The inherent ability of peptides to self-assemble with directional and rationally predictable interactions has fostered a plethora of synthetic two-dimensional (2D) supramolecular biomaterials. However, the design of peptides with hierarchical assembly in different dimensions across mesoscopic lengths remains a challenging task. We here describe the structural exploration of a d /l -alternating cyclic octapeptide capable of assembling one-dimensional (1D) nanotubes in water, which subsequently pack laterally to form giant 2D nanosheets up to 500 μm long with a constant 3.2 nm thickness. Specific amino acid mutations allowed the mapping of structure–assembly relationships that determine 2D self-assembly. Nine peptide modifications were studied, revealing key features in the peptide sequence that nanosheets tolerated, while a total of three peptide variants included modifications that compromised their 2D arrangement. These lessons will serve as guide and inspiration for new 2D supramolecular peptide designs.  相似文献

Water-soluble π-conjugated polymers are increasingly considered for DNA biosensing. However, the conformational rearrangement, supramolecular organization and dynamics upon interaction with DNA have been overlooked, which prevents the rational design of such detection tools. To elucidate the binding of a cationic polythiophene (CPT) to DNA with atomistic resolution, we performed molecular simulations of their supramolecular assembly. Comparison of replicated simulations show a multiplicity of CPT binding geometries that contribute to the wrapping of CPT around DNA. The different binding geometries are stabilized by both electrostatic interactions between CPT lateral cations and DNA phosphodiesters and van der Waals interactions between the CPT backbone and the DNA grooves. Simulated circular dichroism (CD) spectra show that the induced CD signal stems from a conserved geometrical feature across the replicated simulations, i. e. the presence of segments of syn configurations between thiophene units along the CPT chain. At the macromolecular scale, we inspected the different shapes related to the CPT binding modes around the DNA through symmetry metrics. Altogether, molecular dynamics (MD) simulations, model Hamiltonian calculations of the CD spectra, and symmetry indices provide insights into the origin of induced chirality from the atomic to the macromolecular scale. Our multidisciplinary approach points out the hierarchical aspect of CPT chiral organization induced by DNA.  相似文献

A novel supramolecular clipping design for influencing the photophysical properties of functional molecular assemblies, by the preorganization (clipping) of chromophores, is described. Several chromophores end functionalized with molecular recognition units were designed. These molecular recognition units serve as handles to appropriately position these systems upon noncovalent interactions with multivalent guest molecules (supramolecular clippers). Towards this goal, we have synthesized 1,5‐dialkoxynaphthalene (DAN) and naphthalenediimide (NDI) functionalized with dipicolylethylenediamine (DPA) motifs. These molecules could preorganize upon noncovalent clipping with adenosine di‐ or triphosphates, which resulted in preassociated excimers and mixed (cofacial) charge‐transfer (CT) assemblies. Chiral guest binding could also induce supramolecular chirality, not only into the individual chromophoric assembly but also into the heteromeric CT organization, as seen from the strong circular dichroism (CD) signal of the CT transition. The unique ability of this design to influence the intermolecular interactions by changing the binding strength of the clippers furthermore makes it very attractive for controlling the bimolecular photophysical processes.  相似文献

We report the H‐type supramolecular polymerization of two new hydrophobic BODIPY derivatives equipped with ester and amide linkages. Whereas the ester‐containing BODIPY derivative undergoes an isodesmic supramolecular polymerization in which the monomers are parallel‐oriented, the replacement of the ester by amide groups leads to a highly cooperative self‐assembly process into H‐type aggregates with a rotational displacement of the dye molecules within the stack. The dye organization imposed by simultaneous π–π and hydrogen bonding interactions is the driving force for the cooperative supramolecular polymerization, whereas the absence of additional hydrogen bonds for the ester‐containing moiety does not suffice to induce cooperative phenomena.  相似文献

A “chirality driven self‐sorting” strategy is introduced for the controlled supramolecular organization of donor (D) and acceptor (A) molecules in multicomponent assemblies. The trans‐1,2‐bis(amido)cyclohexane (trans‐BAC) has been identified as a supramolecular motif with strong homochiral recognition to direct this chirality controlled assembly process of enantiomers in solution. Stereoselective supramolecular polymerization of trans‐BAC appended naphthalene diimide monomers (NDIs) has been probed in detail by spectroscopic and mechanistic investigations. This chirality‐driven self‐sorting design of enantiomeric components also offers to realize mixed and segregated D‐A stacks by supramolecular co‐assembly of the NDI acceptors with trans‐BAC appended dialkoxynaphthalene (DAN) donor monomers. Such an unprecedented chirality control on D‐A organization paves the way for the creation of supramolecular p‐n nanostructures with controlled molecular‐level organization.  相似文献

Micellar-enhanced ultrafiltration (MEUF) of phenol and a cationic surfactant, cetylpyridinium chloride (CPC), is studied using two polysulfone membranes of 5- and 50-kDa molecular weight cutoff (MWCO) and two ceramic membranes of 15- and 50-kDa MWCO. Filtrations are run under laminar cross-flow and steady-state conditions. The effect of operation variables (pressure and retentate flux) and membrane properties (nature and MWCO) on permeate flux, surfactant, and phenol rejections is analyzed. The permeate flux depends, among other variables, on the fouling favored by membrane-micelle interactions, which are strongest in the 50-kDa MWCO ceramic membrane. On the other hand, surfactant rejection is mainly determined by the pore size and influenced by the pressure for both 50-kDa MWCO membranes. An equilibrium distribution constant, K(s), of phenol between surfactant micelles and water is calculated. Its value is not significantly affected by operation conditions and membrane type. K(s) is also approximately 20% lower than the value determined in a previous work with batch dead-end ultrafiltration.  相似文献

The mesoscopic organization adopted by two primary amphipathic peptides, P(beta) and P(alpha), in Langmuir-Blodgett (LB) films made of either the pure peptide or peptide-phospholipid mixtures was examined by atomic force microscopy. P(beta), a potent cell-penetrating peptide (CPP), and P(alpha) mainly differ by their conformational states, predominantly a beta-sheet for P(beta) and an alpha-helix for P(alpha), as determined by Fourier transform infrared spectroscopy. LB films of pure peptide, transferred significantly below their collapse pressure, were characterized by the presence of supramolecular structures, globular aggregates for P(beta) and filaments for P(alpha), inserted into the monomolecular film. In mixed peptide-phospholipid films, similar structures could be observed, as a function of the phospholipid headgroup and acyl chain saturation. They often coexisted with a liquid-expanded phase composed of miscible peptide-lipid. These data strongly suggest that primary amphipathic CPP and antimicrobial peptides may share, to some extent, common mechanisms of interaction with membranes.  相似文献

Multiple noncovalent interactions can drive self‐assembly through different pathways. Here, by coordination‐assisted changes in π‐stacking modes between chromophores in pyrene‐conjugated histidine (PyHis), a self‐assembly system with reversible and inversed switching of supramolecular chirality, as well as circularly polarized luminescence (CPL) is described. It was found that l ‐PyHis self‐assembled into nanofibers showing P‐chirality and right‐handed CPL. Upon ZnII coordination, the nanofibers changed into nanospheres with M‐chirality, as well as left‐handed CPL. The process is reversible and the M‐chirality can change to P‐chirality by removing the ZnII ions. Experimental and theoretical models unequivocally revealed that the cooperation of metal coordination and π‐stacking modes are responsible the reversible switching of supramolecular chirality. This work not only provides insight into how multiple noncovalent interactions regulate self‐assembly pathways.  相似文献

Hierarchical chiral structures made up of dendritic oligo(L- or D-glutamic acid) moieties of folic acid derivatives induce supramolecular chirality in the self-assembled columnar structures of the folic acids. These folic acids self-assemble through the intermolecular hydrogen bonds of the pterin rings to form disklike tetramers. In the neat states, the stacked tetramers form thermotropic hexagonal columnar phases over wide temperature ranges, including room temperature. Addition of alkali metal salts induces chirality in the columnar phases. In dilute solution states in a relatively polar solvent (chloroform), the folic acid derivatives form non-chiral, self-assembled structures. In the presence of sodium triflate, the folic acid forms chiral columnar assemblies through the oligo(L-glutamic acid) moiety, similar to those formed in the liquid-crystalline (LC) states. The enantiomer of the folic acid induces columnar assemblies with reversed helicity. In the case of the diastereomer, no induced helicity is observed. Application of an apolar solvent (dodecane) drives the folic acid derivatives to form chiral assemblies in the absence of ions. In this case, lipophilic interactions promote nanophase segregation, which enhances the formation of chiral columns. Interestingly, the chiral supramolecular structure of the diastereomer induces the most intense circular dichroism. In both cases, the molecular chirality in the oligo(glutamate) moieties yields supramolecular chirality of the folic acids that self-assemble through cooperative molecular interactions.  相似文献

Host-defense, antibiotic peptides are believed to generate their cytolytic effects by interacting with the membranes of bacterial cells. Direct analyses of peptide interactions with real cellular membranes are difficult, however, due to the high complexity of physiological membranes. This review summarizes experimental work aiming to understand peptide-membrane interactions and their relationships with the peptides' biological actions using specific model systems. Varied model assemblies have been constructed that generally aim to mimic the fundamental lipid bilayer organization of the membrane. The model systems we will describe include multilamellar and unilamellar vesicles, planar lipid bilayers, lipid monolayers and micelles, and colorimetric biomimetic membranes. The different artificial models have facilitated examination of specific biological or chemical parameters affecting peptide action, for example the effect of membrane lipid composition on peptide affinities and membrane penetration, the relationship between membrane fluidity and peptide interactions, the conformations of active peptides, and other factors. We evaluate the strengths and limitations of the various approaches, and point to future directions in the field.  相似文献

Self-assembling peptides form a prominent class of supramolecular materials with in general good biocompatibility. To afford better control over the material properties, tremendous progress has been made in studying the supramolecular organization of the peptide assemblies. This knowledge has helped us to understand the correlation between the molecular structure of the peptide building blocks and the properties of the supramolecular products. However, peptide self-assembly consists of a complex pathway rather than a spontaneous thermodynamic process. This implies that the outcome of the self-assembly is critically governed by the assembly pathway. Here, we are going to discuss how peptide self-assembly can be modulated at the intermediate steps in the self-assembly pathway. The focus will be to demonstrate this engineering approach on the example of zero-dimensional/one-dimensional nanostructure selectivity over the β-sheet assembly pathway. In addition, we provide examples of biomedical applications of such steered peptide assemblies in the field of drug delivery and tissue engineering.  相似文献

Amphiphile supramolecular assemblies result from the cooperative effects of multiple weak interactions between a large number of subcomponents. As a result, prediction of and control over the morphologies of such assemblies remains difficult to achieve. Here, we described the fine-tuning of the shape, size, and morphology transitions of twisted and helical membranes formed by non-chiral dicationic n-2-n gemini amphiphiles complexed with chiral tartrate anions. We have reported that such systems express the chirality of the tartrate components at a supramolecular level and that the mechanism of the chiral induction by counterions involves specific anion cation recognition and the induction of conformationally labile chirality in the cations. Here, we demonstrate that the morphologies and dimensions of twisted and helical ribbons, as well as tubules, can be controlled and that interconversion between these structures can be induced upon modifying temperature, upon introducing small amounts of additives, or slightly modifying molecular structure. Specifically, electron microscopy, IR spectroscopy, and small-angle X-ray scattering show that (i) varying the hydrophobic chain length or adding gemini having bromide counterions (1%) or the opposite enantiomer (10%) leads to an increase of the diameter of membrane tubules from 33 to 48.5 nm; (ii) further addition (1.5%) of gemini bromide or a slight increase in temperature induces a transition from tubules to twisted ribbons; (iii) the twist pitch of the ribbons can be continuously tuned by varying enantiomeric excess; and (iv) it was also observed that the morphologies of these ribbons much evolve with time. Such unprecedented observations over easy tuning of the chiral supramolecular structures are clearly related to the original feature that the induction of chirality is solely due the counterions, which are much more mobile than the amphiphiles.  相似文献

Steroidal bile acids and their derivatives exhibit characteristic inclusion behaviors in the crystalline state. Their crystals present varied assemblies due to asymmetric molecular structures, which relate to supramolecular properties through cooperative weak interactions. An overview indicates that the steroidal assemblies lie in an intermediate position among various molecules and have hierarchical structures such as primary, secondary, tertiary, and host-guest assemblies like proteins. Such an interpretation brought about the idea that the assemblies with dimensionality present supramolecular chirality such as three-axial, tilt, helical, bundle, and complementary chirality. This concept of the supramolecular chirality enables us to understand formation of chiral crystals starting from the molecular chirality of the steroidal molecules.  相似文献

Fouling of nanofiltration membranes is studied during filtration of aqueous surfactant solutions under different conditions. To this purpose, four typical nanofiltration membranes (Desal51HL, NF270, NTR7450 and NFPES10) and three typical surfactants (nonionic neodol, anionic SDBS and cationic cetrimide) are selected. Fouling is studied as a function of the surfactant concentration, with and without addition of an electrolyte (NaCl), at different pH and when filtering a mixture of surfactants. Adsorption experiments and hydrophobicity measurements (to study the orientation of the surfactants on the membrane surface) are also performed under the different conditions. The least membrane fouling is found for the anionic surfactant SDBS, while for the cationic surfactant cetrimide very low relative fluxes are observed. Neodol shows an intermediate degree of fouling. Both hydrophobic and electrostatic interactions (in the case of ionic surfactants) between the membrane surface and the surfactant explain the degree of adsorption and hence fouling, as membrane fouling is correlated with the amount of adsorbed surfactant. The difference between cetrimide and SDBS becomes especially visible when changing the pH: increasing the pH leads not only to an opposite orientation of the adsorbed surfactants, but also to an opposite trend in adsorbed amount and membrane fouling. This study permits selection of an optimal nanofiltration membrane to recycle wastewater containing surfactants in the carwash industry. The optimal choice would be a hydrophilic membrane with a low molecular weight cut-off and a small negative surface charge at neutral pH. Cationic surfactants in the wastewater should also be avoided as much as possible.  相似文献

Monte Carlo simulations are used to model the self-organizing behavior of the biomineralizing peptide KSL (KKVVFKVKFK) in the presence of phosphate. Originally identified as an antimicrobial peptide, KSL also directs the formation of biosilica through a hypothetical supramolecular template that requires phosphate for assembly. Specificity of each residue and the interactions between the peptide and phosphate are considered in a coarse-grained model. Both local and global physical quantities are calculated as the constituents execute their stochastic motion in the presence and absence of phosphate. Ordered peptide aggregates develop after simulations reach thermodynamic equilibrium, wherein phosphates form bridging ligands with lysines and are found interdigitated between peptide molecules. Results demonstrate that interactions between the lysines and phosphate drive self-organization into lower energy conformations of interconnected peptide scaffolds that resemble the supramolecular structures of polypeptide- and polyamine-mediated silica condensation systems. Furthermore, the specific phosphate-peptide organization appears to mimic the zwitterionic structure of native silaffins (scaffold proteins of diatom shells), suggesting a similar template organization for silica deposition between the in vitro KSL and silaffin systems.  相似文献

Cell-penetrating peptides (CPPs) are short polycationic sequences that can translocate into cells without disintegrating the plasma membrane. CPPs are useful tools for delivering cargo, but their molecular mechanism of crossing the lipid bilayer remains unclear. Here we study the interaction of the HIV-derived CPP TAT (48-60) with model membranes by solid-state NMR spectroscopy and electron microscopy. The peptide induces a pronounced isotropic (31)P NMR signal in zwitterionic DMPC, but not in anionic DMPG bilayers. Octaarginine and to a lesser extent octalysine have the same effect, in contrast to other cationic amphiphilic membrane-active peptides. The observed non-lamellar lipid morphology is attributed to specific interactions of polycationic peptides with phosphocholine head groups, rather than to electrostatic interactions. Freeze-fracture electron microscopy indicates that TAT(48-60) induces the formation of rodlike, presumably inverted micelles in DMPC, which may represent intermediates during the translocation across eukaryotic membranes.  相似文献

Understanding the roles of various parameters in orchestrating the preferential chiral molecular organization in supramolecular self‐assembly processes is of great significance in designing novel molecular functional systems. Cyclic dipeptide (CDP) chiral auxiliary‐functionalized naphthalenediimides (NCDPs 1 – 6 ) have been prepared and their chiral self‐assembly properties have been investigated. Detailed photophysical and circular dichroism (CD) studies have unveiled the crucial role of the solvent in the chiral aggregation of these NCDPs. NCDPs 1 – 3 form supramolecular helical assemblies and exhibit remarkable chiroptical switching behaviour (M‐ to P‐type) depending on the solvent composition of HFIP and DMSO. The strong influence of solvent composition on the supramolecular chirality of NCDPs has been further corroborated by concentration and solid‐state thin‐film CD studies. The chiroptical switching between supramolecular aggregates of opposite helicity (M and P) has been found to be reversible, and can be achieved through cycles of solvent removal and redissolution in solvent mixtures of specific composition. The control molecular systems (NCDPs 4 – 6 ), with an achiral or D ‐isomer second amino acid in the CDP auxiliary, did not show chiral aggregation properties. The substantial roles of hydrogen bonding and π–π interactions in the assembly of the NCDPs have been validated through nuclear magnetic resonance (NMR), photophysical, and computational studies. Quantum chemical calculations at the ab initio, semiempirical, and density functional theory levels have been performed on model systems to understand the stabilities of the right (P‐) and left (M‐) handed helical supramolecular assemblies and the nature of the intermolecular interactions. This study emphasizes the role of CDP chiral auxiliaries on the solvent‐induced helical assembly and reversible chiroptical switching of naphthalenediimides.  相似文献

Manipulating the self-assembly pathway is essentially important in the supramolecular synthesis of organic nano- and microarchitectures. Herein, we design a series of photoisomerizable chiral molecules, and realize precise control over pathway complexity with external light stimuli. The hidden single-handed microcoils, rather than the straight microribbons through spontaneous assembly, are obtained through a kinetically controlled pathway. The competition between molecular interactions in metastable photostationary intermediates gives rise to a variety of molecular packing and thereby the possibility of chirality transfer from molecules to supramolecular assemblies.  相似文献
