The Rh2(OAc)4-catalyzed reactions of o-(methoxycarbonyl)-alpha-diazoacetophenone with enantiomerically pure 5-ethoxy-3-p-tolylsulfinylfuran-2(5H)-ones 1a and 1b afford 4,10-epoxybenzo[4,5]cyclohepta[1,2-c]furan-3,9-diones 6a and 6b, in good or moderate yields and in a completely regioselective way. The pi-facial selectivity is complete for 1a, which only yields anti-6a adducts, and very high for 1b. The endo stereoisomers are favored with respect to the exo ones in both reactions. The sulfinyl group significantly increases the reactivity of the dipolarophile as it has been demonstrated by studying the behavior of 5-methoxyfuran-2(5H)-one (3).  相似文献

A new approach to enantiomerically pure 2,8-dialkyl-1,7-dioxaspiro[5.5]undecanes and 2,7-dialkyl-1,6-dioxaspiro[4.5]decanes is described and utilizes enantiomerically pure homopropargylic alcohols obtained from lithium acetylide opening of enantiomerically pure epoxides, which are, in turn, acquired by hydrolytic kinetic resolution of the corresponding racemic epoxides. Alkyne carboxylation and conversion to the Weinreb amide may be followed by triple-bond manipulation prior to reaction with a second alkynyllithium derived from a homo- or propargylic alcohol. In this way, the two ring components of the spiroacetal are individually constructed, with deprotection and cyclization affording the spiroacetal. The procedure is illustrated by acquisition of (2S,5R,7S) and (2R,5R,7S)-2-n-butyl-7-methyl-1,6-dioxaspiro[4.5]-decanes (1), (2S,6R,8S)-2-methyl-8-n-pentyl-1,7-dioxaspiro[5.5]undecane (2), and (2S,6R,8S)-2-methyl-8-n-propyl-1,7-dioxaspiro[5.5]undecane (3). The widely distributed insect component, (2S,6R,8S)-2,8-dimethyl-1,7-dioxaspiro[5.5]undecane (4), was acquired by linking two identical alkyne precursors via ethyl formate. In addition, [(2)H(4)]-regioisomers, 10,10,11,11-[(2)H(4)] and 4,4,5,5-[(2)H(4)] of 3 and 4,4,5,5-[(2)H(4)]-4, were acquired by triple-bond deuteration, using deuterium gas and Wilkinson's catalyst. This alkyne-based approach is, in principle, applicable to more complex spiroacetal systems not only by use of more elaborate alkynes but also by triple-bond functionalization during the general sequence.  相似文献

In this paper, stereocontrolled tandem Michael addition-elimination reaction of the novel chiral source, 5-(l-menthyloxy)-3,4-dichloro-2(5H)-furanone, with various thiols and amines has been investigated. A series of new enantiomerically pure compounds, 5–(l-menthyloxy)-4-substituted-3-chloro-2(5H)-furanones, were obtained in good yields with d. e. ? 98% under mild conditions.  相似文献

Recent work in the preparation of organically templated metal sulfates under hydrothermal conditions has been extended to include the sulfation of alpha-molybdena through the synthesis of [C(5)H(14)N(2)][(MoO(3))(3)(SO(4))].H(2)O. Single crystals were grown under hydrothermal conditions from molybdenum oxide, water, sulfuric acid, and an enantiomerically pure (R)-2-methylpiperazine source and characterized using both single-crystal X-ray diffraction and infrared spectroscopy. One-dimensional [(MoO(3))(3)(SO(4))](n)(2n-) chains, based on a neutral alpha-molybdena backbone, are connected through an extensive hydrogen-bonding network containing [C(5)H(14)N(2)](2+) cations and occluded water molecules. The direction of the hydrogen bonding is primarily dictated by the nucleophilicity of the respective oxide ligands, as determined using bond valence sums.  相似文献

A new series of 1beta-methylcarbapenems 1a-d bearing 5-methyl-4-mercaptopyrrolidinone rings has been prepared and evaluated for in vitro antibacterial activity and pharmacokinetic parameters. Most compounds showed excellent antibacterial activity and high stability to dehydropeptidase-1. We have synthesized optically active 5-methyl-4-hydroxypyrrolidinones from enantiomerically pure aziridine esters.  相似文献

Starting from the commercially available enantiopure (1S,2S)-2-amino-1-phenyl-1,3-propanediol novel enantiomerically pure benzimidazoles were prepared; N-alkylation gave chiral benzimidazolium salts, which were tested in asymmetric benzoin condensations. The synthesis of conceptually new, enantiomerically pure, C2 symmetric bis-thiazolium and bis-benzimidazolium salts was also developed. These new chiral heterocycles were employed as catalysts in the asymmetric dimerisation of benzaldehyde to give benzoin with moderate enantioselectivity.  相似文献

The utility of phosphonothioate methanolysis to degrade organophosphate neurotoxins has prompted the stereochemical investigation of this useful transformation. The methanolysis of enantiomerically pure O,S-diethyl phenylphosphonothioate (5) was studied both in the presence and in the absence of metal ions known to catalyze the phosphonothioate → phosphonate transformation. This report outlines the syntheses of enantiomerically pure 5 and its methanolysis product O-ethyl O-methyl phenylphosphonate (7). Compound 7 results from exclusive P-S scission of 5, which is the desired mode of phosphonothioate methanolysis (E(a) = 14.5 ± 0.5 kcal/mol). The stereochemical analysis of the phosphonothioate methanolysis was done for the first time with β-cyclodextrin, and it shows complete inversion on the phosphorus center upon methoxide displacement of ethanethiolate. The presence of La(III) or Zn(II) complexes do not alter this S(N)2-like substitution which sheds new light on the mechanism of methanolysis of phosphonothioates.  相似文献

Several base variations of 2'- and 3'-deoxy derivatives of (+)-4'-deoxy-5'-noraristeromycin have been prepared from enantiomerically pure precursors following standard purine nucleoside construction. A new and general synthesis of alkyl cyanoformates is presented starting from the appropriate alcohol and oxalyl chloride. This is used to prepare enantiomerically pure cyanoformates from enantiomerically pure primary and secondary alcohols. Optimal conditions for the addition of various achiral cyanoformates to aldehydes catalysed by an enantiomerically pure titanium(salen) catalyst in the presence of potassium cyanide as a cocatalyst are developed. Under these conditions, two chiral cyanoformates also reacted with aldehydes to give cyanohydrin carbonates. The stereochemistry of this process is predominantly determined by the stereochemistry of the titanium(salen) catalyst and the stereochemistry of two of the cyanohydrin carbonates was confirmed by X-ray crystallography. In a further extension of the chemistry, a homogeneous system in which the potassium cyanide/18-crown-6 complex is used as the cyanide cocatalyst has been developed and the kinetics of this reaction show that it displays first order kinetics, provided at least 2 mol % of the potassium cyanide complex are employed.  相似文献

A new method for the synthesis of ramosetron hydrochloride, the most wildly used 5-HT3 receptor antagonist, was reported. The intermediate 8 was obtained in a high e.e. value via lipase catalyzed hydrolytic kinetic resolution. Then enantiomerically pure ramosetron hydrochloride was synthesized in two steps with excellent yield (the total yield is 24.6%). The method provides a new environment friendly and atom economy way to synthesize enantiomerically pure ramosetron.  相似文献

(S)-(+)-N-acetylphenylglycineboronic acid (1) is synthesized in enantiomerically pure form; its reaction with chiral diols quantitatively yields cyclic boronic esters 5a-g. The asymmetric synthesis of (R)-2-isopropyl-5-methylhex-5-enal in 98% ee is described. It was discovered that the key alkylation step employing an Evans chiral auxiliary and 3-methylbut-3-en-1-yl trifluoromethanesulphonate as the alkylating agent led to significant competing O-alkylation, a phenomenon not previously reported. Type II carbonyl ene cyclisation of the aldehyde with a range of Lewis acids led to either the (R,R)- or (R,S)-5-methylidenecyclohexanols without concurrent racemisation of the alpha stereogenic centre of the aldehyde. Conditions for effecting the easy racemisation of a model enantiomerically pure aldehyde, (S)-2-methylbutanal, were developed. In an effort to secure a dynamic kinetic resolution procedure, these conditions were applied to (R)-2-isopropyl-5-methylhex-5-enal. However, a competing and dominant Prins cyclisation occurred instead leading to a mixture of all possible cycloadducts, all of which were obtained in 98% ee. Any unreacted aldehyde was found to be enantiomerically pure.  相似文献

The angular triquinane (?)-silphiperfol-6-en-5-on ((?)- 14 ) has been synthesized for the first time time in enantiomerically pure form. A highly efficient triplet-sensitized photoreaction, the oxadi-π-methane rearrangement of the complex β,γ-enone(+)- 6 to (?)- 7 , constitutes the key step. The know staring material, (1R, 7aR)-3,6,7,7a-tetrahydro-1-hydroxy-7a-methyl-1H-inden-5(2H)-one ((?)- 2 ), is readily accessible in > 97% enantiomeric purity. This new approach should enable access also to other angularly fused triquinanes of the silphinene type and related structures.  相似文献
