Cyclic voltammetry was used to investigate the oxidation of 8‐oxo‐2′‐deoxyguanosine (8‐oxo‐dG) on the glassy carbon (GC), platinum, gold and SnO2 electrodes over a range of the sweep rate, 8‐oxo‐dG concentration and the solution pH. Reaction mechanism that is common to all these electrodes involves the two‐electron two‐proton charge transfer step followed by the irreversible chemical reaction(s). Rate of the charge transfer reaction decreases with the increasing solution pH (GC, Pt, Au), and depends on the nature of the electrode material following the sequence GC>Pt, Au>>SnO2. These effects can be related to the degree of oxidation of the electrode surface (Pt, Au, SnO2), or to the density of the active surface sites (GC). Any of these electrodes can be used for the fabrication of an amperometric detector for 8‐oxo‐dG .  相似文献

A novel and efficient method for the preparation of 1,3‐dihydro‐3‐oxo‐2‐benzofuran‐1‐carboxylates 4 under mild conditions has been developed. Thus, the reaction of [2‐(dimethoxymethyl)phenyl]lithiums, generated easily from 1‐bromo‐2‐(dimethoxymethyl)benzenes 1 , with α‐keto esters gives the corresponding 2‐[2‐(dimethoxymethyl)phenyl]‐2‐hydroxyalkanoates 2 . The TsOH‐catalyzed cyclization of these hydroxy acetals is followed by the oxidation of the resulting cyclic acetals 3 with PCC to give the desired products in satisfactory yields. The reaction of [2‐(dimethoxymethyl)‐4,5‐dimethoxyphenyl]lithium with (MeOC?O)2, followed by treatment with NaBH4 or organolithiums, affords 2‐[2‐(dimethoxymethyl)‐4,5‐dimethoxyphenyl]‐2‐hydroxyalkanoates 6 , which can similarly be transformed into the corresponding 1,3‐dihydro‐3‐oxo‐2‐benzofuran‐1‐carboxylates 7 in reasonable yields.  相似文献

Electrochemical oxidation of 3,4‐dihydroxybenzoic acid ( 1 ) and 4‐tert‐butylcatechol ( 5 ) in the presence of 4‐hydroxy‐6‐methyl‐2‐pyrone ( 2 ) as nucleophile in aqueous solution has been studied using cyclic voltammetry and controlled‐potential coulometry. The results indicate that 1 via Michael reaction under electro‐decarboxylation reaction converts to heterocyclic compound 4 , and the quinone derived from 4‐tert‐butylcatechol ( 5 ) participates in Michael reaction with 2 and through an ECE mechanism converts to the corresponding o‐quinone ( 6a ). The electrochemical synthesis of 4 and 6a has been successfully performed in an undivided cell.  相似文献

A series of oligovalent carbohydrate assemblies (ranging from mono‐ to pentavalent), derived from three structurally different β‐linked or β‐(1→2)‐linked mannosides, has been chemically synthesized, and the respective compounds have been biologically evaluated in order to investigate their immunostimulatory properties. The Crich methodology for β‐mannosylation was successfully utilized to introduce the β‐linkages, and a click chemistry protocol was utilized to generate the oligovalent derivatives. A convenient protecting group strategy involving the simultaneous use of both p‐methoxybenzyl and benzylidene groups was employed, which allowed a simple and cost‐effective global deprotection step. The immunomodulatory properties of the synthesized multivalent mannosides were evaluated by assessing cytokine production in human white blood cell cultures. The Th2‐type cytokines interleukin‐4 and interleukin‐5 (IL‐4 and IL‐5), the Th1 cytokine interferon‐γ (IFN‐γ), the Treg cytokine IL‐10, and the pro‐inflammatory cytokine tumor necrosis factor (TNF) were included in the screening. A single trivalent acetylated mannobiose derivative was identified as a potent inducer of Treg and Th1 immune response, resulting in strong IL‐10 and moderate IFN‐γ productions dose‐dependently, while inducing no Th2 cytokine response. The immunomodulatory properties of this trivalent mannoside were further studied in vitro in allergen (Bet v)‐stimulated human peripheral blood mononuclear cell cultures of birch pollen allergic subjects. Stimulation with birch pollen induced strong IL‐4 and IL‐5 responses, which could be suppressed by the trivalent acetylated mannobiose derivative. The IL‐10 response was also suppressed, whereas the production of IFN‐γ was strongly enhanced. The results suggest that the identified lead compound has suppressive effects on the Th2‐type allergic inflammatory response and shows potential as a possible lead adjuvant for the specific immunotherapy of allergies.  相似文献

Electrochemical oxidation of desipramine (DES) has been studied in the presence of 4,6‐dimethylpyrimidine‐2‐thiol (DMPT) as nucleophile in acidic medium by means of cyclic voltammetry, controlled‐potential electrolysis and spectroscopic data, as diagnostic techniques. Voltammetric studies of electro‐oxidation of desipramine were realized in a range of pH 1.0 to 8.0 in the absence and presence of DMPT. The results indicate the participation of the product of electrochemical oxidation of desipramine in the reaction with DMPT with ECEC electrochemical mechanism. However, the voltammetry and coulometry results imply existence of a catalytic (EC′) electrochemical mechanism in parallel with ECEC electrochemical mechanism. The product has been characterized by IR, 1H NMR, 13C NMR and MS methods.  相似文献

4‐Arylisocoumarins (=4‐aryl‐1H‐2‐benzopyran‐1‐ones) 6 were prepared from 2‐(1‐aryl‐2‐methoxyethenyl)‐1‐bromobenzenes 1 . Successive treatment of these bromo styrenes with BuLi and 1‐formylpiperidine gave a mixture of (E)‐ and (Z)‐2‐(1‐aryl‐2‐methoxyethenyl)benzaldehydes 2 . Hydrolysis of (Z)‐isomers with conc. HBr, followed by pyridinium chlorochromate (PCC) oxidation of the resulting 1H‐2‐benzopyran‐1‐ol derivatives 4 (and 5 ), afforded the desired products.  相似文献

Gold nanoparticle (Au‐NPs)‐Titanium oxide nanotube (TiO2‐NTs) electrodes are prepared by using galvanic deposition of gold nanoparticles on TiO2‐NTs electrodes as support. Scanning electron microscopy and energy‐dispersive X‐ray spectroscopy results indicate that nanotubular TiO2 layers consist of individual tubes of about 60–90 nm diameters and gold nanoparticles are well‐dispersed on the surface of TiO2‐NTs support. The electrooxidation of hydroquinone of Au‐NPs/TiO2‐NTs electrodes is investigated by different electrochemical methods. Au‐NPs/TiO2‐NTs electrode can be used repeatedly and exhibits stable electrocatalytic activity for the hydroquinone oxidation. Also, determination of hydroquinone in skin cream using this electrode was evaluated. Results were found to be satisfactory and no matrix effects are observed during the determination of hydroquinone content of the “skin cream” samples.  相似文献

The kinetics of oxidation of 1‐methoxy‐2‐propanol and 1‐ethoxy‐2‐propanol by ditelluratocuprate(III) (DTC) in alkaline liquids has been studied spectrophotometrically in the temperature range of 293.2–313.2 K. The reaction rate showed first order dependence in DTC and fractional order with respect to 1‐methoxy‐2‐propanol or 1‐ethoxy‐2‐propanol. It was found that the pseudo‐first order rate constant kobs increased with an increase in concentration of OH? and a decrease in concentration of TeO42?. There is a negative salt effect. A plausible mechanism involving a pre‐equilibrium of a adduct formation between the complex and 1‐methoxy‐2‐propanol or 1‐ethoxy‐2‐propanol was proposed. The rate equations derived from mechanism can explain all experimental observations. The activation parameters along with the rate constants of the rate‐determining step were calculated.  相似文献

A convenient one‐pot method for the preparation of (4Z)‐4‐(arylmethylidene)‐5‐ethoxy‐1,3‐oxazolidine‐2‐thiones 2 and 3 from ethyl (2Z)‐3‐aryl‐2‐isothiocyanatoprop‐2‐enoates 1 , which can be easily prepared from ethyl 2‐azidoacetate and aromatic aldehydes, has been developed. Thus, these α‐isothiocyanato α,β‐unsaturated esters were treated with organolithium compounds, including lithium enolates of acetates, to provide 5‐substituted (4Z)‐4‐(arylmethylidene)‐5‐ethoxy‐1,3‐oxazolidine‐2‐thiones, 2 , and 2‐[(4Z)‐(4‐arylmethylidene)‐5‐ethoxy‐2‐thioxo‐1,3‐oxazolidin‐5‐yl]acetates, 3 .  相似文献

Well‐defined macromonomers of poly(ethylene oxide) and poly(tert‐butyl methacrylate) were obtained by anionic polymerization induced directly by the carbanion issued from 2‐methyl‐2‐oxazoline. When ethylene oxide was added to this carbanion with lithium as the counterion, a new compound able to initiate the polymerization of ε‐caprolactone in an anionically coordinated way was synthesized, and this led to well‐defined poly(ε‐caprolactone) macromonomers. © 2005 Wiley Periodicals, Inc. 利用循环伏安法研究了2-硝基苯甲醚在离子液体中的电化学还原行为。以Cu为工作电极,石墨电极为对电极,饱和甘汞电极(SCE)为参比电极,在室温离子液体中电化学还原2-硝基苯甲醚合成2-氨基苯甲醚。优化了电解实验条件,在最佳实验条件(E= -1.0 V, T=50 ℃, c=74.4 mmol·L-1, Q=6 F·mol-1, Y=10:1)下2-氨基苯甲醚的产率最高可达51.3%。在离子液体中电化学合成2-氨基苯甲醚,反应条件温和,避免了有毒易挥发的溶剂、催化剂及其他支持电解质等的使用;并实现了离子液体的重复利用,为2-氨基苯甲醚提供了一条新的绿色化合成路线。  相似文献

An easy‐to‐do paper‐based solid‐phase microextraction (p‐SPME) was developed for determination of 8‐hydroxy‐2’‐deoxyguanosine (8‐OHdG) in urine sample by CE‐LIF. Small piece of filter paper was used as a solid phase to extract 8‐OHdG from urine sample. Its primary mechanism is based on the hydrogen‐bonding interaction between 8‐OHdG and cellulose molecules. The effects of the pH of the sample solution, extraction time, and temperature on the peak area of the analyte were investigated in order to obtain the optimal p‐SPME conditions. Comparing with the untreated sample, the p‐SPME can significantly reduce the interference to the separation of 8‐OHdG by CE‐LIF. Meanwhile, the p‐SPEM can provide more than three times concentrated effect. The developed method was evaluated according to an FDA guideline for biological analysis. The precisions (RSD%, n = 5) of the peak area and migration time of the analyte at three different concentrations were within 3.02–5.82% and 0.92–1.58%, respectively. The limit of identification of the method is about 5 nM according to the significant difference between two sets of the samples with and without spiking the standard (Student's t ‐test, p < 0.05). Good linearity was obtained in the range of 10–1000 nM (R 2>0.99) based on the standard addition. The recoveries at three different concentrations were within 99.8–103.5%. The results of the real sample analysis are consistent with those reported in our previous paper (Electrophoresis 2014, 35, 1873–1879).  相似文献

Voltammetric behavior of two mercaptopyrimidine derivatives (2‐thiouracil and 2‐thiobarbituric acid) has been studied by cyclic voltammetry at a cobalt phthalocyanine (CoPc)‐modified carbon‐paste electrode. The results of voltammetric determinations showed that the CoPc in the matrix of modified electrode acts as catalyst for electrooxidation of these thiols (RSH), lowering the overpotential of the reaction and significantly increasing the sensitivity for detection of thiols in neutral conditions. The results of voltammetric and polarization measurements in solutions with various pHs were used for prediction of the mechanism of electrocatalytic oxidation at the surface of modified electrode. These results showed that at the modified electrode, electrochemical oxidation of thiolate anion (RS?) is the rate‐determining step. It was found that the modified electrode exhibits good selectivity for catalytic oxidation of mercaptopyrimidines over other biologically important mercaptans such as cysteine, glutathione and thioglycolic acid. The results demonstrate that the peak current for thiol oxidation has a linear variation with the concentration in the range of 1×10?2–1×10?5 M. This system can be used for sensitive and selective voltammetric detection of mercaptopyrimidine derivatives.  相似文献

2‐(1‐Aryl‐2‐methoxyethenyl)benzaldehydes 2 , obtained by successive treatment of 1‐(1‐aryl‐2‐methoxyethenyl)‐2‐bromobenzenes 1 with BuLi and 1‐formylpiperidine, were transformed to the corresponding phenylhydrazones 3 on treatment with PhNHNH2. When these hydrazones were allowed to react with conc. HBr, cyclization, followed by dehydrogenation with air occurred, furnished 3‐aryl‐2‐methoxyinden‐1‐one (Z)‐phenylhydrazones 4 .  相似文献

The mechanism of the gas‐phase elimination kinetics of 2‐ethoxypyridine has been studied through the electronic structure calculations using density functional methods: B3LYP/6‐31G(d,p), B3LYP/6‐31++G(d,p), B3PW91/6‐31G(d,p), B3PW91/6‐31++G(d,p), MPW1PW91/6‐31G(d,p), MPW1PW91/6‐31++G(d,p), PBEPBE/6‐31G(d,p), PBEPBE/6‐31++G(d,p), PBE1PBE1/6‐31G(d,p), and PBE1PBE1/6‐31++G(d,p). The elimination reaction of 2‐ethoxypyridine occurs through a six‐centered transition state geometry involving the pyridine nitrogen, the substituted carbon of the aromatic ring, the ethoxy oxygen, two carbons of the ethoxy group, and a hydrogen atom, which migrates from the ethoxy group to the nitrogen to give 2‐pyridone and ethylene. The reaction mechanism appears to occur with the participation of π‐electrons, similar to alkyl vinyl ether elimination reaction, with simultaneous ethylene formation and hydrogen migration to the pyridine nitrogen producing 2‐pyridone. © 2011 Wiley Periodicals, Inc. 研究了室温下间苯二酚和甲基乙烯基酮分别与β-环糊精( β-CD)形成包结物后的几种不同固相反应,结果表明包结物A(间苯二酚/β-CD)与包结物B(甲基乙烯基酮/β-CD)反应能够很好地得到目的产物,产率及ee值分别为82.8%和78.4%;间苯二酚与包结物B反应仅得到低光学活性产物(ee值为19.5%);包结物A与甲基乙烯基酮反应却没有得到手性目的产物。以熔点、X-粉末衍射、固相核磁碳谱及ROESY多种方法对所形成的包结物进行了表征,包结物中主客体的比例(1:1)通过1H NMR (400 MHz)得以确定,文章对固相环加成反应的机制也进行了初步探讨。  相似文献

The 2‐hydroxy‐3,5,5‐trimethylcyclopent‐2‐en‐1‐one ( 1 ) was synthesized in 42% yield by rearrangement of epoxy ketone 10 on treatment with BF3⋅Et2O under anhydrous conditions. Intermediate 10 was available from the known enone 8 , either via direct epoxidation (60% H2O2, NaOH, MeOH; yield 50%), or via reduction to the corresponding allylic alcohol 14 (LiAlH4, THF), followed by epoxidation ([VO(acac)2], tBuOOH) and reoxidation under Swern conditions, in 37% total yield.  相似文献
