The separation of arsenic based on in situ chelation with ammonium diethyl dithiophosphate (ADDTP) has been carried out using methanol-modified supercritical CO2. Aliquots of extract were added to an electroanalytical cell and arsenic was determined by square wave cathodic stripping voltammetry (SWCSV) at a hanging mercury drop electrode (HMDE). Quantitative extractions of As(DDTP)3 were achieved when the experiments were carried out at a pressure of 2500 psi, a temperature of 90 °C, 2.0 mL of methanol, 20.0 min of static extraction and 5.0 min of dynamic extraction in the presence of 18 mg of ADDTP. Analysis of arsenic was made using 150 mg L−1 of Cu(II) in 1 M HCl solution as supporting electrolyte in the presence of ADDTP as ligand. Preconcentration was carried out by deposition at a potential of −0.50 V and the intermetallic compound CuxAsy was reduced at a potential of −0.77 to −0.82 V, depending on ligand concentration. The results showed that the presence of ligand plays an important role, increasing the method's sensitivity and preventing the oxidation of As(III). The calibration graph of the As(DDTP)3 solution was linear from 0.8 to 12.5 μg L−1 of arsenic (LOD 0.5 μg L−1, R = 0.9992, tacc = 60 s). The method was validated using carrot pulp spiked with arsenic solution. This method was applied to the determination of arsenic in samples of carrots, beets and irrigation water. Arsenic in beets was: skin 4.10 ± 0.18 mg kg−1; pulp 3.83 ± 0.19 mg kg−1 and juice 0.71 ± 0.09 mg L−1; arsenic in carrots was: skin 2.15 ± 0.09 mg kg−1; pulp 0.59 ± 0.11 mg kg−1 and juice 0.71 ± 0.03 mg L−1. Arsenic in water were: Chiu-Chiu 0.08 mg L−1, Inacaliri 1.12 mg L−1, and Salado river 0.17 ± 0.07 mg L−1.  相似文献

Aceto Balsamico Tradizionale of Modena (ABTM) is a typical product (PDO denomination) of the province of Modena produced by cooked grape must which undergoes a long ageing period (at least 12 years) in series of wooden casks (batterie). The study of the transformations of this product during ageing is extremely relevant in order to control the authenticity of ABTM towards succedaneous products and mislabelling of age.

This paper presents the results of the investigation of sugars and fixed organic acids in ABTM samples of different ages, coming from different batterie. The analytes were simultaneously determined by a gas chromatographic method optimised for this peculiar matrix.

The method shows good separation and resolution of the investigated chemical species and allows their determination in the concentration ranges reported in brackets: malic (7.6–15.5 g kg−1), tartaric (4.0–9.7 g kg−1), citric (0.6–1.5 g kg−1) and succinic (0.36–0.62 g kg−1) acid and glucose (153–294 g kg−1), fructose (131–279 g kg−1), xylose (011–0.39 g kg−1), ribose (0.078–0.429 g kg−1), rhamnose (0.061–0.195 g kg−1), galactose (0.136–0.388 g kg−1), mannose (0.41–1.46 g kg−1), arabinose (0.33–1.00 g kg−1) and sucrose (0.46–6.84 g kg−1), with mean associated errors ranging from 5 to 19% depending on the analytes.

Moreover, the recovery values are always satisfactory, being close to one for most of the analytes.

Furthermore, in order to assess the degree of variability of the different analytes content with vinegar ageing and the similarity/dissimilarity among series of casks a three-way data analysis method (Tucker3) is proposed. The chemometric technique applied on the data set shows differences between the samples on the bases of their different ageing period, and between the batterie, which traditionally have an own peculiar production procedure.  相似文献

An isotachophoresis (ITP)–capillary zone electrophoresis (CZE) combination was used for the determination of chlorite in drinking waters. No sample preparation is needed and no interfering by other anions in tap water was observed. The reached limits of detection with conductivity detector were 0.012–0.017 mg l−1. By four-fold sample loading with a 30 μl valve, 0.005 mg l−1 of chlorite was determined with R.S.D.=3.3%. The concentrations of 0.05 and 0.20 mg l−1 were measured with R.S.D. of 2.2 and 2.7%, respectively. The recoveries of chlorite from drinking water were 96–106% in the range of 0.02–0.20 mg l−1. The R.S.D. values of migration times (inter-day) were up to 1.3%. The time for analysis is about 15 min.  相似文献

A simple procedure was developed for the direct determination of As(III) and As(V) in water samples by flow injection hydride generation atomic absorption spectrometry (FI–HG–AAS), without pre-reduction of As(V). The flow injection system was operated in the merging zones configuration, where sample and NaBH4 are simultaneously injected into two carrier streams, HCl and H2O, respectively. Sample and reagent injected volumes were of 250 μl and flow rate of 3.6 ml min−1 for hydrochloric acid and de-ionised water. The NaBH4 concentration was maintained at 0.1% (w/v), it would be possible to perform arsine selective generation from As(III) and on-line arsine generation with 3.0% (w/v) NaBH4 to obtain total arsenic concentration. As(V) was calculated as the difference between total As and As(III). Both procedures were tolerant to potential interference. So, interference such as Fe(III), Cu(II), Ni(II), Sb(III), Sn(II) and Se(IV) could, at an As(III) level of 0.1 mg l−1, be tolerated at a weight excess of 5000, 5000, 500, 100, 10 and 5 times, respectively. With the proposed procedure, detection limits of 0.3 ng ml−1 for As(III) and 0.5 ng ml−1 for As(V) were achieved. The relative standard deviations were of 2.3% for 0.1 mg l−1 As(III) and 2.0% for 0.1 mg l−1 As(V). A sampling rate of about 120 determinations per hour was achieved, requiring 30 ml of NaBH4 and waste generation in order of 450 ml. The method was shown to be satisfactory for determination of traces arsenic in water samples. The assay of a certified drinking water sample was 81.7±1.7 μg l−1 (certified value 80.0±0.5 μg l−1).  相似文献

A novel, rapid and specific ultra performance liquid chromatography electrospray ionization tandem mass spectrometry (UPLC-ESI-MS/MS) method has been developed for simultaneous determination and pharmacokinetic studies of three bufadienolides (bufalin, cinobufagin and resibufogenin) in rat plasma. The analytes, bufalin, cinobufagin, resibufogenin and the internal standard, diphenhydramine were extracted from rat plasma samples by a one-step liquid–liquid extraction and separated on an ACQUITY UPLC™ BEH C18 column with gradient elution using a mobile phase composed of acetonitrile and water (containing 0.1% formic acid) at a flow rate of 0.20 mL min−1. Detection was carried out on a triple-quadrupole tandem mass spectrometer in the multiple reaction monitoring (MRM) mode via an electrospray ionization (ESI) interface. The three bufadienolides could be simultaneously determined within 3.0 min. Linear calibration curves were obtained over the concentration ranges of 1.0–200 ng mL−1 for all the analytes. The intra- and inter-day precisions (relative standard deviation (R.S.D.)) were less than 11.35 and 10.87%, respectively. The developed method was applied for the first time to the pharmacokinetic studies of bufadienolides in rats following a single intravenous administration of 2.10 mg kg−1 bufadienolides.  相似文献

Acrylamide levels over a wide range of different food products were analysed using both liquid chromatography–tandem mass spectrometry (HPLC–MS–MS) and gas chromatography–tandem mass spectrometry (GC–MS–MS). Two different sample preparation methods for HPLC–MS–MS analysis were developed and optimised with respect to a high sample throughput on the one hand, and a robust and reliable analysis of difficult matrices on the other hand. The first method is applicable to various foods like potato chips, French fries, cereals, bread, and roasted coffee, allowing the analysis of up to 60 samples per technician and day. The second preparation method is not as simple and fast but enables analysis of difficult matrices like cacao, soluble coffee, molasses, or malt. In addition, this method produces extracts which are also well suited for GC–MS–MS analysis. GC–MS–MS has proven to be a sensitive and selective method offering two transitions for acrylamide even at low levels up to 1 μg kg−1. For the respective methods the repeatability (n=10), given as coefficient of variation, ranged from 3% (acrylamide content of 550 μg kg−1) to 12% (acrylamide content of 8 μg kg−1) depending on the food matrix. The repeatability (n=3) for different food samples spiked with acrylamide (5–1500 μg kg−1) ranged from 1 to 20% depending on the spiking level and the food matrix. The limit of quantification (referred to a signal-to-noise ratio of 9:1) was 30 μg kg−1 for HPLC–MS–MS and 5 μg kg−1 for GC–MS–MS. It could be demonstrated that measurement uncertainties were not only a result of analytical variability but also of inhomogeneity and stability of the acrylamide in food.  相似文献

The determination of phenolic compounds is of great importance owing to their high toxicity. Some of them are present in tobacco smoke and it is important for their monitoring in air of closed room. A simple, rapid and sensitive method was developed for simultaneous determination of hydroquinone, resorcinol and phenol in this kind of samples. Synchronous fluorescence technique was used and the data were processed by using the partial least-squares (PLS) chemometric algorithm. The concentrations for experimental calibration matrix were varied between 0.02 and 0.2 mg L−1 for hydroquinone, between 0.05 and 0.6 mg L−1 for resorcinol and between 0.05 and 0.4 mg L−1 for phenol in accordance with the national legislation. The cross-validation method was used to select the number of factors. To check the accuracy of the proposed method a recovery study on real samples was carried out.  相似文献

A sensitive and specific monoclonal ELISA for the determination of tissue bound furazolidone metabolite 3-amino-2-oxazolidinone (AOZ) is described. The procedure enables the detection of AOZ in matrix supernatant after homogenisation, protease treatment, acid hydrolysis and derivatisation of AOZ released from the tissue by o-nitrobenzaldehyde. The formed p-nitrophenyl 3-amino-2-oxazolidinone (NPAOZ) is determined by ELISA calibrated with matrix-matched standards in the concentration range of 0.05–5.0 μg I−1. The assay was validated according to criteria set down by Commission Decision 2002/657/EC for the performance and validation of analytical methods for chemical residues. Detection capability, set on the basis of acceptance of no false negative results, was 0.4 μg kg−1 for shrimp, poultry, beef and pork muscle. This sensitivity approaches the established confirmatory LC–MS/MS able to quantify tissue-bound AOZ at levels as low as 0.3 μg kg−1. An excellent correlation of results obtained by ELISA and LC/MS–MS within the concentration range 0–32.1 μg kg−1 was found in the naturally contaminated shrimp samples (r = 0.999, n = 8). A similar correlation was found for the incurred poultry samples within the concentration range of 0–10.5 μg kg−1 (r = 0.99, n = 8).  相似文献

The first application of the flow analysis coupled with chemiluminescence detection and based on stopped-flow chemistry to the simultaneous determination of two components, using a two equation system, is described. The proposed method to determine simultaneously morphine and naloxone is based on the chemiluminescence oxidation of these compounds by their reaction with potassium permanganate in an acidic medium. The main feature of the system used is that the recording of the whole chemiluminescence intensity-versus-time profiles can be obtained, using the stopped-flow technique in a continuous-flow system. Then, the chemiluminescent signals obtained at two times of these profiles can be used to determine the concentration of both opiate narcotics. The effect of common emission enhancers on the chemiluminescence emission of these compounds in different acidic media, using the above-mentioned technique, was studied, in order to achieve the best conditions in which, the CL profiles of both compounds should be additive. The parameters selected were sulphuric acid 1.0 mol L−1, permanganate 0.2 mmol L−1 and formaldehyde 0.8 mol L−1. Taken in account the different profiles of the transient CL signal obtained with each compounds, using the selected chemical conditions, two measurement times (1.4 and 4.8 s) of these responses curves were considered with the purpose to establish a simple 2 × 2 matrix calculation. Using the chemiluminiscent signals obtained at these times, a linear calibration graph was obtained for each one of the compounds between 0.01 and 1.00 mg L−1 for morphine and 0.10–1.50 mg L−1 for naloxone. The present chemiluminescence procedure was applied to the determination of both compounds in mixtures and was found to be satisfactory.  相似文献

Enrichment of benzo[a]pyrene in vegetable oils and determination by HPLC-FL   总被引:7,自引:0,他引:7  
We have developed a simple method for the determination of the carcinogen Benzo[a]pyrene (BP) in vegetable oils. The method consists of extraction of the vegetable oil in acetonitrile, concentration to dryness in rotary evaporator and redissolution of the residue in hexane. The purification of the hexane extract was on Sep-Pack Silica Plus cartridges, and the determination of the BP in the isolated extract was by HPLC-FL. Detection and quantification limits were 0.23 and 0.32 μg kg−1 of olive oil, respectively. Recovery (>93%) and RSD (<4%) were satisfactory. When applied to 18 oil samples, BP levels varied from not detected to 1.99 μg kg−1.  相似文献

A simple and rapid on-line spectrophotometric method for the determination of bromate is proposed. The method is based on the reaction of bromate and 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol (5-bromo-PADAP) with SCN, a red complex is formed and monitored at 550 nm. The linear range found is between 0.18 and 3.00 mg l−1 with a detection limit of 0.15 mg l−1. The sampling rate was calculated to be 45 samples per hour. Homocystine was for the first time, chemically linked to a highly cross-linked agarose support (Novarose) to be employed as a chelating adsorbent for preconcentration and AAS determination of nickel in table salt and baking soda. Nickel is quantitatively adsorbed on a small column packed with 0.25 ml of the adsorbent, in a pH range of 5.5–6.5 and simply eluted with 5 ml of a 1 mol l−1 hydrochloric acid solution.

A factorial design was used for optimization of the effects of five different variables on the recovery of nickel. The results indicated that the factors of flow rate and column length, and the interactions between pH and sample volume are significant.

In the optimized conditions, the column could tolerate salt concentrations up to 0.5 mol l−1 and sample volumes beyond 500 ml. Matrix ions of Mg2+ and Ca2+, with a concentration of 200 mg l−1, and potentially interfering ions of Cd2+, Cu2+, Zn2+ and Mn2+, with a concentration of 10 mg l−1, did not have significant effect on the analyte's signal. Preconcentration factors up to 100 and a detection limit of 0.49 μg l−1, corresponding to an enrichment volume of 500 ml, were obtained for the determination of the analyte by flame AAS. Application of the method to the determination of natural and spiked nickel in table salt and baking soda solutions resulted in quantitative recoveries. Direct ETAAS determination of nickel in the same samples was not possible because of a high background observed.  相似文献

A multisyringe flow injection system (MSFIA) with spectrophotometric detection is proposed as a fast, robust and low-reagent consumption system for the determination of chloride (Cl) in waters. The system is based in the classic reaction of Cl with Fe3+ and Hg(SCN)2, but due to the hazardous properties of this last reagent, the proposed methodology has been developed with the aim to minimize the consumption of this one, consuming less than 0.05 mg of Hg for a Cl determination, being the system of this type with the lowest Hg consumption. The linear working range was between 1 and 40 mg L−1 Cl and the detection limit was 0.2 mg L−1 Cl. The repeatability (RSD) was 0.8% for a 10 mg L−1 Cl solution, and the injection throughput was 130 h−1. The proposed system is compared with other chloride monitoring flow systems, this comparison is realized with a point of view of the equilibrium between the obtained analytical features and produced residues toxicity. The proposed system was applied to the determination of Cl in mineral, tap and well water.  相似文献

A highly sensitive cathodic stripping voltammetric method for the determination of naringin is presented. It is based on the formation and accumulation of two naringin–mercury complexes at the electrode surface, followed by reduction of the surface species during a differential pulse voltammetric scan. The cathodic stripping responses at −0.25 V and −0.42 V, are evaluated with respect to various experimental conditions, such as composition and pH of the supporting electrolyte, naringin concentration, accumulation potential and preconcentration time. The new method is suitable for the determination of naringin concentrations between 0.1 mg l−1 (1.72×10−7 mol l−1) and 40 mg l−1 (6.88×10−5 mol l−1). A 3σ limit of detection of 32 μg l−1 (55 nmol l−1) can be reached. The relative standard deviation (r.s.d.) is <1.5%. Recovery experiments yielded a mean recovery of 97% (r.s.d.=4.1%). The application of the procedure to the selective determination of naringin in grapefruit juice is demonstrated.  相似文献

Methods for the determination of aluminium and manganese in human scalp hair samples by electrothermal atomic absorption spectrometry using the slurry sampling technique were developed. Palladium and magnesium nitrate were used as chemical modifiers. Hair samples were pulverized using a zirconia vibrational mill ball, and were prepared as aqueous slurries. Determinations can be performed in the linear ranges of 1.9–150 μg l−1 Al3+ and 0.03–10.0 μg l−1 Mn2+. Limits of detection of 0.9 mg kg−1 and 27.6 μg kg−1 were obtained for aluminium and manganese, respectively. The analytical recoveries were between 99.6 and 101.8% for aluminium and in the 98.3–101.3% range for manganese. The repeatability of the methods (n=11), slurry preparation procedure and ETAAS measurement, was 16.0 and 7.9% for aluminium and manganese, respectively. The methods were finally applied to the aluminium and manganese determination in 25 scalp hair samples from healthy adults. The levels for aluminium were between 8.21 and 74.08 mg kg−1, while concentrations between 0.03 and 1.20 mg kg−1 were found for manganese.  相似文献

A simple and sensitive kinetic-spectrophotometric method was developed for the determination of ultra trace amount of formaldehyde in food samples. The method was based on the oxidation of rhodamine B (RhB) by potassium bromate in sulfuric acid medium (formaldehyde as catalyst). The reaction was monitored by measuring the decrease in absorbance of the dye at 515 nm after 6 min. The developed method allowed the determination of formaldehyde in the range of 10–100 μg L−1 with good precision, accuracy and the detection limit was down to 2.90 μg L−1. The relative standard deviations for the determination of 10 and 60 μg L−1 of formaldehyde were 3.0% and 1.9% (n = 10), respectively. The method was found to be sensitive, selective and was applied to the determination of formaldehyde in foods with satisfactory results.  相似文献

In this paper, a fully software-controlled multisyringe flow injection (MSFIA) spectrophotometric system is proposed for the determination of sulfide in environmental and waste waters. The implementation of ancillary solenoid valves into the flow network allows a multitude of injection modalities to be explored, the selected modality being directly dependent on the aim of the assays. The multicommuted sandwich-type approach is introduced in this work as an efficient means to warrant high sensitivity for the particular assay with excellent repeatabilities and a considerable reagent saving. Moreover, a high injection frequency may be easily attained by carrying out a multiple injection modality during a single forward displacement of the piston driver bar. The interfacing of the robust and versatile multisyringe piston pump with an optical fiber plug-in spectrophotometer furnished with a light emitting diode enables the miniaturization of the flow analyzer, which is thus readily adaptable to in-situ and real-time monitoring schemes. The flow method is based on the coupling Fischer’s reaction of sulfide with N,N-dimethyl-p-phenylenediamine in the presence of Fe(III) as oxidizing reagent in a 0.7 M HCl medium. Careful selection of the physical and chemical variables enabled coefficients of variations better than 1.5% (n = 10) at the 1 mg l−1 level for both injection modalities. Dynamic working ranges of 0.2–2.0 and 0.5–5 mg l−1 sulfide for sandwich and multiple injection techniques, and detection limits of 0.09 and 0.15 mg l−1, respectively, were obtained. Furthermore, the sandwich modality featured an average slope of 0.43 ± 0.02 l mg−1 calculated from 10 day-to-day calibration plots. This result reveals better sensitivity than other flowing stream methods described in the literature. The multiple injection technique allowed an improvement of the injection throughput up to 80 h−1, although a decrease of sensitivity was concomitantly observed (average slope of 0.17 ± 0.01 l mg−1).

The multisyringe flow method was successfully applied to the determination of sulfide in different spiked water matrices (namely, mineral, tap, freshwater, seawater and wastewater) with recoveries ranging from 96 to 104%. Good agreement was also found in water samples between the MSFIA results and those of the batch APHA-standard method.  相似文献

Hg(II)-imprinting thiol-functionalized mesoporous sorbent was prepared by a sol–gel method and characterized by X-ray diffraction (XRD), FT-IR spectroscopy and nitrogen gas adsorption–desorption. The static adsorption capacity of the Hg(II)-imprinted and non-imprinted sorbent was 78.5 and 26.6 mg g−1, respectively. The breakthrough capacity was 4.46 mg g−1, and the relative selectivity coefficient for Hg(II) in the presence of Cd and Pb was 3.3 and 3.9, respectively. A new method using a micro-column packed with Hg(II)-imprinting thiol-functionalized mesoporous sorbent has been developed for preconcentration of trace mercury prior to its determination by inductively coupled plasma optical emission spectrometry (ICP-OES). The effects of pH, sample flow rate and volume, elution solution and interfering ions on the recovery of the analyte have been investigated. The limit of detection was 0.39 ng ml−1 with a concentration factor of 150 times. The developed method has been applied to the determination of trace mercury in some biological and environmental samples with satisfactory results. The accuracy was assessed through recovery experiments and analysis of certified reference material.  相似文献

An improved flow-based procedure is proposed for turbidimetric sulphate determination in waters. The flow system was designed with solenoid micro-pumps in order to improve mixing conditions and minimize reagent consumption as well as waste generation. Stable baselines were observed in view of the pulsed flow characteristic of the systems designed with solenoid micro-pumps, thus making the use of washing solutions unnecessary. The nucleation process was improved by stopping the flow prior to the measurement, thus avoiding the need of sulphate addition. When a 1-cm optical path flow cell was employed, linear response was achieved within 20–200 mg L−1, described by the equation S = −0.0767 + 0.00438C (mg L−1), r = 0.999. The detection limit was estimated as 3 mg L−1 at the 99.7% confidence level and the coefficient of variation was 2.4% (n = 20). The sampling rate was estimated as 33 determinations per hour. A long pathlength (100-cm) flow cell based on a liquid core waveguide was exploited to increase sensitivity in turbidimetry. Baseline drifts were avoided by a periodical washing step with EDTA in alkaline medium. Linear response was observed within 7–16 mg L−1, described by the equation S = −0.865 + 0.132C (mg L−1), r = 0.999. The detection limit was estimated as 150 μg L−1 at the 99.7% confidence level and the coefficient of variation was 3.0% (n = 20). The sampling rate was estimated as 25 determinations per hour. The results obtained for freshwater and rain water samples were in agreement with those achieved by batch turbidimetry at the 95% confidence level.  相似文献

A new assay of nucleic acids at nanogram level was established based on the enhanced resonance light scattering (RLS) signals of two zwitterionics cocamidopropyl hydroxysultaine (HSB) and lauryl betaine (BS-12). Under optimum conditions, the weak RLS signal of HSB is enhanced by nucleic acids, and the enhanced RLS intensity is proportional to the concentration of nucleic acids in the range of 0.02–7.3 mg l−1 for calf thymus DNA and 0.01–8.6 mg l−1 for fish sperm DNA. The detection limits were 1.5 ng ml−1 for calf thymus DNA and 1.9 ng ml−1 for fish sperm DNA. Plasmid DNA extracted from K-12-HB101 colt was determined with satisfactory results.  相似文献
