The electrostatic adsorption onto charged surfaces of comb copolymers comprising a polyelectrolyte backbone and pendent PEG side chains, such as poly(l-lysine)-g-poly(ethylene glycol) (PLL-g-PEG), has in previous studies provided protein-repellent thin coatings, particularly on metal oxide surfaces. A drawback of this approach is, however, the instability of such adsorbed layers under extreme pH values or high ionic strength. We have overcome this limitation in the present study by covalently immobilizing PLL-g-PEG copolymers onto aldehyde plasma-modified substrates. Silicon wafers, optical waveguide chips, and perfluorinated ethylene-co-propylene (FEP) polymer substrates were first coated with a thin plasma polymer layer using a propionaldehyde plasma, followed by covalent immobilization of PLL-g-PEG via reductive amination between amine groups of the PLL backbone with aldehyde groups on the plasma-deposited interlayer. The stability in high salt media and the protein resistance of different molecular architectures of immobilized PLL-g-PEG layers were quantitatively investigated by XPS, an optical waveguide technique (OWLS), and ToF-SIMS. The adsorption of bovine serum albumin was found to be below the detection limit (<2 ng/cm(2)), as for electrostatically adsorbed PLL-g-PEG layers. However, after 24 h of exposure of covalently immobilized layers of PLL-g-PEG to high ionic strength buffer (2400 mM NaCl), no significant change in the protein resistance was observed, whereas under the same conditions electrostatically adsorbed PLL-g-PEG coatings lost their protein resistance. Moreover, covalent immobilization via an aldehyde plasma interlayer enabled the application of PLL-g-PEG layers onto substrates such as FEP onto which electrostatic binding is not possible. These findings create a generic platform for the covalent immobilization of PLL-g-PEG onto a wide variety of substrates.  相似文献

In this study, we grafted water‐soluble biocompatible polymer, poly(N‐(2‐hydroxypropyl)methacrylamide) (PHPMA), onto the surface of multi‐walled carbon nanotubes (MWNTs). The reversible addition‐fragmentation chain transfer (RAFT) agents, dithioesters, were successfully immobilized onto the surface of MWNTs first, PHPMA chains were then subsequently grafted onto MWNTs via RAFT polymerization by using dithioesters immobilized on MWNTs as RAFT agent. FTIR, XPS, 1H NMR, Raman and TGA were used to characterize the resulting products and to determine the content of water‐soluble PHPMA chains in the product. The MWNTs grafted with PHPMA chains have good solubility in distilled water, PBS buffer, and methanol. TEM images of the samples provide direct evidence for the formation of a nanostructure that MWNTs coated with polymer layer. © 2006 Wiley Periodicals, Inc. The objective of this work was to gain a better understanding of the mechanism of resistance to protein adsorption of surfaces grafted with poly(ethylene oxide) (PEO). A polyurethane-urea was used as a substrate to which PEO was grafted. Grafting was carried out by introducing isocyanate groups into the surface followed by reaction with amino-terminated PEO. Surfaces grafted with PEO of various chain lengths (PUU-NPEO) were prepared and characterized by water contact angle and X-ray photoelectron spectroscopy (XPS). XPS data indicated higher graft densities on the PUU-NPEO surfaces than on analogous surfaces prepared using hydroxy-PEO (PUU-OPEO) as reported previously [J.G. Archambault, J.L. Brash, Colloids Surf. B: Biointerf. 33 (2004) 111-120]. Protein adsorption experiments using radiolabeled myoglobin, concanavalin A, albumin, fibrinogen and ferritin as single proteins in buffer showed that adsorption was reduced on the PEO-grafted surfaces by up to 95% compared to the control. Adsorption decreased with increasing PEO chain length and reached a minimum at a PEO MW of 2000. Adsorption levels on surfaces with 5000 and 2000 MW grafts were similar. There was no clear effect of protein size on resistance to protein adsorption. Adsorption on the PUU-NPEO surfaces was significantly lower than on the corresponding PUU-OPEO surfaces, again suggesting higher graft densities on the former. Adsorption of fibrinogen from plasma was also greatly reduced on the grafted surfaces. From analysis (SDS-PAGE, immunoblotting) of the proteins eluted after plasma exposure, it was found that the grafted surfaces and the unmodified substrate adsorbed the same proteins in roughly the same proportions, suggesting that adsorption to the PEO surfaces occurs on patches of bare substrate. The PEO grafts did not apparently cause differential access to the substrate based on protein size.  相似文献

A radio-frequency glow discharge induced graft copolymerization process has prepared a PAAc-grafted PE film in this present study. Poly(ethylene oxide) (PEO) was then covalently immobilized onto PAAc-grafted PE film by a chemical coupling reaction. Carboxylic groups were introduced onto the surface of PE film by exposing the film to an Ar plasma, and the plasma-treated film was then next reacted with a 50% acrylic acid aqueous solution. These functional groups were reacted with bisamino PEO by a coupling agent CMC. ESCA, IR, and water contact angle studies were conducted for characterizing the surfaces of these films during the varied stages of the reaction. Increasing the molecular weight of the PEO attached to the PAAc-grafted PE resulted in an increased wettability of the films. © 1993 John Wiley & Sons, Inc.  相似文献

The surface structures of three kinds of poly(ethylene oxide)‐segmented nylon (PEO‐Ny) molten films were investigated using a scanning electron microscopy (SEM), an electron spectroscopy for a chemical analysis (ESCA), and a static secondary ion mass spectrometry (SSIMS). The PEO‐Ny's used were high semicrystalline PEO‐segmented poly(iminosebacoyliminohexamethylene) (PEO‐Ny610), low semicrystalline PEO‐segmented poly(iminosebacoylimino‐m‐xylene) (PEO‐NyM10), and amorphous PEO‐segmented poly(iminoisophthaloyliminomethylene‐1,3‐cyclohexylenemethylene) (PEO‐NyBI). SEM observations show that the surfaces of the PEO‐Ny610 and PEO‐NyM10 films are composed of spherulite, and that PEO‐NyBI film has a smooth surface. The results of ESCA and SSIMS exhibit the significant enrichments of PEO segment at the surfaces of all the films regardless of the crystallinity. The reason for the enrichment of PEO segment was discussed in terms of the surface tension of the corresponding homopolymers in the melting state. © 2000 John Wiley & Sons, Inc. Hybrid nanoarchitecture of tailor‐made Poly(ethyl acrylate)/clay was prepared by surface‐initiated atom transfer radical polymerization (SI‐ATRP), by tethering ATRP initiator on active hydroxyl group, present in surface as well as in the organic modifier of the clay used. Extensive exfoliation was facilitated by using these initiator modified clay platelets. Poly(ethyl acrylate) chains with controlled polymerization and narrow polydispersities were forced to be grown from within the clay gallery (intergallery) as well as from the outer surface (extragallery) of the clay platelets. The polymer chains attached onto clay surfaces might have the potential to provide the composites with enhanced compatibility in blends with common polymers. Attachment of the initiator on clay platelets was confirmed by Fourier transform infrared spectroscopy (FTIR), X‐ray photoelectron spectroscopy (XPS), elemental analysis, Wide‐angle X‐ray diffraction (WAXD), and microscopic analysis. Finally, end group analysis (by Matrix‐Assisted Laser Desorption Ionization Mass Spectrometry, and chain extension experiment) of the cleaved polymer and morphological study (by WAXD, Transmission Electron Microscopy), performed on the polymer grafted clays examined the effect of grafting on the efficiency of polymerization and the degree of dispersion of clay tactoids in polymer. © 2008 Wiley Periodicals, Inc. Epoxide and aldehyde end‐functionalized poly(ethylene glycol)s (PEGs) (Mw = 400, 1000, 3400, 5000, and 20,000) were grafted to poly(ethylene terephthalate) (PET) film substrates that contained amine or alcohol groups. PET‐PAH and PET‐PEI were prepared by reacting poly(allylamine) (PAH) and polyethylenimine (PEI) with PET substrates, respectively; PET‐PVOH was prepared by the adsorption of poly(vinyl alcohol) (PVOH) to PET substrates. Grafting was characterized and quantified by the increase of the intensity of the PEG carbon peak in the X‐ray photoelectron spectra. Grafting yield was optimized by controlling reaction parameters and was found to be substrate‐independent in general. Graft density consistently decreased as PEG chain length was increased. This is likely due to the higher steric requirement of higher molecular weight PEG molecules. Water contact angles of surfaces containing long PEG chains (3400, 5000, and 20,000) are much lower than those containing shorter PEG chains (400 and 1000). This indicates that longer PEG chains are more effective in rendering surfaces hydrophilic. Protein adsorption experiments were carried out on PET‐ and PEG‐modified derivatives using collagen, lysozyme, and albumin. After PEG grafting, the amount of protein adsorbed was reduced in all cases. Trends in surface requirements for protein resistance are: surfaces with longer PEG chains and higher chain density, especially the former, are more protein resistant; PEG grafted to surfaces containing branched or network polymers is not effective at covering the underlying substrate, and thus does not protect the entire surface from protein adsorption; and substrates containing surface charge are less protein‐resistant. © 2004 Wiley Periodicals, Inc. Surface modification using living radical polymerization (LRP) chemistry is a powerful technique for surface modification of polymeric substrates. This research demonstrates the ability to use LRP as a polymer substrate surface‐modification platform for covalently grafting polymer chains in a spatially and temporally controlled fashion. Specifically, dithiocarbamate functionalities are introduced onto polymer surfaces using tetraethylthiuram disulfide. This technique enables integration of LRP‐based grafting for the development of an integrated, covalent surface‐modification method for microfluidic device construction. The unique photolithographic method enables construction of devices that are not substrate‐limited. To demonstrate the utility of this approach, both controlled fluid flow and cell patterning applications were demonstrated upon modification with various chemical functionalities. Specifically, poly(ethylene glycol) (375) monoacrylate and trifluoroethyl acrylate were grafted to control fluidic flow on a microfluidic device. Before patterning, surface‐functionalized samples were characterized with both goniometric and infrared spectroscopy to ensure that photografting was occurring through pendant dithiocarbamate functionalities. Near‐infrared results demonstrated conversion of grafted monomers when dithiocarbamate‐functionalized surfaces were used, as compared to dormant control surfaces. Furthermore, attenuated total reflectance/infrared spectroscopy results verified the presence of dithiocarbamate functionalities on the substrate surfaces, which were useful in grafting chains of various functionalities whose contact angles ranged from 7 to 86°. © 2006 Wiley Periodicals, Inc. This work reports the surface‐initiated atom transfer radical polymerization (ATRP) from hydrogen plasma‐treated porous poly(tetrafluoroethylene) (PTFE) membranes using the C? F groups as initiators. Hydrogen plasma treatment on PTFE membrane surfaces changes their chemical environment through defluorination and hydrogenation reactions. With the hydrogen plasma treatment, the C? F groups of the modified PTFE membrane surface become effective initiators of ATRP. Surface‐initiated ATRP of poly(ethylene glycol) methacrylate (PEGMA) is carried out to graft PPEGMA chains to PTFE membrane surfaces. The chain lengths of poly(PEGMA) (PPEGMA) grafted on PTFE surfaces increase with increasing the reaction time of ATRP. Furthermore, the chain ends of PPEGMA grown on PTFE membrane surfaces then serve as macroinitiators for the ATRP of N‐isopropylacrylamide (NIPAAm) to build up the PPEGMA‐b‐PNIPAAm block copolymer chains on the PTFE membrane surfaces. The chemical structures of the modified PTFE membranes are characterized using X‐ray photoelectron spectroscopy. The modification increases the surface hydrophilicity of the PTFE membranes with reductions in their water‐contact angles from 120° to 60°. The modified PTFE membranes also show temperature‐responsive properties and protein repulsion features owing to the presence of PNIPAAM and PPEGMA chains. © 2010 Wiley Periodicals, Inc. Surface-initiated atom-transfer radical polymerization (ATRP) of poly(ethylene glycol) monomethacrylate (PEGMA) was carried out on the hydrogen-terminated Si(100) substrates with surface-tethered alpha-bromoester initiator. Kinetic studies confirmed an approximately linear increase in polymer film thickness with reaction time, indicating that chain growth from the surface was a controlled "living" process. The "living" character of the surface-grafted PEGMA chains was further ascertained by the subsequent extension of these graft chains, and thus the graft layer. Well-defined polymer brushes of near 100 nm in thickness were grafted on the Si(100) surface in 8 h under ambient temperature in an aqueous medium. The hydroxyl end groups of the poly(ethylene glycol) (PEG) side chains of the grafted PEGMA polymer were derivatized into various functional groups, including chloride, amine, aldehyde, and carboxylic acid groups. The surface-functionalized silicon substrates were characterized by reflectance FT-IR spectroscopy and X-ray photoelectron spectroscopy (XPS). Covalent attachment and derivatization of the well-defined PEGMA polymer brushes can broaden considerably the functionality of single-crystal silicon surfaces.  相似文献

A method was adopted to fix a series of polymers of PE‐b‐PEO with different PEO/PE segments on the chains of LLDPE. Maleic anhydride (MA) reacting with hydroxyl group of PE‐b‐PEO (mPE‐b‐PEO) was used as the intermediate. The structures of intermediates and graft copolymers were approved by 1H NMR and FTIR. XPS analysis revealed a great amount of oxygen on the surface of grafted copolymers although the end group of PEO was fixed on the LLDPE chains through MA. Thermal properties of the graft copolymers as determined by differential scanning calorimetry (DSC) showed that PE segments in the grafted monomers could promote the heterogeneous nucleation of the polymer, increase Tc, and crystal growth rate. While the amorphous PEO segments which attached to the crystalline PE segments in LLDPE, impaired their ability to fit the crystal lattice, and depressed the crystallization of LLDPE backbones. In this study, it was also verified through the dynamic rheological data that increasing Mn of grafted monomers significantly increased the complex viscosity and enhanced the shear‐thinning behavior. Long‐branched chains formed by grafted monomers enhanced the complex moduli (G′ and G″) value and retarded relaxation rate. However, there were little influence on the rheological properties when increasing the amounts of PEO segments (or decreasing PE segments) of grafted monomers with similar molecular weight. © 2008 Wiley Periodicals, Inc. The UV‐induced and catalyst‐free photoiniferter surface grafting using xanthate RAFT agents is studied. First, a novel silane containing a xanthate RAFT agent moiety is synthesized and successfully grafted onto a silicon wafer as confirmed by secondary ion mass spectrometry (SIMS). Next, using only methyl acrylate, solvent and UV light (365 nm) polymer brushes are grafted rapidly from the surface with film thicknesses up to 25 nm reached within half an hour. The photograft polymerization of various vinyl monomers onto nanosized silica surfaces was investigated. It was initiated by eosin moieties introduced onto the silica surface. The preparation of the silica with eosin moieties was achieved by the reaction of eosin with benzyl chloride groups on the silica surface.These were introduced by the reaction of surface silanol groups with 4‐(chloromethyl)phenyltrimethoxysilane in the presence of t‐butyl ammonium bromide as a phase‐transfer catalyst. The photopolymerization of various vinyl monomers, such as styrene, acrylamide, acrylic acid, and acrylonitrile was successfully initiated by eosin moieties on the silica surface in the presence of ascorbic acid as a reducing agent and by oxygen. The corresponding polymers were grafted from the silica surface. The grafting efficiency (percentage of grafted polymer to total polymer formed) in the photoinitiation system was much larger than that in the radical polymerization initiated by surface radicals; these radicals were formed by the thermal decomposition of azo groups introduced onto the silica surface. It was found that the polymer‐grafted silica gave stable dispersions in good solvents of grafted polymer and the wettability of the surfaces can be easily controlled by grafting of polymers. © 2004 Wiley Periodicals, Inc. Poly(ethylene terephthalate) (PET) films were treated by argon plasma following by graft copolymerization with acrylic acid (AAc). The obtained PET-surface grafted PAA (PET-g-PAA) was coupled with chitosan (CS) and o-carboxymethylchitosan (OCMCS) molecules, respectively. Their surface physicochemical properties were characterized by X-ray photoelectron spectroscopy (XPS), water contact angle and streaming potential measurements. The PET-g-PAA surface containing carboxylic acid, CS immobilized PET surface containing amino and OCMCS immobilized PET surface containing both carboxylic acid and amino groups, make the PET surface exhibited a hydrophilic character. The blood compatibility was evaluated by platelet contacting experiments and protein adsorption experiments in vitro. The results demonstrate that the PET surface coupling OCMCS shows much less platelet adhesive and fibrinogen adsorption compared to the other surface modified PET films. The anticoagulation of PET-OCMCS is ascribed to the suitable balance of hydrophobicity/hydrophilicity, surface zeta potential and the low adsorption of protein.  相似文献

Living anionic surface‐initiated polymerization on flat gold substrates has been conducted to create uniform homopolymer and diblock copolymer brushes. A 1,1‐diphenylethylene (DPE) self‐assembled monolayer was used as the immobilized precursor initiator. n‐BuLi was used to activate the DPE in tetrahydrofuran at –78 °C to initiate the polymerization of different monomers (styrene, isoprene, ethylene oxide, and methyl methacrylate). Poly(styrene) (PS) and poly(ethylene oxide) (PEO) in particular were first investigated as grafted homopolymers, followed by their copolymers, including poly(isoprene)‐b‐poly(methylmethacrylate) (PI‐b‐PMMA). A combined approach of spectroscopic (Fourier transform infrared spectroscopy, surface plasmon spectroscopy, ellipsometry, X‐ray photoelectron spectroscopy) and microscopic (atomic force microscopy) surface analysis was used to investigate the formation of the polymer brushes in polar solvent media. The chemical nature of the outermost layer of these brushes was studied by water contact angle measurements. The effect of the experimental conditions (solvent, temperature, initiator concentration) on the surface properties of the polymer brushes was also investigated. © 2005 Wiley Periodicals, Inc. A novel tri‐component copolymer, polycaprolactone/poly(ethylene oxide)/polylactide (PCEL) was synthesized. The effect of the chemical composition on physical properties was investigated by using NMR, differential scanning calorimetry (DSC) and X‐ray diffraction. Both the soft segment poly(ethylene oxide) (PEO) and polycaprolactone (PCL) could enhance the mobility of polymer chains and decrease the crystallizability of the copolymers. The polymeric microspheres, which are of interest for drug delivery systems, were prepared using an emulsification‐solvent evaporation technique. By scanning electron microscopy (SEM) and atomic force microscopy (AFM), the surface morphology of the microspheres was studied. It was found that the presence of PEO segment could improve the hydrophilicity of the copolymers and the morphology of the polymeric microspheres could be altered by adjusting the chemical composition. The accumulation of PEO segments on the outer surface of the polymeric microspheres was proven by X‐ray photoelectron spectroscopy (XPS). Biomimetic coatings offer exciting options to modulate the biocompatibility of biomaterials. The challenge is to create surfaces that undergo specific interactions with cells without promoting nonspecific fouling. This work reports an innovative approach toward biomimetic surfaces based on the covalent immobilization of a carboxylate terminated PEGylated hyaluronan (HA-PEG) onto plasma functionalized NiTi alloy surfaces. The metal substrates were aminated via two different plasma functionalization processes. Hyaluronan, a natural glycosaminoglycan and the major constituent of the extracellular matrix, was grafted to the substrates by reaction of the surface amines with the carboxylic acid terminated PEG spacer using carbodiimide chemistry. The surface modification was monitored at each step by X-ray photoelectron spectroscopy (XPS). HA-immobilized surfaces displayed increased hydrophilicity and reduced fouling, compared to bare surfaces, when exposed to human platelets (PLT) in an in vitro assay with radiolabeled platelets (204.1 +/- 123.8 x 10 (3) PLT/cm (2) vs 538.5 +/- 100.5 x 10 (3) PLT/cm (2) for bare metal, p < 0.05). Using a robust plasma patterning technique, microstructured hyaluronan surfaces were successfully created as demonstrated by XPS chemical imaging. The bioactive surfaces described present unique features, which result from the synergy between the intrinsic biological properties of hyaluronan and the chemical composition and morphology of the polymer layer immobilized on a metal surface.  相似文献

采用125I放射标记技术研究了血浆白蛋白和纤维蛋白原在聚甲基丙烯酸甲酯-接枝-十八烷基聚氧乙烯(PMMA-g-SPEO)、聚甲基丙烯酸甲酯-接枝-乙基聚氧乙烯(PMMA-g-EPEO)和聚甲基丙烯酸甲酯-甲基丙烯酸十八酯共聚物(PMMA-co-SMA)表面的竞争吸附行为.结果表明,十八烷基聚氧乙烯复合修饰的PMMA-g-SPEO可高选择性地形成白蛋白可逆吸附层,有效地阻抗血小板的粘附,延长材料的凝血时间,是一种理想的白蛋白原位复合的生物医用功能材料.  相似文献

Poly(ethylene oxide) (PEO) and low molecular weight poly(ethylene glycol) (PEG) were covalently immobilized on silicon wafers and gold films by way of the CH insertion reaction of perfluorophenyl azides (PFPAs) by either photolysis or thermolysis. The immobilization does not require chemical derivatization of PEO or PEG, and polymers of different molecular weights were successfully attached to the substrate to give uniform films. Microarrays were also generated by printing polymer solutions on PFPA-functionalized wafer or Au slides followed by light activation. For low molecular weight PEG, the immobilization was highly dependent on the quality of the film deposited on the substrate. While the spin-coated and printed PEG showed poor immobilization efficiency, thermal treatment of the PEG melt on PFPA-functionalized surfaces resulted in excellent film quality, giving, for example, a grafting density of 9.2×10(-4)?(-2) and an average distance between grafted chains of 33? for PEG 20,000. The anti-fouling property of the films was evaluated by fluorescence microscopy and surface plasmon resonance imaging (SPRi). Low protein adsorption was observed on thermally-immobilized PEG whereas the photoimmobilized PEG showed increased protein adsorption. In addition, protein arrays were created using polystyrene (PS) and PEG based on the differential protein adsorption of the two polymers.  相似文献

Low‐density polyethylene (LDPE) and polystyrene (PS) films with hydrophilic surface were prepared by photochemical grafting of sulfobetaine‐based copolymer containing photolabile moiety, and long‐term stability of the hydrophilic nature of the surfaces in seawater was proved. The sulfobetaine‐based copolymer was prepared by copolymerization of N,N‐dimethyl‐N‐(3‐(methacryloylamino)propyl)‐N‐(3‐sulfopropyl) ammonium betaine with 2 or 5 mol% of N‐methacryloyl‐4‐azidoaniline, and the resulted polymers were grafted onto the plasma pretreated LDPE and PS films. The contact angle measurements were used to prove the modification as well as to follow the changes in the hydrophilicity during storage at room temperature under air atmosphere as well as in seawater at 32°C. The stability of the polymer layer was confirmed also by FTIR and AFM. Polysulfobetaine‐modified LDPE and PS surfaces exhibited significantly higher long‐term hydrophilicity compared with only plasma treated LDPE and PS surfaces.  相似文献
