The treatment of a β3‐amino acid methyl ester with 2.2 equiv. of lithium diisopropylamide (LDA), followed by reaction with 5 equiv. of N‐fluorobenzenesulfonimide (NFSI) at ?78° for 2.5 h and then 2 h at 0°, gives syn‐fluorination with high diastereoisomeric excess (de). The de and yield in these reactions are somewhat influenced by both the size of the amino acid side chain and the nature of the amine protecting group. In particular, fluorination of N‐Boc‐protected β3‐homophenylalanine, β3‐homoleucine, β3‐homovaline, and β3‐homoalanine methyl esters, 5 and 9 – 11 , respectively, all proceeded with high de (>86% of the syn‐isomer). However, fluorination of N‐Boc‐protected β3‐homophenylglycine methyl ester ( 16 ) occurred with a significantly reduced de. The use of a Cbz or Bz amine‐protecting group (see 3 and 15 ) did not improve the de of fluorination. However, an N‐Ac protecting group (see 17 ) gave a reduced de of 26%. Thus, a large N‐protecting group should be employed in order to maximize selectivity for the syn‐isomer in these fluorination reactions. 相似文献

Convenient methods for the synthesis of functionalized organophosphorus compounds containing carbonyl groups as well as di‐ or trialkoxymethyl fragments attached to phosphorus, and their derivatives, starting from the available derivatives of trivalent phosphorus acids, are proposed, and some properties of the new functionalized organophosphorus compounds are presented. So, the alkylation and acylation reaction of (dialkoxymethyl) phosphonites and their analogs have been studied. It is found that the Arbuzov rearrangement of these compounds was accompanied by the phosphorus–carbon bond cleavage with unique retention of the three‐coordinate phosphorus. © 2012 Wiley Periodicals, Inc. A concise synthesis of α‐benzylidene‐γ‐methyl‐γ‐butyrolactones 5a – g from substituted benzaldehydes is described. Compounds 1a – g on reaction with phosphorane 2 , provide the pentenoates 3a – g , which can be hydrolyzed to the acids 4a – g . The latter are cyclized to the corresponding butyrolactones 5a – g in excellent yields. The pentenoates 3a – g , on acid catalyzed cyclization, also provide 5a – g in very high yields. 相似文献

The reactivity of the enamino compounds 4-amino-3-phenylamino(thio)carbonyl-3-penten-2-one 1 and 2 and ethyl 3-amino-2-phenylamino(thio)carbonyl-2-butyrate 7 and 8 was studied using the reaction with hydrazine hydrate and hydrazine hydrochloride to evaluate the 1,3 electrophilic center of the compounds by the formation of the pyrazole rings. The reactions of α,β‐unsaturated nitriles ( 1, 9, 12 ) as bielectrophiles with aminoazoles ( 2, 4, 6 ) as binu‐cleophiles were investigated. Acrylonitrile ( 1 ) reacts almost exclusively in a chemoselective Michael‐type addition yielding the substituted azoles 3, 5 and 7 , respectively. Cinnamonitriles 9a,b behave in a similar way, but the free CN group adds a second molecule 4 yielding 10a,b and its cyclocondensation product 11a,b as minor component. The attempted formation of azolopyrimidines is best achieved by the reaction of the benzylidenemalononitriles 12a ‐ f with 2 or 4 . The process is chemo‐ and regioselective. The structure determinations were based on NMR measurements including DEFT, COSY, ROESY, HMQC and HMBC techniques and correct earlier suggestions. 相似文献

Five‐coordinate bicycloazastannoxides containing a β‐hydroxy‐α‐amino acid moiety were synthesized. By hydrolysis under the action of 10% HCl, five β‐hydroxy‐α‐amino acids were prepared, and their structures were characterized by 1H NMR spectroscopy, MS, and elemental analyses. The mechanism of the hydrolysis of the organotin (IV) complexes is discussed, and the yields of diastereomers are investigated by the HPLC method.
