The crystal structure of title compound, 4 , shows that the 10-aryl group is parallel to the plane bisecting the pyridobenzothiazine ring. This structure is in contrast to that normally found for phenothiazines substituted with electron-withdrawing substituents on the 10-phenyl ring. In those compounds, the 10-aryl group is perpendicular to the plane bisecting the phenothiazine ring. The esr spectrum of the cation radical of 4 shows that the radical is located on the hetero ring system which is opposite to that of the cation radical of 4′-dimethylamino-10-phenylphenothiazine in which the radical is located on the 10-aryl ring.  相似文献

Novel piperidine carboxylic acid derivatives of 10H-pyrazino[2,3-b][1,4]benzothiazine were prepared and evaluated for their inhibitory activity on the upregulation of adhesion molecules such as intercellular adhesion molecule-1 (ICAM-1). Replacement of the methanesulfonyl group on the piperidine ring of previously prepared derivatives with a carboxylic acid-containing moiety resulted in a number of potent adhesion molecule inhibitors. Of these, (anti) [3-(10H-pyrazino[2,3-b][1,4]benzothiazin-8-yl)methyl-3-azabicyclo[3.3.1]non-9-yl]acetic acid 2q (ER-49890), showed the most potent oral inhibitory activities against neutrophil migration in an interleukin-1 (IL-1) induced paw inflammation model using mice, and leukocyte accumulation in a carrageenan pleurisy model in the rat, and therapeutic effect on collagen-induced arthritis in rats.  相似文献

The compound 10-phenylpyrido[3,2-b][1,4]benzothiazine 5-oxide, 1 , has been obtained in nearly quantitative yield oxidation of 10-phenylpyrido[3,2-b][1,4]benzothiazine with oxygen in dioxane solution. The 13C nmr chemical shift assignments of 1 are reported. Its structure has been determined by X-ray single crystal methods. The crystals of 1 are monoclinic, space group P21/n. There are four molecules in a unit-cell of dimensions a = 12.347(3), b = 12.947(3), c = 8.987(1)Å, β = 106.73(1)° and V = 1375.8(5) Å3. The central ring is in a boat conformation and the sulfoxide oxygen atom occupies the axial position. The folding angle between the planes of the pyrido and the benzo planes is 161.55(9)°.  相似文献

During a search for novel, orally-active inhibitors of upregulation of adhesion molecules such as intercellular adhesion molecule-1 (ICAM-1), we found a new series of 10H-pyrazino[2,3-b][1,4]benzothiazine derivatives to be potent ICAM-1 inhibitors. Of these compounds, N-[1-(10H-Pyrazino[2,3-b][1,4]benzothiazin-8-ylmethyl)piperidin-4-yl]-N',N'-dimethylsulfamide 7p showed the potent oral inhibitory activities against neutrophil migration in a murine interleukin-1 (IL-1) induced paw inflammation model. The reaction of 4-methoxy-5-amino-6-mercaptopyrimidine with 2-oxo-1-chlorocyclopentyl(hexyl)glyoxalate esters gave derivatives of the previously unknown tetracyclic systems 1,2-dioxocyclopenta (hexa)[g]oxazolidino[3,2-f]pyrimido[4,5-b][1,4]thiazines, which are transformed by ammonium acetate into derivatives of 1,2-dioxocyclopenta(hexa) [g]imidazolidino[3,2-f]pyrimido[4,5-b]-[1,4] thiazines. Derivatives of the new tricyclic 1-oxazino [5,4-g]pyrimido[4,5-b][1,4]thiazine system were obrtained by reaction of 6-carbethoxy-7-acetylpyrimido [4,5-b] [1,4] thiazines with hydroxylamine.  相似文献

Deeply colored cyanine dyes have been synthesized from the previously unknown 3-methyl-12H-naphtho[1,2-b]thiazolo[3,2-d][1,4]thiazinium salts. A new group of 3-substituted-5H-benzo[5,6][1,4]thiazino[3,2-e][1,2,4]triazines was designed, synthesized and evaluated as inhibitors of 15-lipoxygenase (15-LO), and the results were compared with those of standard ligand 4-methyl-2-(4-methylpiperazin-1-yl)pyrimido[4,5-b][1,4]benzothiazine (4-MMPB). Among the newly designed ligands, compound 9e showed the best IC50 of 15-LO inhibition (IC50 = 38 µM). The docking calculations were performed in MOE software based on the function of force-field scoring, in order to study the interaction of these new compounds and standard ligand with 15-LO. The docking study implied that these ligands have hydrogen bond interaction with the residue of active site of 15-LO.  相似文献

Several [1,2,4]triazolobenzothiadiazocin-11-ones 3 were prepared via ring expansion of [1,2,4]triazolo[3,2-b][2,4]benzothiazepin-10(5H)-ones 1 in presence of sodium azide. The reaction intermediate was isolated and characterized as an aryl isocyanate 2b . A possible pathway for formation of novel triazolobenzothiadiazocin-11-one 3 is described.  相似文献

Reaction of 5-hydroxy-6-aminopyrimidines with 2-bromohydroresorcinol and bromodimedone in DMF in the presence of sodium hydride gave 6,7,8,9-tetrahydro-10H-pyrimido[5,4-b][1,4]benzoxazines. When 4-chloro-5-hydroxy-6-aminopyrimidine was treated with bromodimedone the principal products were 6,7,8,9-tetrahydropyrimido[4,5-b][1,4]benzodioxanes. The nucleophilic substitution of the chlorine atom in position 4 of the tetrahydropyrimidobenzoxazines by amines, sodium alkanoates, and thiourea has been studied and the corresponding amines and alkoxy- and thio derivatives of this tricyclic system obtained. Thiation of [1,2,4]triazino[3,2-b]quinazoline-3,10-dione 1 proceeds selectively to give the 3-thioxo-analog 3 . The latter was converted to the corresponding 3-methylthio derivative 4 which was reacted with aniline and hydrazine to give the corresponding anilino- and hydrazino derivatives 5 and 7 . Compound 7 was converted to the hydrazones 8a,b and into the novel heterocyclic ring systems [1,2,4]triazolo[4′,3′:4,5][1,2,4]triazino-[3,2-b]quinazolin-7-ones 9, 10a,b and tetrazolo[1′,5′:4,5][1,2,4]triazino[3,2-b]quinazolin-7-one 11 .  相似文献

In the present study, the experimentally observed regioselectivity in pyrimido[4,5-b][1,4]benzothiazine synthesis has been modeled using density functional theory method at M06/6-311++G** level. Also the tautomerism of all four tautomers of 6-ethyl-2-thio(1H) pyrimidin-(3H)-4-one as an important intermediate through the reaction path in pyrimido[4,5-b][1,4]benzothiazine synthesis has been investigated extensively in the gas and solution phase. The stability order of tautomers was found as A > D > B > C, in gas and chloroform solvent; however, changing to more polar solvents this order was changed as A > B > D > C using polarized continuum model. In the next step, we focused on another significant feature of this synthesis which has major role in the observed regioselectivity. Two proposed reaction paths with two different transition states that it seems the mode of its intermolecular cyclization has a special role in regioselectivity of the final product have been considered. Comparison of our calculated NMR and IR spectrum with those already reported for some pyrimido[4,5-b][1,4]benzothiazines demonstrates a reliable agreement. Moreover, all obtained results in the gas and solution phase confirm that the synthesis of above-mentioned compound is thermodynamically more favorable than the possible regioisomeric product.  相似文献

We have obtained 5-(2-pyridyl)[1,3]dithiolo[4,5-b][1,4]dithiine-2-thione for the first time by cycloaddition of 2-ethynylpyridine to 4,5-dihydro-1,3-dithioltrithione (isotrithionedithiol). We have studied this thione, 5-(2-pyridyl)- and 5-(4-pyridyl)-5,6-dihydro[1,3]dithiolo[4,5-b][1,4]dithiine-2-thiones by mass spectroscopy and also IR, UV, 1H and 13C NMR spectra. Sixteen pyrrolo[3,2- e][1,4]diazepin-2-ones 1 were synthesized in 4-5 steps and 5-48% overall yields from 4-oxoproline 8 by a route featuring a diastereoselective Pictet-Spengler reaction to close the seven-membered diazepinone ring. Crystallographic analysis of pyrrolo[3,2- e][1,4]diazepin-2-one 1b by X-ray diffraction demonstrated that the alpha-amino acid residue adopted dihedral angle geometry similar to an ideal gamma-turn, illustrating the potential for employing these novel heterocycles as peptide turn mimics.  相似文献

A facile synthesis of the 4H-imidazo[5,1-c][1,4]benzothiazine ring system is described. The synthesis utilized the annulation of an imidazole ring onto a 2H-1,4-benzothiazin-3(4H)-one via the addition of the anion of ethyl isocyanoacetate. [reaction: see text] A novel methodology was developed for the efficient synthesis of 4-chloro-pyrimido[4,5-b][1,4]benzodiazepines. The key is the intramolecular Friedel-Crafts cyclization of 5-amino-4-(N-substituted)anilino-6-chloropyrimidine with either a carboxylic acid or its derivatives to construct the 4-chloro-pyrimido[4,5-b][1,4]benzodiazepine core. Subsequent nucleophilic substitution allows the introduction of one more diversity point in the target molecules. This strategy provides an efficient method to access a library of compounds based on privileged substructures that are of great interest in drug discovery.  相似文献

Treatment of 5-methylthio-2,3-dihydrothieno[3,2-f]-1,4-thiazepine ( 9 ) with acylhydrazines gave 5,6-dihydrothieno[3,2-f]-1,2,4-triazolo[4,3-d][1,4]thiazepines 10, 11 , and that of 9 with ethyl anthranilate gave 5,6-dihydrothieno[3′,2′:6,7][1,4]thiazepino[5,4-b]quinazolin-8-one ( 14 ). Reaction of 9 with hydrazine hydrate or 4-chlorophenylhydrazine afforded 5-hydrazino compounds 12, 15 , which were subsequently cyclized to ethyl 5,6-dihydrothieno[3,2-f]-1,2,4-triazolo[4,3-d][1,4]thiazepine-3-carboxylate ( 13 ), 2-(4-chlorophenyl)-5,6-dihydrothieno[3,2-f]-1,2,4-triazolo[4,3-d][1,4]thiazepin-3(2H)-one ( 16 ) and 2-(4-chlorophenyl)-6,7-dihydro-2H-thieno[3,2-f][1,2,4]triazino[4,3-d][1,4]thiazepine-3,4-dione ( 17 ). New representatives of heterocyclic systems, imidazolidino[3,2-f]-pyrido-[2,3-b]- and imidazolidino[3,2-f]pyrimido[4,5-b]-1,4-thiazines, have been obtained.
