HPLC with UV and acidified potassium permanganate chemiluminescence detection, combined with multivariate data analysis techniques, were used for the geographical classification of some Australian red (Cabernet Sauvignon) and white (Chardonnay) wines from two regions (Coonawarra and Geelong). Identification of the wine constituents prominent in the chromatography was performed by mass spectrometry. Principal components analysis and linear discriminant analysis were used to classify the wines according to region of production. Separation between regions was achieved with both detection systems and key components leading to discrimination of the wines were identified. Using two principal components, linear discriminant analysis with UV detection correctly classified 100% of the Chardonnay wines and, overall 91% of the Cabernet Sauvignon wines. With acidified potassium permanganate chemiluminescence detection, 75% of the Chardonnay wines and 94% of the Cabernet Sauvignon wines were correctly classified using two factors.  相似文献

The potential of a vanguard technique as is the ion mobility spectrometry with ultraviolet ionization (UV-IMS) coupled to a continuous flow system (CFS) have been demonstrated in this work using a gas phase separator (GPS). This vanguard system (CFS-GPS-UV-IMS) has been used for the analysis of different types of white wines to obtain a characteristic profile for each type of wine and their posterior classification using different chemometric tools. Precision of the method was 3.1% expressed as relative standard deviation. A deep chemometric study was carried out for the classification of the four types of wines selected. The best classification performance was obtained by first reducing the data dimensionality by principal component analysis (PCA) followed by linear discriminant analysis (LDA) and finally using a k-nearest neighbour (kNN) classifier. The classification rate in an independent validation set was 92.0% classification rate value with confidence interval [89.0%, 95.0%] at 95% confidence level.The same white wines analyzed using CFS-GPS-UV-IMS were analyzed using gas chromatography with a flame detector (GC-FID) as conventional technique. The chromatographic method used for the determination of superior alcohols in wine samples shown in the Regulation CEE 1238/1992 was selected to carry out the analysis of the same samples set and later the classification using appropriate chemometrics tools. In this case, strategies PCA-LDA and kNN classifier were also used for the correct classification of the wine samples. This combination showed similar results to the ones obtained with the proposed method.  相似文献

In this study, the potential of high performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry (HPLC–QTOFMS) for metabolomic profiling of red wine samples was examined. Fifty one wines representing three varieties (Cabernet Sauvignon, Merlot, and Pinot Noir) of various geographical origins were sourced from the European and US retail market. To find compounds detected in analyzed samples, an automated compound (feature) extraction algorithm was employed for processing background subtracted single MS data. Stepwise reduction of the data dimensionality was followed by principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA) which were employed to explore the structure of the data and construct classification models. The validated PLS-DA model based on data recorded in positive ionization mode enabled correct classification of 96% of samples. Determination of molecular formula and tentative identification of marker compound was carried out using accurate mass measurement of full single MS spectra. Additional information was obtained by correlating the fragments obtained by MS/MS accurate mass spectra using the QTOF with collision induced dissociation (CID) of precursor ions.  相似文献

This study outlines the use of mid-infrared (MIR) spectroscopy combined with principal component analysis (PCA) and linear discriminant analysis (LDA) for the varietal classification of commercial red and white table wines. Three red varieties (Cabernet Sauvignon, Shiraz and Merlot) and four white varieties (Chardonnay, Riesling, Sauvignon Blanc and Viognier) were sourced from different wine regions in Australia. Wine samples were scanned in transmission on a FOSS WineScan FT 120 from wave numbers 926 to 5012 cm−1. All samples were sourced from the 2006 vintage and had not been blended with any other variety or wine from other regions. Spectral data were reduced to a small number of principal components (PCs) and LDA was then performed to successfully separate the wines into the different varieties. To test the robustness of the LDA models developed for the red wines, a set of red wines scanned in 2005 were used. Correct classification of over 95% was achieved for the validation set.  相似文献

Abstract

Studying wine mineral profile has been proven as a valuable tool in geographical origin discrimination and authenticity for both producers and consumers. Adulteration of wines, in terms of geographical origin or variety, is considered a major topic of extensive research. Traceability and authenticity of wines have been previously studied on the basis of typical mineral element patterns by means of chemometric methods. In this context, analytical methods were developed for the determination of mineral elements in wines by inductively coupled plasma–mass spectrometry. This study aimed at classifying selected varietal Greek wines from various regions by employing instrumental analysis. Preliminary data of wine mineral content enabled for the classification of samples according to geographical origin and variety. However, further work is required in order to draw more valid conclusions and to obtain a detailed map of the mineral element content of Greek wines according to their geographical origin and/or variety.  相似文献

An electronic tongue (ET) based on potentiometric chemical sensors was assessed as a rapid tool for the quantification of bitterness in red wines. A set of 39 single cultivar Pinotage wines comprising 13 samples with medium to high bitterness was obtained from the producers in West Cape, South Africa. Samples were analysed with respect to a set of routine wine parameters and major phenolic compounds using Fourier transform infrared-multiple internal reflection spectroscopy (WineScan) and high-performance liquid chromatography, respectively. A trained sensory panel assessed the bitterness intensity of 15 wines, 13 of which had a bitter taste of medium to high intensity. Thirty-one wine samples including seven bitter-tasting ones were measured by the ET. Influence of the chemical composition of wine on the occurrence of the bitter taste was evaluated using one-way analysis of variance. It was found that bitter-tasting wines had higher concentrations of phenolic compounds (catechin, epicatechin, gallic and caffeic acids and quercetin) than non-bitter wines. Sensitivity of the sensors of the array to the phenolic compounds related to the bitterness was studied at different pH levels. Sensors displayed sensitivity to all studied compounds at pH 7, but only to quercetin at pH 3.5. Based on these findings, the pH of wine was adjusted to 7 prior to measurements. Calibration models for classification of wine samples according to the presence of the bitter taste and quantification of the bitterness intensity were calculated by partial least squares-discriminant analysis (PLS-DA) regression. Statistical significance of the classification results was confirmed by the permutation test. Both ET and chemical analysis data could discriminate between bitter and control wines with the correct classification rates of 94% and 91%, respectively. Prediction of the bitterness intensity with good accuracy (root mean square error of 2 and mean relative error of 6% in validation) was possible only using ET data.  相似文献

The diagnosis of Duchenne muscular dystrophy (DMD) has historically utilized either PCR or requires Southern blot analysis, a southern blot analysis, however, is not amenable to incorporation in a microdevice format. A PCR amplification-based method has been developed, and we have previously coupled this amplification with microchip separation of the PCR fragments for DMD diagnosis. Diagnoses of affected patients were performed by comparing exon concentrations to those of control samples amplified at the same time. To accurately identify mutations in patient samples, this work established normal ranges for the concentration of each amplified exon fragment using control samples amplified over successive days. Our studies show that the number of cycles used in the amplification process affects this range. Affected patient samples were analyzed using these normal ranges and the mutations detected by Southern blot analysis were also diagnosed using the microchip separation method.

Employing the microchip separation method decreases the time required for the analysis, but the time required for DNA purification and PCR amplification must also be decreased for faster total analysis of patient samples. Development of microchip methods for these processing steps is one approach for reducing the individual times, while also providing the possibility of integrating these steps in a single device. Here we report on the microchip extraction of genomic DNA from whole blood using a novel sol–gel matrix that is easily formed in microdevices. IR-mediated PCR amplification of a β-globin fragment from genomic DNA followed by electrophoretic analysis on a single integrated microdevice is presented for the first time. Work towards the development of a micro-total analysis device for DMD diagnosis, through integration of all processing steps on a single device, is also discussed.  相似文献

Abstract

An HPLC method is described which enables the qualitative and quantitative analysis of residues of pharmacologically active substances in food stuffs of animal origin. Egg samples were spiked with the therapeutic drugs sulfapyridine and its N-acetyl metabolite, with ethopabat, chloramphenicol, meticlorpindol, metronidazol, ipronidazol, furazolidone and nicarbazin and with pyrazon and benzothiazuron as internal standards in the range of 0.02 to 0.1 mg/kg. The drugs were extracted with acetonitrile and purified by liquid-liquid partition steps. They were separated on a reversed phase column and detected with a multi-signal UV/Vis diode-array detector at 3 different wavelengths: 275, 315 and 360 nm. The peak's identity was confirmed by comparing retention times and UV spectra with those of standards. Peaks were checked for homogenity by comparing spectra at the peak's upslope, apex and downslope. The detection limit was in the range of 0.001 to 0.05 mg/kg.  相似文献

We examined 144 samples of red dry wines of varieties Cabernet and Merlot, produced in the territory of the main wine-making enterprises of the geographical zones of Krasnodar krai, to consider the possibility of determining the regional and varietal identity of red wines. The concentration of micro- and macroelements in the wines was determined by inductively coupled plasma atomic emission spectrometry. Chemometric studies were performed in the Statistica package environment, using discriminant analysis and classification trees. Adequate mathematical models were proposed for wines to identify the grape variety and the region where it grows. A software product was developed that automates the necessary calculations.  相似文献

The analysis of mitochondria by capillary electrophoresis usually takes longer than 20 min per replicate which may compromise the quality of the mitochondria due to degradation. In addition, low sample consumption may be beneficial in the analysis of rare or difficult samples. In this report, we demonstrate the ability to analyze individual mitochondrial events in picoliter-volume samples (approximately 80 pL) taken from a bovine liver preparation using microchip capillary electrophoresis with laser-induced fluorescence detection (micro-chip CE-LIF). Using a commercial "double-T" glass microchip, the sample was electrokinetically loaded in the "double-T" intersection and then subjected to electrophoretic separation along the main separation channel. In order to decrease interactions of mitochondria with channel walls during the analysis, poly(vinyl alcohol) was used as a dynamic coating. This procedure eliminates the need for complicated covalent surface modifications within the channels that were previously used in capillary electrophoresis methods. For analysis, mitochondria, isolated from bovine liver tissue, were selectively labelled using 10-nonyl acridine orange (NAO). The results consist of electropherograms where each mitochondrial event is a narrow spike (240 +/- 44 ms). While the spike intensity is representative of its NAO content, its migration time is used to calculate and describe its electrophoretic mobility, which is a property still largely unexplored for intracellular organelles. The five-fold decrease in separation time (4 min for microchip versus 20 min for capillary electrophoresis) makes microchip electrophoretic separations of organelles a faster, sensitive, low-sample volume alternative for the characterization of individual organelle properties and for investigations of subcellular heterogeneity.  相似文献

Wine composition depends on many factors which are especially important for quality wines from specific regions, such as protected designation of origin (PDO) wines. Nineteen analytical parameters were analysed in commercial rosé wines from different Spanish PDOs (Ribera del Duero (RD), Rioja (Rj), Valdepeñas and La Mancha (M-V)), and from two consecutive vintages. Stepwise linear discriminant analysis (SLDA) was used to differentiate and classify these wines by their geographical origin. All the wines were made from “similar” varieties of grapes. The final model selected 12 variables, being ethanol and calcium the most important variables for the differentiation of the three PDOs. The evaluation of the model was done by crossvalidation, obtaining a global percentage of correct classification of 98.8% and of global prediction of 97.3%. These results can be considered satisfactory and acceptable, and the selected variables can be useful to differentiate these wines by their origin.  相似文献

Twenty-one almond samples from three different geographical origins (Sicily, Spain and California) were investigated by determining minerals and fatty acids compositions. Data were used to discriminate by chemometry almond origin by linear discriminant analysis. With respect to previous PCA profiling studies, this work provides a simpler analytical protocol for the identification of almonds geographical origin. Classification by using mineral contents data only was correct in 77% of the samples, while, by using fatty acid profiles, the percentages of samples correctly classified reached 82%. The coupling of mineral contents and fatty acid profiles lead to an increased efficiency of the classification with 87% of samples correctly classified.  相似文献

Classical multivariate analysis techniques such as factor analysis and stepwise linear discriminant analysis and artificial neural networks method (ANN) have been applied to the classification of Spanish denomination of origin (DO) rose wines according to their geographical origin. Seventy commercial rose wines from four different Spanish DO (Ribera del Duero, Rioja, Valdepeñas and La Mancha) and two successive vintages were studied. Nineteen different variables were measured in these wines. The stepwise linear discriminant analyses (SLDA) model selected 10 variables obtaining a global percentage of correct classification of 98.8% and of global prediction of 97.3%. The ANN model selected seven variables, five of which were also selected by the SLDA model, and it gave a 100% of correct classification for training and prediction. So, both models can be considered satisfactory and acceptable, being the selected variables useful to classify and differentiate these wines by their origin. Furthermore, the casual index analysis gave information that can be easily explained from an enological point of view.  相似文献

An integrated system of a silicon-based microfabricated polymerase chain reaction (microPCR) chamber and microfabricated electrophoretic glass chips have been developed. The PCR chamber was made of silicon and had aluminum heaters and temperature sensors integrated on the glass anodically bonded cover. Temperature uniformity in the reaction chamber was +/-0.3 degrees C using an improved novel "joint-heating" scheme. Thermal cycling was digitally controlled with a temperature accuracy of +/- 0.2 degrees C. Small operating volumes together with high thermal conductivity of silicon made the device well suited to rapid cycling; 16 s/cycle were demonstrated. For analysis of the PCR products, the chamber output was transferred to the glass microchip by pressure. Analysis time of PCR amplified genomic DNA was obtained in the microchip in less than 180 s. The analysis procedure employed was reproducible, simple and practical by using viscous sieving solutions of hydroxypropylmethylcellulose and dynamically coated microchip channels with poly(vinylpyrrolidone). DNA fragments that differ in size by 18 base pairs (bp) were resolved. Analysis of genomic male and female amplified DNA by microPCR was achieved in microchip, and application of the integrated microPCR-microchip for the identification of bird sex was tested. Genomic DNA samples from several bird species such as pigeon and chicken were analyzed. Hence, the system could be used as well to determine the sex of avian species.  相似文献

The first report of pulsed amperometric detection (PAD) on an electrophoretic microchip is presented. A hybrid poly-(dimethylsiloxane)/glass device was coupled with a platinum working electrode for the electrochemical detection of glucose, maltose, and xylose. Under optimized detection conditions, glucose was found to respond linearly from 20 to 500 microM with a measured detection limit of 20 microM. The coupling of PAD with a microchip provides a straightforward approach to the analysis of a wide range of carbohydrates using microfluidics.  相似文献

This work presents a novel electrophoretic microchip design which is capable of directly coupling with flow-through analyzers for uninterrupted sampling. In this device, a 3 mm wide sampling channel (SC) was etched on quartz substrate to create the sample inlet and outlet and the 75 microm wide electrophoretic channels were also fabricated on the same substrate. Pressure was used to drive the sample flow through the external tube into the SC and the flow was then split into outlet and electrophoretic channels. A gating voltage was applied to the electrophoretic channel to control the sample loading for subsequent separations and inhibit the sample leakage. The minimum gating voltage required to inhibit the sample leakage depended on the solution buffer and increased with the hydrodynamic flow-rate. A fluorescent dye mixture containing Rhodamine B and Cy3 was introduced into the sample stream at either a continuous or discrete mode via an on-line injection valve and then separated and detected on the microchip using laser-induced fluorescence. For both modes, the relative standard deviation of migration time and peak intensity for consecutive injections was determined to be below 0.6 and 8%, respectively. Because the SC was kept floating, the external sampling equipment requires no electric connection. Therefore, such an electrophoresis-based microchip can be directly coupled with any pressure-driven flow analyzers without hardware modifications. To our best knowledge, this is something currently impossible for reported electrophoretic microchip designs.  相似文献

Young and aged wines from two viticole zones in the Andalusian province of Córdoba (southern Spain) were analysed for their content in Ca, Mg, Fe, Cu, Mn and Zn by flame atomic absorption spectrophotometry, and Na, K, Al and Sr by flame atomic emission spectrophotometry. Significant differences in mean content were found for Na, Mn, Mg, Fe and Zn between wines from Montilla–Moriles and Villaviciosa. Linear discriminant analysis using those variables gave 97.9% recognition ability and 95.7% prediction ability. Cluster and principal component analysis show some differences in wines according to geographical origin and to the ageing of wines. Significant differences between young and aged wines were found in the mean content for Mg, K, Sr, Zn and Mn, obtaining 93.62% recognition ability and prediction ability by using linear discriminant analysis and leave-one-out cross-validation test, respectively. Finally, linear discriminant analysis could also be able to classify the samples according to their provenance and to their ageing simultaneously, obtaining 93.6% of the wines correctly classified.  相似文献

Automation of electrophoretic microchips for sequential analysis of different samples is demonstrated. This system used an autosampler, which was on-line connected to the microchip and the whole process including sample loading and injection, analysis and data acquisition as well as washing were all automated. Rhodamin B at different concentrations was first loaded into a hydrodynamic flow stream by an autosampler, delivered to the microchip, and then sequentially injected into the electrophoretic microchannel for analysis and detection. Automation was achieved by running two independent programs, one for sample loading by an autosampler and the other one for electrophoretic injection by voltage switching, on the same computer. Using this sampling chip, each loaded volume (0.2-1 microL) can be injected for dozens of electrophoretic analyses (1-10 nL for each injection). The variances caused by the external connections, which did not affect the electrophoretic analysis but would cause band broadening of the loaded sample in the hydrodynamic flow stream, were theoretically deduced. Results indicate that the dead volume (approximately 300 nL) due to the connection fitting on the chip could lead to dilution of the loaded sample by a factor of one when 0.2 microL of sample was loaded. Such a design allows sequential analysis of a series of samples while the running buffer is continuously pumped into the connection capillary as well as microchannels for washing between two loaded samples to minimize cross contamination without human intervention. Using this sampling chip, the required sample amount and handling time can be greatly reduced compared to the manual method.  相似文献

Analysis of inorganic ions in cerebrospinal fluid (CSF) is used mainly in the diagnostics of central nervous system diseases, such as Alzheimer’s disease or multiple sclerosis. A new analytical method for fast determination of inorganic cations (ammonium, calcium, magnesium, sodium and potassium) and anions (chloride, sulfate, nitrite and nitrate) in CSF on an electrophoretic microchip was developed in this context. Zone electrophoresis (ZE) separations were performed on the microchip with coupled channels (CC) and contact conductivity detection. Two different propionate background electrolytes were used for the sequential determination of cations at pH 3.1 and anions at pH 4.3. ZE was used for the determination of cationic constituents while ZE–ZE approach was employed for the determination of chloride in the first separation channel on the CC microchip and other anionic micro-constituents in the second channel. LOD values were in the range of 0.003–0.012 mg L?1 and 0.019–0.047 mg L?1 for cations and anions, respectively. Repeatability of migration time was up to 1.2 % for both cations and anions. Repeatability of peak area ranged from 0.3 to 5.6 % for cations and from 0.6 to 6.0 % for anions. Recovery of both cations and anions was in the range 90–106 %. CSF samples were only diluted appropriately without other sample pretreatment prior to analysis. Developed sequential method is suitable for fast determination of the studied cations and anions in CSF with total analysis time <15 min.  相似文献

In this paper, we describe a method for size-based electrophoretic separation of sodium dodecyl sulfate (SDS)-protein complexes on a polymethyl methacrylate (PMMA) microchip, using a separation buffer solution containing SDS and linear polyacrylamide as a sieving matrix. We developed optimum conditions under which protein separations can be performed, using polyethylene glycol (PEG)-coated polymer microchips and electrokinetic sample injection. We studied the performance of protein separations on the PEG-coated PMMA microchip. The electrophoretic separation of proteins (21.5-116.0 kDa) was completed with separation lengths of 3 mm, achieved within 8 s on the PEG-coated microchip. This high-speed method may be applied to protein separations over a large range of molecular weight, making the PEG-coated microchip approach applicable to high-speed proteome analysis systems.  相似文献
