Conventional matrix-assisted laser desorption/ionization mass spectrometry is limited to analyses of higher molecular weight compounds due to high background noise generated by the matrix in the lower mass region. Surface-assisted laser desorption/ionization (SALDI) mass spectrometry is an alternative solution to this problem. Nanoparticles, structured silicon surfaces and carbon allotropes are commonly used as SALDI surfaces. Here, for the first time, we demonstrate the application of silicon nitride nanoparticles as a suitable medium for laser desorption/ionization of small drug molecules.  相似文献

Silver nanoparticle synthesized from chemical reduction has been successfully utilized as a matrix in matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) of peptides. Acting as a substrate to adsorb analytes, as well as a transmission medium for UV laser, silver nanoparticle was found to assist in the desorption/ionization of peptides with little or no induced fragmentation. The size of the nanoparticle was typically in the range of 160 ± 20 nm. One of the key advantages of silver nanoparticle for peptides analysis is its simple step for on-probe sample preparation. In addition, it also minimizes the interferences of sodium dodecyl sulfate (SDS) surfactant background signal, resulting in cleaner mass spectra and more sensitive signal, when compared to α–cyano-4-hydroxycinnamic acid (CCA) matrix.  相似文献

Proteolytic processing of the amyloid precursor protein (APP) by β-secretase and γ-secretase leads to the generation and deposition of amyloid β (Aβ) in Alzheimer’s disease (AD). N-terminally or C-terminally truncated Aβ variants have been found in human cerebrospinal fluid and cultured cell media using immunoprecipitation and mass spectrometry. Unfortunately, the profile of plasma Aβ variants has not been revealed due to the difficulty of isolating Aβ from plasma. We present here for the first time studies of Aβ and related peptides in human plasma. Twenty-two Aβ-related peptides including novel peptides truncated before the β-secretase site were detected in human plasma and 20 of the peptides were identified by tandem mass spectrometry. Using an internal standard, we developed a quantitative assay for the Aβ-related peptides and demonstrated plasma dilution linearity and the precision required for their quantitation. The present method should enhance the understanding of APP processing and clearance in AD progression.  相似文献

We propose a convenient technique applicable for investigations of various functions of nanoparticles produced by laser ablation in liquids. It was demonstrated that nanoparticles of anatase-TiO2, a electrode material for lithium secondary batteries, produced by laser ablation in acetone could be efficiently deposited on a substrate by using an electrophoresis technique. Analysis of the electrochemical properties of nanoparticles become much more facile with those deposited nanoparticles than with dispersed nanoparticles. In addition, it was demonstrated that comparison of the electrochemical properties between nanoparticles and microparticles were possible by means of this technique.  相似文献

Small molecule analysis is one of the most challenging issues in matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. We have developed a cobalt coated substrate as a target for matrix-free analysis of small molecules in laser desorption/ionization mass spectrometry. Cobalt coating of 60-70 nm thickness has been characterized by scanning electron microscopy, energy dispersive X-ray analysis, X-ray diffraction, and laser induced breakdown spectroscopy. This target facilitates hundreds of samples to be spotted and analyzed without mixing any matrices, in a very short time. This can save a lot of time and money and can be a very practical approach for the analysis of small molecules by laser desorption/ionization mass spectrometry.  相似文献

利用实验室烟雾腔,在不同的实验条件下研究了由羟基启动的三甲苯光氧化反应所产生的二次有机气溶胶,采用激光解吸附技术和气溶胶飞行时间质谱仪测量了产物的化学组分和粒子的粒径分布。结果表明:随着反应物三甲苯浓度的增加,所生成的二次有机气溶胶粒子数量增多;随着反应时间的延长,生成的粒子数目也增加,但是反应到一段时间后,检测到的粒子数目几乎保持不变;光照强度的增加有利于二次有机气溶胶的生成;通过对产物的化学成分分析,说明了在二次有机光氧化产物中粒子相产物主要是芳香的环保留产物、非芳香环保留产物和环断裂产物。  相似文献

In this paper, nanodiamonds have been successfully used in matrix-assisted laser desorption/ionization mass spectrometric analysis of human serum and urine. Excimer laser ablation of superconductive borocarbide material (YNi2B2C) in typical conditions for the deposition of superconductive thin films has been investigated using time-of-flight mass spectrometry. The mass spectra show the presence of all the target elemental ionized atoms as well as diatomics. The ablation yield of the metal ions is a strongly increasing function of the laser fluence, while the contrary is true for non-metal ions. The dependence of non-metal light mass diatomic ions on laser fluence indicates the presence of aggregation processes as the laser fluence is increased. Moreover, evidence of aggregation processes involving metallic ions at high laser fluence is also obtained by the mass spectra. An interesting aspect of our results is the observation of an ion spatial distribution characterized by the presence of the lighter species at the plume edges, while the heavier ones are concentrated at the plume center.  相似文献

Synthetic organic pigments are widely used in modern and contemporary works of art. They have been examined by a variety of techniques including spectroscopic methods such as Fourier transform infrared spectroscopy, Raman, and X‐ray powder diffraction as well as chromatographic or mass spectrometric techniques such as pyrolysis‐gas chromatography/mass spectrometry and laser desorption ionization mass spectrometry (LDI–MS). Often, a combination of techniques has been used to examine these pigments. This paper describes use of Raman spectroscopy to create a database of colorants including two pigments not previously reported, PO1 and PO2. Then, using Raman spectroscopy in combination with LDI–MS, samples from modern works of art by artists including Mark Rothko, Barnett Newman, and José de Rivera were examined in order to identify the pigments present. One finding was that Rothko used a variety of red pigments over the course of his career including PR11 which has not been previously reported in artwork, and PO2 found with its positional isomer PR1. Knowledge of the colorants serves to inform conservators about display and treatment decisions. Published 2014. We have developed a method for the determination of melamine (MEL), ammeline (AMN), and ammelide (AMD) by surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) using gold nanoparticles (Au NPs). The major peaks for MEL, AMN, and AMD at m/z 127.07, 128.05, and 129.04 are assigned to the [MEL + H](+), [AMN + H](+), and [AMD + H](+) ions. Because the three tested compounds adsorb weakly onto the surfaces of the Au NPs through Au-N bonding, they can be easily concentrated from complex samples by applying a simple trapping/centrifugation process. The SALDI-MS method provides limits of detection of 5, 10, and 300 nM for MEL, AMN, and AMD, respectively, at a signal-to-noise ratio of 3. The signal variation for 150-shot average spectra of the three analytes within the same spot was 15%, and the batch-to-batch variation was 20%. We have validated the practicality of this approach by the analysis of these three analytes in infant formula and grain powder. This simple and rapid SALDI-MS approach holds great potential for screening of MEL in foods.  相似文献

We report on the formation of self-organized nanostructures (NS) on bulk Al under its ablation in air and liquids with femtoseconds (fs) laser pulses. In case of exposure into liquids, NS are regularly formed on the Al surface with an average period of about 200 nm, independent of the laser polarization. A dispersion of Al nanoparticles (NPs) into the liquid additionally occurs. Irregular nano-bumps are produced when the irradiation is performed in air. NP dispersions as well as NS formed on Al surface show a characteristic absorption peak in the near UV which has been attributed to plasmon oscillation of electrons. The wings of this peak extending to the visible, lead to a distinct yellow coloration of the processed Al surface and the liquid dispersions. Ultrafast laser processing of bulk Al in liquids may be potentially a promising technique for efficient production of nanosized aluminum.  相似文献

Nanoparticle and metal phthalocyanine (MPc) transparent colloidal aqueous solutions were directly obtained by 355 nm YAG laser ablation. We found that too long an irradiation time does not contribute to producing nanoparticles and their generation efficiency increases with a low solution temperature. We believe this due to nanoparticle reassociation which is caused by hydrophobicity. To prevent generated nanoparticles from reassociating we performed experiments adding two kinds of ionic and nonionic surfactants into solution. We found five characteristics of nanoparticle generation from adding surfactants to a solution regardless of the type of surfactant used. These characteristics are that: (1) production efficiency increases; (2) stability is better after irradiation; (3) irradiation intensity needed to induce nanoparticle generation becomes lower; (4) mean size of the generated nanoparticles becomes smaller; and (5) crystalline structures of oxo(phthalocyaninato) vanadium (IV) (VOPc) are controllable by changing the surfactant concentration.  相似文献

Significant advances have been made in the past decade in the field of mass spectrometry imaging (MS imaging). It is a relatively unestablished technique aimed at direct, high-sensitive and spatially exclusive detection of biological molecules from the surface of tissue sections, so that semi-quantitative distribution map of the analyte can be reconstituted from the mass spectra obtained. There is tremendous potential in its application especially in clinical field, such as biomarker discovery or pharmacokinetic study. However, vast majority of the work has been performed on frozen tissue sections, while it remains generally unpractical to produce frozen sections with clinically resected tumor samples. Here we report our novel sample preparation technique that enabled MS imaging from formalin-fixed paraffin embedded (FFPE) tissue section, including retrospective archive as old as 11 years. FFPE sections were first dewaxed with pre-warmed xylene, and exposed tissue surface was enzymatically digested in nanoliter scale droplets to retain analyte localization. As a result, we succeeded in obtaining MS images of peptide peaks derived from several proteins, identified by MS/MS analysis, using ovarian cancer FFPE sections. The qualities of mass spectra obtained by this method were not significantly different from those obtained from frozen sections. By this, we opened the door to retrospective study of past clinical cases in aim to discover molecular biomarker.  相似文献

We performed laser ablation of a silver plate in polyvinylpyrrolidone (PVP) aqueous solutions to prepare silver nanoparticles. Secondary laser irradiation onto the prepared colloidal solutions was also carried out. It was revealed that the formation efficiency was increased by addition of PVP as well as the stability of nanoparticles. The result of shadowgraph measurements suggested that the increased ablation efficiency by PVP is attributable to increased secondary etching efficiency by the solvent-confined plasma toward the silver plate. On the other hand, the size decrease of the nanoparticles by addition of PVP was more remarkable during the secondary irradiation process than in the laser ablation (nanoparticle preparation) process. This result indicates that emitted materials interact less sufficiently with PVP molecules in the laser ablation process than in the secondary laser irradiation process.  相似文献

在自行研制的气溶胶飞行时间激光质谱仪(ATOFLMS)上实时探测单个悬浮土壤颗粒的粒径和化学成分。利用双束连续激光对单个粒子的空气动力学粒径进行测量,并用266 nm的Nd∶YAG激光器对气溶胶单粒子进行解吸附电离作用,产物离子通过飞行时间质谱仪的无场漂移区后完成单粒子化学成分的检测。本实验中使用了4个不同地区的土壤样本,在实验室内对分析的样本进行预处理和再悬浮,通过导管引入ATOFLMS进行测量,得到大量单粒子的粒径和质谱数据。发现在众多的单粒子的阳离子质谱中,金属成分以地壳元素(Fe, K, Al, Ca)为主,在其他阳离子质谱中包含了Mg和Na等。对悬浮土壤粒子的粒径进行实时检测的结果表明这些粒子多以粗粒子为主, 粒径主要集中在1~2 μm。实验结果表明该仪器在大气气溶胶环境监测及相关研究领域具有重要的实用价值。  相似文献

A polypropylene (PP) film was ablated using a femtosecond laser with a center wavelength of 785 nm, a pulse width of 184 fs and a repetition rate of 1 kHz. Increments of both the pulse energy and the shot number of pulses lead to co-occurrence of photochemical and thermal effect, demonstrated by the spatial expansion of rim on the surface of PP. The shapes of the laser-ablated PP films were imaged by a scanning electron microscope (SEM) and measured by a 3D optical measurement system (NanoFocus). And, the gas and water vapor transmission rate, mechanical properties of PP film micropatterned by fs laser pulses was characterized. Our results demonstrate that a femtosecond pulsed laser is an efficient tool for breathable packaging films in modifying the flow of air and gas, where the micropatterns are specifically tailored in size, location and number of which is easily controlled by laser processing conditions.  相似文献

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) was used for the characterization of a partially transesterified poly(beta-hydroxyalkanoate), PHA, polymer produced by the bacterial strain Alcaligenes eutrophus using saponified vegetable oils as the sole carbon sources. The transesterification was carried out separately under acidic and basic conditions to obtain PHA oligomers weighing less than 10 kDa. The intact oligomers were detected in their cationized [M + Na](+) and [M + K](+) forms by MALDI-TOFMS. A composition analysis, using the MALDI-TOF spectra, indicate that the oligomers obtained via acid catalysis were terminated with a methyl 3-hydroxybutyrate end group, and those obtained by base catalysis had a methyl crotonate (olefinic) termination. In addition to HB (hydroxy butyrate), the oligomers were found to contain a small percentage of HV (hydroxy valerate). This was independently confirmed using gas chromatography/mass spectrometry (GC/MS). In comparison, the analysis of a commercial PHA polymer, transesterified under identical conditions, only showed the presence of HB, i.e. a pure PHB homopolymer.
