A one-step procedure was developed for the synthesis of 5-acetyl-2-amino-4-aryl-3-cyano-4H-pyrano[3,2-b]indoles involving the three-component reaction of 1-acetylindol-3(2H)-one with aromatic aldehydes and malononitrile in ethanol in the presence of triethylamine as the catalyst. The structure of 5-acetyl-2-amino-4-(4"-chloro-3"-nitrophenyl)-3-cyano-4H-pyrano[3,2-b]indole was established by X-ray diffraction analysis.  相似文献

Substituted 2-amino-4-aryl-3-cyano-5-oxo-5,6-dihydro-4H-pyrano[2,3-d]pyrido[3",2":4,5]thieno[3,2-b]pyridines were synthesized by the reactions of 4-hydroxy-1H-thieno[2,3-b;4,5-b]dipyridin-2-ones with arylidenemalononitriles or by the three-component reactions of hydroxythienodipyridinones with aldehydes and malononitrile in DMF in the presence of triethylamine. Methods for syntheses of substituted 3-alkoxycarbonyl-6-amino-4-aryl-2-(3-cyanopyridin-2-ylthiomethyl)-4H-pyrans were developed on the basis of the reactions of 4-(3-cyanopyridin-2-ylthio)acetoacetates and arylidenemalononitriles or aldehydes and malononitrile. Ethyl 4-(3-cyanopyridin-2-ylthio)acetoacetate and 4-methoxybenzylidenecyanothioacetamide were used for the synthesis of 6-(pyridin-2-ylthiomethyl)-3-cyanopyridine-2(1H)-thione.  相似文献

The reactions of aroylacetonitriles with the nickel chelate of ethyl acetoacetate afforded new block heterocyclic reagents, viz., 3-acetyl-4-amino-6-arylpyran-2-ones. The reactions of the latter with diethyl oxalate gave rise to ethyl 7-aryl-4,5(1H,5H)-dioxopyrano[4,3-b]pyridine-2-carboxylates.  相似文献

4-Methoxy-1,1,1-trifluoropent-3-en-2-one reacts with cyanothioacetamide to give 3-cyano-4-methyl-6-trifluoromethylpyridine-2(1H)-thione, which was transformed into 3-amino-4-methyl-6-trifluoromethylthieno[2,3-b]pyridine-2-carboxanilide by the reaction with chloroacetanilide. Summary By condensation of 2,2-difluoro-4-methyl-benzo[d]-1,3,2-2H-dioxaborines (12) with cyano acetic acid derivatives in presence of weak bases, 3-cyano-4-methyl-benzo[b]pyran-2-ones (13) or their 3-cyano-4-methyl-benzo[b]pyran-2-imine precursors (14) are available in satisfactory yields.

Summary The Claisen rearrangement of 7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (2 a) gave 7-hydroxy-8-(1-phenyl-2-propenyl)-3-phenyl-(4H)-1-benzopyran-4-one (3 a) and 2,3-dihydro-2,6-diphenyl-3-methyl-(7H)furo[2,3-h]-1-benzopyran-7-one (7 a). 2-Methyl-7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (2 b) afforded4 b and7 b. 8-Methyl-7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (12) gave only the alkali soluble product 7-hydroxy-8-methyl-6-(1-phenyl-2-propenyl)-3-phenyl-(4H)-1-benzopyran-4-one (13).3 a,4 b, and13 were further cyclized in acidic medium to9 a,10 b, and14 followed by dehydrogenation.This paper is dedicated to Dr. F. The three-component reaction of 4-hydroxy-6-methylpyran-2(2H)-one with cyanoacetic acid derivatives and carbonyl compounds in EtOH or in the ionic liquid, viz., 1-butyl-3-methylimidazolinium hexafluorophosphate ([bmim][PF6]), affords substituted 2-amino-7-methyl-5-oxo-4,5-dihydropyrano[4,3-b]pyrans. The yield of substituted pyrano[4,3-b]pyrans in [bmim][PF6] is by 10—14% higher than that in EtOH.  相似文献

Abstract  A new route to the synthesis of 2-arylthieno[2,3-d]pyrimidin-4(3H)-ones has been developed through heterocyclization of 2-amino-4,5-dimethylthiophene-3-carboxamide with aromatic aldehydes in boiling glacial acetic acid followed by air oxidation. The unoxidized intermediates, 2-aryl-2,3-dihydrothieno[2,3-d]pyrimidin-4(1H)-ones, are isolated when the reactions are carried out either at room temperature or under a nitrogen atmosphere. Summary Condensation of 3-aryl-5-thio-1,2,4-4H-trizoles (2a–i) and 2-bromodimedone (3) inTHF/benzene gave 2-aryl-6,6-dimethyl-8-oxo-5a-hydroxy-5,5a,6,7,8,8a-hexahydro-1,2,4-4H-triazolo[3,2-b]benzothiazoles (5a–i). These were also obtained by a one step synthesis on heating a mixture of dimedone,NBS, and2a–i in benzene containing a trace of benzoyl peroxide. Thermal dehydration of5a–i inPPA/anhydrous ethanol yielded the corresponding 2-aryl-6,6-dimethyl-8-oxo-5,6,7,8-tetrahydro-1,2,4-4H-triazolo[3,2,b]benzothiazoles (6a–i). The formation of [3,4-b] fused isomers (4a–i) during the reaction of2 with3 was ruled out by an unambiguous synthesis of8a–i. Antibacterial screening of selected compounds againstEscherichia coli andStaphylococcus aureus was not encouraging.

Zusammenfassung Die Kondensation von 3-Aryl-5-thio-1,2,4-4H-triazolen (2a–i) mit 2-Bromdimedon (3) inTHF/Benzol ergab 2-Aryl-6,6-dimethyl-8-oxo-5a-hydroxy-5,5a,6,7,8,8a-hexahydro-1,2,4,-4H-triazolo[3,2-b]benzothiazole (5a–i). Dasselbe Ergebnis wurde durch Erhitzen einer Mischung von Dimedon,NBS und2a–i in Benzol unter Zusatz einer Spur Benzoylperoxid in einer einstufigen Synthese erreicht. Thermische Dehydrierung von5a–i inPPA/Ethanol(abs.) ergab die entsprechenden 2-Aryl-6,6-dimethyl-8-oxo-5,6,7,8-tetrahydro-1,2,4-4H-triazolo[3,2-b]benzothiazole (6a–i). Die Bildung [3,4-b]-kondensierter Isomere (4a–i) während der Reaktion von2 mit3 konnte durch eine eindeutige Synthese von8a–i ausgeschlossen werden. Antibakterielles Screening ausgewählter Verbindungen gegenüberEscherichia coli undStaphylococcus aureus brachte keine ermutigenden Resultate.

3-Amino-2-(isoxazol-3-yl)-4-methoxymethyl-6-methylthieno[2,3-b]pyridine was synthesized by the reaction of 2(1H)-thioxopyridine-3-carbonitrile with 3-chloromethylisoxazole in the presence of two equivalents of KOH. Boiling of 3-amino-2-(isoxazol-3-yl)-4-methoxymethyl-6-meth-ylthieno[2,3-b]pyridine with Raney nickel results in 4-aminothieno[2,3-b;4,5-b′]dipyridine or 5-(4-amino-2-pyridyl)pyridine depending on the reaction conditions. Condensation of lactim ethers of 3,4-dihydrocarbostyril and 1H-2,3,4,5-tetrahydrobenz[b]azepin-2-one with malonodinitrile, cyanoacetamide, and ethyl cyanoacetate gave the corresponding 2-methylidene derivatives. A green, efficient, and rapid procedure for the synthesis of 2-amino-3-cyano-1,4,5,6-tetrahydropyrano[3,2-c]quinolin-5-one derivatives has been developed by one-pot condensation of 4-hydroxyquinolin-2(1H)-one, aldehyde, and malononitrile in the presence of ammonium acetate in EtOH. The synthesis of pyrano[3,2-a]acridines is presented, where 7-chloro-9-phenyl-2,3-dihydroacridin-4(1H)-one reacts with arylaldehydes and malononitrile in the presence of piperidine in ethanol, giving with high yield via a multicomponent method. Also a new synthesis of pyrazolo[4,3-a]acridines is reported, where 7-chloro-9-aryl-2,3-dihydroacridin-4(1H)-one on Claisen condensation with ethylformate followed by hydrazine hydrate treatment through the intermediate 7-chloro-4-hydroxy-9-aryl-1,2-dihydroacridin-3-carbaldehyde. The structures of newly synthesized compounds were deduced by spectroscopic techniques, elemental analysis, and single-crystal x-ray diffraction.  相似文献

A method for the synthesis of substituted 3-amino-2-(organylsulfinyl)thieno[2,3-b]pyridines by the Thorpe—Ziegler intramolecular cyclization of substituted 3-cyano-2-[(organyl-sulfinyl)methylthio]pyridines was proposed. 3-Amino-2-(organylsulfonyl)thieno[2,3-b]pyridines were obtained by reactions of substituted 3-cyanopyridine-2-thiones with chloromethyl organyl sulfones. Oxidative amination of 3-chloro-6,8-dimethylpyrimido[4,5-c]pyridazine-5,7(6H,8H)-dione with primary alkylamines and potassium amide in liquid ammonia gives rise to the corresponding 4-amino derivatives as the major products. The reactions with acyclic secondary amines are accompanied by annelation of the pyrrole moiety to the starting heterosystem to form 1-R-3-R"-6,8-dimethylpyrrolo[2",3";3,4]pyrimido[4,5-c]pyridazine-7,9(6H,8H)-diones. The reaction with piperidine as the aminating agent occurs exclusively as aminodehalogenation. The Sonogashira cross-coupling of 4-amino-3-chloro-6,8-dimethylpyrimido[4,5-c]pyridazine-5,7(6H,8H)-diones with terminal alkynes affords 1-R-2-R"-6,8-dimethylpyrrolo[3",2";3,4]pyrimido[4,5-c]pyridazine-7,9(6H,8H)-diones.  相似文献
