Summary A new and simple HPLC method is described for the separation and quantitative determination of the (+)-and (−)-enantiomers of lansoprazole. The analytes were extracted from serum as previously described for whole lansoprazole [K. Borner, Chromatographia 45, 450–452 (1997)]. The enantiomers were separated by chromatography on a CHIRAL-AGPR column which contained covalently bound acid α1-glycoprotein as chiral selector. In the pure drug the (−)/(+) ratio was 0.99:1.01. In serum of twelve human volunteers the concentration of the (−)-enantiomer was 3 to 5 times higher than that of the (+)-enantiomer. Both enantiomers differ remarkably in their pharmacokinetics.  相似文献

Summary A human serum albumin (HSA)-based HPLC column has been modified in situ by disulfiram, an alcohol-deterrent drug reported to bind cys34, the only free cysteine in HSA, under physiological conditions. The reversible and covalent binding of disulfiram was found to change the binding properties of the protein, giving rise to a new selector which performed differently from the native albumin-based stationary phase. When low concentrations of disulfiram were used as mobile phase modifier, reversible binding resulted in a cooperative allosteric effect with improved selector performance. Covalent modification resulted in markedly reduced affinity for binding of the drugs to sites I or II, while still maintaining enantioselectivity. This study has enabled the monitoring of interactions of disulfiram with potentially coadministered drugs, and the preparation of a chiral selector with different drug affinity and enantioselectivity.  相似文献

Summary A high-performance liqid chromatographic method is reported for the measurement of miconazole in systemic, fungal infectious patients. Pharmacokinetic data are presented for a single patient receiving miconazole therapy. Sample preparation involves protein precipitation by acetonitrile (1:1, vol/vol). Analyses are carried out on a reversed-phae chromatographic system using octadecylsilane stationary phase: a mobile phase consisting of 0.05 M acetate buffer (pH 7.4)acetonitrile (20:80, vol/vol) is used to elaute miconazole is quantified on the basis of ultraviolet absorption at 220 nm. The precision of the method ranged from 3.21% at 0.5 mg/L to 0.85% at 2.0 mg/L. The limit of quantification was established as 0.1 mg/L. Interference from other drugs that are co-adimistered such as amphotericin B, 5-fluorocytosine of ketoconazole and most other comonly encountered drugs was not observed.  相似文献

Summary A high-performance liquid gel-permeation chromatographic method is described for the determination of human serum immunoglobulin G (IgG) by separating the fluorescent immuno complex from the free fluorescence-labeled antibody. Fluorescence-labeled antibody used in this study was fluorescein isothiocyanate (FITC)-labeled Fab fragment goat anti-human IgG (anti-IgG Fab). Immuno complexes and antibody of different molecular sizes can be separated. FITC-labeled anti-IgG Fab was added to the serum and the mixture is passed through the column. An immuno complex separates as well-delineated peak in the column void volume, and was measured by the fluorescence of the column eluate (Ex=490nm, Em=520nm). The total analysis time for a serum sample was approximately 15min. The minimum detection limit was 25 mg/dl. The relative standard deviation was below 2% (peak area). The results of the HPL-GPC analysis correlate well with those obtained by laser nephelometric assay (r=0.992).  相似文献

Summary Polarities of the carotenoids in human serum are very different; many nonpolar carotenoid hydrocarbons (e.g. -carotene, lycopene) and highly polar hydroxycarotenoids (e. g. -cryptoxanthin, zeaxanthin, lutein) can be found among them.Gradient elution chromatography was used for the separation of -carotene, lycopene, -cryptoxanthin, lutein and zeaxanthin in serum samples applying amino and cyano packings (Chromsil-NH2, Chromsil-CN). The effects of different stationary phases on the selectivity were compared. The method is particularly suitable for the direct determination of serum levels of carotenoid in serum extracts.  相似文献

Summary A new HPLC stationary phase was synthesized by thein situ covalent immobilization of human serum albumin (HSA). The protein was immobilized on a commerically available diol column which had been activated with 1,1-carbonyldiimidazole. Initial chromatographic studies show that this phase can be used for chiral separations of enantiomeric solutes and that these separations may reflectin vitro binding to the HSA. The effects of mobile phase composition and temperature on the stereochemical resolutions are reported.  相似文献

Summary A simple and rugged SPE method for the determination of tetracycline (TC), minocycline (MC) and demeclocycline (DCC) in porcine serum by high performance liquid chromatography (HPLC) was developed. The spiked serum sample was pretreated with 2% phosphoric acid followed by a simple and rugged solid-phase extraction procedure using the OasisTM HLB extraction cartridges. High and reproducible recoveries were obtained even though the cartridges were run dry. The extracted sample analytes were injected onto a Waters SymmetryShieldTM RP8 column. The mobile phase was a simple volatile solution containing 0.1% TFA, 2% methanol and 7% acetonitrile in Water. The antibiotics were detected at 350 nm. The calibration curves were linear from 2.0 to 25.0 μg mL−1 of TC and MC with DCC as the internal standard at a concentration of 25.0 μg mL−1. For six replicate analyses, the average recoveries of TC and MC from porcine serum sample fortified at the level of 2.5 μg mL−1 were 96.1% with 1.3% RSD and 101% with 0.54% RSD; at level of 0.5 μg mL−1 the average recoveries were 88% with 1.6% RSD and 97.8% with 1.4% RSD.  相似文献

Summary A very rapid, sensitive and reproducible HPLC method was developed for simultaneous determination of eight anti-epileptic drugs (AEDs): lamotrigine, primidone, ethosuximide, sulthiame, felbamate, phenobarbital, carbamazepine, phenytoin and oxcarbazepine-metabolite (10-hydroxy-carbazepine) in human serum. Sample purification requires only protein precipitation with an appropriate reagent. Separation was by reversed-phase HPLC, using a C18 column, 20% acetonitrile and 40 mM phosphoric acid buffer as mobile phase. Column temperature was set at 50°C, and measurement was by UV detection at 205 nm. The inter and intra-assay coefficients of variation (CV) ranged 1.13–7.10% and 1.14–8.49%, respectively. The absolute (measured) and relative (analytic) recoveries of the drugs ranged 96.7%–104.4% and 97.3%–106.1%, respectively. No interference with other common antiepileptic drugs and analgesics were observed. The method requires only 100 μl serum or less. It is very fast (sample preparation and analysis time approx. 23 min for all 9 AEDs), and suitable for routine clinical use, especially for epileptic patients on polytherapy.  相似文献

Summary A simple and rapid, high performance liquid chromatographic (HPLC) procedure for determination of mebeverine in dosage forms (tablet and liquid) is described. Reversed-Phase chromatography was carried out using a mobile phase containing 0.05 M ammonium acetate buffer and acetonitrile, [(45%, v/v) pH 5.2] with UV-detection (263 nm). Replicate regression analyses of three standard plots in the concentration range of 0.5–10 mcg mL−1 obtained on three different days gave a correlation coefficient >0.9995 and the coefficient of variation of the slopes <2.2%. The assay was precise within day and between days as indicated by ANOVA test. The recoveries from 10 replicate tablets of two commercial mebeverine brands and liquid were in order 99.3, 100.5 and 100.1% of the label amount and their coefficient of variations were 1.41, 0.89 and 0.69%, respectively. The limit of quantitation of mebeverine was 5 ng mL−1.  相似文献

Summary A method for the quantitative determination of L-alanine in triglycine sulphate crystals, in the presence of excess amounts of glycine, is described. L-alanine is determined after derivatization with 4-N,N-dimethyl-aminoazobenzene-4-sulfonyl chloride (DABS-Cl) and in the presence of DABS-valine used as the internal standard. The results obtained indicate that the method permits the determination of alanine in triglycine sulphate crystals at concentrations of about 1%.  相似文献

Summary An HPLC method with fluorescence detection has been developed for the determination of fluoxetine and its main metabolite norfluoxetine in human plasma. Pretreatment of the biological samples by liquid-liquid extraction was used to improve the sensitivity of a previously published SPE procedure. The method uses 200 μL plasma and recovery is good for both analytes. On a C8 column with a mixture of perchlorate buffer and acetonitrile as mobile phase fluoxetine, norfluoxetine and the internal standard (paroxetine) were eluted in less than 9 min, without interference from the biological matrix. Response for both analytes was linearly dependent on concentration over the range 2.5–500 ng mL−1, and repeatability (RSD%) was <4%. The limit of detection was 1 ng mL−1 for both fluoxetines. Application to plasma samples from depressed patients treated with fluoxetine gave good results. There was no interference from other common CNS drugs. This method seems to be a useful tool for clinical monitoring, because it requires small plasma samples and is highly sensitive and highly selective.  相似文献

Summary An HPLC method has been developed for the determination of fat-soluble vitamins. Ten fat-soluble vitamins were separated simultaneously on a 25 cm×4.6 mm i.d. Hypersil C18 column with acetonitrile-dichloromethane-methanol, 60:20:20 (v/v) as mobile phase at 1.0 mL min−1 with wavelength-programmed ultraviolet-visible-absorbance detection. Total analysis time was 12 min. The limits of detection were 0.03, 0.01, 0.55, 1.84, 0.02, 0.02, 0.01, 0.16, 0.33, 0.01, and 0.01 ng mL−1 for retinol, retinyl acetate, retinyl palmitate, β-carotene, ergocalciferol, cholecalciferol, tocopherol, tocopherol acetate, phylloquinone, menatetrenone, and menadione, respectively. Analysis of human serum 2–7 days after ingestion of oral vitamins and Chinese herbs led to the conclusion that the concentration of vitamins was higher than for control serum.  相似文献

Summary An HPLC method was developed for determination of amoxicillin, penicillin G, penicillin V, ampicillin, oxacillin, cloxacillin, nafcillin and dicloxacillin in serum from pigs and cattle. Serum was cleaned up by solid-phase extraction (SPE), ultra-filtered and derivatised. The method was linear in the range tested up to 2000 ng mL−1 of individual penicillins in serum. Limits of detection were 11–14 ng mL−1. Mean recoveries were 90–103% in the range 20–2000 ng mL−1. The relative repeatability, standard deviation was <10% at 20 ng mL−1 level and <6% in the range 100–2000 ng mL−1.  相似文献

Summary A rapid and sensitive high performance liquid chromatographic method is described for the determination of azlocillin in serum. This method involves a short manual protein precipitation of the sample followed by an injection into a PR 18 column for separation and quantitation. The mobile phase was a 22% (V/V) solution acetonitrile in phosphate buffer pH 4.8 at a flow rate of 2,5 ml/min. The spectrophotometer detector was set at 220 nm with a sensitivity of 0.08 AUFS.  相似文献

Summary A simple and very sensitive HPLC method, for the simultaneous determination in human plasma of adriamycin and its metabolite adriamycinol, is described. Plasmas from patients were stored frozen. Thawed samples were extracted by absorption of anthracyclin onto a small C18 column. After evaporation of the eluate and reconstitution of the residue with methanol (100L), 30 to 40L of the mixture were injected into the chromatograph. Separation was obtained using an RP 8 column with a mobile phase of formate buffermethanol-acetonitrile (502327, v/v). A spectrofluorimeter was used as detector. The limit of sensitivity of the assay was 50 pcg/ml of plasma.  相似文献

Summary An alternative to the traditional solvent extraction method used to extract and rapidly quantify ethyl-and vinylphenol and ethyl-and vinylgaiacol from wine is presented. The method is based on retention of volatile phenols on adsorbants. Among the tested resins, the most efficient, AG 2-X8 (anion exchange resin), worked as well with a synthetic solution as with wines. The percolation of clarified wine adjusted to pH 9 on this resin permits, in particular, the elimination of organic acids. Phenols are not eluted after rinsing the column with 1N HCl, but are eluted with methanol after this treatment. Good recovery (91 %) and good repeatability are observed. The eluate is directly analysed by HPLC on an RP18 column after two-fold dilution in water. The four volatile phenols were completely separated and detected by UV at 280 nm with high sensitivity (20–40 ppb). No interference with other compounds were noted in the different wines analysed.  相似文献

Summary A fluorometric method for determining indomethacin in serum by reversed-phase high performance liquid chromatography has been developed. Deproteinized serum containing indomethacin was injected directly onto a C18-bonded vinyl alcohol copolymer column with an alkaline mobile phase (pH 10.0, 35% acetonitrile in phosphate buffer), and was detected fluorometrically (Ex. 298 nm and Em. 375 nm) via postcolumn in-line alkaline hydrolysis at high temperature (140°C). The calibration curve was linear over the range of 0.1–10.0 g/ml when injecting a volume of 10 l of deproteinized serum. The detection limit (signal-to-noise ratio=3) for indomethacin in serum was 10 ng/ml using a 20-l aliquot of deproteinized serum.  相似文献

Isocratic separation was achieved using a Spherisorb 5m ODS column with an eluent consisting of water-orthophosphoric acid to pH 3.2: acetonitrile: methanol (523513). Summary A simple, fast, fully automated method for plasma serotonin determination is described. Full automation is obtained by coupling two devices: a sample processing station and a solid-phase autosampler. The sample processing station dilutes the plasma sample and is then connected, on-stream, with the solid-phase autosampler. It firstly fills a loop with all the solvents necessary for the sample clean-up, then, inverting the flow, pumps these solvents through the silica-bonded cation-exchange disposable extraction cartridge positioned on the autosampler. For the elution, the cartridge is switched on-stream with the HPLC analytical column and serotonin is eluted by the HPLC mobile-phase. The HPLC separation is performed by ion-pairing reversed-phase liquid chromatography. The column effluent is completely reduced by an electrochemical reactor and serotonin is detected in an oxidation-mode by a dual-cell electrochemical detector. The plasma sample is 50 l, the plasma sensitivity is 40 ng/l, the retention time is 6 min and the recovery is 95%. The repeatibility, the normal ranges for platelet-poor and for platelet-rich plasma have been established and correlation with manual HPLC calculated.Presented at the 17th International Symposium on Chromatography, September 25–30, 1988, Vienna, Austria.  相似文献

Summary A simple, low-cost, sensitive and selective HPLC method was developed for the determination of phenazopyridine in human plasma. The method employs UV detection of phenazopyridine and of the internal Standard at 2 different wavelengths. Calibration curves were linear over a large dynamic range, i.e., within 0.05–10.0 μg mL−1 with limit of quantification of 0.05 μg mL−1, and a limit of detection of 0.01 μg mL−1.  相似文献
