Aryllithiums prepared by bromine-lithium interchange in chiral 2-(o-bromophenyl)-substituted perhydro-1,3-benzoxazines participate in the intramolecular 6-exo carbolithiation reaction with unactivated double bonds attached to the nitrogen substituent of the heterocycle. When the reaction time is extended or no TMEDA is used, the cyclized lithium intermediates react intramolecularly with the N,O-acetal system leading to 2-azabenzonorbornane derivatives. The reactions are highly stereoselective and constitute a high-yielding synthesis of enantiopure 4-substituted tetrahydroisoquinolines or 7-substituted 2-azabenzonorbornanes.  相似文献

Atom transfer radical cyclization reactions of N-(4-pentenyl)iodoacetamides were investigated. The reactions were efficiently promoted by BF3.OEt2. For N-alkenyl-substituted iodoamides, excellent regioselectivity in favor of 8-endo cyclization was observed, while both 7-exo and 8-endo cyclization products were formed with the 8-endo cyclization preferred in the cases of N-(2-allylphenyl)-substituted iodoamides. Density functional theory calculations at the B3LYP/6-31G level revealed that both the s-trans and the s-cis conformational transition structures were feasible for the 8-endo cyclization of N-alkenyl-substituted alpha-carbamoyl radicals while 7-exo transition structures were much less stable. For the cyclization of N-(2-allylphenyl)-substituted alpha-carbamoyl radicals, the transition structures for 8-endo and 7-exo cyclizations were of comparable energy. These results were in excellent agreement with the experimental observations.  相似文献

Reaction of 2-methyl-endo-tricyclo[3.2.1.0(2,4)]octane (1) with bromine in CCl(4) gave 2-exo,3-endo-dibromo-2-endo-methylbicyclo[3.2.1]octane (3) which rearranges on silica to 2-exo,6-endo-dibromo-1-methylbicyclo[2.2.2]octane (4). Reaction in methanol gave 4-endo-bromo-2-exo-methoxy-2-endo-methylbicyclo[3.2.1]octane (10) and 4-endo-bromo-2-endo-methoxy-2-exo-methylbicyclo[3.2.1]octane (11) formed by corner attack of the bromine electrophile with C2-C4 bond rupture and inversion of configuration at the site of electrophilic attack. Reaction of 2-methyl-endo-tricyclo[3.2.1.0(2,4)]oct-6-ene (2) with bromine in CCl(4) and methanol gave products of reaction at the alkene site.  相似文献

A general procedure is described for converting primary amines to N-substituted piperazines. Reaction of an amine with an N-substituted iminodiacetic acid anhydride (V) yields an iminodiacetic acid monoamide (VI) which closes to a 2,6-piperazinedione (VII) upon treatment with acetic anhydride. The diones are reduced to piperazines with borane-THF. Fourteen examples of this process, using twelve aliphatic or aromatic amines and three iminodiacetic acids, are presented. Yields of piperazines, based upon starting amine, ranged from 21 to 52%. The procedure is rapid and no purification problems were encountered. Alternate methods for preparing the 2,6-piperazinedione intermediates are discussed.  相似文献

Two stereocenters are created by the reaction, and a single isomeric product was obtained from each of the five substrates examined. (E) and (E,E)-conjugated dienoates and dienamides of high stereoisomeric purities are prepared via thermal SO2 extrusion from cis-2,5-disubstituted-2,5-dihydrothiofene-1,1-dioxides generated by a retro Diels-Alder reaction. Applications of this method to the syntheses of two insecticidal natural dienamides : pellitorine and pipercide and of methyl tetradeca-2E,4,5-trienoate, an insect sex pheromone, are described.  相似文献

An efficient synthesis of 2-substituted (+/-)-(2R,3R,5R)-tetrahydrofuran-3,5-dicarboxylic acid derivatives has been developed. Starting from 5-norborne-2-ol, the key intermediate (+/-)-methyl 5,6-exo,exo-(isopropylidenedioxy)-2-oxabicyclo[2.2.1]heptane-3-exo-carboxylate (15) was synthesized in an efficient six-step sequence. The key transformation is the base-catalyzed methanolysis-rearrangement of (+/-)-6,7-exo,exo-(isopropylidenedioxy)-4-exo-iodo-2-oxabicyclo[3.2.1]octan-3-one (14). Further manipulation of the 3-substituent of (+/-)-methyl 5,6-exo,exo-(isopropylidenedioxy)-2-oxabicyclo[2.2.1]heptane-3-exo-carboxylate (15) followed by deprotection of the diol moiety and ring opening catalyzed by RuCl(3)/NaIO(4) gave the title compounds in good yield.  相似文献

The Bu(3)Sn radical-induced reaction of o-iodobenzylvinylsilanes and o-iodoallylsilanes leads to cyclic products in yields of 40-60%. These regioselective cyclizations occur with high preference for a 5-exo and a 7-endo mode with a 6-exo mode being absent. An example for ring closure via a 7-exo mode is described.  相似文献

Stereodefined multisubstituted dienamides could be concisely prepared in high yields by direct addition of 1-lithiobutadiene derivatives to both N-aryl and N-alkyl isocyanates. Electrophilic cyclization of these dienamides was achieved to generate substituted cyclic iminoethers in excellent yields with perfect selectivity. When treated with 12 N aqueous HCl, dienamides underwent efficient and selective electrophilic cyclization to afford cyclic imidate derivatives. When treated with NBS, monobrominated or double-brominated cyclic iminoethers were formed.  相似文献

Calculations of the activation energy of cyclization of alpha-sulfenyl-, alpha-sulfinyl-, and alpha-sulfonyl-5-hexenyl radicals and their respective 5-methyl-5-hexenyl counterparts at the G3MP2B3 level agree quite well with experimental data. The alpha-sulfinyl-5-hexenyl radical exhibits unexpected regioselectivity (93.99:6.01) via the 5-exo mode, whereas the alpha-sulfenyl- and alpha-sulfonyl-5-hexenyl radicals show increasing branching ratios of the 6-endo product. In contrast, the cyclization of the alpha-sulfur-based 5-methyl-substituted counterparts yields essentially the 6-endo products in all cases; in particular, the alpha-SO2-5-CH3-5-hexenyl radical gives high regioselectivity (98.85:1.15) via the 6-endo mode. Several other 5-substituted moieties, including the electron-withdrawing (CN and NO2) or electron-donating substituents (NH2), also proceed preferentially to 6-endo closure. The alpha-sulfonyl-5-amine-5-hexenyl radical is calculated to proceed to exclusively the 6-endo product, a demonstration of the high synthetic value of this reactant.  相似文献

A facile solid phase synthesis of 2-substituted 1,3-oxazin-6-ones using polymer-supported Meldrum‘s acid has been reported. Reaction of the resin-bound cyclic malonic acid ester with triethyl orthoformate and subsequent double substitution with amide, afforded the corresponding polymer-supported acylaminomethylene cyclic malonic acid ester, which upon thermal treatment led to 1, 3-oxazin-6-ones in good yields and with high purity.  相似文献

Reaction of (R)-phenylglycinol with methyl 5-oxopentanoate gave either bicyclic lactam cis-1 (the kinetic product) or its isomer trans-1 (under equilibrating conditions) as the major products, which were converted to the corresponding (cis or trans) unsaturated lactams 4 and 5. On treatment with lithium alkyl (or aryl) cyanocuprates, these chiral building blocks undergo conjugate addition to give enantiopure trans-3,4-substituted 2-piperidone derivatives in high yield and stereoselectivity. The synthetic potential of this transformation is illustrated by the synthesis of (+)-femoxetine and the two enantiomers of the known antidepressant paroxetine.  相似文献

An efficient method for the preparation of polymer-bound cyclic malonic acid ester starting from Merrifield resin has been developed. Reaction of the resin-bound cyclic malonic acid ester with triethyl orthoformate and subsequent double substitution with nucleophilic reagents, such as arylamine, urea, thiourea, 2-aminobenzothiazoles, or isothiosemicarbazones, afforded the corresponding polymer-bound substituted aminomethylene cyclic malonic acid esters, which upon thermal treament led to 4(1H)-quinolones, 3-substituted uracils and thiouracils, 4H-pyrimido[2,1-b]benzothiazol-4-ones, and 1-(N-alkylidene or benzylideneamino)-1,6-dihydro-2-methylthio-6-oxo-pyrimidines, depending on the structures of the nucleophilic reagents.  相似文献

Reaction of ethyl or methyl 3-oxoalkanoates with N,N-dimethylformamide dimethyl acetal gave, generally in excellent yields, a series of ethyl or methyl 2-dimethylaminomethylene-3-oxoalkanoates II which reacted with phenylhydrazine to afford the esters of 5-substituted 1-phenyl-1H-pyrazole-4-carboxylic acids III in high yields. Esters III were hydrolyzed to the relative 5-substituted 1-phenyl-1H-pyrazole-4-carboxylic acids which were converted by heating to 5-substituted 1-phenyl-1H-pyrazoles in excellent yields. Reaction of II with methylhydrazine afforded in general a mixture of 3- and 5-substituted ethyl 1-methyl-1H-pyrazole-4-carboxylates with the exception of IIg , which gave in high yield methyl 5-benzyl-1-methyl-1H-pyrazole-4-carboxylate, which was hydrolyzed to the relative pyrazolecarboxylic acid. This afforded by heating 5-benzyl-1-methyl-1H-pyrazole in quantitative yield.  相似文献

Staudinger/aza-Wittig reaction of 6,6,6-trifluoro-5-oxohexanoyl azide with PPh(3) or PBu(3) was examined. A reactive intermediate acyl imine 1 was trapped by methanol. Without nucleophile, isomerized enamide form 3 was obtained. N-Iodobenzoylation and N-haloalkylation of 3 and following radical cyclization via the 5-exo or 6-exo mode gave benzoindolizidinone, indolizidinone, and quinolizidinone derivatives 10-14, which have a trifluoromethyl group at the bridgehead position adjacent to nitrogen. Although LiAlH(4) reduction of 10 and 11 gave a mixture of saturated benzoindolizidine 15 and amino alcohol 16, reduction with BH(3)/THF selectively gave the desired 15 and indolizidine 17 from 13.  相似文献

Reaction of 5-substituted 2-thioxo-1,3-thiaselenoles with ethyl diazoacetate, phenyl azide and ethyl azidoformate afforded 2-substituted ω-carbethoxy-1,4-thiaselenafulvenes (II) , 5-substituted 2-phenylimino-1,3-thiaselenoles (IV) and 5-substituted 2-carbethoxyimino-1,3-thiaselenoles (V) , respectively.
