In this work, a commercially available diol-silica stationary phase was converted in situ to a chiral stationary phase by dynamically coating it with hydroxypropyl-beta-cyclodextrin (HP-beta-CD). This stationary phase was shown useful for the capillary electrochromatography (CEC) separation of neutral and anionic enantiomers such as some organochlorine pesticides and dansyl amino acids, respectively. The inclusion of HP-beta-CD in the mobile phase to produce the in situ chiral stationary phase allowed the rapid separation of the anionic dansyl amino acid enantiomers at relatively low electroosmotic flow (EOF). The formation of host-guest complexes between the dansyl amino acids and the neutral HP-beta-CD in the mobile phase lowered the actual charge-to-mass ratios of the anionic solutes, thus speeding up their transport by the EOF across the packed capillary column. Several parameters affecting enantioseparation were investigated, including the concentration of HP-beta-CD, ionic strength, pH, and organic modifier content of the mobile phase.  相似文献

采用C18固定相,以羟丙基-β-环糊精为手性流动相添加剂,建立了奥昔布宁对映体的高效液相色谱拆分方法。考察了手性添加剂、有机极性调节剂、缓冲盐的种类和浓度以及流动相的pH值和流速及柱温等因素对对映体分离的影响。在最佳分离条件下,奥昔布宁对映体的分离度为1.54,检测限为1.0 ng。该方法简便,重复性好,比手性固定相法更加经济。  相似文献

This paper reviews the mixed chelation approach to resolution of the optical isomers of D and L dansyl amino acids by high performance liquid chromatography. The use of eluants containing Cu(II) complexes of L-proline, L-arginine, L-histidine, and L-histidine methyl ester effected the separation of many D and L amino acids, including those with aliphatic, polar, and aromatic substituents. The mechanism of separation, which is based on the preferential ternary complex formation of the analyte amino acid and the chiral chelate with Cu(II) in the mobile phase, is discussed. The stereoselectivity depends mainly on the different steric interactions between the alkyl side chains of the amino acid analytes and the chiral ligands coordinating around Cu(II), although such parameters as pH, temperature, organic modifier, and concentration of the chiral additive also affect the chromatographic separation. Among the chiral ligands studied, L-histidine methyl ester is unique in that it possesses both achiral selectivity for the dansyl amino acids and chiral selectivity for the respective D and L enantiomers. With a mobile phase gradient of acetonitrile in a buffer containing Cu(II) L-histidine methyl ester complex, a stereoselective procedure was devised for the analysis of D and L amino acid enantiomers, achieving the separation that the current amino acid analyzer could not perform. Finally, the use of the mixed chelation approach in two biomedical studies is described. In the first application, the histidine methyl ester gradient was adapted for analyzing amino acids in cerebrospinal fluid; in the second, an L-aspartame Cu(II) complex eluant was developed for measuring the urine concentration of D and L pipecolic acid (piperidine-2-carboxylic acid), a metabolite of lysine.  相似文献

A novel chiral stationary phase based on chitosan covalently bonded onto silica gels has been prepared and used for the separation of various alpha-amino acid enantiomers as well as alpha-hydroxycarboxylic acid enantiomers by chiral ligand-exchange chromatography with copper(II) as a complexing ion. The methanol content and copper(II) ion concentration in the eluent affected retentivity and enantioselectivity. Furthermore, a plausible chiral recognition mechanism for resolution of alpha-amino acids was proposed.  相似文献

A chiral silica-based stationary phase having surface-bound hydroxypropyl-beta-cyclodextrin (HP-beta-CD) with a relatively strong electroosmotic flow (EOF) was introduced for enantioseparation by capillary electrochromatography (CEC). The stationary phase contained a hydrophilic sulfonated sublayer to which a chiral top layer of HP-beta-CD was immobilized. While the sulfonated sublayer was to provide a relatively strong EOF, the top HP-beta-CD was to confer the desired chiral recognition towards enantiomeric solutes. This HP-beta-CD sulfonated silica (CDSS) stationary phase proved useful for the rapid separation of anionic enantiomers such as dansyl amino acids and phenoxy acid herbicides. The effects of the organic modifier content, pH, and ionic strength of the mobile phase on enantioseparation were investigated. Under the optimized separation conditions, ten dansyl amino acids and six phenoxy acid herbicides were enantioseparated with a resolution greater than unity.  相似文献

A cellulose tris‐(3,5‐dimethylphenylcarbamate)‐based chiral stationary phase was studied as a tool for the enantioselective separation of 21 selected analytes with different pharmaceutical and physicochemical properties. The enantioseparations were performed using supercritical fluid chromatography. The effect of the mobile phase composition was studied. Four different additives (diethylamine, triethylamine, isopropylamine, and trifluoroacetic acid) and isopropylamine combined with trifluoroacetic acid were tested and their influence on enantioseparation was compared. The influence of two different mobile phase co‐solvents (methanol and propan‐2‐ol) combined with all the additives was also evaluated. The best mobile phase compositions for the separation of the majority of enantiomers were CO2/methanol/isopropylamine 80:20:0.1 v/v/v or CO2/propan‐2‐ol/isopropylamine/trifluoroacetic acid 80:20:0.05:0.05 v/v/v/v. The best results were obtained from the group of basic β‐blockers. A high‐performance liquid chromatography separation system composed of the same stationary phase and mobile phase of similar properties prepared as a mixture of hexane/propan‐2‐ol/additive 80:20:0.1 v/v/v was considered for comparison. Supercritical fluid chromatography was found to yield better results, i.e. better enantioresolution for shorter analysis times than high‐performance liquid chromatography. However, examples of enantiomers better resolved under the optimized conditions in high‐performance liquid chromatography were also found.  相似文献

在纤维素-三(3,5-二甲基苯基氨基甲酸酯)(CDMPC)手性固定相上,分别采用正相、反相及极性有机相色谱模式对氟环唑外消旋体进行了拆分,并考察了流动相组成在手性识别中对手性分离的影响。氟环唑在Chiralcel OD-H手性色谱柱(填充CDMPC手性固定相)上采用反相色谱模式,以甲醇-水(体积比为80∶20)为流动相,获得了最佳的拆分,其两对对映异构体的分离度Rs分别为1.64和6.50。  相似文献

The influence of tetrabutylammonium (TBA) as a hydrophobic charged additive of the mobile phase was investigated in a chromatographic system involving d,l-dansyl amino acids as the test solute enantiomers and immobilized human serum albumin as the chiral stationary phase. By varying the column temperature, van't Hoff plots for solute retention and enantioselectivity were performed and thermodynamic parameters were calculated. An enthalpy-entropy compensation study revealed that the type of interaction between the analyte and stationary phase was independent of TBA concentration in the eluent. The counterion dependence on retention indicated that the coulombic interactions between dansyl amino acid and the site II binding cavity were of crucial importance in this association process. Also, the increasing variations of chiral discrimination with a TBA addition were attributed, by an analysis of the thermodynamic parameter trends, to a great facilitation of enantioselective H-bonding between solute and polar residues at the cavity rim.  相似文献

采用纤维素-三(4-甲基苯甲酸酯)(CTMB)手性固定相,利用反相色谱法研究了氟比洛芬、普拉洛芬、布洛芬、萘普生、洛索洛芬5种芳基丙酸类手性药物的色谱拆分行为。考察了流动相组成、酸碱添加剂及柱温对上述5种药物对映体分离的影响,并通过热力学研究及对映体结构分析对CTMB固定相的手性拆分机理进行了探讨。结果表明,除萘普生采用乙腈-0.1%(v/v)甲酸溶液外,以甲醇-0.1%(v/v)甲酸水溶液为流动相可使普拉洛芬、洛索洛芬、氟比洛芬和布洛芬的对映体间的分离度均大于1.5,CTMB固定相对这5种芳基丙酸类药物的手性拆分能力依次为普拉洛芬>洛索洛芬>氟比洛芬>布洛芬>萘普生。在各自的优化色谱条件下,将方法应用于上述5种药物制剂的含量测定,结果令人满意。  相似文献

In order to develop a chiral stationary phase (CSP), which has even higher separation ability than the corresponding commercially available crown ether based CSP (OA-8000 having a pseudo-18-crown-6 ether with an OMe group as a selector), chemically bonded type CSP having a phenolic OH group on a crown ring was developed. Normal mobile phases with or without acid additive can be used with this OH type CSP in contrast to the conventional OMe type CSP which has a neutral chiral selector. Enantiomers of 25 out of 27 amino compounds, including 20 amino acids, 5 amino alcohols, and 2 lipophilic amines, were efficiently separated on a column with this CSP. Nine amino compounds out of 27 were separated with better separation factors than the corresponding OMe type CSP. It is noteworthy that the chromatography on this CSP exhibited excellent enantiomer-separations for amines and amino alcohols when triethyl amine was used as an additive in the mobile phase. Comparison of enantiomer separation ability on this OH type of CSP and on the OMe type of CSP and correlation between the enantioselectivity in chiral chromatography and that of the corresponding model compounds in solution imply that the chiral separation arose from chiral recognition in host guest interactions.  相似文献

High-performance liquid chromatography (HPLC) coupled to atmospheric pressure chemical ionization (APCI) mass spectrometry was used for the separation and detection of amino acid and peptide enantiomers. With detection limits as low as 250 pg, 25 amino acids enantiomers were baseline resolved on a Chirobiotic T chiral stationary phase. APCI demonstrated an order of magnitude better sensitivity over electrospray ionization (ESI) for free amino acids and low molecular mass peptides at the high LC flow-rates necessary for rapid analysis. As the peptide chain length increased (peptides with M(r) > or = 300 Da), however, ESI proved to be the more ideal atmospheric pressure ionization source. A mobile phase consisting of 1% (w/w) ammonium trifluoroacetate in methanol and 0.1% (w/w) formic acid in water increased the sensitivity of the APCI method significantly. A step gradient was then used to separate simultaneously all 19 native protein amino acid enantiomers in less than 20 min using extracted ion chromatograms.  相似文献

以纤维素三-(3,5-二甲基苯基氨基甲酸酯)为手性固定相(Lux Cellulose-1),建立了在正相色谱条件下直接分离盐酸川丁特罗对映体的高效液相色谱法。考察了乙醇、异丙醇等有机改性剂,三氟乙酸、二乙胺等流动相添加剂和柱温对对映体分离的影响。结果显示,酸性和碱性添加剂对对映体分离的影响最为显著: 添加二乙胺时两对映体无分离趋势;添加三氟乙酸时对映体保留强,且分离趋势明显;而同时添加三氟乙酸和二乙胺则两对映体分离显著改善,分离度可达4.0。优化后的色谱条件: 色谱柱为Lux Cellulose-1手性柱(250 mm×4.6 mm, 5 μm),流动相为正庚烷-乙醇-三氟乙酸-二乙胺(88:12:0.3:0.05, v/v/v/v),流速为1.0 mL/min,紫外检测波长为246 nm,柱温为25 ℃。该方法简便,快速,可用于左旋盐酸川丁特罗原料中右旋异构体杂质的检查。  相似文献

Ligand-exchange micellar electrokinetic capillary chromatography was used for the chiral resolution of underivatized and dansyl amino acid enantiomers simultaneously. The separation was achieved by chiral copper(II)-L-valine complexes incorporated in micelles of sodium dodecyl sulfate (SDS). The enantioresolution was strongly affected by SDS and a concentration of 20 mM SDS was shown to be necessary for the separation. Other impacting factors were investigated including pH, the molar ratio of copper(II) to L-valine and the total concentration of complex. Using the proposed method, 11 different dansyl amino acids and two underivatized amino acids were separated successfully with a running electrolyte of 20 mM NH4OAc, 4 mM CuSO4, 8 mM L-valine and 20 mM SDS at pH 9.0 in less than 25 min. Experiments were also performed with other amino acid ligands in order to vary the stability and the sterical arrangement of the copper(II) complexes and the possible chiral recognition mechanism was also discussed briefly.  相似文献

合成了一种新型奎宁-冠醚组合型手性固定相(QN-CR CSP)并用于氨基酸手性对映体的直接拆分,该固定相对12种氨基酸对映体有良好的手性拆分能力。基于氨基酸手性识别中离子交换和络合的协同作用,建立了一种新型的等温吸附模型。通过迎头特殊点洗脱法(FACP)测定色氨酸(Trp)在不同金属离子添加剂条件下的等温吸附线,验证了模型的合理性。流动相中的Li+、Na+、K+等金属离子与氨基酸竞争固定相中的冠醚络合位点,随着金属离子与冠醚的络合作用力和络合吸附平衡常数增大,固定相对Trp的手性拆分能力下降。该模型的建立对理解氨基酸在此类固定相中的手性保留行为以及固定相结构的进一步优化具有重要意义。  相似文献

建立了新型抗抑郁药米那普仑在环糊精手性固定相上的高效液相色谱拆分方法。在反相色谱条件下采用未衍生化β-环糊精(Cyclobond I 2000)、乙酰基-β-环糊精(AC-β-CD)、2,3-二甲基-β-环糊精(DM-β-CD)、3,5-二甲基苯基氨基甲酸酯-β-环糊精(DMP-β-CD)4种手性柱分离米那普仑对映体。考察了固定相、流动相比例、pH、流速和柱温对拆分的影响。利用分子对接和结合能计算方法,研究米那普仑分子与AC-β-CD的对接过程,探讨其可能的分离机制。优化后的拆分条件如下:固定相为乙酰基-β-环糊精手性柱Astec CYCLOBONDTMI 2000 AC(25 cm×4.6 mm,5μm),流动相为乙腈-0.1%(体积分数)pH 5.0醋酸三乙胺溶液(TEAA)(5∶95,v/v),流速为0.4mL/min,柱温为25℃,检测波长为220 nm。在此条件下,米那普仑对映体获得快速拆分,分离度(Rs)为1.74,理论塔板数为10 125。分子模拟结果表明引起手性识别的作用力主要是环糊精衍生化的乙酰基导致的氢键作用差异。该方法快速、高效、重现性好。  相似文献

A new chiral stationary phase (CSP) was prepared by reacting MDL 63,246 (Hepta-Tyr), a glycopeptide antibiotic belonging to the teicoplanin family, with 5-μm diol-silica particles. The CSP mixed with 5-μm amino silica particles (3:1) was packed into 75-μm fused-silica capillaries for only 6.6 cm and used for electrochromatographic experiments analyzing several hydroxy acid enantiomers. A reversed electroosmotic flow carried both analytes and mobile phase towards the anode in a short time (1–3 min), being baseline resolved all the studied analytes. In order to achieve the fastest enantiomeric resolution of the studied hydroxy acids, the effect of several experimental parameters such as mobile phase composition (organic modifier type and concentration, pH of the buffer and ionic strength), capillary temperature and applied voltage on enantioresolution factor, retention time, enantioselectivity were evaluated. The packed capillary column allowed the separation of mandelic acid enantiomers in less than 72 s with resolution factor Rs=2.18 applying a voltage of 30 kV and eluting with a mobile phase composed by 50 mM ammonium acetate (pH 6)–water–acetonitrile (1:4:5, v/v). The CSP was also tested in the capillary liquid chromatography mode resolving all the studied enantiomers applying 12 bar pressure to the mobile phase [50 mM ammonium acetate (pH 6)–water–methanol–acetonitrile, 1:4:2:3, v/v)], however, relatively long analysis times were observed (12–20 min).  相似文献

合成了三-(4-甲基苯甲酸)纤维素酯(MCTB)手性固定相,用反相高效液相色谱法在该手性固定相上对2-(4-羟基苯氧基)丙酸酯对映体进行拆分.实验结果表明,在三-(4-甲基苯甲酸)纤维素酯手性固定相上,以甲醇与水的体积分数为75:25做流动相能较好的拆分2-(4-羟基苯氧基)丙酸甲酯、乙酯和丁酯对映体,其分离因子分别为1.38、1.49、0.98;同时还发现2-(4-羟基苯氧基)丙酸酯中酯基团的大小对其对映体的分离也有明显的影响,其中以乙酯的拆分效果最佳。  相似文献

Unique stationary phases of octadecylsilica (ODS) coated with acylcarnitines have been developed for liquid chromatographic columns. The ODS column coated with acylcarnitine was readily prepared by recycling the solution containing acylcarnitine through an ODS column in a closed loop. Acylcarnitine was adsorbed on the ODS surfaces by hydrophobic interaction between the acyl group of acylcarnitine and the octadecyl group of the ODS phases. The ODS column coated with stearoylcarnitine (CN-18 column) was the most stable among the four columns coated with acylcarnitines of various acyl chain lengths (decanoylcarnitine, lauroylcarnitine, myristoylcarnitine, and stearoylcarnitine) under the condition of delivery of the mobile phase, indicating that adsorption of acylcarnitine on the ODS surfaces depended on the length of acyl chains. The CN-18 column was usable for delivering the mobile phase contained less than 20% (v/v) acetonitrile, retaining almost the same separation efficiency as the intact ODS column. The retention behavior of ionic solutes on the CN-18 column could be explained by both ionic and electrostatic interactions between the solutes and the stationary phase. The CN-18 column enabled efficient separation of inorganic anions, nicotinic acids, amino acids, and nucleotides. The chiral ODS column coated with enantiomer of stearoylcarnitine, L-stearoylcarnitine (L-CN-18 column) could achieve direct enantiomeric separation of DL-tryptophan, alpha-methyl-DL-tryptophan and DL-3-indolelactic acid using 100% water as the mobile phase. The L-CN-18 column could also separate enantiomers of amino acids and alpha-hydroxycarboxylic acids by ligand-exchange chromatographic mode using a mobile phase containing copper(II) ion. The chiral recognition is discussed for enantiomeric separation on the L-CN-18 column.  相似文献

Thirteen pairs of enantiomers belonging to the same structural family (phenylthiohydantoin‐amino acids) were analyzed on two polysaccharide chiral stationary phases, namely, tris‐(3,5‐dimethylphenylcarbamate) of amylose (Chiralpak AD‐H) or cellulose (Chiralcel OD‐H) in supercritical fluid chromatography with a carbon dioxide/methanol mobile phase (90:10 v/v). Five different temperatures (5, 10, 20, 30, 40°C) were applied to evaluate the thermodynamic behavior of these enantioseparations. On the cellulose stationary phase, the retention, and separation trends were most similar among the set of probe analytes, suggesting that the chiral cavities in this stationary phase have little diversity, or that all analytes accessed the same cavities. Conversely, the retention and separation trends on the amylose phase were much more diverse, and could be related to structural differences among the set of probe analytes (carbon chain length in the amino acid residue, secondary amine in proline, existence of covalent rings, or formation of pseudo‐rings via intramolecular hydrogen bonds). The large variability of behaviors on the amylose phase suggests that the chiral‐binding sites in this chiral stationary phase have more variety than on the cellulose phase, and that the analytes did access different cavities.  相似文献

采用直链淀粉手性固定相高效液相色谱法在正相条件下直接拆分了比索洛尔对映异构体。分别以异丙醇、乙醇为有机改性剂,考察了流动相的组成与配比、流速及柱温等因素对比索洛尔对映体分离的影响。确定了比索洛尔对映体的最佳拆分条件:流动相正己烷-乙醇-二乙胺(体积比为88∶12∶0.1),流速0.6 mL/min,检测波长270 nm,柱温20 ℃。该方法可快捷、简便地拆分比索洛尔对映体。  相似文献
