We explored the CE with (bpy) (2+) (3) electrochemiluminescence detection for the kinetic study of drug-enzyme interaction. Effects of four nonsteroidal anti-inflammatory drugs including aspirin, paracetamol, sodium salicylate and phenacetin on prolidase (PLD) activity in erythrocytes were investigated. Aspirin enhanced PLD activity whereas the other three had inhibiting effects. This may reveal their different effects on the collagen biosynthesis and catabolism that influence tumor invasiveness. Kinetic study of paracetamol on PLD showed that the value of Michaelis constant K(m) for PLD was 1.23 mM. The mechanism of PLD inhibition by paracetamol is noncompetitive inhibition, and the inhibitor constant K(i) value obtained in our research was 9.73 x 10(3) microg/L.  相似文献

Electrochemiluminescence (ECL) detection with capillary electrophoresis (CE) separation system was used to the rapid analysis of mefenacet within 7 min. The linear response range of mefenacet was from 1.07 × 10−8 to 5.0 × 10−7 M with a detection limit of 4.0 × 10−9 M. Recent advances and key strategies in capillary electrophoresis and microchip CE with electrochemical detection (ECD) and electrochemiluminescence (ECL) detection are reviewed. This article consists of four main parts: CE-ECD; microchip CE-ECD; CE-ECL; and microchip CE-ECL. It is expected that ECD and ECL will become powerful tools for CE microchip systems and will lead to the creation of truly disposable devices. An easy and universal wall-jet configuration for microchip CE-ECL detection system was constructed and investigated in this work. Two detection modes of pre-column and post-column were applied to the above system. TPA, tramadol and lidocaine were chosen as model analytes to estimate the system in both modes. The important operational parameters such as the concentration of luminescent reagent and the distance between the separation outlet and the working electrode were optimally obtained and compared for the first time.  相似文献

A new method for the determination of selenomethionine (SeMet) was developed using capillary electrophoresis on-line coupled with electrochemiluminescence detection. The effects of several factors such as detection potential, concentration and pH of phosphate buffer, separation voltage and injection voltage were investigated. Under optimal conditions, the linear concentration range for SeMet was 0.001 to 0.8 μg mL?1 with a correlation coefficient of 0.9996. The limit of detection (S/N?=?3) was 0.39 ng mL?1. The recovery of SeMet in selenium-enriched yeast (SEY) was 98.6% on an average. The method presented is simple, quick, sensitive and can be reliably applied to the determination of SeMet in SEY.  相似文献

A new method to estimate the leucine aminopeptidase (LAP, EC 3.4.11.1) activity using capillary electrophoresis coupled with electrochemiluminescence (ECL) is described. The liberated proline produced by LAP catalyzing the hydrolysis reaction of leucin–proline was used as an ECL coreagent to enhance Ru(bpy)32+ ECL signals efficiently. The detection limit for proline was 2.88 × 10?6 m (signal‐to‐noise ratio 3), which was equal to 9.60 × 10?8 units of LAP being used to catalyze leucin–proline for 1 min. The Michaelis constant Km (2.07 × 10?2 mol/L) and the maximum reaction velocity Vmax (1.06 × 10?5 mol/L/min) of LAP for leucin–proline are reported. The reaction conditions including the concentration of metal ions, incubation temperature and pH were optimized. This method was successfully applied to detect LAP activity in plasma and the results were in good agreement with that obtained by the clinical method. 基于稀土掺杂类普鲁士蓝化学修饰电极对Ru(bpy)32 的电催化氧化可增敏电致发光信号,建立了一种毛细管电泳-电致化学发光测定洛贝林的新方法。研究了工作电极电位、缓冲液的酸度及其浓度、分离电压和进样时间等实验参数对洛贝林测定的影响。在优化的实验条件下,其线性范围为1.5×10-7mol/L~1.5×10-4mol/L,检出限(S/N=3)为5.0×10-8mol/L。本法可直接用于注射液和空白人尿中洛贝林的测定,回收率为98.3%~101.2%。  相似文献

The paper presents a rapid method for the determination of dioxopromethazine hydrochloride (DPZ), an antihistamine drug, by the capillary electrophoresis with electrochemiluminescene detection (CE–ECL) using tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) reagent. This CE–ECL detection method has high sensitivity, good selectivity and reproducibility for DPZ analysis. Under the optimized conditions: separation capillary, 38 cm length (25 μm i.d.); sample injection, 10 s at 8 kV; separation voltage, 12.5 kV; running buffer, 20 mmol L−1 sodium phosphate of pH 6.0; detection potential, 1.15 V; 50 mmol L−1 of phosphate buffer (pH 7.14) containing 5 mmol L−1 of Ru(bpy)32+ in ECL detection cell, the detection limit of DPZ was 0.05 μmol L−1 (S/N = 3). The linear range extended from 5 to 100 μmol L−1. The linear curve obtained was Y = 181.62 + 9.28X with a correlation coefficient of 0.9970. The relative standard deviations of the ECL intensity and the migration time for six continuous injections of 5 μmol L−1 DPZ were 3.7% and 0.92%, respectively. The CE–ECL method was applied to analyze DPZ in real samples including tablets, rat serum and human urine, and satisfactory results were obtained without interference from samples matrix. The CE–ECL technique was proved to be a potential method for the detection of DPZ in clinic analysis.  相似文献

A capillary electrophoresis with tris(2,2'-bipyridyl)ruthenium(II)-based electrogenerated chemiluminescence detection method was investigated for the determination of monosaccharides using 2-diethylaminoethanethiol as a derivatizing reagent. Xylose, rhamnose, glucose and glucosamine were selected for the demonstration of the method. Under optimal conditions, the calibration curve of glucose was linear over the range of 1.0 x 10(-4) to 1.0 x 10(-7) mol/L (R = 0.999) and the detection limit was 6.0 x 10(-8) mol/L (S/N = 3). A capillary electrophoresis (CE) coupled with electrochemiluminescence (ECL) detection method for the analysis of ethambutol (EB) and methoxyphenamine (MP) has been investigated. Complete separation of EB and MP was achieved in 8 min using a background electrolyte of 20 mM sodium phosphate at pH 10.0 and a separation voltage of 9 kV. ECL detection was performed with an indium/tin oxide (ITO) working electrode biased at 1.4 V (versus a Pt wire reference) in a 200 mM sodium phosphate buffer (pH 8.0) containing 3.5 mM Ru(bpy)3(2+) (where bpy = 2,2'-bipyridyl). Linear correlation (r > or = 0.993) between ECL intensity and drug concentration was obtained in the range 2-50 ng/ml. The limits of detection (LODs) for EB and MP in water were 1.0 and 0.9 ng/ml, respectively. The relative standard deviation values on peak size (10 ng/ml level) and migration time for the two drugs were in the ranges 5-8 and 0.2-0.7% (n = 7), respectively. Applicability of the CE-ECL method to the analysis of human plasma spiked with EB and MP was examined. The LODs for EB and MP in plasma were 0.4 and 0.3 microg/ml, respectively.  相似文献

The tricyclic antidepressants (TCA) imipramine (Imi) and trimipramine (Tri) were successfully analyzed by capillary electrophoresis (CE) coupling with Tris(2,2-bipyridyl) ruthenium(II)-based (Ru(bpy)32+) end-column electrochemiluminescence (ECL) detection. The addition of β-CD to the running buffer was found to enable baseline separation of the two analytes and the addition of acetonitrile (ACN) as an organic additive to improve the repeatability and sensitivity of the CE method. Under the optimized separation and detection conditions (50 mM PBS (pH = 7.0) and 2 mM Ru(bpy)32+ in the ECL detection cell, 20 mM Tris (pH = 2.0), 0.2 mM β-CD and 20% ACN (v/v) as running buffer), wide linear ranges of 0.1-5 μM and 0.1-5 μM were achieved, with the correlation coefficients of 0.9990 (n = 8) and 0.9980 (n = 8) for Imi and Tri, respectively. Detection limits 5 nM and 1 nM (S/N = 3) were obtained for Imi and Tri, respectively. The method was also successfully applied for the determination of Imi in pharmaceutical dosage form.  相似文献

A Ru(bpy)32+-based electrochemiluminescence (ECL) detection coupled with capillary electrophoresis (CE) has been established for the determination of sinomenine for the first time. Optimum separation was achieved with a fused-silica capillary column (50 cm × 25 μm i.d.) and a background electrolyte of 50 mM sodium phosphate (pH 5.0) at a separation voltage of 15 kV. The content of sinomenine was detected by ECL at the detection voltage of 1.15 V (versus Ag/AgCl) with 5 mM Ru(bpy)32+ in 75 mM phosphate solution (pH 8.0) when a chemically modified platinum electrode by europium(III)-doped prussian blue analogue (Eu-PB) was used as a working electrode. Under the optimized conditions, the ECL intensity was in proportion to sinomenine concentration in the range from 0.01 to 1.0 μg mL−1 with a detection limit of 2.0 ng mL−1 (3σ). The relative standard derivations of migration time and ECL intensity were 0.93 and 1.11%, respectively. The level of sinomenine in Sinomenium acutum Rehd. et Wils was easily determined with recoveries between 98.6 and 102.7%.  相似文献

以铕离子(Ⅲ)掺杂类普鲁士蓝(Eu-PB)化学修饰铂电极为工作电极,基于铜(Ⅱ)-土霉素配合物对三联吡啶钌(Ⅱ)电致化学发光强度的增敏作用,建立了用毛细管电泳电致化学发光法测定土霉素的新方法。实验对毛细管电泳分离条件和电化学发光检测条件进行了优化。在最佳实验条件下,电致化学发光峰面积与铜(Ⅱ)-土霉素配合物的浓度在0.138~46.1μg/mL范围内呈良好的线性关系,检出限为57.0ng/mL(3σ)。本法用于牛奶样中土霉素残留量的测定,加标回收率为95.5%(n=5)。  相似文献

A new and simple capillary electrophoresis with electrochemiluminescence detection was developed for the separation and the quantification of a pair of diastereoisomenc alkaloids(ephedrine and pseudoephedrine).The limits of detection(S/N = 3) were 4.5×10-8 mol/L for ephedrine and 5.2×10-8 mol/L for pseudoephedrine,respectively.The RSDs of migration time and peak area were less than 1.3 and 2.5%(n = 5),respectively.The applicability of the propose method was illustrated in the determination of ephedrine and pseudoephedrine in human urine,ephedrine in nasal drops,and the monitoring of pharmacokinetics for pseudoephedrine.  相似文献

A fast and sensitive approach that can be used to detect norfloxacin in human urine using capillary electrophoresis with end-column electrochemiluminescence (ECL) detection of is described. The separation column was a 75-μm i.d. capillary. The running buffer was 15 mmol L−1 sodium phosphate (pH 8.2). The solution in the detection cell was 50 mmol L−1 sodium phosphate (pH 8.0) and 5 mmol L−1 The ECL intensity varied linearly with norfloxacin concentration from 0.05 to 10 μmol L−1. The detection limit (S/N=3) was 0.0048 μmol L−1, and the relative standard deviations of the ECL intensity and the migration time for eleven consecutive injections of 1.0 μmol L−1 norfloxacin (n=11) were 2.6% and 0.8%, respectively. The method was successfully applied to the determination of norfloxacin spiked in human urine without sample pretreatment. The recoveries were 92.7–97.9%.   相似文献

A new technique for investigating drug-protein binding was developed employing capillary electrophoresis (CE) coupled with tris(2,2'-bipyridyl) ruthenium(II) [Ru(bpy)(3) (2+)] electrochemiluminescence (ECL) (CE-ECL) detection after equilibrium dialysis. Three basic drugs, namely pridinol, procyclidine and its analogue trihexyphenidyl, were successfully separated by capillary zone electrophoresis with end-column Ru(bpy)(3) (2+) ECL detection. The relative drug binding to human serum albumin (HSA) for each single drug as well as for the three drugs binding simultaneously was calculated. It was found that the three antiparkinsonian drugs compete for the same binding site on HSA. This work demonstrated that Ru(bpy)(3) (2+) CE-ECL can be a suitable technique for studying drug-protein binding.  相似文献

A new end-column electrochemiluminescence (ECL) detection technique coupling to capillary electrophoresis (CE) is characterized. A 300 microm diameter Pt working electrode was used to directly couple with a 75 microm inner diameter separation capillary without an electric field decoupler. The hydrodynamic cyclic voltammogram (CV) of Ru(bpy) 3 2+ showed that electrophoretic current did not affect the ECL reaction. The presence of high-voltage (HV) field only resulted in the shift of the ECL detection potential. The distance of capillary to electrode was an important parameter for optimizing detection performance as it determined the characteristics of mass transport toward the electrode and the actual concentration of Ru(bpy) 3 2+ in the detection region. The optimum distance of capillary to electrode was decided by the inner diameter of the capillary, too. For a 75 microm capillary, the working electrode should be placed away from the capillary outlet at a distance within the range of 220-260 microm. The effects of pH value of ECL solution and molecular structure of analytes on peak height and theoretical plate numbers were discussed. Using the 75 microm capillary, under the optimum conditions, the method provided a linear range for tripropylamine (TPA) between 1 x 10(-10) and 1 x 10(-5) mol/L with correlation coefficient of 0.998. The detection limit (signal-to-noise ratio S/N = 3) was 5.0 x 10(-11) mol/L. The relative standard deviation in peak height for eight consecutive injections was 5.6%. By this new technique lidocaine spiked in a urine sample was determined. The method exhibited the linear range for lidocaine from 5.0 x 10(-8) to 1.0 x 10(-5) mol/L with correlation efficient of 0.998. The limit of detection (S/N = 3) was 2.0 x 10(-8) mol/L.  相似文献

Discrete wavelets transform (DWT) was applied to noise on removal capillary electrophoresis-electrochemiluminescence (CE-ECL) electropherograms. Several typical wavelet transforms, including Haar, Daublets, Coiflets, and Symmlets, were evaluated. Four types of determining threshold methods, fixed form threshold, rigorous Stein's unbiased estimate of risk (rigorous SURE), heuristic SURE and minimax, combined with hard and soft thresholding methods were compared. The denoising study on synthetic signals showed that wave Symmlet 4 with a level decomposition of 5 and the thresholding method of heuristic SURE-hard provide the optimum denoising strategy. Using this strategy, the noise on CE-ECL electropherograms could be removed adequately. Compared with the Savitzky-Golay and Fourier transform denoising methods, DWT is an efficient method for noise removal with a better preservation of the shape of peaks.  相似文献

A capillary electrophoresis (CE) method coupled with electrochemiluminescence (ECL) detection for the analysis of glyphosate (GLY) and its major metabolite aminomethylphosphonic acid (AMPA) is presented. Complete separation of GLY and AMPA was achieved in 8 min using a background electrolyte of 20 mM sodium phosphate (pH 9.0) and a separation voltage of 21 kV. ECL detection was performed with an indium tin oxide (ITO) working electrode bias at 1.6 V (vs. a Pt-wire reference) in a 30 0mM sodium phosphate buffer (pH 8.0) containing 3.5mM Ru(bpy)3 2+ (where bpy=2.2'-bipyridyl). Linear correlation (r>or=0.997) between ECL intensity and analyte concentration was obtained in the ranges 0.169-16.9 and 5.55-111 microg ml(-1) for GLY and AMPA, respectively. The limits of detection (LODs) for GLY and AMPA in water were 0.06 microg ml(-1) and 4.04 microg ml(-1), respectively. The developed method was applied to the analysis of GLY in soybeans. The LOD of GLY in soybean was 0.6 microg g(-1). Total analysis time including sample pretreatment was less than 1h.  相似文献
