本文采用分子荧光光谱法对阿莫西林与牛血清白蛋白(BSA)之间的相互作用进行了研究,得知阿莫西林对BSA有猝灭作用,其猝灭方式为静态猝灭,并求出了猝灭常数,结合常数及结合位点数.通过298K和310K时的实验数据求出热力学参数△H、△G和△S,结果表明阿莫西林主要通过氢键和范德华力与BSA之间结合.  相似文献

本文提出了阿莫西林[(2S,5R,6R)-3,3-二甲基-6-[(R)-(-)-2-氨基-2-(4-羟基苯基)乙酰氨基]-7-氧化-4-硫杂-1-氮杂双环[3,2,0]庚烷-2-甲酸三水化合物]荧光分析的方法。并对影响阿莫西林荧光性质(光谱和光谱强度)的各种因素,包括pH值、表面活性剂的增效作用、无机离子的配合作用以及其他介质条件等进行了较为详细的研究。实验结果表明,阿莫西林具有良好的荧光性质,在微碱性条件下荧光发射最强,CTAB对其有增效作用,与无机离子(Mg  相似文献

通过电化学氧化肾上腺素,使其生成羟基吲哚类荧光物质(λem=490nm),实现了药物中肾上腺素含量的电化学氧化-荧光检测。研究结果表明,荧光强度与肾上腺素浓度分别在2.00×10-7—1.00×10-6mol/L和1.00×10-6—4.00×10-5mol/L范围内呈良好的线性关系,检出限为6.70×10-8mol/L。该方法用于药物制剂中肾上腺素含量的测定,相对标准偏差小于1.88%(n=7),加标回收率为88.4%—116.6%。  相似文献

微量元素对B族维生素同步荧光分析法影响的研究   总被引:3,自引:1,他引:3  
本文探讨了钙、镁、铜等 7种微量元素对维生素 B1、B2 、B6的同步荧光光谱的影响。实验结果表明 ,当Ca2 + 的浓度为 1.0× 10 -3mol/ L时 ,VB2 、VB6的同步荧光光谱有显著的增强作用 ;而其他微量元素对VB1、VB2 、VB6的同步荧光有不同程度的猝灭或增强作用。利用 Ca2 + 对 3种维生素荧光强度的增敏作用 ,可以有效地提高同步荧光分析法同时测定 VB1、VB2 、VB6的灵敏度 ,VB1、VB2 、VB6的校准曲线的线性范围分别为 2 .4× 10 -7— 9.0× 10 -6、1.1× 10 -7— 4 .6× 10 -6、7.1× 10 -7— 2 .0× 10 -6mol/ L。采用本方法测定混合样品 ,样品不需分离 ,操作简便、快速  相似文献

甲磺酸培氟沙星是一种新型的抗菌素。本对其荧光分析进行了研究。该物质在pH=3的三酸缓冲溶液中有很强的荧光发射光谱。线性范围较宽,检出限较低,在浓度为1×10^-9 ̄1×10^-6mol/L间均成直线。同时,本对甲磺酸培氟沙星在人血清和尿液中进行了回收测定,回收率在95 ̄105%之间,相对标准偏差小于6%,  相似文献

以快速、简便和有效的荧光分析法研究了 5种环糊精 (cyclodextrins,CDs)对溶解态蒽在水溶液中光降解的影响。发现 5种 CDs都能够促进蒽在水溶液中的光降解过程 ,促进作用的能力的顺序为 :HPCD(hydroxypropyl- β- CD) >DMCD(heptakis(2 ,6 - di- o- methyl) - β- CD) >β- CD>γ- CD>α- CD。这是因为蒽进入 CDs空腔后 ,抑制了蒽三重态的自猝灭过程以及延长了单重态的寿命 ,这使三重态氧猝灭蒽的激发态的几率增加 ,生成的单重态的氧的量增加。探讨了 CDs空腔大小及空间位阻对蒽光降解的影响。  相似文献

平行因子分析法在太湖水体三维荧光峰比值分析中的应用   总被引:5,自引:0,他引:5  
以太湖水样三维荧光光谱数据为例,提出在采用平行因子分析法(PARAFAC)处理后的荧光数据中提取荧光峰强度计算荧光峰比值进行水环境分析评价的方法,较直接在水样原始荧光谱图中获取的荧光峰强度更加准确客观。天然水体中各水样间由于受荧光团复杂多样性等因素的影响,某类荧光物质荧光峰的激发发射波长位置并不是固定不变的,就同一水样而言各类荧光峰之间的相互重叠干扰也将影响到荧光峰强度和位置的准确判断。而在PARAFAC模型各因子中提取相应荧光峰值可以保证各水样间同类荧光物质荧光峰在同一位置又有效减弱同一水样中各类荧光物质荧光峰之间的相互干扰,更加高效准确的利用荧光峰比值进行水环境分析。区域差异性分析时水样因子得分比值的区域变化与原始荧光峰比值变化趋势是一致的。  相似文献

The denaturation of Bovine Serum Albumin (BSA) by a chaotropic agent, guanidinium hydrochloride (GuH+Cl-) was studied by fluorescence lifetime analysis. The BSA was labelled with 1-anilino-8-naphthalene sulfonate (ANS) at two different molar ratios (1:1) and (1:10). The non-exponential fluorescence kinetics of the BSA-ANS complex at different stages of denaturation is analysed using three different models: a discrete tri-exponential sum, stretched exponential, and Gaussian lifetime distribution. In all cases, the fluorescence decay times decreased with protein denaturation. The results from the models show that there are at least two different binding sites located in the BSA protein with different water accessibility.  相似文献

It was found that ethanol and water molecules mixed together can form new clusters. Based on the steady state spectral characteristic and the mole ratio method, three possible bonding constants of ethanol-water clusters were deduced. To confirm the structure of these clusters, the fluorescence lifetimes of different emission bands were investigated, and the average lifetime of the entire decay process was found to vary as the volume percent of ethanol changed. Furthermore, the possible bonding constant n of molecular clusters was determined to be 2 based on the time-resolved spectra, and the cluster was concluded to be a chain structure formed by one ethanol molecule and two water molecules with hydrogen bonds.  相似文献

There is a growing interest in functional membrane heterogeneity on the mesoscopic (several tens to hundreds of molecular dimensions) scale. However, the physical-chemical basis for this sort of heterogeneity in membranes is not entirely clear. Unambiguous methods to demonstrate that the cell plasma membrane and other cellular membranes are in fact heterogeneous on the mesoscopic level are also not generally available. Fluorescence techniques do, however, provide excellent tools for this purpose. In particular, the emerging techniques of scanning near-field optical microscopy and single-molecule fluorescence microscopy hold a great deal of promise for the near-future. All these methods require the use of fluorescent probes (lipids and/or proteins) and a clear definition of how these probes partition between domains of coexisting membrane phases. The development of the concept of membrane heterogeneity over the years since the first proposal of the fluid mosaic model is reviewed briefly. The use of lipid-binding proteins in experimental protocols for the labeling of membranes with fluorescent lipid amphiphiles as monomers in aqueous solutions at concentrations well above their critical aggregation concentrations is discussed. The methods of fluorescence spectroscopy available to the cell biologist for determining probe partition coefficients for partitioning between coexisting membrane phases are reviewed in some detail, as is the relevant theoretical and experimental work reported in the literature.  相似文献

The Cu-Lβ/Cu-Lα x-ray emission intensity ratio of the following compounds have been found to show an interesting consistency: Cu, Cu2O, CuO, La1.85Ba.15CuO4 (vacuum annealed), La1.85Ba.15CuO4 (air annealed, TC = 26K), La1.85Sr.15CuO4 (TC = 35K), YBa2Cu3O7-Δ (TC = 91K), ErBa2Cu3O7-Δ (TC = 81K), and La2CuO4. The interesting consistency is that all of the last six compounds, all superconductors or superconductor related, have the same Lβ/Lα intensity ratio, namely, 0.33±0.01. The Cu-Lα,β spectra of CuO, air and vacuum annealed La1.85 Ba.15CuO4 and YBa2Cu3O7-Δ are presented. Also presented are O-K x-ray fluorescence spectra of air and vacuum annealed La1.85 Ba.15CuO4, and of “new” and “old” YBa2Cu3O7-Δwhich seem to show a correspondence between spectral sharpness and high TC behavior.  相似文献

用氯化稀土与苯甲酸铵在水溶液中反应,合成了苯甲酸稀土(Eu,La)三元固体系列配合物,其化学组成式为(Eu1-xLax)L3(其中L为C6H5COO-,x=0.0~1.0)。研究了它们的红外光谱和荧光光谱。由红外光谱数据表明,配合物中羧基的对称和反对称伸缩振动吸收峰发生了分裂,而且羧基伸缩振动随着La3+离子含量的增加发生了规律性的变化。荧光光谱实验结果表明,用不发光的La3+离子取代苯甲酸铕中部分Eu3+离子可以提高Eu3+离子的特征荧光发射强度。  相似文献

由邻苯二甲酸氢钾,2,2’-联吡啶分别与EuCl3和TbCl3在乙醇水溶液中反应,合成了铕配合物Eu2L3L’和铽配合物Tb2L3L’(L=邻苯二甲酸根,L’=2,2’联吡啶);以不同摩尔比混合两种配合物,经机械研磨,得混合固体铕铽配合物。测试了配合物的荧光光谱及红外光谱,结果表明,与单一铕配合物和铽配合物相比,混合固态配合物无论是荧光发射峰位还是荧光强度均发生了明显变化,铽对铕的荧光强度有敏化作  相似文献

Molecular engineering of polymer surfaces that includes the attachment of functional molecules to existing or previously generated reactive chemical groups like e.g., − OH, − NH2, or − CHO requires simple strategies and tools for the controlled generation of surface functionalities and their derivatization as well as for their identification and eventually quantification. Here, we systematically investigate the plasma-chemical surface modification of polypropylene films in combination with dansyl labeling chemistry and conventional, yet costly, XPS and highly sensitive fluorescence spectroscopy for the detection of surface groups. Based on these results, the potential of and requirements on the fluorometric characterization and quantification of surfaces functionalities are discussed.  相似文献

用荧光光谱法测定Ⅳ-月桂酰基-Ⅳ'-羟乙基乙二胺乙酸盐(两性乙酸钠)的微极性.芘的荧光强度随着两性乙酸钠浓度的增加而减小,其第一峰(I1)和第三峰(I3)的强度随两性乙酸钠浓度的增加先降低,到达一定浓度后,趋于稳定.由芘的I1/I3与浓度关系可以得出两性乙酸钠的临界胶束浓度(CMC值大约为11.03mmol/L),与表面张力法(1.02mmol/L)测定结果一致.同时研究了无机盐的加入对芘的I1/I3值的影响,无机盐的加人使芘的I1/I3值减小,即无机盐的加入使芘所处的微环境极性减小.  相似文献
