5β-androstan-3-ones carrying a 6α-OH group show in their mass spectra a key-ion indicating the loss of water and C-1 to C-4 as C4H5O? particle. 6β-OH isomers lose instead C-1 to C-4 in form of C4H7O?.In 6α-hydroxy-androstan-3-ones differentiation between the connection of the A/B-ring system is possible, because in 5α-isomers the loss of C-3 to C-7 occurs as a C5H6O2 particle, while the 5β-isomers lose the same C atoms as a C5H7O? unit.Compounds with a 6β-OH group in an A/B trans connected ring system show a tendency for thermal water elimination. After rearrangement of the double bond in 4,5 position the typical fragments for 3-keto-Δ4-steroids are obtained.Occasionally a strong influence of a 6-OH group on fragmentation reactions in the D-ring system is observed: The presence of a 6α-OH group in an androstan-3,17-dione enhances the loss of C-16 and C-17 in the form of acetaldehydenol. Also the connection of the A/B-ring system may have a considerable influence on this type of reaction: In 6,17β-dihydroxy-androstan-3-ones only by trans connection of the A/B-ring system, C-16 and C-17 are lost with high probability after water elimination.  相似文献

Six new limonoids, munropins A?F (16), were isolated from the aerial parts of a Chinese medicinal plant, Munronia pinnata (Meliaceae). The structures of 16 were assigned by detailed analyses of their spectroscopic data. Munropins A (1) and B (2) are limonoids possessing a prieurianin skeleton with α,β-unsaturated γ-lactam moieties at C-17. Munropins C (3), D (4), and E (5) are also prieurianin type limonoids with an α,β-unsaturated γ-lactone moiety, an acetyl group, and an acetoxyacetyl group at C-17, respectively. Munropin F (6) was assigned as a nimbolinin type limonoid with a γ-hydroxy-α,β-unsaturated γ-lactone moiety.  相似文献

Five new fawcettimine-type alkaloids (15) and three new natural products (68), along with four known analogues, fawcettimine (9), fewcettidine (10), lycopoclavamine B (11), and lycopladine B (12), were isolated from the whole plant of Lycopodium complanatum var. glaucum Ching. The structures of lycogladines A-H (18) were determined based on HRESIMS, 1D and 2D NMR spectroscopic analysis, as well as single-crystal X-ray diffraction. Compound 1 possesses a new ring system that is formed through the linkage of C-13-OC-2, which is rarely present in Lycopodium alkaloids (LAs). Compounds 18 were tested for their β-site amyloid precursor protein (APP)-cleaving enzyme 1 (BACE1) and acetylcholinesterase (AChE) inhibitory activities.  相似文献

A novel taxane with an unprecedented hemiacetal ring between C-13 and C-9 was isolated from the needles of Taxus cuspidata. The structure was characterized as (12αH)-2α,10β-diacetoxy-5α-cinnamoyloxy-9α,13α-epoxytax-4(20)-ene-11β,13β-diol (1). This is the first example of a natural taxane with a C-13 and C-9 oxygen bridge to form an unusual 6/8/6/6-membered ring system.  相似文献

(Z)-3β-Acetoxy- and (Z)-3 α-acetoxy-5, 10-seco-1 (10)-cholesten-5-one ( 6a ) and ( 7a ) were synthesized by fragmentation of 3β-acetoxy-5α-cholestan-5-ol ( 1 ) and 3α-acetoxy-5β-cholestan-5-ol ( 2 ), respectively, using in both cases the hypoiodite reaction (the lead tetraacetate/iodine version). The 3β-acetate 6a was further transformed, via the 3β-alcohol 6d to the corresponding (Z)-3β-p-bromobenzoate ester 6b and to (Z)-5, 10-seco-1 (10)-cholestene-3, 5-dione ( 8 ) (also obtainable from the 3α-acetate 7a ). The 1H-and 13C-NMR. spectra showed that the (Z)-unsaturated 10-membered ring in all three compounds ( 6a , 7a and 8 ) exists in toluene, in only one conformation of type C 1, the same as that of the (Z)-3β-p-bromobenzoate 6b in the solid state found by X-ray analysis. The unfavourable relative spatial factors (interdistance and mutual orientation) of the active centres in conformations of type C 1 are responsible for the absence of intramolecular cyclizations in the (Z)-ketoesters 6 and 7 ( a and c ).  相似文献

We present in this report the development of a convergent and highly stereocontrolled cycloaddition strategy toward the synthesis of C-1, C-6, and C-14 tris-oxygenated eudesmane sesquiterpenoids. This approach was demonstrated in the first total synthesis of (±)-6β,14-epoxyeudesm-4(15)-en-1β-ol (1), a structurally unique ethereal eudesmane sesquiterpenoid, via an effective Diels-Alder construction of a compact functionalized tricycle intermediate from readily available N-benzylfurfurylamine (2) and homoprenyl maleic anhydride (3) as the C(5) and C(10) building blocks, respectively.  相似文献

Three new sesquiterpenes, namely 3β,11-dihydroxy-4,14-oxideenantioeudesmane (1), 1β,10β,12,14-tetrahydroxy-allo-aromadendrane (2) and 1β,10β,13,14-tetrahydroxy-allo-aromadendrane (3), along with six known sesquiterpenes (49), were isolated from the roots of Solanum torvum. Compound 4 and 5 are epimers, their main difference lies in the C-11 configulation. Normally, epimers do not make a huge difference in C NMR spectra, but in this kind of structure of A, B, C rings, and C ring is sterically strained structure, stericall effects influence strongly the 13C NMR chemical shifts, when C-11 configulation changed, it makes a huge difference in the three ring of structure, such as C-6, C-7, C-11. New compound 2 and 3 are epimers and similar to compound 4 and 5, their just increase a hydroxy in C-1 and have a same regular pattern in C NMR spectra, otherwise, compound 5 was firstly confirmed by single-crystal X-ray diffraction.  相似文献

A new solvent of cellulose (1.5 mol/L NaOH/0.5 mol/L urea aqueous solution) was used as one of the homogeneous reaction media of polysaccharides for methylation, hydroxyethylation and hydroxypropylation. A water insoluble β-(1→3)-D-glucan, sample PCS3-Ⅱ, isolated from fresh sclerotium of Poria cocos was sulfated in dimethyl sulfoxide (Me2SO), carboxymethylated in NaOH, isopropanol solution, as well as methylated, hydroxyethylated and hydroxypropylated in the new solvent system, respectively, to obtain five water-soluble derivatives coded as S-PCS3-Ⅱ, C-PCS3-Ⅱ, M-PCS3-Ⅱ, HE-PCS3-Ⅱand HP-PCS3-Ⅱ. Their chemical structure and distribution of substitution were characterized by infrared spectroscopy (IR), elementary analysis (EA), ^1H-NMR, ^13C-NMR, 2D-COSY, 2D-TOCSY and 2D-^1H-detected ^1H ^13C HMQC spectra. The results reveal that the relative reactivity of hydroxyl groups of the β(1→3)-D-glucan is in the order C-6 > C-4 > C-2 on the whole. The substitution of the samples S-PCS3-Ⅱ, C-PCS3-Ⅱ and M-PCS3-Ⅱ occurred mainly at C-6 position and secondly at C-4 and C-2 positions, and that of HE-PCS3-Ⅱ occurred at C-6 and C-4 positions and of HP-PCS3-Ⅱ almost completely occurred at C-6 position. The degrees of substitution (DS) obtained from ^13C-NMR range from 0.23 to 1.27. The water solubility of the derivatives is in the order S-PCS3-Ⅱ > C-PCS3-Ⅱ > M-PCS3-Ⅱ > HE-PCS3-Ⅱ > HP-PCS3-Ⅱ. This work provides a novel and nonpolluting process for the methylation, hydroxyethylation and hydroxypropylation of β-(1→3)-D-glucan.  相似文献

Three C-14 oxygenated taxanes, 2α,5α,10β,14β-tetraacetoxytaxa-4(20),11-diene (1), 2α,5α,10β-triacetoxy-14β-(2-methylbutyryloxy)taxa-4(20),11-diene (2), and yunanaxane (3), major products of callus cultures of Taxus spp., were regio- and stereoselectively hydroxylated at the 7β position by a fungus, Absidia coerulea IFO 4011. Intriguingly, when 1 was co-administered with β-cyclodextrin and incubated with the fungus cell cultures, three other compounds 5α,9α,10β,13α-tetraacetoxytaxa-4(20),11-dien-14β-ol (7), 5α,9α,10β,13α-tetraacetoxytaxa-4(20),11-dien-1β-ol (8) and 5α,9α,10β,13α-tetraacetoxy-11(15→1) abeotaxa-4(20),11-dien-15-ol (9) were obtained.  相似文献

Mass spectra of steroids with a 3β-configurated hydroxy-group and a keto-function in position 6 are characterized in the case of cis-connection of the A/B-ring system by a key ion at M-71, whereas if the rings A and B are trans-connected the production of a key ion of mass 139 is preferred. The substitution of the 3-hydroxy group by a keto-function alters the main fragmentation routes: In the case of cis-connection of the A/B-ringsystem a key ion corresponding to the loss of C-3 to C-7 is observed, while by trans-connection of the A/B-ringsystem C-1 and C-2 are lost as C2H5-radical.  相似文献

New C-1 oxidized eremophilane-type sesquiterpenoids, subspicatins A, B, C, and D, and subspicatolide, were isolated from the root of Ligularia subspicata and their structures were established by spectroscopic and X-ray analyses. Subspicatins A and B were the major components. The species were found to be diverse both in the composition of the root chemicals and in the nucleotide sequences of the internal transcribed spacers (ITS) of the ribosomal RNA gene and the atpB-rbcL intergenic region, but the difference among the samples was not distinct but continuous.  相似文献

Ternifolide A (1), a new diterpenoid featuring a unique 10-membered lactone ring formed between C-6 and C-15, along with ternifolide B (2), a nor-diterpenoid, and ternifolide C (3) were isolated from the leaves of Isodon ternifolius. Both H-8 and H-9 being α-orientations in compound 1 were found for the first time. The absolute configurations of 1 and 3 were confirmed by X-ray diffraction study. The degree of substitution (DS) and distribution of O-acetyl groups of water-soluble cellulose acetate (CA) were investigated by 13C-NMR. For this purpose, three different series of CA samples with low DS were prepared by respective homogeneous reaction, i.e., (1) deacetylation of cellulose triacetate (CTA) in acetic acid—water solution (D-series), (2) reaction of CTA with hydrazine (H-series), and (3) acetylation of cellulose with acetic anhydride in a 10% LiCl-dimethylacetamide solution (A-series). It was found that (i) water-soluble CA can be obtained only from D-series products, (ii) the DS value of water-soluble CA ranges from 0.5 to 1.1, (iii) the D-series products exhibit little difference between the relative DS values at C-2, C-3 and C-6 hydroxyl groups, and (iv) the relative DS at C-6 hydroxyl groups is very high compared to those at C-2 and C-3 hydroxyl groups in H- and A-series products. Aqueous solution of water-soluble CA (D-series sample) showed no gel—sol transition, even when the temperature was raised to 95°C. X-ray diffraction observations revealed that the water-soluble D-series samples were essentially noncrystalline, but the water-insoluble A-series samples were crystalline. It was also found that the relative ease of acetylation is C-6 > C-2 > C-3.  相似文献

Three isopimarane diterpenes [fladins B (1), C (2), and D (3)] were isolated from the twigs and leaves of Chinese folk medicine, Isodon flavidus. The chemical structures were determined by the analysis of the comprehensive spectroscopic data, and the absolute configuration was confirmed by X-ray crystallographic analysis. The structures of 1–3 were formed from isopimaranes through the rearrangement of ring A by the bond break at C-3 and C-4 to form a new δ-lactone ring system between C-3 and C-9. This structure type represents the first discovery of a natural isopimarane diterpene with an unusual lactone moiety at C-9 and C-10. In the crystal of 1, molecules are linked to each other by intermolecular O-H···O bonds, forming chains along the b axis. Compounds 1–3 were evaluated for their bioactivities against different diseases. None of these compounds displayed cytotoxic activities against HCT116 and A549 cancer cell lines, antifungal activities against Trichophyton rubrum and T. mentagrophytes, or antiviral activities against HIV entry at 20 µg/mL (62.9–66.7) µM. Compounds 1 and 3 did not show antiviral activities against Ebola entry at 20 µg/mL either; only 2 was found to show an 81% inhibitory effect against Ebola entry activity at 20 µg/mL (66.7 µM). The bioactivity evidence suggested that this type of compound could be a valuable antiviral lead for further structure modification to improve the antiviral potential.  相似文献

Caesaldecins A and B (1a/1b), a pair of epimeric cassane-type diterpenoids containing an ether linkage between C-6 and C-20 and an α,β-butenolide ring in the northern hemisphere, were isolated from the leaves of the medicinal plant Caesalpinia decapetala (Roth) Alston, along with a new labdane-type diterpenoid, termed as 8(17),11(Z),13(E)-trien-15,18-dioic acid (2). Structures of 1 and 2 were established by extensive spectroscopic analyses, including HRESIMS, 1D and 2D NMR, and electronic circular dichroism (ECD) calculations. The in vitro antibacterial and inhibitory activities against a panel of bacteria and cancer cell lines were evaluated, respectively. Compound 2 displayed moderate antibacterial activity against the methicillin-resistant Staphylococcus aureus subsp. aureus (MRSA) with an MIC50 value of 5.99?μg?mL?1.  相似文献

The biosynthetic origins of amphidinoide B (1) were investigated on the basis of 13C-NMR data of 13C-enriched samples obtained by feeding experiments with [1-(13)C], [2-(13)C], and [1,2-(13)C2] sodium acetates in cultures of a dinoflagellate Amphidinium sp. These incorporation patterns suggested that 1 was generated from three successive polyketide chains, an isolated C1 unit from C-2 of acetates, six branched C1 units from C-2 of acetates, and an "m-m" and an "m-m-m" unit derived only from C-2 of acetates. The labeling patterns of amphidinolide B (1) were different from those of amphidinolide H (2), a 26-membered macrolide closely related to 1.  相似文献

Three novel pentacyclic triterpenoid dienolides, phyllenolide A (=3β‐acetoxyglutina‐5(10), 6‐dien‐27,8α‐olide; 1 ), phyllenolide B (=3β‐(benzoyloxy)glutina‐5(10),6‐dien‐27,8α‐olide; 2 ), and phyllenolide C (=3β‐(2‐hydroxybenzoyloxy)glutina‐5(10),6‐dien‐27,8α‐olide; 3 ), were isolated from the aerial parts of Phyllanthus myrtifolius Moon . (Euphorbiaceae). These three compounds possess an endocyclic γ‐lactone moiety across ring C and a homo‐annular diene system in ring B. Their structures were established by analyses of CD, NOED, and 2D‐NMR spectra.  相似文献

Three new nitrogen-containing diterpenoids, designated cespitulactams A (1), B (2), and C (3), were isolated from Cespitularia taeniata May. Compounds 1-3 are novel structures having a phenylethyl amino side at C-10 and with an amide function at C-20. Their structures were determined on the basis of extensive spectroscopic analysis and chemical correlation.
