An efficient synthesis of bioactive chiral flavanones (1) was achieved through the Rh-catalyzed asymmetric 1,4-addition of arylboronic acid to chromone. The reaction in toluene proceeded smoothly at room temperature in the presence of 0.5% Rh catalyst with electron-poor chiral diphosphine MeO-F(12)-BIPHEP. In this reaction, the 1,2-addition to (S)-1 frequently occurred to yield (2S,4R)-2,4-diaryl-4-chromanol as a byproduct, which could be reduced by changing the reaction solvent to CH(2)Cl(2) to deactivate the Rh catalyst (3% required).  相似文献

Six new chromones, 6-methoxy-2-[2-(3-methoxy-4-hydroxyphenyl)ethyllchromone (2), 6,8-dihydroxy-2-(2-phenylethyl)chromone (3), 6-hydroxy-2-[2-(4-hydroxyphenyl)ethyl]chromone (4), 6-hydroxy-2-[2-(2-hydroxyphenyl)ethyl]chromone (5), 7-hydroxy-2-(2-phenylethyl)chromone (6), and 6-hydroxy-7-methoxy-2-(2-phenylethyl)chromone (7) were isolated from the ether extract of agarwood in addition to a known compound, 2-(2-phenylethyl)chromone or flidersiachromone (1). Their structures were determined by spectroscopic methods including UV, IR, and NMR spectral data and comparisons with the calculated values using the hydroxyl and methoxyl substituent increments of the chromone ring.  相似文献

芦荟色酮是在芦荟叶中特有的一类具有抗炎和抑制酪氨酸酶等作用的活性物质。以芦荟全叶为原料,经过一系列的预处理手段,从脱色剂活性炭中获得芦荟色酮粗提物,再经过溶剂分配和富集后采用高速逆流色谱(HSCCC)对其中的色酮成分进行分离纯化。研究结果表明,采用氯仿-甲醇-水(体积比为4∶3∶2)混合溶液和二氯甲烷-甲醇-水(体积比为5∶4∶2)混合溶液作为溶剂分离系统,经过两步HSCCC可以分离纯化出色谱纯度在95%以上的芦荟色酮单体。经过紫外检测、快原子轰击质谱及核磁共振等方法的结构分析鉴定,证实分离所得物质为肉桂酰基-C-葡萄糖甙芦荟色酮。  相似文献

Another aspect concerning chromone chemistry leading to the one-pot synthesis of functionalized novel spirobenzofuranones has been described. The synthesis involves reaction of the zwitterionic intermediates formed by the 1:1 interaction between isocyanides and acetylenecarboxylates with 3-cyanochromones, whereupon through an unexpected and unprecedented reaction of the chromone moiety the isolated benzofuranones are formed. The regioselectivity of the reaction was investigated by DFT calculations. The geometries of the intermediates, transition structures, and intermediate products, leading to the final products, were optimized using the B3LYP functional with the 6-31G(d) basis set. The structures of the products were elucidated by 1D and 2D NMR experiments. Full assignment of all (1)H and (13)C NMR chemical shifts has been achieved. A plausible mechanistic rationale is proposed.  相似文献

A new synthesis of chromones and flavones based on the reaction of 2-hydroxy-1-acetonaphthone (1) with cinammonitriles (2 a-h) is described. Structures of the compounds are established by chemical and spectral data. This synthetic approach appears general in its applicability. It has been applied to the synthesis of a series of polycyclic chromone and flavone compounds containing the naphthalene and pyrene ring systems that hold promise as agents for the chemo preventation of cancer.  相似文献

The synthesis of some new 1,3,2-oxazaphosphinine 18, 1,3,2-diazaphosphinine 19, acyclic, and cyclic α-aminophosphonate derivatives 3–17 containing the chromone moiety have been achieved via reaction of 3-(phenyliminomethyl)chromone (1), 3-(phenylaminomethylene)-2-hydroxychromanone (4), and/or 3-(phenylamino-methylene)-2-(phenylamino)chromanone (5) with diethyl phosphite, tris(2-chloroethyl)phosphite, and phenylphosphonic dichloride. Structures of the products were verified on the basis of their elemental analyses, IR, 1H, and 31P NMR spectral data.  相似文献

The chromone epoxide ring system has been synthesised. Base-catalysed cyclization and dehydrobromination of -bromo-o-acyl (aroyl) oxyacetopheones or 2-bromo-1, 3-diones yielded 3-substituted chromone epoxides. Acid-catalysed rearrangement of a 2-methylchromone epoxide in an aprotic solvent gave a 2-methylenech-romanonol, while a 3-methoxymethylchromone epoxide fragmented to chromonol under these conditions; in alchols, chromone epoxides gave 2-alkoxychromanonols.  相似文献

Four new chromone derivatives, 5-hydroxy-6-methoxy-2-(2-phenylethyl)chromone (1), 6-hydroxy-2-(2-hydroxy-2-phenylethyl)chromone (2), 8-chloro-2-(2-phenylethyl)-5,6,7-trihydroxy-5,6,7,8-tetrahydrochromone (3), 6,7-dihydroxy-2-(2-phenylethyl)-5,6,7,8-tetrahydrochromone (4) were isolated from the MeOH extract of withered wood of Aquilaria sinensis, together with seven known constituents of agarwood.  相似文献

The chromone epoxide ring system has been synthesised. Base-catalysed cyclization and dehydrobromination of α-bromo-o-acyl (aroyl) oxyacetopheones or 2-bromo-1, 3-diones yielded 3-substituted chromone epoxides. Acid-catalysed rearrangement of a 2-methylchromone epoxide in an aprotic solvent gave a 2-methylenech-romanonol, while a 3-methoxymethylchromone epoxide fragmented to chromonol under these conditions; in alchols, chromone epoxides gave 2-alkoxychromanonols.  相似文献

Currently, aloesin is used in the cosmetic industry as a whitening agent because it inhibits tyrosinase activity. Aloesin is a C-glycosylated chromone compound isolated from aloe, and it is difficult to synthesize because of C-glycosyl moiety in the molecule. The purpose of this study is to search for a new chromone compound which is easy to synthesize and which posesses stronger tyrosinase inhibitory activity than aloesin. Fourteen chromone derivatives were synthesized and screened for their mushroom-tyrosinase inhibitory activity. 5-Methyl-7-methoxy-2-(2'-benzyl-3'-oxobutyl)chromone (15) showed the strongest activity among tested compounds. Its activity was not only stronger than aloesin, but also stronger than arbutin and kojic acid. The kinetic analysis revealed a competitive inhibition of 15 with tyrosinase for the L-tyrosine binding site.  相似文献

The first fluorous variants of the Boc (tert-butyloxycarbonyl) group have been prepared and tested for their suitability as nitrogen protecting groups. A group with two fluorous chains and an ethylene spacer, (RfCH2CH2)2(CH3)COC(O)-, was readily attached to a representative amine but was difficult to cleave. In contrast, groups with two fluorous chains and a propylene spacer, (RfCH2CH2CH2)2(CH3)COC(O)-, or one fluorous chain and an ethylene spacer, (RfCH2CH2)(CH3)2COC(O)-, were readily formed and cleaved. The fluorous alcohol component of the (F)Boc group can be removed by evaporation and can be recovered and reused. The utility of the new (F)Boc group (C8F17CH2CH2)(CH3)2COC(O)- was demonstrated in 16 and 96 compound library synthesis exercises. Separations can be achieved either by manual, parallel fluorous solid-phase extraction, or automated, serial fluorous chromatography. The results provide additional confirmation of the value of "light" fluorous synthesis techniques, and the new fluorous Boc groups expand the applicability of fluorous synthesis techniques to many classes of nitrogen-containing organic compounds.  相似文献

A new Chromone, 7,8-dimethoxy-2-[2-(3'-acetoxyphenyl)ethyl]chromone (1) was isolated from an acetone extract of the Cambodian agarwood along with two known chromones, 6-methoxy-2-(2-phenylethyl)chromone (2) and 6,7-dimethoxy-2-(2-phenylethyl)chromone (3). In addition, an abietane ester (4) and the sesquiterpene dehydrofukinone (5) were isolated from the agarwood oil of the same origin. Structural elucidation of all isolated compounds was made based on IR, 1H and 13C NMR spectroscopic data.  相似文献

Chromone and coumarin derivatives exhibit a wide spectrum of biological activity, including spasmolytic, antiarrhythmic, cardiothonic, antiviral, and anticancer properties. Phosphorus-containing chromone and coumarin derivatives form a novel group of compounds, possessing remarkable cytotoxicity and alkylating and anticancer activity against selected tumor cell lines. Derivatives containing a phosphorus atom at position 2 of a γ-pyrone ring are known to be efficient antibacterial agents.

This review presents methods developed for the synthesis of derivatives of chromone and coumarin that contain a phosphonate moiety. Among them, the reaction of derivatives of 2-hydroxyacetophenone with phosphonic compounds is the one most frequently used. Some analogues were characterized by X-ray crystallography.  相似文献

甲硫醇(CH_3SH)是医药、农药、饲料和合成材料等领域的重要有机合成中间体,以硫化氢合成气(CO/H_2/H_2S)一步催化合成CH_3SH具有广泛的应用前景.目前,钾改性的二硫化钼材料是一步法合成CH_3SH最高效和广泛的催化剂,但是对催化剂活性相以及K和MoS_2之间物理化学行为认知的不足严重限制了其催化性能.我们主要综述了钾钼基催化剂上3类活性相(MoS_2相、 K-MoS_2相和K_xMoS_2相)在一步法合成CH_3SH过程的研究现状,简要介绍了可控合成3类活性相的关键因素,重点关注了K对MoS_2相的调控作用及其对合成CH_3SH反应机理的影响,并对钾钼基催化剂一步法合成CH_3SH的未来发展进行了展望.  相似文献

Furo[2, 3-b]chromone ( 1a ) was conveniently prepared by the PPE-promoted cyclization of 5-(2′-carboxy-phenoxy)furan-2-carboxylic acid ( 8 ) which was made from methyl salicylate and ethyl 5-nitrofuran-2-carboxyl-ate. Furo[2, 3-b]chromon-2-acrylic acid ( 13 ) was obtained by a similar cyclization. Direct acylation of 1a afforded 2-acetylfuro[2, 3-b]chromone ( 10 ). Several derivatives of furo[2, 3-b]chromone were found to display activity in the rat passive cutaneous anaphylaxis (PGA) test for antiallergic activity.  相似文献

In an attempt to find new antitumor agents, a novel class of chromone compounds with a benzimidazoleor a benzoxazole ring in positions 2 or 6 were synthesized νia condensation in polyphosphoric acid(PPA) byusing chromone acids as the starting materials. During the preparation process, it was found that PPA couldcleave the chromone ring to produce a ring-opening compound(6). The molar ratio of the chromone com-pound (5) to the ring-opening compound (6) varied with the change of reaction temperature and time. Basedon MTT protocol, the antitumor activity of each of the compounds obtained was evaluated against three hu-man cancer cell lines: KB(oral epidermal), A2780(ovary) and Be17402 (liver). The IC50 varied from 54.7μmol/L to more than 180 μmol/L.  相似文献

We present a robust protocol based on iterations of free energy perturbation (FEP) calculations, chemical synthesis, biophysical mapping and X‐ray crystallography to reveal the binding mode of an antagonist series to the A2A adenosine receptor (AR). Eight A2AAR binding site mutations from biophysical mapping experiments were initially analyzed with sidechain FEP simulations, performed on alternate binding modes. The results distinctively supported one binding mode, which was subsequently used to design new chromone derivatives. Their affinities for the A2AAR were experimentally determined and investigated through a cycle of ligand‐FEP calculations, validating the binding orientation of the different chemical substituents proposed. Subsequent X‐ray crystallography of the A2AAR with a low and a high affinity chromone derivative confirmed the predicted binding orientation. The new molecules and structures here reported were driven by free energy calculations, and provide new insights on antagonist binding to the A2AAR, an emerging target in immuno‐oncology.  相似文献
