Fourier transform infrared spectroscopy (FTIR) is a well-established, non-destructive method of obtaining chemical information from biological samples such as tissues and cells. This review focuses specifically on the development of infrared spectroscopic microanalysis at the single-cell level. Technological developments, including that of the infrared microscope, synchrotron radiation FTIR, and focal plane array detectors, and their impact on the field are discussed along with the various data processing procedures that are currently used to extract meaningful information. There is then an emphasis on live cell infrared (IR) imaging, including developments in water correction and microfluidic device design. The review concludes with look to future directions, highlighting the potential impact of quantum cascade lasers.  相似文献

应用FTIR-ATR光谱技术与多元统计分析相结合的方法对镰刀菌、立枯丝核菌、核盘菌、瓜果腐霉菌和辣椒疫霉菌等重要的土传致病真菌进行鉴别.对测得的FTIR光谱进行了归一化和二阶求导处理,选择了适合于真菌分类识别的14个光谱特征波段.在此基础上,采用典型判别分析和系统聚类分析对不同菌株进行了识别.结果表明,不同菌种间识别准确率达到100%,而镰刀菌种内不同菌株间识别准确率为95.56%,说明傅里叶变换红外光谱技术在植物病原真菌分类识别方面具有较大的应用潜能.  相似文献

Abstract

Honey authenticity is of great importance for commercial and health reasons. The most common adulteration methods are overfeeding of bees with sugar or by adding sucrose. In the study, a Fourier transform infrared spectroscopy based on chemometric method, a partial least squares, for determination of sucrose sirup adulteration of Turkish honeys was improved. For this purpose, honey samples were adulterated with known amounts of sucrose sirup in concentrations ranging between 5.94 and 27.80%. A calibration curve was constructed within 992–923?cm?1 with the regression coefficient of 0.999. Obtained results from the spectra were confirmed by high performance liquid chromatography-refractive index detector and a good correlation was found between the results of the spectroscopic and chromatographic methods. Additionally, some method validation parameters (linearity, repeatability, limit of detection, and quantification) of the spectroscopic method were studied and obtained results were compared with chromatographic method. To prove the usefulness of the method, twenty different honey samples were collected from markets in Turkey and analyzed for sucrose contents. The results indicated that predicted sucrose concentration of honey samples by the spectroscopic method ranged between 4.52 and 15.16% and results were confirmed by the chromatographic method with high correlation (correlation coefficient = 92%). The results of the present indicated showed that adulteration ratio of honey samples by sugar addition can be detected using Fourier transform infrared spectroscopy technique in a short time with high accuracy. In conclusion, this method could be regarded as a new alternative technique for routine analysis in quality control of honey samples.  相似文献

Bladder cancer is one of the most common cancers in Africa. It takes several days to reach a diagnosis using histological examinations of specimens obtained by endoscope, which increases the medical expense. Recently, spectroscopic analysis of bladder cancer tissues has received considerable attention as a diagnosis technique due to its sensitivity to biochemical variations in the samples. This study investigated the use of Fourier transform infrared (FTIR) spectroscopy to analyze a number of bladder cancer tissues. Twenty-two samples were collected from 11 patients diagnosed with bladder cancer from different hospitals without any pretreatment. From each patient two samples were collected, one normal and another cancerous. FTIR spectrometer was used to differentiate between normal and cancerous bladder tissues via changes in spectra of these samples. The investigations detected obvious changes in the bands of proteins (1650, 1550 cm–1), lipids (2925, 2850 cm–1), and nucleic acid (1080, 1236 cm–1). The results show that FTIR spectroscopy is promising as a rapid, accurate, nondestructive, and easy to use alternative method for identification and diagnosis of bladder cancer tissues.  相似文献

Fourier transform infrared (IR) diffuse reflectance optics have been applied to study molecular orientation of the surfaces of nylon, glass fibers and needle-like organic crystals. Samples were arranged parallel to the incidence first, and then perpendicular to it. The relative intensity changes between bands in the spectra show the molecular level orientation rather than the macroscopic geometry of the sample. This technique has the advantage over internal reflectance in that there is no optical contact problem.  相似文献

Fourier transform infrared emission spectra of MnH and MnD were observed in the ground X7Σ+ electronic state. The vibration-rotation bands from v = 1 → 0 to v = 3 → 2 for MnH and from v = 1 → 0 to v = 4 → 3 for MnD were recorded at an instrumental resolution of 0.0085 cm−1. Spectroscopic constants were determined for each vibrational level and equilibrium constants were found from a Dunham-type fit. The equilibrium vibrational constant (ωe) for MnH was found to be 1546.84518(65) cm−1, the equilibrium rotational constant (Be) is 5.6856789(103) cm−1 and the eqilibrium bond distance (re) was determined to be 1.7308601(47) Å.  相似文献

Application of a high temperature wall-stabilized argon arc source (HTAAS) for high resolution near- and mid-infrared Fourier transform infrared spectroscopy (FTIR) has been investigated. Spectra of N2O, NH3 and the complex OC-HCl were recorded at maximum apodized instrumental resolution (0.004 cm−1). The signal to noise (S/N) ratio of these molecular absorption transitions were compared with corresponding spectra recorded using a 1520(30) K globar source. At 7.6 μm, recorded 14N2 16O transitions are consistent with a HTAAS at approximately 10,000 K and an enhancement of 12.6 in recorded S/N ratios with respect to the latter globar source. Such results demonstrate a significantly enhanced performance for specific FTIR applications such as high resolution static gas phase spectroscopy and molecular spectroscopy in supersonic jet expansions, and potentially for infrared microscopy.  相似文献

Fake liquors have caused severe body injuries or even deaths worldwide, rapid detection of such lethal drinks is thus quite necessary. Methanol has been identified as a primary cause of the problem, so methanol monitoring is critical to the detection of fake liquors. The present work provides an effective strategy for rapid detection of different lethal fake liquors. Using gas-phase Fourier transform infrared spectroscopy in a digitally labeled approach, the spectral bands of methanol were extracted by the iterative discrete wavelet transform for classification, which is named as digitally labeled gas-phase Fourier transform infrared spectroscopy. In digitally labeled gas-phase Fourier transform infrared spectroscopy, principal component analysis and least square support vector machine were combined to discriminate problematic samples using the iterative discrete wavelet transform filtered signals. As a result, the method could cleverly extract spectral features of methanol from the alcoholic drinks in the presence of uncontrolled matrix effects. The recognition accuracy was higher than 97.0%, and each measurement was done within 3?min. The results illustrate that the digitally labeled gas-phase Fourier transform infrared spectroscopy method serves well to rapidly discriminate fake liquors as an efficient and promising tool, and could be well extended to detection of any other targeted volatile substance in complicated systems.  相似文献

The first field measurements of volcanic gases using mid-IR difference frequency laser spectroscopy are reported. The results were obtained at the summit crater of Masaya volcano, Nicaragua, with the gases being drawn into a multi-pass cell and measured at reduced pressure. Automated sensitive and selective detection of CO2, SO2, H35,37Cl, H2O, and CH4 was achieved. Simultaneous measurements obtained with open-path Fourier transform spectroscopy provide a useful comparison of the two optical techniques. We also consider the potential measurement of CO2 isotopic ratios in volcanic gases using laser-based spectroscopy.  相似文献

Kidney stone is the most painful and prevalent urological disorder of the urinary system throughout the world. Thus, analysis of kidney stones is an integral part in the evaluation of patents having stone disease. Spectroscopic investigations of stones provide an idea about the pathogenesis of stones for its better cure and treatment. Hence, the present work targets multispectroscopic investigations on kidney stones using Fourier transform infrared (FTIR) and wave dispersive X‐ray fluorescence (WD‐XRF) spectroscopy which are the most useful analytical methods for the purpose of bio‐medical diagnostics. In the present study, FTIR spectral method is used to investigate the chemical composition and classification of kidney stones. The multicomponents of kidney stones such as calcium oxalate, hydroxyl apatite, phosphates, carbonates, and struvite were investigated and studied. Qualitative and quantitative determination of major and trace elements present in the kidney stones was performed employing WD‐XRF spectroscopy. The wide range of elements determined in the kidney stones were calcium (Ca), magnesium (Mg), phosphorous (P), sodium (Na), potassium (K), chlorine (Cl), sulfur (S), silicon (Si), iodine (I), titanium (Ti), iron (Fe), ruthenium (Ru), zinc (Zn), aluminum (Al), strontium (Sr), nickel (Ni), copper (Cu), and bromine (Br). For the first time, ruthenium was detected in kidney stone samples employing WD‐XRF in very low concentration. Our results revealed that the presence and relative concentrations of trace elements in different kinds of stones are different and depend on the stone types. From the experiments carried out on kidney stones for trace elemental detection, it was found that WD‐XRF is a robust analytical tool that can be useful for the diagnosis of urological disorders. We have also compared our findings with the results reported using XRF technique. A technique for image encryption using fractional Fourier transform (FRT) and radial Hilbert transform (RHT) is proposed. The spatial frequency spectrum of the image to be encrypted is first segregated into two parts/channels using RHT, and image subtraction technique. Each of these channels is encrypted independently using double random phase encoding in the FRT domain. The different fractional orders and random phase masks used during the process of encryption and decryption are the keys to enhance the security of the proposed system. The algorithms to implement the proposed encryption and decryption scheme are discussed, and results of digital simulation are presented.  相似文献

提出一种基于微光学元件的空间调制微型傅里叶变换红外光谱仪,通过引入红外微结构衍射光学元件、多级微反射镜和微透镜阵列,实现仪器的微型化.介绍了微型傅里叶变换红外光谱仪的结构及基本原理,分析了微型准直系统和聚焦耦合光学系统的设计理论,研究了单片折衍混合准直透镜的残存像差、衍射面的衍射效率、多级微反射镜的衍射、微透镜阵列的孔径衍射和中继系统的轴向装配误差对光谱复原的影响.最后,对中波红外微型傅里叶变换光谱仪进行了建模仿真,得到的复原光谱与理想的光谱曲线比较符合,实际的光谱复原误差为2.89%.该中波红外微型静态傅里叶变换光谱仪无可动部件,且采用了微光学元件取代了传统的红外镜头,不仅稳定性良好,而且体积小、重量轻,有利于在线监测应用.  相似文献

This article reviews some of the recent advances on FTIR spectroscopy in areas related to natural tissues and cell biology. It is an update on our previously published review on the applications of spectroscopic methods employed for the analysis of biological molecules, and summarizes some of the most widely used peak frequencies and their assignments. The aim of this review is to update and consolidate our previously published spectral database, which will help researchers in defining the chemical structure of the biological tissues introducing most of the important peaks present in the natural tissues more precisely and accurately. In spite of applying different methods, there seems to be a considerable similarity in defining the peaks of identical areas of the FTIR spectra. As a result, it is believed that preparing a unique collection of the frequencies encountered in FTIR spectroscopic studies can provide substantial help for future studies. In this article, we have included recent studies that have been reported since previous publication that will be of considerable assistance and added value to those who are focusing their research on defining chemical pathway to the progression of different diseases.  相似文献
