A new ternary monocopper(II) complex with co‐ligands of 2,2′‐diamino‐4,4′‐bithiazole (dabt) and picrate (pic), namely [Cu(dabt)(pic)2], has been synthesized and characterized using elemental analyses, molar conductance measurements, infrared and electronic spectral studies and single‐crystal X‐ray diffraction. The crystal structure analyses revealed that the copper(II) ion has a {CuN2O4} distorted octahedral coordination environment. The hydrogen bonding interactions contribute to a three‐dimensional supramolecular structure in the crystal. The reactivity towards herring sperm DNA showed that the copper(II) complex can interact with DNA in the mode of intercalation. The molecular docking of the complex with DNA sequence d(ACCGACGTCGGT)2 demonstrated that the copper(II) complex is stabilized by hydrogen bonding interaction. The in vitro anticancer activities suggested that the copper(II) complex is active against selected tumor cell lines. The effects of the two co‐ligands in the copper(II) complex on DNA‐binding events and in vitro anticancer activity are preliminarily discussed. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

A water soluble chloro bridged binuclear copper(II) complex (3) and mononuclear complex (4) have been synthesized from chloro substituted 2‐oxo‐1,2‐dihydroquinolin‐3‐yl‐methylene‐2 hydroxybenzohydrazide 1 and 2 and CuCl2·2H2O. The structures of the complexes have been determined by single crystal X‐ray diffraction. The binding interactions of the ligands and complexes with CT‐DNA and protein have been evaluated by absorption and emission spectroscopic method. CT‐DNA and ethidium bromide (EB) competitive studies revealed that the compounds could interact with CT‐DNA through intercalation binding mode. Interactions of the compounds with BSA were also studied by UV−visible, fluorescence and synchronous fluorescence spectroscopic methods which showed that the compounds had a strong binding affinity with BSA through static quenching process. The cytotoxic effect of the compounds examined on cancer cell lines, such as A549 (lung cancer) and MCF7 (breast cancer) cell lines showed that all four compounds exhibited substantial cytotoxic activity.  相似文献

Three novel compounds, (η6‐p‐cymene)RuCl2(2‐fluoro‐5‐aminopyridine) (compound 1), (η6‐p‐cymene)RuCl2(5‐amino‐2‐chlorpyridine) (compound 2) and (η6‐p‐cymene)RuCl2(2‐bromo‐ 5‐aminopyridine) (compound 3), were synthesized and characterized. The compound 1 and 3 were determined by X‐ray diffraction, showing a distorted piano‐stool type of geometry with similar bond lengths and angles around the ruthenium. Compound 2 exhibited moderate in vitro activity against A549 and MCF‐7 human cancer cells, the other two lower activities. The UV–vis and fluorescent absorption titrations showed that three compounds binded with CT‐DNA in a minor groove. The intrinsic binding constants (Kb) were calculated to be 2.13(±0.03) × 105 M?1, 2.89(±0.03) × 105 M?1 and 2.45(±0.03) × 105 M?1 for compound 1, 2 and 3, respectively, by using UV–vis absorption titrations data. Among the three compound, the highest value of intrinsic binding constant of compound 2 was consistent with its highest cytoxicity against A549 and MCF‐7 human cancer cells in vitro.  相似文献

Azole compounds have attracted commercial interest due to their high bactericidal and plant‐growth‐regulating activities. Uniconazole [or 1‐(4‐chlorophenyl)‐4,4‐dimethyl‐2‐(1H‐1,2,4‐triazol‐1‐yl)pent‐1‐en‐3‐ol] is a highly active 1,2,4‐triazole fungicide and plant‐growth regulator with low toxicity. The pharmacological and toxicological properties of many drugs are modified by the formation of their metal complexes. Therefore, there is much interest in exploiting the coordination chemistry of triazole pesticides and their potential application in agriculture. However, reports of complexes of uniconazole are rare. A new cobalt(II) complex of uniconazole, namely dichloridotetrakis[1‐(4‐chlorophenyl)‐4,4‐dimethyl‐2‐(1H‐1,2,4‐triazol‐1‐yl‐κN4)pent‐1‐en‐3‐ol]cobalt(II), [CoCl2(C15H18ClN3O)4], was synthesized and structurally characterized by element analysis, IR spectrometry and X‐ray single‐crystal diffraction. The crystal structural analysis shows that the CoII atom is located on the inversion centre and is coordinated by four uniconazole and two chloride ligands, forming a distorted octahedral geometry. The hydroxy groups of an uniconazole ligands of adjacent molecules form hydrogen bonds with the axial chloride ligands, resulting in one‐dimensional chains parallel to the a axis. The complex was analysed for its antifungal activity by the mycelial growth rate method. It was revealed that the antifungal effect of the title complex is more pronounced than the effect of fungicide uniconazole for Botryosphaeria ribis, Wheat gibberellic and Grape anthracnose.  相似文献

Three new binudear cobalt (II) complexes with extended te-tracarboxylato- bridge have been synthesized and characterized, namely [Co2 (PMTA) (bpy)4] (1), [Co2(PMTA)-(phen)4] (2) and [Co2(PMTA) (NO2phen)4] (3), where PMTA represents the tetraanion of pyroniellitic acid, and bpy, phen, NO2-phen denote 2,2'-bipyridine, 1,10-phenan-throline; 5-nirto-1, 10-plienanthroline, respectively. Based on elemental analyses, molar conductivity measurements, IR and electronic spectra studies, it is proposed that these complexes have PMTA-bridged structures and consist of two cobalt (II) ions, each in a distorted octahedral environment. These complexes were further characterized by variable temperature magnetic susceptibility measurements (4-300 K) and the observed data were successfully simulated by the equation based on the spin Hamiltonian operator, giving the exchange integral J = - 1.02 cm-1 for 1, J = -1.21 cm-1 for 2 and J = - 1.18 cm-1 for 3, respectively. A series of cobalt(II) complexes containing tridentate 2‐pyrazolyl‐substituted 1,10‐phenanthroline ligands (L) with the general formula [LCoCl2] have been successfully synthesized and fully identified by IR spectroscopy, elemental analysis and mass spectroscopy. Cobalt complexes Co4–Co8 were further confirmed by X‐ray crystallographic analysis, and all the complexes adopted distorted trigonal pyramid geometries around the cobalt center. In combination with methylaluminoxane, the complexes exhibit high cis‐1,4‐selectivity for 1,3‐butadiene polymerization. The catalytic activities of the complexes mainly depend on the nature of the substituent and its position at the pyrazolyl ring of the ligand. Complexes having a bulkier substituent on the pyrazolyl ring of the ligand show lower catalytic activity and the incorporation of electron‐withdrawing substituent enhances the activity. Polymerization behaviors were almost not affected with varying [Al]/[Co] ratio, but both activity and the cis‐1,4 content decrease slightly as polymerization temperature increasing. The crystal structure of the title complex {[Co(TCB)2/2-(IMI)2(H2O)2][Co(IMI)4(H2O)2] } (H2O)4 (where TCB = 1,2,4,5-benzenetetracarboxylic anion; IMI = imidazole) has been determined by X-ray diffraction method. Crystal data for {[Co(TCB)2/2(IMI)2(H2O)2][Co(IMI)4(H2O)2]}-(H2O)4: triclinic, space group P 1, a = 1.0647(2) nm, b = 1.1165(1)nm,c = 1.00361(1)nm,α = 91.56(1)°,β = 111.34(1)°, γ = 115.642(10)°, V = 0.9772(3) nm5, Z = 1. The polymer cobalt (II) complex has a novel three-dimension network structure. Co(1) atom and Co(2) atom both are coordinated in an octahedral arrangement and located in the center of the coordination anion and the center of the coordination cation, respectively. Moreover four carboxyl groups of TCB are divided into two types, two para-carboxyl groups bridge Co(1) atom in monodentate fashion and other two para-carboxyl groups are in free.  相似文献

Three water‐soluble complexes, [Cu2L2Cl2] ( 1 ), [CoL2(im)2] ( 2 ) and [ZnLClH2O] ( 3 ) (HL = 8‐hydroxyquinoline‐5‐sulphonic acid; im = N ‐methylimidazole), were prepared and characterized using various spectral techniques. The DNA binding behaviour of complexes 1 – 3 was studied using UV–visible and circular dichroism (CD) spectra and cyclic voltammetry. All three complexes exhibit hypochromism but complexes 1 and 3 alone give a red shift of 4 nm with a significant binding constant of K b = 2.1 × 104 and 1.0 × 104 M−1, respectively, but complex 2 shows no red shift with lower K b of 4.1 × 103 M−1. The voltammetric E 1/2 of complex 1 on interaction with herring sperm DNA shifts to a more positive potential, as expected, than complex 2 due to higher DNA affinity. Additionally, analysis of electrochemical data yields a value of K +/K 2+ greater than one suggesting that complex 1 binds to DNA through intercalation in the M(I) state. Evidently in CD spectral analysis, complex 1 exhibits a decrease in molar ellipticity with a red shift of 10 nm and a significant decrease in intensity compared to complexes 2 and 3 . This clearly indicates that complex 1 induces the B → A transition to a greater extent than 2 and 3 . Oxidative cleavage using circular plasmid pUC18 DNA with complex 1 was investigated using gel electrophoresis. Interestingly, complex 1 displays a strong DNA binding affinity and is efficient in cleaving DNA in the presence of H2O2 at pH = 8.0 at 37 °C.  相似文献

A copper(II) complex was evaluated for its anti‐tumor activity. Firstly, electrophoretic studies were applied on the complex. These studies revealed the binding of the complex to calf thymus DNA, leading to a delay in electrophoretic mobility of the DNA molecule. Secondly, spectroscopic data pointed out that the λmax of DNA was shifted to a longer wavelength, which was accompanid by a hyperchromic shift. Moreover, the λmax of copper(II) complex was shifted to a shorter wavelength. The favorable reaction conditions between the DNA molecule and the copper(II) complex were studied. Thirdly, The effects of the ligand and the Cu(II) ion were tested separately on the DNA molecule by electrophoresis technique. Furthermore, the fluorescence quenching of DNA bound ethidium ion by Cu(II)‐Girard's T complex was noticed. The IR spectral data of DNA before and after the reaction with the copper(II) complex indicated that the interaction takes place through the carbonyl group of DNA nucleobases. Finally, a significant increase in the mean survival of EAC (Ehrlich ascites carcinoma) tumor‐bearing mice was observed when treated with the copper(II) complex. The tumor volume was also significantly reduced (p < 0.0001). Electrophoretic studies showed that the DNA pattern extracted from EAC cells of tumor‐bearing mice was affected after treatment with the copper(II) complex. Preparation and crystal structure of {4-chloro-2-[(2-morpholinoethylimino)methyl]phenolato} methanolchlorozinc(II) are reported. The X-ray structure reveals highly distorted square pyramidal geometry around zinc, binding to one phenolate O and two imine N atoms of the Schiff base, one methanol and one chloride. The complex and its ligand were tested in vitro for antibacterial and cytotoxic activity with a wide range of bactericidal activity and significant cytotoxic activity.  相似文献

A complex of formula [Ni(pobb)2](pic)2, (pobb = 1,3‐bis(1‐propylbenzimidazol‐2‐yl)‐2‐oxapropane, pic = 2,4,6‐trinitrophenol), has been synthesized and structurally characterized by physico‐chemical and spectroscopic methods. The crystals crystallize in the monoclinic system, space group C2/c, a = 25.766(11) Å, b = 14.943(7) Å, c = 19.543(14) Å, α = 90°, β = 129.722(4)°, γ = 90°, Z = 4. The coordination environment around nickel(II) atom can be described as a distorted octahedral geometry. The interactions of the ligand pobb and the nickel (II) complex with calf thymus DNA (CT‐DNA) are investigated by using electronic absorption titration, ethidium bromide‐DNA displacement experiments and viscosity measurements. The experimental evidence indicated the compounds interact with calf thymus DNA through intercalation.  相似文献

The synthesis and characterization of Ru (II) terpyridine complexes derived from 4′ functionalized 2,2′:6′,2″‐terpyridine (tpy) ligands are reported. The heteroleptic complexes comprise the synthesized ligands 4′‐(2‐thienyl)‐ 2,2′:6′,2″‐terpyridine) or (4′‐(3,4‐dimethoxyphenyl)‐2,2′:6′,2″‐terpyridine and (dimethyl 5‐(pyrimidin‐5‐yl)isophthalate). The new complexes [Ru(4′‐(2‐thienyl)‐2,2′:6′,2″‐terpyridine)(5‐(pyrimidin‐5‐yl)‐isophthalic acid)Cl2] ( 9 ), [Ru(4′‐(3,4‐dimethoxyphenyl)‐2,2′:6′,2″‐terpyridine)(5‐(pyrimidin‐5‐yl)‐isophthalic acid)Cl2] ( 10 ), and [Ru(4′‐(2‐thienyl)‐2,2′:6′,2″‐terpyridine)(5‐(pyrimidin‐5‐yl)‐isophthalic acid)(NCS)2] ( 11 ) were characterized by 1H‐ and 13C‐NMR spectroscopy, C, H, N, and S elemental analysis, UPLC‐ESI‐MS, TGA, FT‐IR, and UV‐Vis spectroscopy. The biological activities of the synthesized ligands and their Ru (II) complexes as anti‐inflammatory, antimicrobial, and anticancer agents were evaluated. Furthermore, the toxicity of the synthesized compounds was studied and compared with the standard drugs, namely, diclofenac potassium and ibuprofen, using hemolysis assay. The results indicated that the ligands and the complex 9 possess superior anti‐inflammatory activities inhibiting albumin denaturation (89.88–100%) compared with the standard drugs (51.5–88.37%) at a concentration of 500 μg g?1. These activities were related to the presence of the chelating N‐atoms in the ligands and the exchangeable chloro‐ groups in the complex. Moreover, the chloro‐ and thiophene groups in complex 9 produce a higher anticancer activity compared with its isothiocyanate derivative in the complex 11 and the 3,4‐dimethoxyphenyl moiety in complex 10 . Considering the toxicity results, the synthesized ligands are nontoxic or far less toxic compared with the standard drugs and the metal complexes. Therefore, these newly synthesized compounds are promising anti‐inflammatory agents in addition to their moderate unique broad antimicrobial activity.  相似文献

Two transition metal complexes, [Cu(FH)3]⋅2Cl⋅2H2O and [Ni(FH)3]⋅2Cl⋅2H2O, were synthesized from the reactions of furan‐2‐carboxylic acid hydrazide with CuCl2⋅2H2O and NiCl2⋅6H2O. The synthesized complexes were characterized using analytical and various spectral techniques. The structures of the complexes were determined using single‐crystal X‐ray diffraction. The interactions of the complexes with calf thymus DNA (CT‐DNA) were studied using absorption, fluorescence, cyclic voltammetric and viscosity measurements. The experimental results showed that the complexes could interact with CT‐DNA through intercalation. A gel electrophoresis assay demonstrated the ability of the complexes to cleave pBR322 DNA. The binding interaction of the complexes with bovine serum albumin was investigated using a fluorescence spectroscopic method. The radical scavenging ability, assessed using a series of antioxidant assays involving 2,2‐diphenyl‐2‐picrylhydrazyl radical, hydroxyl radical and nitric oxide radical, showed that the complexes possess significant radical scavenging properties. Further, the in vitro cytotoxic effect of the complexes examined on cancerous cell lines, such as human cervical cancer cells (HeLa) and human breast cancer cell line (MCF‐7), showed that the complexes exhibit significant anticancer activity.  相似文献

A novel bis[benzyl‐N′‐hydrazinecarbodithioato‐κ2 N′,S]nickel(II) complex was synthesized and characterized by means of various physical, chemical, and spectroscopic techniques. The X‐ray single crystal diffraction analysis indicated two independent close comparable bis‐chelated square planar complexes of trans‐configuration, where S‐benzyl dithiocarbazate (SBDTC) ligand is coordinated via N,S‐donor set. The complex is able to inhibit Ehrlich ascites carcinoma (EAC) cell proliferation by 51.81% and 75.75%, with 0.3 and 50 mg kg?1 (dose adjusted) dose, respectively, administered intraperitoneally for five successive days in mice model. Apoptotic cell morphological changes were examined using optical and fluorescence microscopy techniques. Expression pattern of apoptosis regulatory genes in EAC cells treated with the synthesized nickel(II) complex for five consecutive days showed an increased expression of P53, Bax, Cas‐8, Cas‐9, Cas‐3, Cyt‐c, and TNF‐α proapoptotic genes and decreased expression of antiapoptotic Bcl‐2 gene. The Ni(II) complex and Bleomycin (standard drug) were used in molecular docking coupled with molecular dynamics simulation studies with the aim to support the experimental results and to investigate the apoptotic effect towards the targeting apoptotic genes. Both experimental and computational studies reveal that the nickel(II) complex inhibits EAC cells growth successfully, suggesting a potential compound for cancer treatment.  相似文献

Complexes CuL3Cl2, PdL2Cl2 and PtL2Cl2, where L is a novel ligand from the series of 2-substituted 5-aminotetrazoles, namely 5-amino-2-tert-butyltetrazole (1), have been synthesized by the reaction of metal(II) chlorides with 1 and characterized by IR spectroscopy, thermal and X-ray analyses. The crystallographic structural analysis of these complexes revealed that 1 acts as a monodentate ligand coordinated to the metal via endocyclic N4 atom. Platinum complex demonstrates promising cytotoxicity against human cervical carcinoma cells with IC50 value average between those of cisplatin and carboplatin.  相似文献

Two isomeric Zinc (II) complexes constructed by 3,5‐bis(1‐imidazoly) pyridine has been synthesized and characterized by single crystal X‐ray diffraction, elemental analyses and infrared spectroscopy. The binding mode and ability of complex 1–2 with CT‐DNA were studied by UV and fluorescence spectra. The intrinsic binding constant Kb (Kb1 = 2.305 × 104 M?1, Kb2 = 3.095 × 104 M?1) and the observed association constant Kobs (Kobs1 = 1.523*106 M?1, Kobs2 = 2.057*106 M?1) indicated that the insertion ability of complex 2 with CT‐DNA is stronger than complex 1. Gel electrophoresis showed that complexes have a good ability to hydrolyze cleavage pBR322 plasmid DNA. The cytotoxicity and apoptosis studies showed that complexes exhibited excellent cytotoxic activity against HeLa cells, especially complex 2 had better growth inhibition than Cisplatin. Molecular docking study simulated the binding model of complexes with DNA (PDB:4av1), showing an imidazole plane of complex 2 can be inserted into a DNA base pair in relative parallel. A coordination polymer, [Cd(L)2(H2O)2]n (HL = 2-methyl-1H-4,5-imidazoledicarboxylic acid), was prepared from reactions of Cd(NO3)2 with L at room temperature. It was characterized by IR spectra and elemental analysis. The complex was structurally characterized by X-ray single-crystal diffraction revealing that the complex crystallizes in monoclinic with P2(1)/c space group, a = 12.203(10)?Å, b = 9.332(8)?Å, c = 7.554(7)?Å, β = 100.894(2)?Å, V = 844.7(13)?Å3, Z = 2. Fluorescence and UV absorption spectroscopy indicated that the complex can bind to fish sperm DNA. Gel electrophoresis assay demonstrated the ability to cleave the HL-60 plasmid DNA. Apoptotic study showed that the complex exhibited significant cancer cell (JEKO and KB) inhibitory rate.  相似文献

Three new mononuclear cobalt(II) complexes containing ligands with extended planar quinoxaline moieties, {dipyrido[3,2‐a:2′,3′‐c]phenazine (dppz) or dipyrido[3,2‐d:2′,3′‐f]quinoxaline (dpq)}, viz. [Co(dppz)(acac)2] · CH3OH ( 1 ), [Co(dpq)(tfnb)2] ( 2 ) and [Co(dpq)(dbm)2] ( 3 ), where acac = acetylacetonate, tfnb = benzoyltrifluoroacetone and dbm = dibenzoylmethane, have been synthesized and structurally characterized as octahedral complexes. The binding ability of the complexes with calf thymus (CT)‐DNA has been investigated by spectroscopic and viscosity measurements. Results indicate that all complexes bind to CT‐DNA via intercalative mode, and the DNA binding affinity of dppz complex 1 is apparently stronger than that of dpq complexes 2 and 3 . Furthermore, DNA photocleavage experiments indicate that these complexes are efficient DNA cleaving agents in UV‐A (365 nm) and hydroxyl radical (HO·), singlet oxygen (1O2) and superoxide anion (1O2?) serve as the major cleavage active species. In addition, interaction of the complexes with bovine serum albumin (BSA) was investigated using UV ? visible and fluorescence methods, which indicated that all complexes could quench the intrinsic fluorescence of BSA in a static quenching process. Two new Pd(II) complexes of N′‐(4‐(diethylamino)‐2‐hydroxybenzylidene)furan‐2‐carbohydrazide were synthesized and characterized using various spectral methods. The structure of one of the complexes was determined using single‐crystal X‐ray diffraction. DNA and protein binding affinities of the synthesized compounds were examined using UV–visible and fluorescence titration method. Four different mononuclear palladium(II) complexes of 3‐acetyl‐8‐methoxycoumarin Schiff bases were synthesized and characterized by spectrochemical techniques. Further analysis through X‐ray crystallography confirmed the structures of the complexes. Their interactive ability with Calf Thymus DNA and protein (Bovine Serum Albumin and Human Serum Albumin) were investigated by means of absorption and emission methods. The intercalative mode of binding with DNA was supported by EB displacement studies and viscosity measurements. Configurational changes that occurred in the proteins have been analysed with the help of 3D fluorescence studies. The complexes were shown to have good antimicrobial activity against the tested bacterial and fungal pathogens. In addition, antiproliferative activity of the complexes was evaluated on A549 and MCF‐7 cell lines and the complexes were comparatively more active than the standard drug cisplatin. Among the compounds, complex 3 was the most effective against MCF‐7 (IC50 value of 5.20 ± 0.15 μM) and A549 (5.09 ± 0.13 μM) compared with the other complexes 1 (6.48 ± 0.17 μM; 5.98 ± 0.09 μM), 2 (5.53 ± 0.12 μM; 5.85 ± 0.11 μM), 4 (6.73 ± 0.19 μM; 6.63 ± 0.16 μM) and cisplatin (16.79 ± 0.08 μM; 15.10 ± 0.05 μM) respectively. LDH and NO release assays confirmed the cytotoxic potential of the synthesized complexes.  相似文献
