters affecting extraction efficiency were systematically studied and optimized. In order to enhance the sensitivity further, field-amplified sample injection was applied. Under optimum extraction and stacking conditions, enrichment factors of up to 140 and 1750 as compared to conventional capillary zone electrophoresis were obtained resulting in limits of detection of 12-33 μg/L and 1.0-2.8 μg/L with dispersive liquid-liquid microextraction and ultrasound-assisted emulsification microextraction when combined with field-amplified sample injection. Calibration graphs showed good linearity for urine samples by both methods with coefficients of determination higher than 0.9973 and percent relative standard deviations of the analyses in the range of 3.4-8.2% for (n = 5). The results showed that the use of ultrasound to assist microextraction provided higher extraction efficiencies than disperser solvents, regarding the hydrophilic nature of the investigated analytes.  相似文献

A magnetic molecularly imprinted polymer (MMIP) was fabricated and used as the sorbent for the MMIP-dispersive solid-phase microextraction of fenitrothion prior its determination by high-performance liquid chromatography equipped with an ultraviolet detector. The MMIP was prepared using functionalized Fe3O4 nanoparticles as the magnetic supporter. Methacrylic acid, ethylene glycol dimethacrylate and fenitrothion were used as the functional monomer, the cross-linker and the template, respectively. The properties of the resultant MMIP were evaluated using X-ray diffraction spectroscopy, Fourier transform infrared spectroscopy and scanning electron microscopy. The sorbent exhibited high selectivity and affinity toward fenitrothion compared to other organophosphate pesticides with the maximum adsorption capacity of 31.5 mg g?1. The effective variables on the extraction were optimized by univariable and MultiSimplex methods. The calibration curve exhibited linearity over the concentration range of 0.3–50.0 μg L?1 with the limit of detection of 0.1 μg L?1. The relative standard deviations at 10.0 μg L?1 level of FNT (n = 5) for intra- and inter-day assays were 1.6 and 3.1%, respectively. The proposed method was successfully used for the determination of trace amounts of FNT in food and water samples.  相似文献

In this work, the suitability of the combination of molecular imprinting and capillary electrochromatography (MIP-CEC) to be used as powerful tool in environmental or food analysis has been for the first time studied and successfully demonstrated. A molecularly imprinted monolith (MIM) has been synthesised and evaluated as stationary phase for the selective determination of the fungicide thiabendazole (TBZ) in citrus samples by non-aqueous capillary electrochromatography. The influence of the mobile phase composition, the voltage of the power supply and the separation temperature on the recognition of TBZ by the imprinted polymer has been evaluated, and the imprint effect in the MIM was clearly demonstrated. Once optimum recognition conditions were established, other variables affecting mechanical properties and chromatographic performance of MIM were adjusted using computational approach. The high selectivity achieved by the MIP-CEC developed procedure allowed unambiguous detection and quantification of TBZ in citrus samples by direct injection of the crude sample extracts, without any previous clean-up, in less than 6 min. The developed method was properly validated and the calculated detection limits were below the established maximum residue limits (MRLs), clearly demonstrating the suitability of the method to be used for the control of the selected fungicide.  相似文献

A novel molecularly imprinted polymer solid-phase extraction (MISPE) with flow-injection chemiluminescence (CL) was developed for the determination of pazufloxacin mesilate (PZFX). The molecularly imprinted polymer (MIP) was synthesized by using PZFX as the imprinting molecule. A glass tube packed the particles of the MIP was employed as MISPE micro-column, which was connected into the sampling loop of the eight-way injection valve for on-line selective preconcentration and extraction of PZFX. The eluent of acetonitrile:acetic acid (9:1, v:v) was used as carrier for eluting the adsorbed PZFX to react with the mixture of cerium(IV) and sodium sulfite in the flow cell to produce strong CL. The relative intensity of CL was linear to PZFX concentration in the range from 2.5 × 10−9 to 2.5 × 10−7 g mL−1. The limit of detection was 7 × 10−10 g mL−1 (3 σ) and the relative standard deviation for 5 × 10−8 g mL−1of PZFX solution was 3.7% (n = 7). A molecularly imprinted polymer solid phase extraction (MISPE) method combined with flow-injection chemiluminescence (FI-CL) for the determination of residual tetracycline (TC) in fish samples is presented. The molecularly imprinted polymer (MIP) of TC was synthesized and particles of this MIP were packed into a polytetrafluoroethylene (PTFE) tube, which was connected into the sampling loop of an eight-way injection valve and served as the MISPE column for on-line selective adsorption of TC. The eluent (CH3CN : HNO3 (0.01 mol L(-1)) = 4 ratio 1, v ratio v) was used for extracting the adsorbed TC, which could be detected by its good enhancing effect on the CL reaction between Ce(iv) and rhodamine B. The CL intensity is linear to TC concentration in the range from 4 x 10(-9) to 4 x 10(-7) g mL(-1). The detection limit is 1 x 10(-9) g mL(-1) (3 sigma) and the relative standard deviation is 2.4% (n = 9). The conditions of preconcentration, extraction and CL reaction were carefully studied. The selectivity experiment shows that the selectivity and sensitivity of the CL method could be improved greatly when MIP was used as a recognition material in SPE. However, the MISPE column interacted indiscriminately with oxytetracycline (OTC) with a 49 +/- 2% binding. An intermediate differential pulsed elution (DPE) step using 3% acetic acid as eluent was employed to remove OTC and other interfering substances. The proposed MISPE-CL method has been applied successfully to the determination of TC in fish samples. At the same time, the binding characteristics of the polymer to tetracycline were evaluated by batch and dynamic methods.  相似文献

A method was developed for the determination of ractopamine in pig urine using molecularly imprinted solid-phase extraction (MISPE) as the sample clean-up technique combined with high-performance liquid chromatography. The molecularly imprinted polymer (MIP) was synthesized in acetonitrile-triethylamine system using ractopamine (RAC) as the template and acrylamide as the monomer. The binding capacity of the polymer toward RAC was found to be about 2.57 mg of ractopamine/g of polymer. The optimal procedures for MISPE consisted of conditioning with 3 mL methanol, equilibrating with 3 mL of water, loading volume of <10 mL of aqueous sample (pH 7), washing with 3 mL water and 3 mL methanol, and eluting with 5 mL of 5% ammonia in methanol. In the four spiked samples with the levels of 0.01, 0.1, 1.0 and 5.0 μg/mL, the mean recoveries of analyte on the MIP were higher than 90% with relative standard deviation <10%, and significant differences between imprinted and non-imprinted materials were observed. The MIP selectivity was evaluated by checking 11 drugs with similar and different molecular structures to that of RAC. The characteristics of three-dimensional cavities and hydrogen bond interaction were regarded as the main factors that dominated the retention of RAC on the MISPE cartridge.  相似文献

建立了悬浮固化分散液液微萃取-毛细管电泳法同时测定水中磺酰脲类除草剂残留的方法。以十二醇为萃取剂、甲醇为分散剂,采用悬浮固化分散液液微萃取技术对水样进行分离提取,并结合毛细管电泳法进行测定。该方法可以有效提取、分离、检测水中残留的微量苯磺隆、吡嘧磺隆、苄嘧磺隆等9种磺酰脲类除草剂,各待测物在10.0~1000 μg/L范围内线性关系良好,相关系数r≥0.992,方法检出限为2.40~7.50 μg/L,方法精密度为6.55%~13.9%。将该方法用于实际水样的测定,取得了较满意的结果,加标回收率为82.0%~104%。该方法简便快速,适合水中磺酰脲类除草剂的同时测定。  相似文献

A molecularly imprinted polymer (MIP) with metolachlor as template was firstly coated on stainless steel fiber through chemical bonding strategy to solve the fragility problem of silica fiber substrate for solid-phase microextraction. The surface pretreatment of stainless steel fiber and the polymerization conditions were investigated systematically to enhance the preparation feasibility and MIP coating performance, and then a porous and highly cross-linked MIP coating with 14.8-μm thickness was obtained with over 200 times re-usability which was supported by non-fragile stainless steel fiber adoption. The MIP coating possessed specific selectivities to metolachlor, its metabolites and other chloroacetanilide herbicides with the factors of 1.1–4.6. Good extraction capacities of metolachlor, propisochlor and butachlor were found with MIP coating under quick adsorption and desorption kinetics, and the detection limits of 3.0, 9.6 and 38 μg L−1 were achieved, respectively. Moreover, the MIP-coated stainless steel fiber was evaluated for trace metolachlor, propisochlor and butachlor extraction in the spiked soybean and corn samples, and the enrichment factors of 54–60, 27–31 and 15–20 were obtained, respectively.  相似文献

A simple, sensitive and reproducible molecularly imprinted solid-phase extraction (MISPE) coupled with high performance liquid chromatographic method was developed for monitoring tilmicosin in feeds. The polymers were prepared using tylosin as mimic template molecule, methacrylic acid as functional monomer and ethylene glycol dimethacrylate as cross-linking monomer, and chloroform as a solvent by bulk polymerization. Under the optimum MISPE conditions, the novel polymer sorbent can selectively extract and enrich tilmicosin from variety of feeds. The MISPE cartridge was better than non-imprinted, C18 and HLB cartridges in terms of both recovery and precision. Mean recoveries of tilmicosin from five kinds of feeds spiked at 1, 10 and 50 mg kg−1 ranged from 76.9% to 95.6%, with intra-day and inter-day relative standard deviation less than 7.6%. The linearity was ranged from 1.0 to 100 mg L−1 for matrix standard solution (r = 0.9990). The limit of detection was approximately 0.35 mg kg−1 and the limit of quantification was approximately 0.98 mg kg−1. There was cleaner chromatogram by using MISPE than C18 and HLB SPE.  相似文献
