Synthesis and Taste Properties of 4,1′,4′,6′‐Tetrahalodeoxysucrose Analogues

The synthesis of a series of 1,4,6‐trideoxy‐1,4,6‐trihalo‐β‐d‐hexulofuranosyl 4‐deoxy‐4‐halo‐α‐d‐hexopyranosides is described. The 4‐chloro‐, 4‐bromo‐ and 4‐iodo‐4‐deoxy‐β‐d‐fructofuranosyl analogues were synthesized from a 3′,4′‐lyxo‐epoxide using the respective alkali metal halides. The corresponding 4‐halodeoxytagatofuranosyl analogues, on the other hand, were obtained by direct halide displacement of the 4′‐O‐trifluoromethanesulfonyl derivative, which was derived by regioselective sulfonylation of 1,6‐di‐O‐trityl‐β‐d‐fructofuranosyl 6‐O‐trityl‐α‐d‐glucopyranoside via its stannylene acetal. The sweetness intensities of these tetrahalodeoxy compounds strongly suggest that both size and configuration of the halogen substituents at C‐4 and C‐4′ are critical for sweetness enhancement.     

Diastereoselective and enantioselective synthesis of 4′-aza analogues of 2′,3′-dideoxynucleosides

A diastereo- and enantioselective synthesis of 4′-aza analogues of 2′,3′-dideoxynucleosides has been designed by the strategy of the 1,3-dipolar cycloaddition reaction of a Vasella-type nitrone. The reaction leads to (1′R)- and (1′S)-4′-aza analogues of 2′,3′-dideoxythimidine and fluorouridine, in enantiomerically pure forms.     

Synthesis of 3,4-Biheterylthieno[2,3-b]-thiophenes. Part I: Synthesis of 3,4-Bi(1′,3′,4′-thiadiazolyl-, s-triazolyl-,1′,3′,4′-thiadiazinyl-, 1′,3′,4′-triazinyl-, thiazolyl-, 1′,3′-thiazinyl- and primidinyl)-thieno[2,3-b]thiophenes

3,4-Diamino-2,5-dicarbethoxythieno[2,3-b]thiophene ( 1 ) was allowed to react with NaNO 2 and active methylenes to afford the corresponding azo compounds 2a-c . Hydrazonyl chloride 2a was treated with carbon disulfide, phenyl isothiocyanate, benzonitrile, benzyl cyanide, malononitrile, benzalaniline, ethyl mercaptoacetate, and ethyl glycinate to give 1,3,4-thiadiazolyl-, s-triazolyl-, 1,3,4-thiadiazinyl-, 1,3,4-triazinylthieno[2,3-b]thiophenes 3-6 respectively. The reaction of 2b,c with urea, thiourea, and guanidine afforded pyrimidinyl- and thiazinylazothieno [2,3-b]thiophenes 7-10 respectively. Bithiazolylthieno[2,3-b]thiophenes 11 and 13 were synthesized by treating compound 1 with CS 2 along with halo compounds. The addition of S,S-, N,S-, and N,O-acetals to the Schiff base 14 afforded compounds 15-17 respectively.     

Synthesis and mesomorphic properties of 4-methoxyphenyl 4′- n -alkoxy-2′,3′,5′,6′-tetrafluorobiphenyl-1-carboxylates

A novel type of 4-methoxyphenyl 4′-n-alkoxy-2′,3′,5′,6′-tetrafluorobiphenyl-1-carboxylates have been synthesized. Textural observations by polarizing microscopy and DSC measurements of the phase transitions show that all of these compounds are thermotropic liquid crystals with only a nematic mesophase. The relationship between the properties and chemical structures of these compounds is discussed.     

Isolation and Crystal Structure of 1′,4′-Trans-diol of Abscisic Acid

1 ′,4′-Trans-diol of abscisic acid was isolated from botrytis cinerea as a colorless crystal. The molecular and crystal structures have been determined by X-ray diffraction analysis. It crystallizes in orthorhombic system, space group P212121 with a = 6.724(3), b = 17.559(6), c =12.265(2) (A), a = β = y = 90°, V = 1448.1(8) (A)3, Z = 4, Dx = 1.222 g/cm3, F(000) = 576 and μ(MoKa) = 0.087 mm-1. The final R = 0.0628 and wR = 0.1604 for 2501 independent reflections with Rint = 0.0160 and 1679 observed reflections with I ＞2σ(Ⅰ). There are three intermolecular hydrogen bonds in a unit cell.     

Synthesis of Steroidal Spiro-1′,2′,4′-triazolidine-3′-thiones

Steroidal ketone thiosemicarbazones (4–6), obtained from the corresponding ketones(1–3), on oxidative cyclization with H₂O₂ at 0°C provide title compounds (7–9), respectively. The structures of these compounds have been established on the basis of their elemental analytical and spectral data.     

An Improved Synthesis of 2′-Hydroxy-3′,4′,6′-trimethoxyacylophenones

A wide variety of 2′-hydroxypolymethoxyacetophenones and propiophenones, e.g. 1a and 1b are used in the synthesis of flavone and ehromone natural products.¹ Repeated attempts to prepare 1a and 1b by reacting 1,2,3,5-tetramethoxybenzene 2 with acetyl or propionyl chlorides and AlCl₃ in ether² gave products which were difficult to purify.³ We traced the problem to ring-ethoxy contaminants which were isolated and tentatively identified as 3a and 3b.     

Synthesis of 3′,4′-Dimethoxyisoflavone Derivaties

2-(Un)substituted-3,4-dimethoxy-7-hydroxyisoflavones were synthesized. Their derivatives substituted at the phenolic hydroxyl were prepared by alkylation and acylation. Aminomethyl derivatives were also prepared.     

Crystal structure of 1,1′-biphenyl-2,2′,3,3′-tetracarboxylic and 1,1′-biphenyl-2,2′,3,3′,5,5′,6,6′-octacarboxylic acids: solid-state chiralization and dissociation

X-ray diffraction analysis revealed unexpected stereochemical features accompanying crystal self-assembly of the two title oligocarboxylic acids.     

Synthesis and spatial structure of methyl substituted 2-phenylpiperidine-4-spiro-5′-imidazolidine-2′,4′-diones

Piperidine-4-spiro-5-irnidazolidine-2, 4-diones have been synthesized from the appropriate methyl substituted 2 phenyl-4-hydroxypiperidines and 4-cyano-4-aminopiperidines by the Bucherer—Bergs and Strecker methods. Synthesis,characterization, and investigations of antimicrobial activity of 6,6-dimethyl-3-aryl-3′,4′,6,7-tetrahydro-1′H,3H-spiro[benzofuran-2,2′-naphthalene]-1′,4(5H)-dione

Chalcone-like compounds 3a–l, 2-(benzylidene)-3,4-dihydronaphthalen-1(2H)-one, were synthesized from the addition of different benzaldehyde derivatives (2a–l) to 1,2,3,4-tetrahydro-1-napthalone (1) in basic medium. Mn(OAc)₃-mediated addition of dimedone (4) to chalcone-like compounds gave the spirobenzofuran derivatives (5a-l), 6,6-dimethyl-3-aryl-3′,4′,6,7-tetrahydro-1′H,3H-spiro[benzofuran-2,2′-naphthalene]-1′,4 (5H)-dione, in good yields. The structures of synthesized compounds 5a–l were elucidated on basis of spectral data (NMR, IR) and elemental analysis. In addition, their antibacterial activities were screened against some human pathogenic microorganisms.     

Synthesis of 1′-substituted 4′,4′-dimethyl-6′-methoxy-4′H-spiro[cyclohexane-1,3′-isoquinolines]

The reaction of 1-(4-methoxyphenyl)-1-(1-methylcyclohexyl)ethanol with nitriles in concentrated sulfuric acid afforded 1′-substituted 6′-methoxy-4′,4′-dimethyl-4′H-spiro[cyclohexane-1,3′-isoquinolines] as a result of consecutive Wagner-Meerwein rearrangement and Ritter reaction.     

Preparation,structures, and properties of nickel complexes containing 2,6-bis[4′,4′-dimethyloxazolin-2′-yl]pyridine,a pincer ligand

Five- and six-coordinate nickel complexes [(Dm-Pybox)NiCl₂], [(Dm-Pybox)Ni(H₂O)Cl₂], [(Dm-Pybox)Ni(H₂O)₂Cl]Cl (2a), and [(Dm-Pybox)Ni(MeOH)₂Cl]Cl (2b), where Dm-Pybox is 2,6-bis[4′,4′-dimethyloxazolin-2′-yl]pyridine, have been isolated and structurally characterized by single crystal X-ray diffraction. The solid state structures of 2a and 2b feature different modes of non-covalent interactions, C–H?Cl, C–H?O and O–H?Cl interactions. Spectroscopic and analytical methods, UV–vis and thermogravimetric analyses were done to further investigate chemical properties of the complexes.     

Synthesis and antiviral evaluation of 2′,2′,3′,3′-tetrafluoro nucleoside analogs

Herein, we report the synthesis of novel 2′,2′,3′,3′-tetrafluorinated nucleoside analogs along with their phosphoramidate prodrugs. A tetrafluoro ribose moiety was coupled with different Boc/benzoyl-protected nucleobases under Mitsunobu conditions. After deprotection, tetrafluorinated nucleosides 13b, 14b, 20b-22b were reacted with phenyl-(isopropoxy-l-alaninyl)-phosphorochloridate to afford corresponding monophosphate prodrugs 24b–28b. All synthesized compounds were evaluated against several DNA and RNA viruses including HIV, HBV, HCV, Ebola and Zika viruses.     

Synthesis and Crystal Structure of 3′-Ethyl 5′-Methyl 1-Benzyl-4′-methyl-2-oxo-3′,4′-dihydrospiro-[indoline-3,2′-pyrrole]-3′,5′-dicarboxylate

The crystal structure of the title compound(C24H24N2O5, Mr = 420.45) has been determined by single-crystal X-ray diffraction. The crystal is of triclinic, space group P1 with a = 8.991(1), b = 11.166(1), c = 11.169(1) , α = 91.413(2), β = 105.887(2), γ = 90.992(2)°, V = 1077.8(2) 3, Z = 2, Dc = 1.296 g/cm3, F(000) = 444, μ(MoKα) = 0.091 mm-1, the final R = 0.0466 and wR = 0.1507 for 4185 observed reflections(I 2σ(I)). The single-crystal X-ray diffraction data indicated intermolecular C(17)–H(17)O(4), C(5)–H(5)… O(4) hydrogen bonds and C–H…π interaction in the structure.

Binuclear complexes of chromium,manganese and rhodium withN-hydroxyethyl-N,N′,N′-tribenzimidazolylmethylethylenediamine

Summary Three novel binuclear complexes, Cr₂Cl₆L·EtOH, Mn₂OAc₄L·EtOH, Rh₂Cl₆L.2H₂O, where L =N-hydroxyethyl-N,N,N-tribenzimidazolylmethylethylenediamine, have been prepared and characterized by elemental analyses, i.r., u.v.-vis. and e.s.r. spectra, and by magnetic susceptibility measurements. A weak antiferromagnetic spin exchange interaction exists between the metal ions in a binuclear unit for the dimanganese complex. Preliminary bioassays show that the manganese complex possesses antitumour activityin vitro.
