Mass-transfer systems based on electrokinetic phenomena (i.e., capillary electrochromatography (CEC)) have shown practical potential for becoming powerful separation methods for the biotechnology and pharmaceutical industries. A dynamic mathematical model, consisting of the momentum balance and the Poisson equations, as well as the unsteady-state continuity expressions for the cation and anion of the background electrolyte and of a positively charged analyte (adsorbate), is constructed and solved to determine quantitatively the electroosmotic velocity, the electrostatic potential, the concentration profiles of the charged species in the double layer and in the electroneutral core region of the fluid in the interstitial channels for bulk flow in the packed chromatographic column, and the axial current density profiles as the adsorbate adsorbs onto the negatively charged fixed sites on the surface of the nonporous particles packed in the chromatographic column. The frontal analysis mode of operation is simulated in this work. The results obtained from model simulations provide significant physical insight into and understanding of the development and propagation of the dynamic profile of the concentration of the adsorbate (analyte) and indicate that sharp, highly resolved adsorption fronts and large amounts of adsorbate in the adsorbed phase for a given column length can be obtained under the following conditions: (i) The ratio, gamma(2, 0), of the electroosmotic velocity of the mobile liquid phase at the column entrance after the adsorption front has passed the column entrance to the electrophoretic velocity of the anion is very close to -1. The structure of the equations of the model and model simulations indicate that a stable adsorption front cannot develop when gamma(2, 0) is less than -1 unless the value of the mobility of the cation is less than the value of the mobility of the analyte, which may be a rare occurrence in practical CEC systems. (ii) The ratio of the mobility of the cation to the mobility of the analyte is less than two orders of magnitude. This effect becomes more significant as the value of the equilibrium adsorption constant, K(A, 3), of the analyte increases. (iii) The concentration of the analyte relative to the concentration of the cation is increased (feed solutions with less dilute concentrations of the analyte are employed). Therefore, to obtain good performance for CEC systems operated in the frontal analysis mode (well-resolved adsorption fronts and high adsorbate amounts in the adsorbed phase), one can choose an electrolyte whose cation has a mobility that is not more than one or two orders of magnitude greater than the mobility of the analyte and whose anion has a mobility such that the value of gamma(2, 0) is close to -1; one can then bring the value of gamma(2, 0) closer to -1 by decreasing the particle diameter, d(p), and/or making the value of the surface charge density, delta(0), of the particles more negative (in effect, making the value of the zeta potential, zeta(p), at the surface of the particles more negative at time t=0) to change the value of the velocity, |(x=0), of the electroosmotic flow (EOF) at the column entrance (|(x=0) is determined after the adsorption front has passed the column entrance). This approach could provide conditions in the column that avoid overloading of the adsorbate. One can obtain faster breakthrough times at the sacrifice of resolution and utilization of the adsorptive capacity of the packed bed if one employs a cation whose mobility is very large relative to the mobility of the analyte and/or an anion that provides a value of gamma(2, 0) significantly greater than -1. If it is possible, one can increase the concentration of the analyte in the feed stream to avoid sacrificing resolution and adsorptive capacity of the packed bed and still decrease the time at which breakthrough occurs. Also, the dynamic behavior of the axial current density, i(x), profiles indicates that the magnitude of i(x) and/or the change in the value of i(x) across the adsorption front could serve as a measurement for the rate of propagation of the adsorption front through the column. Furthermore, the effect of the decreased magnitude of the velocity of the EOF in the region of the column where the analyte is present in the adsorbed phase could act to decrease the effect of tailing when CEC systems are operated in the pulse injection mode (analytical electrochromatography) because the higher velocity of the fluid upstream of the migrating adsorption zone may compress the tail of the peak. Monolithic silica columns with surface-bound octadecyl (C18) moieties have been prepared by a sol-gel process in 100 microm ID fused-silica capillaries for reversed-phase capillary electrochromatography of neutral and charged species. The reaction conditions for the preparation of the C18-silica monoliths were optimized for maximum surface coverage with octadecyl moieties in order to maximize retention and selectivity toward neutral and charged solutes with a sufficiently strong electroosmotic flow (> 2 mm/s) to yield rapid analysis time. Furthermore, the effect of the pore-tailoring process on the silica monoliths was performed over a wide range of treatment time with 0.010 M ammonium hydroxide solution in order to determine the optimum time and conditions that yield mesopores of narrow pore size distribution that result in high separation efficiency. Under optimum column fabrication conditions and optimum mobile phase composition and flow velocity, the average separation efficiency reached 160 000 plates/m, a value comparable to that obtained on columns packed with 3 microm C18-silica particles with the advantages of high permeability and virtually no bubble formation. The optimized monolithic C18-silica columns were evaluated for their retention properties toward neutral and charged analytes over a wide range of mobile phase compositions. A series of dimensionless retention parameters were evaluated and correlated to solute polarity and electromigration property. A dimensionless mobility modulus was introduced to describe charged solute migration and interaction behavior with the monolithic C18-silica in a counterflow regime during capillary electrochromatography (CEC )separations. The mobility moduli correlated well with the solute hydrophobic character and its charge-to-mass ratio.  相似文献

A mathematical model has been constructed and solved to describe quantitatively the profiles of the electrostatic potential, pressure, and velocity of the electroosmotic flow (EOF) in charged cylindrical capillaries and in capillary columns packed with charged particles. The results obtained from model simulations (i) provide significant physical insight and understanding with regard to the velocity profile of the EOF in capillary columns packed with charged porous particles which represent systems employed in CEC, (ii) provide the physical explanation for the experimental results which indicate that the velocity of the EOF in capillary columns packed with charged porous particles is a very weak function (it is almost independent) of the diameter of the particles, and (iii) indicate that the intraparticle velocity, nu(p,i), of the EOF can be greater than zero. The intraparticle Peclet number, Pe(int rap), for lysozyme was found to be greater than unity and this intraparticle convective mass transfer mechanism could contribute significantly, if the appropriate chemistry is employed in the mobile liquid phase and in the charged porous particles, in (a) decreasing the intraparticle mass transfer resistance, (b) decreasing the dispersive mass transfer effects, and (c) increasing the intraparticle mass transfer rates so that high column efficiency and resolution can be obtained. Furthermore, the results from model simulations indicate that for a given operationally permissible value of the applied electric potential difference per unit length, Ex, high values for the average velocity of the EOF can be obtained if (1) the zeta potential, zeta(p), at the surface of the particles packed in the column has a large negative magnitude, (2) the value of the viscosity, mu, of the mobile liquid phase is low, (3) the magnitude of the dielectric constant, epsilon, of the mobile liquid phase is reasonably large, and (4) the combination of the values of the concentration, C(infinity), of the electrolyte and of the dielectric constant, epsilon, provide a thin double layer. The theoretical results for the velocity of the EOF obtained from the solution of the model presented in this work were compared with the experimental values of the velocity of the EOF obtained from a fused-silica column packed with charged porous silica C8 particles. Systems with four different particle diameters and three different concentrations of the electrolyte were considered, and the magnitude of the electric field was varied widely. Novel general expressions are constructed and presented that describe the behavior of the height equivalent of a theoretical plate (plate height), H, as a function of the linear velocity, Vx, along the axis, x, of the column and the kinetic parameters that characterize the mass transfer and adsorption mechanisms in chromatographic columns. Open tube capillaries as well as columns packed with either non-porous or porous particles are studied. The porous particles could have unimodal or bimodal pore-size distributions and intraparticle convective fluid flow and pore diffusion are considered. The expressions for the plate height, H, presented in this work could be applicable to high-performance liquid chromatography (HPLC) and capillary electrochromatography (CEC) systems, and could be used together with experimental plate height, H, versus linear velocity, Vx, data to determine the values of the parameters that characterize intraparticle convective fluid flow and pore diffusion. Furthermore, chromatographic systems under unretained as well as under retained conditions are examined. The experimental values of the plate height, H, versus the linear velocity, Vx, for a CEC system involving charged porous silica C8 particles and an uncharged analyte are compared with the theoretical results for the plate height, H, obtained from the expressions presented in this work. The agreement between theory and experiment is good, and the results indicate that the magnitude of the intraparticle electroosmotic flow (EOF) in the pores of the particles is substantial while the pore diffusion coefficient was of small magnitude. But the overall intraparticle mass transfer resistance in these particles was low because of the significant contribution of the intraparticle EOF. Simulation results are also presented (i) for a hybrid HPLC-CEC system, and (ii) for different CEC systems involving open capillaries as well as packed columns having non-porous or porous particles. The analysis of the results indicates (a) the reasons for the superior performance exhibited by the hybrid HPLC-CEC system over the performance obtained when the system is operated only in the HPLC mode, and (b) the operational configuration and the properties that the structure of the porous particles would have to have in CEC systems involving uncharged or charged analytes under unretained or retained conditions in order to obtain high CEC efficiency (low values of the plate height, H).  相似文献

Column technologies for capillary electrochromatography (CEC) are reviewed. To achieve high efficiency, the inner diameters of open-tubular and packed columns should be less than 25 and 200 μm, respectively. To obtain acceptable separation speed under typical CEC conditions (e.g. 30 kV, 1 mm s−1 electroosmotic flow velocity, and 2–4×10−8 m2 V−1 s−1 electroosmotic mobility) the column lengths for open-tubular and packed columns should be less than 120 and 60 cm, respectively. Capillary CEC columns are generally classified into three types: packed, open-tubular, and continuous-bed or monolithic. The various column preparation procedures and the advantages and disadvantages of each column type are discussed in detail.  相似文献

The results obtained from the pore network model employed in this work, clearly show that the magnitudes of the intraparticle electroosmotic volumetric flow-rate, Qintrap, and velocity, (v(intrap,x)), in the pores of the charged porous silica particles considered in this study are greater than zero. The intraparticle Peclet number, Pe(intra, of a solute in these charged porous silica particles would be greater than zero, and, in fact, the magnitude of the intraparticle Peclet number, Pe(intrap), of lysozyme is greater than unity for all the values of the pore connectivity, nT, of the intraparticle pores and of the applied electric potential difference per unit length, Ex, along the axis of the capillary column considered in this work. Furthermore, the values of the intraparticle electroosmotic volumetric flow-rate, Qintrap, and velocity, (v(intrap,x)), as well as the magnitude of the pore diffusion coefficient, Dp, of the solute increase as the value of the pore connectivity, nT, of the intraparticle pores increases. The intraparticle electroosmotic flow can contribute significantly, if the appropriate chemistry is employed in the mobile liquid phase and in the charged porous particles, in (i) decreasing the intraparticle mass transfer resistance, (ii) decreasing the dispersive mass transfer effects, and (iii) increasing the intraparticle mass transfer rates so that high column efficiency and resolution can be obtained.  相似文献

The aim of this study was to evaluate the applicability of ODS-AQ packing material as a stationary phase in capillary electrochromatography (CEC). The electroosmotic flow created on an ODS-AQ stationary phase was measured at different mobile phase compositions and at different column temperatures. It was observed that the electroosmotic flow generated in the column increased by 50% when the temperature of the system was raised from 20 degrees C to 60 degrees C, while all other conditions were kept constant. The electroosmotic flow produced by the ODS-AQ stationary phase was found to be comparable to the flow generated in a column packed with Nucleosil bare-silica material. In addition, a set of polar compounds (D-lysergic acid diethylamide derivatives) was utilized to determine the influence of temperature and mobile phase composition on their chromatographic behavior on an ODS-AQ stationary phase in a CEC mode. A linear relationship between the solute retention factor and column temperatures was seen over the temperature range studied (20 degrees C to 60 degrees C). A quadratic function was used to describe the changes in the solute retention factors with variation of acetonitrile concentration in the mobile phase.  相似文献

Separation of enantiomers was performed by applying packed capillary electrochromatography (CEC). Fused-silica capillaries of different lengths with an inner diameter of 100 microm were packed with a cellulose derivative immobilized onto macroporous silica gel. Parameters such as content of modifier in the mobile phase, concentration and pH of the buffer were varied for a set of test capillaries to determine their influence on enantioselectivity. In packed CEC the highest influence on resolution of the test racemates was found by changing the acetonitrile content, while variation of the buffer concentration mostly affects the electroosmotic velocity. The performance of packed CEC and nano-LC was also compared. Packed CEC showed much better column efficiency and enantioselectivity under similar flow/electroosmotic velocity.  相似文献

A new in-house designed and constructed injection valve for capillary electrochromatography (CEC) based on a rotating injection part with compartments for the eluent as well as for the sample has been coupled to a mass spectrometer via a sheath flow electrospray ionisation (ESI) interface, using short capillary columns of 15 cm length. The CEC columns were packed with 3 microm C(18) bonded silica particles, and a mixture of peptides was analysed using an ammonium acetate/acetonitrile eluent. A significant increase in the signal-to-noise ratio was obtained when the peptides were dissolved in water with the same content of organic modifier as in the eluent with an addition of 0.5% (v/v) acetic acid. When the CEC analysis was performed without any additional pressure, the separation current sometimes dropped tremendously due to bubble formation, caused by different permeability in the first and packed part of the column causing an extremely low electroosmotic flow. The separation current was restored to its original value by applying only 7 bar at the inlet of the CEC column, and the separation performance for the test peptides was recovered. A comparison of the CEC performance of peptides in pure CEC mode and in low-pressure CEC mode is reported.  相似文献

A silica-based monolithic stationary phase prepared by the sol-gel process in a 100 microm I.D. fused-silica (FS) capillary has been modified chemically with 3-mercaptopropyl trimethoxysilane followed by immobilization of a strong cation-exchange (SCX) type chiral selector, (S)-N-(4-allyloxy-3,5-dichlorobenzoyl)-2-amino-3,3-dimethylbutane phosphonic acid, by radical addition reaction onto the reactive sulfhydryl surface. After a fine-tuning of the mobile phase composition, the enantioselective capillary column was evaluated for the separation of various chiral basic drugs by enantioselective non-aqueous capillary electrochromatography (CEC), in comparison to capillary column analogs packed with 3.5 microm silica particles having attached the same selector. The performance of the monolithic silica column was further compared to corresponding polymethacrylate-based organic polymer monoliths. The study indicated that strong counter-ions such as 2-aminobutanol or N,N,N',N'-tetramethylethylenediamine are needed, although they reduce the electroosmotic flow velocity and separation factors in comparison to less efficient counter-ions, in order to allow the elution of the oppositely charged solutes in the ion-exchange retention mode within reasonable run time and as sharp zones. In contrast, weak counter-ions such as N,N-diisopropylethylamine (Huenig base) provided stronger electroosmotic flow and much better separation factors, but relatively poor peak efficiencies. Overall, with the chemically functionalized monolithic silica column the high quality separations of packed column analogs could be approximated, with regards to both separation factors and peak performances. On the other hand, the monolithic capillary column certainly outperformed the packed column in terms of system robustness under capillary electrochromatography conditions and showed excellent column longevity. The enantioselective strong cation-exchange-type monolithic silica column performed also well in comparison to the organic polymer monolith.  相似文献

Summary A packing procedure was adopted for capillary electrochromatography (CEC) that produces capillary columns with high separation efficiencies. The columns were fully packed, 50 cm long, with UV detection being performed through the packed section 30 cm from the inlet end. The CEC experiments were run at ambient pressure, with no additional pressure applied to the ends of the column. The stationary phase (octadecyl silica (ODS), 5 μm) promoted a high velocity electroosmotic flow (EOF), enabling rapid and efficient separation of a hydrocarbon test mixture. Some 1-phenyl-3-methyl-5-pyrazolone (PMP) derivatives of mono- and disaccharides were baseline separated, using a 5 mM NaH2PO4 in 80% acetonitrile and 20% water (v/v) buffer solution. A major impediment to enhancing the speed of separation in capillary electrochromatography (CEC) is the upper limit on the electroosmotic flow (EOF) velocity by the maximal zeta potential of the chromatographic surface. Here, a new approach to speeding up EOF, suggested by Yang and El Rassi (Electrophoresis 1999, 20,18-23), is examined critically. It entails the use of a tandem arrangement of a separating column and an auxiliary column, the sole function of which is to boost EOF velocity in the separating column and thus facilitate faster analysis by CEC. Based on the principle of conservation of mass and current and using experimental data obtained in a wide range of conditions, the flow velocities in the separating and auxiliary columns were evaluated. The results show that an equidiameter open tubular auxiliary column offers a greater enhancement of EOF velocity than a packed column. Nevertheless, within the scope of the experiments the enhancement of EOF velocity by as much as 50% by using open tubular auxiliary columns has been obtained.  相似文献

毛细管电色谱(CEC)以内含色谱固定相的毛细管为分离柱,以电渗流为驱动力,既可以分离带电物质也可以分离中性物质。它结合了毛细管电泳和高效液相色谱两者的优点,兼具高柱效、高分辨率、高选择性和高峰容量的特点,同时具有色谱和电泳的双重分离机理。然而,“纯粹”的电色谱在实际应用中有着天然的弱点,即: 在电流通过毛细管柱中的流动相时容易产生气泡(焦耳热作用),从而使电流中断和电渗流停止,毛细管柱必须被重新用流动相润湿后方能再次使用。加压毛细管电色谱(pCEC)将液相色谱中的压力流引入CEC系统中,不仅解决了气泡、干柱等问题,而且实现了定量阀进样和二元梯度洗脱。CEC和pCEC作为微分离领域的两种前沿技术,满足了当前复杂样品分析和分析仪器微型化的需求,近年来获得了广泛的关注。本文综述了这两种技术近来的发展,包括仪器、色谱固定相的发展,总结了其在生命科学、药物分析、食品安全以及环保样品分析等方面的应用进展,评述了各方法的特点,并展望了CEC和pCEC今后的发展和应用前景。  相似文献

The influence of temperature, T, on the retention times, peak widths, peak symmetry coefficients and theoretical plate numbers of two small linear peptides, [Met5]enkephalin and [Leu5]enkephalin, has been studied with capillary electrochromatography (CEC) capillary columns of 100 microm I.D. and 250 mm packed length with a total length of 335 mm, containing 3 microm Hypersil n-octadecyl bonded silica. With increasing column temperature from 15 to 60 degrees C, the electroosmotic flow (EOF) and the column efficiencies increased, whereas the retention coefficients (Kcec) of both peptides decreased. A linear relationship was found between the EOF value and the square root of the temperature over this temperature range, with a linear regression correlation of 0.998. Non linear Van 't Hoff plots (In Kcec versus 1/T) were observed for these peptides between 15 and 60 degrees C, suggesting that a phase-transition occurred with the n-octadecyl chains bonded on the silica surface, affecting the CEC retention behaviour of these peptides. In CEC systems, the Kcec values of peptides incorporate contributions from both electrophoretic migration and chromatographic retention. Positive and negative Kcec values can, in principle, thus arise with these charged analytes. However, the Kcec values of the enkephalin peptides under all temperature conditions studied were positive with an eluent composed of water-50 mM NH4OAc/AcOH, pH 5.2-acetonitrile (5:2:3, v/v) and therefore the chromatographic component dominates the retention process with these small peptides under these conditions.  相似文献

To investigate how fluid compressibility affects efficiency in supercritical fluid separations, band dispersion along a packed capillary column was measured from on-column elution rate profiles obtained under solvating gas chromatography (SGC) conditions; this allowed efficiency to be determined with respect to position along the column. Theoretical efficiency was also modeled. The model indicates that the primary cause of band broadening in SGC is high mobile phase velocity near the column outlet. However, the experimental results show that significant band broadening also occurs near the column inlet in a region that corresponds to high elution rates of the analyte. On-column detection also revealed spatial focusing of the analyte as it moves down the column density gradient.  相似文献

A linear polymer-coated capillary was prepared by in-capillary copolymerization of N-tert-butylacrylamide (TBAAm) with a charged monomer, 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS), after the capillary pretreatment with a bifunctional reagent. The coated capillaries were applied in capillary electrochromatographic (CEC) separation of small neutral compounds. Hydrophobic groups in the linear polymer, which were immobilized onto the capillary surface, functioned as the stationary phase in reversed-phase CEC separation, and charged groups in the linear polymer generated electroosmotic flow (EOF) along the column. The coated capillaries were prepared by a simple procedure. Moreover, the reproducibility with respect to EOF rate and migration times of the solutes was excellent. The results for CEC separation of small molecules using the linear polymer-coated capillaries are presented.  相似文献

In this article, we present an analysis on the transport of charged samples through micro- and nanofluidic channels with large zeta potentials (|zeta| > (kBT)/e). Using the Method of Moments formulation, the diffusion-convection equation has been solved to evaluate the mean velocity and the dispersion of analyte bands in a parallel-plate device under electrokinetically- and pressure-driven flow conditions. The effect of electromigration induced by the lateral electric field within the Debye layer has been quantified in our work using a Peclet number (Pe t) based on the characteristic electrophoretic velocity of the solute molecules in the transverse direction. It has been shown that while the effects of transverse electromigration on analyte transport only depends on the product Pe t zeta* for |zeta*| = (ezeta)/kBT < 1, both these parameters independently affect the flow of charged species in large zeta potential systems. For a given value of Pe t zeta*, the mean velocity and the slug dispersivity can vary by as much as an order of magnitude in going from a small zeta potential system (|zeta*| < 1) to a channel with |zeta*| = 4.  相似文献

This work describes initial investigations of strong anion-exchange (SAX) packing materials for capillary electrochromatography (CEC). The use of SAX phases in CEC is theoretically appealing for the analysis of negatively charged species. The reversed direction of the electroosmotic flow (EOF) generated by SAX phases (in comparison to reversed phases and strong cation-exchange phases) means that negative species can migrate with the EOF, not against it, hence the analysis times, of such species should be decreased and efficiencies improved. Duplex CEC columns (the standard for instruments using UV detection) consist of a packed and an unpacked section. Using common reversed-phase packing materials the direction of the EOF in both sections is co-linear, however when normal fused-silica capillaries are packed with SAX material the direction of the EOF in the two sections oppose one another. It has been shown, using conventional duplex CEC columns and fully packed CEC-MS columns that the opposing direction of EOF causes a massive degradation in column performance. Consequentially, it is demonstrated that if the EOF in the open section of the duplex SAX column can be controlled via pH or capillary derivatisation then good, reproducible CEC can be performed on anionic species using SAX packed CEC columns.  相似文献
