In the presence of a catalytic amount of ytterbium(III) triflate and under microwave irradiation, mixtures of 2-amino-3-thiophenecarbonitriles, ketones, and silica gel afforded smoothly the corresponding amino-thieno[2,3-b]pyridine derivatives in one step. A wide variety of ketones were tested under these conditions. The reactions proceeded rapidly and afforded the desired products in good to excellent yields.  相似文献

A convenient modular synthesis for the construction of densely functionalized thieno[3,2-b]pyrroles, allosteric inhibitors of the Hepatitis C virus NS5B polymerase, is described. The route allows the introduction of substituents in positions 4, 5, and 6 of the thienopyrrole scaffold and can also be applied to the regioisomeric thieno[2,3-b]pyrrole core.  相似文献

The synthesis of two classes of bisheterocyclic compounds, pyrrolothiazoles and thiopyranopyridines, is reported. The 4-dimethylamino-2-dimethylaminomethylenamino-1-thiabuta-1,3-diene 1 is used like a useful building block for the chemoselective synthesis of these heterocycles. Indeed, synthon 1 can react as thiazabutadiene or thiabutadiene form to afford either five- or six-membered ring monocyclic azadienes, themselves being precursors to the bicyclic structures. Semi-empirical calculations were undertaken to explain this efficient chemoselectivity.  相似文献

The uncatalyzed reaction of N-(tert-butoxycarbonyl)-2-tert-butyldimethylsilyloxypyrrole 3 with 1,4-quinones bearing an electron withdrawing group at C-2 has been studied. Use of 1,4-quinones 4, 5 bearing an ester group at C-2 provided an efficient synthesis of the respective pyrrolidinobenzofuran adduct 9 or pyrrolidinonaphthofuran adduct 10 whereas use of 1,4-quinones 6, 7 and 8 bearing an acetyl group at C-2 afforded silyloxypyrroles 11, 12 and 13 resulting from direct electrophilic substitution of the silyloxypyrrole by the electrophilic quinone. Addition of Eu(fod)3 to the reaction of 2-acetyl-1,4-naphthoquinone 7 and 3-acetyl-5-methoxy-1,4-naphthoquinone 8 with N-(tert-butoxycarbonyl)-2-tert-butyldimethylsilyloxypyrrole 3 provided a method for obtaining the pyrrolidinonaphthofuran adducts 14 and 15 together with silyloxypyrroles 12 and 13. Oxidative rearrangement of pyrrolidinonaphthofuran adduct 15 to pyrrolidino pyranonaphthoquinone 16 using ceric ammonium nitrate in acetonitrile provided a novel approach for the synthesis of an aza-analogue of the pyranonaphthoquinone antibiotic kalafungin.  相似文献

In a three-step sequence starting from readily available starting materials, 2,3-carbon disubstituted furo[2,3-b]pyridines can be accessed in good yields and purity. Furo[2,3-b]pyridines bearing ester, amide and ketone groups at the 2-position can be prepared with a variety of aryl and alkyl groups at the 3-position.  相似文献

Double Fischer cyclisation was used to prepare indolo[3,2-b]carbazole and its 2,8-di-OMe, OH, Br and F-derivatives. N-monosubstituted derivatives of indolo[3,2-b]carbazole (methyl, hydroxymethyl, dimethylaminoethyl) were obtained starting from 5,11-di-Boc-indolo[3,2-b]carbazole.  相似文献

Synthesis of the strained 2H-thieto[3,2-b]quinoline ring system is reported for the first time. Treatment of (Z)-2-benzyliden-2H-thieto[3,2-b]quinoline derivatives of this heterocycle with base, at reflux in ethanol, causes a novel rearrangement to 2-phenylthieno[3,2-b]quinolines. Indeed, the one-pot reaction of 2-aminobenzaldehydes and (Z)-2-benzylidenethietan-3-one in refluxing basic ethanol leads directly to 2-phenylthieno[3,2-b]quinolines in good yields.  相似文献

Irradiation of N-phenyl substituted isoquinolines in acetonitrile containing 1 M NaOH in a multilamp reactor (MLR) furnished benzoxazolo[3,2-b]isoquinolin-11-ones. In contrast, irradiation of the N-benzyl substituted isoquinoline derivative under the same conditions afforded the hydrolysed N-benzylbenzamide derivative. The isoquinolinobenzoxazine was obtained by irradiating the N-benzyl substituted isoquinoline derivative at higher basic conditions. The required isoquinolines were synthesized under solvent-free, solid supported microwave conditions.  相似文献

Polysubstituted 2,3-dihydrofuro[2,3-b]pyridines and 3,4-dihydro-2H-pyrano[2,3-b]pyridines have been synthesized from 1,2,4-triazines using the inverse electron Diels-Alder reaction. For this purpose, 3-methylsulfanyl-1,2,4-triazines were reacted with several nucleophiles allowing the formation of appropriately substituted alkynes to undergo the intramolecular inverse electron demand Diels-Alder reaction. Sealed-tube microwave activation of the cycloaddition reaction has proved to be very efficient and allowed shorter reaction times. This strategy enabled an efficient synthesis of 3-hydroxy-2,3-dihydrofuro[2,3-b]pyridines and 4-hydroxy-3,4-dihydro-2H-pyrano[2,3-b]pyridines with several points of diversity on the bicyclic scaffold.  相似文献

A new aspect concerning chromone chemistry leading to the one-pot synthesis of functionalized novel ABCD-fused chromenopyrazolopyridines has been described. The synthesis involves a multicomponent reaction of chromone-3-benzoylhydrazones with isocyanides and acetylenedicarboxylates, whereupon novel complex tetracyclic benzopyrone derivatives containing three stereogenic centres were formed. The structure elucidation of the products was accomplished by 1D and 2D NMR experiments and confirmed by X-ray crystallographic analysis. Full assignment of all 1H and 13C NMR chemical shifts has been unambiguously achieved. The reaction mechanism is also discussed. In addition, eight chromenopyrazolopyridine derivatives were tested for possible biological activity (antioxidant and lipid peroxidation inhibition).  相似文献

A series of imidazo[2,1-b]thiazole and benzo[d]thiazolo[3,2-a]imidazole analogues were synthesized by stirring an equimolar mixture of dibenzoylacetylene with imidazole/thiazole derivatives in toluene or acetonitrile at room temperature. The products were generated in good yields and characterized by standard analytical techniques such as IR, 1H NMR, 13C NMR and mass spectrometry. The structure of products 19, 20, 22, 24 and 25 were also unambiguously confirmed by single crystal X-ray analysis.  相似文献

A facile synthesis of dimethyl 3-hydroxythieno[3,2-b]furan-2,5-dicarboxylate is reported from the available methyl thioglycolate and dimethyl acetylenedicarboxylate starting materials. An efficient and rapid synthesis of furo[3,2-b]pyridine 1 is described using a one-pot Sonogashira coupling/heteroannulation sequence. Regioselective lithiation of this synthon was performed leading to various 2-substituted furo[3,2-b]pyridines. In this work, we described an easy preparation of substituted 4-amino-5-cyano-1,3-thiazoles. These compounds have been used as starting materials to obtain two classes of compounds. New substituted [1,3]thiazolo[4,5-e]pyridines were synthesized in one step via Friedländer reaction. Diazotation of 4-amino-5-cyano-1,3-thiazoles afforded 4-chloro[1,3]thiazolo[4,5-d][1,2,3]triazines in one step. The later was substituted by a secondary amine to obtain substituted 4-amino[1,3]thiazolo[4,5-d][1,2,3]triazines.  相似文献

Bis(2-, and 3-thienylammonium) hexachlorostannates were condensed with ethoxymethylene derivatives of active methylene compounds in pyridine. The resulting condensation products IVa-IVc and Va-Vc on heating under reflux in Dowtherm or diphenyl ether provided various 4-hydroxythieno[2,3-b] pyridines (Ia-Ic) and 7-hydroxythieno[3,2-b]pyridines (IIa-IIc). Furan[3,2-b]pyrroles are important isosteres for the indole scaffold in which the benzene ring is replaced by the furan ring. A series of novel tetra-substituted furan[3,2-b]pyrroles was synthesized from a simple furaldehyde. The divergent synthesis allows for substitution on multiple positions on the scaffold, creating the potential for the formation of large libraries.  相似文献
