CdTe/CdS量子点-蛋白质与头孢曲松钠的相互作用   总被引:2,自引:2,他引:2

以3-巯基丙酸为稳定剂,采用水热法合成了具有优异光学性质的CdTe/CdS核壳型量子点,量子产率为13.8%。量子点与牛血清白蛋白(BSA)偶联后荧光强度明显增大。通过测定荧光强度确定了偶联的最佳反应条件:pH7.4,反应温度37℃,反应时间40min。研究了溶液pH值和NaCl浓度对量子点及量子点-BSA溶液荧光强度的影响。对牛血清白蛋白测定的线性范围为1.1~19.5mg/L;检出限为0.47mg/L。头孢曲松钠对量子点-BSA的荧光有明显猝灭作用,荧光猝灭程度与其浓度有良好的线性关系。实验结果表明为静态猝灭,头孢曲松钠与BSA按1∶1结合,结合常数为6.31×103L/mol。用圆二色光谱技术探讨了其对BSA构象的影响。  相似文献

本文在水热法合成水溶性CdTe及核壳结构CdTe/CdS量子点的基础上,分别研究了细胞色素c对CdTe量子点及CdTe/CdS核壳量子点荧光的猝灭效应和CdTe量子点对牛血清白蛋白荧光的猝灭效应,并阐述了猝灭机理。结果显示,细胞色素c对CdTe量子点的荧光猝灭效应具有一定的粒径依赖性,粒径越小,猝灭效应越强;细胞色素c对CdTe/CdS核壳量子点的猝灭效应比对CdTe量子点的更强,揭示了受激电子的表面传递机理。CdTe量子点通过松散牛血清白蛋白的螺旋结构而猝灭其荧光。  相似文献

采用均匀沉淀法,以无水氯化锌为原料,以氢氧化钠为沉淀剂,在室温条件下搅拌24h,制备了ZnO量子点,并用荧光光谱法和UV-Vis吸收光谱法研究了该量子点与吖啶橙的相互作用。结果表明:①ZnO量子点和吖啶橙主要以范德华作用力和氢键作用力相互结合,且其结合常数较大,两者间有较强的结合力;②吖啶橙与ZnO量子点之间形成了复合物导致ZnO量子点在349nm处的荧光发生静态猝灭;③根据Forster荧光共振能量转移原理计算得ZnO量子点与吖啶橙分子间距离(r)为1.98nm(小于7nm),判断其分子内发生非辐射能量转移。  相似文献

以巯基乙酸修饰的CdTe量子点为荧光探针,考察了不同条件下槲皮素对CdTe量子点的猝灭作用。发现在磷酸缓冲溶液中,其猝灭作用最强。在此基础上建立了基于荧光猝灭法检测槲皮素含量的新方法,该方法的线性范围为0~3.5×10-5 mol/L,线性方程为△F=42.86+36.98 c(×10-6mol/L),相关系数r为0.9985,检测限(3SD/斜率)为1.7×10-7mol/L。方法应用于降脂宁颗粒和洋葱中槲皮素含量的测定,相对标准偏差为2.1%和2.3%,回收率在98%~102%之间。  相似文献

在水相中合成了巯基乙酸包被的CdTe量子点(CdTe ODs),培氟沙星(PEF)对该量子点的荧光具有显著的猝灭,基于该猝灭作用,实现了用CdTe ODs作为荧光探针对痕量PEF的定量检测.实验发现,在pH为9.5的B-R缓冲溶液中,当CdTeODs的浓度为2.0×10-4 mol/L时,PEF的浓度在6.40×10-5~1.28×10-3mg/mL范围与CdTe ODs的荧光猝灭强度呈良好的线性关系,相关系数r为0.9917,检出限(S/N=3)为1.30×10-5 mg/mL.用该方法对犬猫倍福康和注射用PEF的含量进行了测定,回收率为94.6%~106%.  相似文献

以巯基乙酸修饰的CdTe量子点(QDs)为荧光探针, 考察了不同条件下烟酸诺氟沙星对CdTe QDs荧光的猝灭作用. 发现在磷酸缓冲液中(PBS, pH 7.40, 0.10 mol/L), 其猝灭作用最强, 随后对其猝灭机理进行了讨论. 在此基础上建立了基于荧光猝灭法检测烟酸诺氟沙星含量的新方法, 该方法的线性范围为0.10~90 μg/mL, 线性方程为DF=26.6206c-20.8273 (μg/mL), 相关系数r为0.997, 检测限(3S/N)为2.50×10-2 μg/mL. 应用此法检测兽药雏乐宝、烟酸氟哌酸和强效氟哌酸中烟酸诺氟沙星的含量, 百分标示量在93.0%~104.0%范围内变化, 相对标准偏差(RSD)≤4.2%|与常用的紫外分光光度法相比, 该法具有准确度高、灵敏度高且检测范围宽等优势, 显示出潜在的应用前景.  相似文献

以巯基乙酸为稳定剂,在水溶液中合成了具有特殊光学性质的水溶性CdSe/CdS量子点。以该量子点为荧光探针,基于荧光猝灭法对芹黄素进行了定量检测。考察了缓冲体系、反应时间、量子点浓度等多种因素的影响。实验结果表明,在0.001 mol/L、pH为6.80的KH2PO4-Na2HPO4缓冲液中,当量子点浓度为1.2×10^-4mol/L、反应时间为20 min时,该方法的线性范围为0.16-27.02μg/mL,其线性回归方程为F0/F=0.99665+0.11067ρ(μg/mL),相关系数r=0.998,检出限为0.13μg/mL,并用于合成样品的分析。  相似文献

It was found that oxolinic acid (OA) at high concentration can self-assemble into nano- to micro- meter scale OA aggregates in Tris-HCl (pH 7.48) buffer solution. The nanoparticles of OA were adopted as fluorescence probes in the quantitative analysis of proteins. Under optimum conditions, the fluorescence quenching extent of nanometer scale OA aggregates was in proportion to the concentration of albumins in the range of 3.0 × 10−8 to 3.0 × 10−5 g mL−1 for bovine serum albumin (BSA) and 8.0 × 10−8 to 8.0 × 10−6 g mL−1 for human serum albumin (HSA). The detection limits (S/N = 3) were 3.4 × 10−9 g mL−1 for BSA, and 2.6 × 10−8 g mL−1 for HSA, respectively. Samples were satisfactorily determined. The interaction mechanism of the system was studied using fluorescence, UV-vis, resonance light scattering (RLS) and transmission electron microscope (TEM) technology, etc., indicating that the nonluminescent complex was formed between serum albumin molecular and OA, to disaggregate the self-association of OA, which resulted in the dominated static fluorescence quenching in the system.  相似文献

Heavy metal ions such as Hg and Pb are hazardous due to very high toxicity, mobility, and ability to accumulate through the food chain or atmosphere in the environment system. Therefore, ultrasensitive determination of mercury and lead is important to provide an evaluation index of ions in aqueous environment. This paper describes the investigation of surface modified quantum dots (QDs) as a sensing receptor for Hg2+ and Pb2+ ion detection by optical approach. Water-soluble L-cysteine-capped CdS QDs have been synthesized in aqueous medium. These functionalized nanoparticles were used as a fluorescence sensor for Hg2+ and Pb2+ ions, involved in the fluorescence quenching. The effect of foreign ions on the intensity of CdS QDs showed a low interference response toward other metal ions except Cu2+ and Fe2+ ions. The limit of detection (LOD) of this system is found to be 1.0 and 3.0 nM for Hg2+ and Pb2+ ions, respectively.  相似文献

In Vitro Binding of Furadan to Bovine Serum Albumin   总被引:1,自引:0,他引:1  
Under physiological conditions, the interaction between furadan (FRD) and bovine serum albumin (BSA) was investigated by spectroscopy including fluorescence emission, UV-visible absorption, scattering, circular dichroism (CD) spectra, synchronous and three-dimensional fluorescence spectra. The observed binding constant K b and the number of binding sites n were determined by the fluorescence quenching method. The distance r between donor (BSA) and acceptor (FRD) was obtained according to the Förster theory of non-radiation energy transfer. The enthalpy change (ΔH θ ), Gibbs energy change (ΔG θ ) and entropy change (ΔS θ ) at four different temperatures were calculated. The process of binding was proposed to be a spontaneous process since the ΔG θ values were negative. The positive ΔS θ and ΔH θ values indicated that the interaction of FRD and BSA was driven mainly by hydrophobic interactions. The addition of FRD to BSA solutions leads to enhancement in scattering intensity, exhibiting the formation of an aggregate in solution. CD spectra, synchronous and three-dimensional fluorescence spectra were used to measure the structural change of BSA molecules with FRD present.  相似文献

In this paper, two flavonoid aglycones (baicalein, quercetin) and their glycosides (baicalin, quercitrin) were studied for their ability to bind protein by quenching the protein intrinsic fluorescence. From the spectra obtained, the bimolecular quenching constants, the apparent static binding constants, and binding sites values were calculated. The glycosylation of flavonoids decreases the binding affinity with protein. For quercetin and quercitrin, the binding constants for BSA were 3.65 × 107 and 6.47 × 103 L mol−1, respectively. For baicalein and baicalin, the binding constants were 4.54 × 108 and 1.63 × 106 L mol−1, respectively.  相似文献

运用荧光光谱和紫外-可见吸收光谱研究了在缓冲溶液中不同温度下孔雀石绿(MG)与牛血清白蛋白(BSA)之间的相互作用. 实验结果表明, MG对BSA的内源荧光猝灭为静态猝灭过程. 测定了该反应在不同温度下的结合常数KA, KA分别为7.69×104 L·mol-1(10 ℃)、5.31×104 L·mol-1(20 ℃)和4.85×104 L·mol-1(37 ℃), MG与BSA以摩尔比1:1结合. 根据Forster非辐射能量转移理论, 求出了37 ℃时给体(MG)和受体(BSA)之间能量转移效率和结合距离分别为E=0.1635 和r=2.30 nm. 计算出的热力学参数表明, MG 和BSA之间的作用力主要是通过氢键和范德华力相互作用.  相似文献

The interaction between gatifloxacin mesylate (GM) and salmon sperm DNA was studied by fluorescence spectrometry and ultra‐violet (UV) spectrometry. Additions of salmon sperm DNA to GM solution resulted in its strong fluorescence quenching and UV absorbance decrease due to the strong interaction between GM and salmon sperm DNA. Both the maximum fluorescence emission wavelength and the maximum UV absorbance wavelength of GM did not change with the increasing concentration of salmon sperm DNA, indicating no intercalative binding existed between them. The Stern‐Volmer plot indicated that the fluorescence‐quenching constant at different temperatures or different salmon sperm DNA concentration ranges was different. Effects of ionic strength and I? on the fluorescence quenching of GM by salmon sperm DNA indicated that electrostatic interaction and groove binding coexisted between them.  相似文献

利用紫外吸收、荧光、同步荧光光谱及圆二色谱研究了吡蚜酮与牛血清白蛋白(BSA)的相互作用. 结果发现, 吡蚜酮使BSA的紫外吸收峰强度降低, 峰位红移; BSA的特征荧光峰猝灭, 荧光猝灭常数KSV随着温度的升高而降低, 表明吡蚜酮与BSA发生了较强的相互作用, 且吡蚜酮对BSA的荧光猝灭机制属于静态猝灭. 计算了不同温度下的结合常数和结合位点数; 由van′t Hoff方程计算出体系的ΔH和ΔS值, 得出二者之间的作用力主要为氢键和范德华力; 根据非辐射能量转移理论确定了给体-受体间的结合距离r=2.4 nm. 采用同步荧光光谱和圆二色谱考察了吡蚜酮对牛血清白蛋白构象的影响.  相似文献
