Based on their versatility with respect to amino acid type and sequence, polypeptides have become attractive for a number of biological applications such as drug delivery, biomineralization, and drugs. N‐carboxy anhydride (NCA) polymerization is a convenient way to rapidly prepare high‐molecular weight polypeptides with good control over molecular weight and polydispersity. However, the kinetics of the incorporation of NCA monomers into copolypeptides during random copolymerization are poorly understood. Here, kinetic data is presented that allows insight into the NCA polymerization of a terpolymer composed of three commercially relevant amino acids, namely, glutamic acid, lysine, and tyrosine. Furthermore, kinetic data and copolymerization parameters from the copolymerization of binary mixtures of these three amino acid NCAs is used to make predictions of the terpolymer composition. This study provides access to the information necessary to prepare functional copolypeptides with better‐defined sequence architecture that will be essential for the future development of polypeptide‐based materials. © 2014 Wiley Periodicals, Inc. We report a novel approach for fabrication of multifunctional conjugated polymers, namely poly(p‐phenylene)s (PPPs) possessing polypeptide (poly‐l ‐lysine, PLL) and hydrophilic poly(ethylene glycol) (PEG) side chains. The approach is comprised of the combination of Suzuki coupling and in situ N‐carboxyanhydride (NCA) ring‐opening polymerization (ROP) processes. First, polypeptide macromonomer was prepared by ROP of the corresponding NCA precursor using (2,5‐dibromophenyl)methanamine as an initiator. Suzuki coupling reaction of the obtained polypeptide and PEG macromonomers both having dibromobenzene end functionality using 1,4‐benzenediboronic acid as the coupling partner in the presence of palladium catalyst gave the desired polymer. A different sequence of the same procedure was also employed to yield polymer with essentially identical structure. In the reverse sequence mode, low molar mass monomer (2,5‐dibromophenyl)methanamine, and PEG macromonomer were coupled with 1,4‐benzenediboronic acid in a similar way followed by ROP of the L‐Lysine NCA precursor through the primary amino groups of the resulting polyphenylene. © 2015 Wiley Periodicals, Inc. N‐Carboxyanhydride ring‐opening polymerization (NCA ROP) is a synthetically straightforward methodology to generate homopolypeptides. Extensive control over the polymerization permits the production of highly monodisperse synthetic polypeptides to a targeted molecular weight in the absence of unfavorable side reactions. Sequential NCA ROP permits the creation of block copolypeptides composed of individual polypeptide blocks boasting different functionalities, secondary structures, and desirable chemical properties. Consequently, a plethora of novel materials have been generated that have found wide‐range applicability. This review offers an insight into contemporary synthetic approaches toward NCA ROP before highlighting a number of block copolypeptide architectures generated.  相似文献

A controlled metal‐free synthetic methodology toward well‐defined thermoresponsive polypeptides by decreasing the reaction temperature to 0 °C has been developed. Good control over the molecular weight in the polymerization of a trithiocarbonate‐functionalized N‐carboxyanhydride (MES‐l ‐Glu‐NCA) monomer was obtained using n‐hexylamine as the initiator at 0 °C. It yielded homopolypeptide macro‐transfer agent (PMESLG) with narrow molecular weight distribution (PDI < 1.3) and controllable chain length. Detailed 1H NMR and MALDI‐TOF‐MS analysis clearly confirmed that frequently occurring side‐reactions was absent at 0 °C, and the polymerization was controlled. The resultant PMESLG was applied to mediate the reversible addition‐fragmentation chain transfer (RAFT) polymerization of oligo‐ethylene‐glycol acrylate (OEGA) for the metal‐free synthesis of thermoresponsive polypeptides. These thermoresponsive polypeptides have well‐controlled molecular weight, adopted regular α‐helical conformation, and exhibited a lower critical solution temperature between 23 °C and 55 °C. To the best of our knowledge, there are very few reports about the synthesis of well‐defined thermoresponsive graft polypeptides via NCA polymerization and RAFT. Design and synthesis of biodegradable stimuli-responsive polypeptides are important areas considering their promising applications in biomedical fields. This article summarizes the most recent progresses in the development of stimuli-responsive polypeptide materials prepared via ring-opening polymerization of α-amino acid N-carboxyanhydrides. We discuss the design, synthesis and structure-property correlation of emerging materials including thermo-responsive, redox-responsive, photo-responsive and biomolecule responsive polypeptides. The synthesis of polypeptide‐containing block copolymers combining N‐carboxyanhydride (NCA) ring‐opening polymerization and atom transfer radical polymerization (ATRP) was investigated. An amide initiator comprising an amine function for the NCA polymerization and an activated bromide for ATRP was used. Well‐defined polypeptide macroinitiators were obtained from γ‐benzyl‐L ‐glutamate NCA, O‐benzyl‐serine NCA, and N‐benzyloxy‐L ‐lysine. Subsequent ATRP macroinitiation from the polypeptides resulted in higher than expected molecular weights. Analysis of the reaction products and model reactions confirmed that this is due to the high frequency of termination reactions by disproportionation in the initial phase of the ATRP, which is inherent in the amide initiator structure. In some cases selective precipitation could be applied to remove unreacted macroinitiator to yield well‐defined block copolymers. © 2009 Wiley Periodicals, Inc. The NCAs of the following five amino acids were polymerized in bulk at 120 °C without addition of a catalyst or initiator: sarcosine (Sar), L ‐alanine (L ‐Ala), D ,L ‐phenylalanine (D ,L ‐Phe), D ,L ‐leucine (D ,L ‐Leu) and D ,L ‐valine (D,L ‐Val). The virgin reaction products were characterized by viscosity measurements 13C NMR spectroscopy and MALDI‐TOF mass spectrometry. In addition to numerous low molar mass byproducts cyclic polypeptides were formed as the main reaction products in the mass range above 800 Da. Two types of cyclic oligo‐ and polypeptides were detected in all cases with exception of sarcosine NCA, which only yielded one class of cyclic polypeptides. The efficient formation of cyclic oligo‐ and polypeptides explains why high molar mass polymers cannot be obtained by thermal polymerizations of α‐amino acid NCAs. Polyamides (PA) constitute one of the most important classes of polymeric materials and have gained strong position in different areas, such as textiles, fibers, and construction materials. Whereas most PA are synthesized by step‐growth polycondensation, PA 6 is synthesized by ring opening polymerization (ROP) of ε‐caprolactam (ε‐CLa). The most popular ROP methods involve the use of alkaline metal catalyst difficult to handle at large scale. In this article, we propose the use of organic acids for the ROP of ε‐CLa in bulk at 180 °C (below the polymer's melting point). Among evaluated organic acids, sulfonic acids were found to be the most effective for the polymerization of ε‐CLa , being the Brønsted acid ionic liquid: 1‐(4‐sulfobutyl)?3‐methylimidazolium hydrogen sulfate the most suitable due to its higher thermal stability. End‐group analysis by 1H nuclear magnetic resonance and model reactions provided mechanistic insights and suggested that the catalytic activity of sulfonic acids was a function of not only the acid strength, but of the nucleophilic character of conjugate base as well. Finally, the ability of sulfonic acid to promote the copolymerization of ε‐CLa and ε‐caprolactone is demonstrated. As a result, poly(ε‐caprolactam‐co‐ε‐caprolactone) copolymers with considerably randomness are obtained. Polypeptides have received noticeable attention in the biomedical field due to their structural versatility and biomimetic properties. Particularly, polypeptides that are responsive to biological stimuli, such as mildly acidic extracellular and intracellular conditions, have great potential as delivery carriers for therapeutics. However, synthesis of high-molecular-weight acid-labile peptides is often daunting due to highly restrictive polymerization conditions and limitations in preserving acid-degradable functional groups. For instance, the popular N-carboxyanhydride (NCA) ring-opening polymerization (ROP) is efficient, but acid-labile NCA monomers are difficult to synthesize and store. In this study, acid-labile polypeptides with high molecular weights were synthesized under mild, permissive conditions using carboxylated urethane derivative monomers which are stable for ease of handling. The polymerization was successful in various organic solvents at room temperature, and did not require additional energy or initiation to drive the formation of NCA intermediates. The polymerization was also rapid enough to be independent of inert atmosphere. The strategy explored here to synthesize high-molecular-weight acid-labile polypeptides offers significant advantages including facile synthesis of acid-labile urethane derivative monomers that are stable, even in contact with moisture, and fast polymerization under easily achievable conditions. © 2014 Wiley Periodicals, Inc. Poly(o‐aminophenethyl alcohol) and its copolymers containing the aniline unit were synthesized in aqueous hydrochloric acid medium by chemical oxidative polymerization. The chemical composition of these novel polymers was determined spectroscopically, and their viscosities were measured. These polymers exhibit good solubility in organic solvents that is attributed mainly to the polar hydroxyethyl side groups. Their structures (chain conformation and morphological structure) and properties (conductivity, electrochemical characteristics, glass transition, and degradation behavior) were characterized and then interpreted on the basis of the chemical composition along with the electronic and steric hindrance effects associated with the hydroxyethyl side group. Overall, the side group has a significant effect on the polymerization and influences the structure, chain conformation, and properties of the resultant polymer. The poly(aniline‐coo‐aminophenethyl alcohol)s containing 20–40 mol % o‐aminophenethyl alcohol units are potential conducting materials for microelectronic and electromagnetic shielding applications because they are easier to process than polyaniline but retain its beneficial properties. These polymers can also be used as a functional conducting polymer intermediate owing to the reactivity of the side group. © 2002 Wiley Periodicals, Inc. Data on new modifications of supported titanium‐magnesium catalysts (TMCs) with improved performance in ethylene polymerization are reported. These catalysts possess a high and stable activity, an enhanced ability to regulate molecular weight of the polymer by hydrogen, a controllable particle size at a narrow particle size distribution, and the ability to produce the polymer with an increased bulk density. Various physicochemical methods were used to obtain data on the chemical composition of novel supports and catalysts, their phase composition and crystal structure as well as the pore structure. Poly(γ‐methyl L ‐glutamate)s with Ser, His, Asp, and Glu residues at the amino terminal as the serine protease catalytic site were prepared. The number‐average degree of polymerization of the polypeptides was 51. A dipalmitoylphosphatidylcholine monolayer containing the polypeptides was formed at the air–water interface and was transferred onto gold‐deposited glass plates. The binding of N‐acetyltyrosine ethyl ester, a typical substrate of the serine protease, to the monolayer was characterized by surface plasmon resonance measurements. The four‐polypeptide–lipid monolayer system conditioned on an aqueous solution containing the substrate N‐acetyltyrosine ethyl ester exhibited Langmuir‐type binding of the substrate. Its binding constant of 6.1 × 104 M−1 was about 20 times larger than that observed for a monolayer prepared on pure water. The behavior may have arisen from a substrate‐induced rearrangement of the four kinds of polypeptides in the monolayer, forming a substrate‐binding structure similar to that found in serine protease. © 2000 John Wiley & Sons, Inc. A series of thermoresponsive polypeptides bearing 1‐butyl, 1‐hexyl, or 1‐dodecyl side‐chains (i.e., 6a ‐ 6c ) were synthesized by copper‐mediated 1,3‐dipolar cycloaddition with high grafting efficiency (>95%) between side‐chain “clickable” polypeptide, namely poly(γ‐4‐(propargoxycarbonyl)benzyl‐L‐glutamate) ( 5 ) and 1‐azidoalkanes. 5 with different degree of polymerization (DP = 48–86) were prepared from triethylamine initiated ring‐opening polymerization of γ‐4‐(propargoxycarbonyl)benzyl‐L‐glutamic acid based N‐carboxyanhydride ( 4 ). 1H NMR, FTIR, and GPC results revealed the successful preparation of the resulting polypeptides. 6a ‐ 6c showed reversible UCST‐type phase behaviors in methanol, ethanol, and ethanol/water solvent mixtures depending on the polymer main‐chain length, alkyl side‐chain length, weight percentage of ethanol (fw) in the binary solvent, and so forth. FTIR analysis revealed the presence of the van der Waals interaction between the alkyl pendants of polypeptides and alkyl groups of alcoholic solvents. Variable‐temperature UV‐vis spectroscopy revealed that the UCST‐type phase transition temperature (Tpt) increased as polymer main‐chain length or concentration increased. In ethanol/water solvent mixtures, polypeptide with short alkyl pendant (i.e., 1‐butyl group) and short main‐chain length (DP = 41) showed the widest fw range and Tpts in the range of 61.0–71.1 °C. © 2016 Wiley Periodicals, Inc. A modified polysulfone containing benzylamine groups was synthesized as a reactive membrane material. Polysulfone was activated at the ortho‐sulfone site by direct lithiation with n‐butyllithium, and the resulting lithiated polysulfone was then reacted with benzonitrile; this yielded a polymer with pendant benzimine groups. The structure was confirmed by NMR and IR spectroscopy and by the transformation of imine to ketone by acid hydrolysis. The polymeric benzimine was also reduced to benzylamine with sodium cyanoborohydride in an acidic medium. The structure and degree of substitution of both benzylamine derivatives were determined by NMR and IR spectroscopy. The modified polysulfone containing benzylamine groups initiated the polymerization of N‐carboxyanhydride of γ‐benzyl‐L ‐glutamate [Glu(OBzl)–NCA]. The side‐chain oligopeptide of Glu(OBzl)–NCA attached to polysulfone was converted into molecular recognition sites. © 2003 Wiley Periodicals, Inc. Polymer topologies exert a significant effect on its properties, and polymer nanostructures with advanced architectures, such as cyclic polymers, star‐shaped polymers, and hyperbranched polymers, are a promising class of materials with advantages over conventional linear counterparts. Cyclic polymers, due to the lack of polymer chain ends, have displayed intriguing physical and chemical properties. Such uniqueness has drawn considerable attention over the past decade. The current review focuses on the recent progress in the design and development of cyclic polymer with an emphasis on its synthesis and bio‐related properties and applications. Two primary synthetic strategies towards cyclic polymers, that is, ring‐expansion polymerization and ring‐closure reaction are summarized. The bioproperties and biomedical applications of cyclic polymers are then highlighted. Thermosensitive diethylene glycol‐derived poly(L ‐glutamate) homopolypeptides (i.e., poly‐L ‐EG2‐Glu) with different molecular weights (MW) (Mn,GPC = 5380–32520) were synthesized via the ring‐opening polymerization (ROP) of EG2‐L ‐glutamate N‐carboxyanhydride (EG2‐Glu‐NCA) in N,N‐dimethylformamide solution at 50 °C. Their molecular structure, conformation transition, liquid crystal (LC) phase behavior, lower critical solution temperature (LCST) transition, and morphology evolution were thoroughly characterized by means of FTIR, 1H NMR, gel permeation chromatography, differential scanning calorimetry, wide angle X‐ray diffraction, polarized optical microscope, transmission electron microscope, and dynamic light scattering. In solid state, the homopolypeptide poly‐L ‐EG2‐Glu presented a conformation transition from α‐helix to β‐sheet with increasing their MW at room temperature, while it mainly assumed an α‐helix of 80–86% in aqueous solution. Poly‐L ‐EG2‐Glu showed a thermotropic LC phase with a transition temperature of about 100 °C in solid state, while it gave a reversible LCST transition of 34–36 °C in aqueous solution. The amphiphilic homopolypeptide poly‐L ‐EG2‐Glu self‐assembled into nanostructures in aqueous solution, and their critical aggregation concentrations decreased with increasing MW. Interestingly, their morphology changed from spherical micelles to worm‐like micelles, then to fiber micelles with increasing MW. This work provides a simple method for the generation of different nanostructures from a thermosensitive biodegradable homopolypeptide. © 2012 Wiley Periodicals, Inc. Novel thermoplastic elastomers based on multi‐block copolymers of poly(l ‐lysine) (PLL), poly(N‐ε‐carbobenzyloxyl‐l ‐lysine) (PZLL), poly(ε‐caprolactone) (PCL), and poly(ethylene glycol) (PEG) were synthesized by combination of ring‐opening polymerization (ROP) and chain extension via l ‐lysine diisocyanate (LDI). SEC and 1H NMR were used to characterize the multi‐block copolymers, with number‐average molecular weights between 38,900 and 73,400 g/mol. Multi‐block copolymers were proved to be good thermoplastic elastomers with Young's modulus between 5 and 60 MPa and tensile strain up to 1300%. The PLL‐containing multi‐block copolymers were electrospun into non‐woven mats that exhibited high surface hydrophilicity and wettability. The self‐assembling nature and phase‐transition behavior of a novel class of triarm, star‐shaped polymer–peptide block copolymers synthesized by the combination of atom transfer radical polymerization and living ring‐opening polymerization of α‐amino acid‐N‐carboxyanhydride are demonstrated. The two‐step synthesis strategy adopted here allows incorporating polypeptides into the usual synthetic polymers via an amido–amidate nickelacycle intermediate, which is used as the macroinitiator for the growth of poly(γ‐benzyl‐L ‐glutamate). The characterization data are reported from analyses using gel permeation chromatography and infrared, 1H NMR, and 13C NMR spectroscopy. This synthetic scheme grants a facile way to prepare a wide range of polymer–peptide architectures with perfect microstructure control, preventing the formation of homopolypeptide contaminants. Studies regarding the supramolecular organization and phase‐transition behavior of this class of polymer‐block‐polypeptide copolymers have been accomplished with X‐ray diffraction, infrared spectroscopy, and thermal analyses. This Highlight presents an overview of the rapidly growing field of dynamic covalent polymers. This class of polymers combines intrinsic reversibility with the robustness of covalent bonds, thus enabling formation of mechanically stable, polymer‐based materials that are responsive to external stimuli. It will be discussed how the inherent dynamic nature of the dynamic covalent bonds on the molecular level can be translated to the macroscopic level of the polymer, giving access to a range of applications, such as stimuli‐responsive or self‐healing materials. A primary distinction will be made based on the type of dynamic covalent bond employed, while a secondary distinction will be based on the consideration whether the dynamic covalent bond is used in the main chain of the polymer or whether it is used to allow side chain modification of the polymer. Emphasis will be on the chemistry of the dynamic covalent bonds present in the polymer, in particular in relation to how the specific (dynamic) features of the bond impart functionality to the polymer material, and to the conditions under which this dynamic behavior is manifested. © 2016 The Authors. Differing from the moisture‐sensitive α‐amino acid N‐carboxyanhydrides (AA‐NCAs) monomers, N‐phenoxycarbonyl α‐amino acids (AA‐NPCs) can be prepared and stored in open air. In this contribution, we report that the controlled polymerizations of AA‐NPC monomers of Otert‐butyl‐dl ‐serine (BRS‐NPC), Nε‐benzyloxycarbonyl‐l ‐lysine (ZLL‐NPC) and Nε‐trifluoroacetyl‐l ‐lysine (FLL‐NPC) initiated by amines are surprisingly able to tolerate common nucleophilic impurities such as water and alcohols at a level of monomer concentration. The structures of polypeptides synthesized in the presence of water or alcohols agree well with the designed ones in the case of repeated chain extensions. Detailed mechanism study and density functional theory calculation reveal that the low concentration of AA‐NCA and the high activity of amines are the key factors to the controllability of AA‐NPC polymerizations. The water‐ and alcohol‐tolerant property in polymerizations of AA‐NPCs encourages the following studies on unprotected (phenolic) hydroxyl groups containing AA‐NPCs. The controllable polymerizations of N‐phenoxycarbonyl l ‐tyrosine (LT‐NPC) and N‐phenoxycarbonyl S‐(3‐hydroxypropyl)‐l ‐cysteine (HLC‐NPC) initiated by amines are confirmed and reported for the first time, which extends the library of AA‐NPCs and polypeptides as well. All the universality of library, the convenience of monomer preparation, and the controllability and water‐ and alcohol‐tolerant property of polymerization of AA‐NPCs significantly enhance the feasibility of polypeptide synthesis, making AA‐NPC approach a promising synthetic method of polypeptides. © 2019 Wiley Periodicals, Inc.
