We report a cyclic sample pooling technique devised in two‐dimensional liquid chromatography–electrospray ionization mass spectrometry (LC‐ESI‐MS) shotgun proteomics that renders deeper proteome coverage; we combined low pH reversed‐phase (RP) LC in trifluoroacetic acid in the first dimension, followed by cyclic sample pooling of the eluate and low‐pH RP‐LC in formic acid in the second dimension. Liquid‐crystal (LC) droplet patterns are formed on a glass slide by evaporating a solution of nematic LC dissolved in heptane. In the presence of an anionic phospholipid, 1,2‐dioleoyl‐sn‐glycero‐3‐phospho‐rac‐(1‐glycerol) (DOPG), the LCs display a dark cross pattern, indicating a homeotropic orientation. When LC patterns are incubated with an aqueous mixture of DOPG and poly‐L ‐lysine (PLL), there is a transition in the LC pattern from a dark cross to a bright fan shape due to the electrostatic interaction between DOPG and PLL. Known to catalyze the hydrolysis of PLL into oligopeptide fragments, trypsin is preincubated with PLL, significantly decreasing the interactions between PLL and DOPG. LCs adopt a perpendicular orientation at the water–LC droplet interface, which gives rise to a dark cross pattern. This optical response of LC droplets is the basis for a quick and sensitive biosensor for trypsin.  相似文献

Atenolol, nadolol, metoprolol, bisoprolol and betaxolol were simultaneously determined in groundwater samples by large‐volume injection coupled‐column reversed‐phase liquid chromatography with fluorescence detection (LVI‐LC‐LC‐FD) and liquid chromatography‐time‐of‐flight mass spectrometry (LC‐TOF‐MS). The LVI‐LC‐LC‐FD method combines analyte isolation, preconcentration and determination into a single step. Significant reductions in costs for sample pre‐treatment (solvent and solid phases for clean up) and method development times are also achieved. Using LC‐TOF‐MS, accurate mass measurements within 3 ppm error were obtained for all of the β‐blockers studied. Empirical formula information can be obtained by this method, allowing the unequivocal identification of the target compounds in the samples. To increase the sensitivity, a solid‐phase extraction step with Oasis MCX cartridge was carried out yielding recoveries of 79–114% (n=5) with RSD 2–7% for the LC‐TOF‐MS method. SPE gives a high purification of β‐blockers compared with the existing methods. A 100% methanol wash was allowed for these compounds with no loss of analytes. Limit of quantification was 1–7 ng/L for LVI‐LC‐LC‐FD and 0.25–5 ng/L for LC‐TOF‐MS. As a result of selective extraction and effective removal of coextractives, no matrix effect was observed in LVI‐LC‐LC‐FD and LC‐TOF‐MS analyses. Three kinds of chiral saccharide‐containing liquid crystalline (LC) acetylenic monomers were prepared by click reaction between 2‐azidoethyl‐2,3,4,6‐tetraacetyl‐β‐D ‐galactopyranoside and 1‐biphenylacetylene 4‐alkynyloxybenzoate. The obtained monomers were polymerized by WCl6‐Ph4Sn to form three side‐chain LC polyacetylenes containing 1‐[2‐(2,3,4,6‐tetraacetyl‐β‐D ‐galactopyranos‐1‐yl)‐ethyl]‐1H‐[1,2,3]‐triazol‐4′‐biphenyl 4‐alkynyloxybenzoate side groups. All monomers and polymers show a chiral smectic A phase. Self‐assembled hiearchical superstructures of the chiral saccharide‐containing LCs and LCPs in solution state were studied by field‐emission scanning electron microscopy. Because of the LC behavior, the LC molecules exhibit a high segregation strength for phase separation in dilute solution (THF/H2O = 1:9 v/v). The self‐assembled morphology of LC monomers was dependent upon the alkynyloxy chain length. Increasing the alkynyloxy chain length caused the self‐assembled morphology to change from a platelet‐like texture ( LC‐6 ) to helical twists morphology ( LC‐11 and LC‐12 ). Furthermore, the helical twist morphological structure can be aligned on the polyimide rubbed glass substrate to form two‐dimensional ordered helical patterns. In contrast to LC monomers, the LCP‐11 self‐assembled into much more complicate morphologies, including nanospheres and helical nanofibers. These nanofibers are evolved from the helical cables ornamented with entwining nanofibers upon natural evaporation of the solution in a mixture with a THF/methanol ratio of 3:7. © 2009 Wiley Periodicals, Inc. The preparation of two liquid crystals composed of a redox‐active tetraazanaphthacene (TANC) framework is reported. The materials form smectic A (SmA) thin‐film liquid‐crystalline (LC) phases over a wide temperature range. Cyclic voltammetry analysis revealed that LC TANCs behave as organic electron acceptors. The electron mobilities of the thin films were determined by time‐ of‐flight (TOF) measurements, which are the order of 10?4 cm2 V?1 s?1 in the SmA LC phase. This value is two orders of magnitude larger than those of amorphous organic semiconductors. To the best of our knowledge, very few reports exist on the electron‐transporting behaviors of LC N‐heteroacene semiconductors.  相似文献

Metabolism studies play an important role at various stages of drug discovery and development. Liquid chromatography combined with mass spectrometry (LC/MS) has become a most powerful and widely used analytical tool for identifying drug metabolites. The suitability of different types of mass spectrometers for metabolite profiling differs widely, and therefore, the data quality and reliability of the results also depend on which instrumentation is used. As one of the latest LC/MS instrumentation designs, hybrid ion trap/time‐of‐flight MS coupled with LC (LC‐IT‐TOF‐MS) has successfully integrated ease of operation, compatibility with LC flow rates and data‐dependent MSn with high mass accuracy and mass resolving power. The MSn and accurate mass capabilities are routinely utilized to rapidly confirm the identification of expected metabolites or to elucidate the structures of uncommon or unexpected metabolites. These features make the LC‐IT‐TOF‐MS a very powerful analytical tool for metabolite identification. This paper begins with a brief introduction to some basic principles and main properties of a hybrid IT‐TOF instrument. Then, a general workflow for metabolite profiling using LC‐IT‐TOF‐MS, starting from sample collection and preparation to final identification of the metabolite structures, is discussed in detail. The data extraction and mining techniques to find and confirm metabolites are discussed and illustrated with some examples. This paper is directed to readers with no prior experience with LC‐IT‐TOF‐MS and will provide a broad understanding of the development and utility of this instrument for drug metabolism studies. In the present study, we report the application of LC‐MS based on two different LC‐MS systems to mycotoxin analysis. The mycotoxins were extracted with an ACN/water/acetic acid mixture and directly injected into a LC‐MS/MS system without any dilution procedure. First, a sensitive and reliable HPLC‐ESI‐MS/MS method using selected reaction monitoring on a triple quadrupole mass spectrometer (TSQ Quantum Ultra AM) has been developed for determining 32 mycotoxins in crude extracts of wheat and maize. This method was operated both in positive and in negative ionization modes in two separate chromatographic runs. The method was validated by studies of spiked recoveries, linearity, matrix effect, intra‐assay precision and sensitivity. Further, we have developed and evaluated a method based on accurate mass measurements of extracted target ions in full scan mode using micro‐LC‐LTQ‐Orbitrap as a tool for fast quantitative analysis. Both instruments exhibited very high sensitivity and repeatability in positive ionization mode. Coupling of micro‐LC to Orbitrap technology was not applicable to the negatively ionizable compounds. The LC triple quadrupole MS method has proved to be stable in quantitation, as it is with respect to the matrix effects of grain samples.  相似文献

In this communication, we describe the design of an online multi-chromatographic approach to the routine NMR analyses of low-level components ( approximately 0.1%) in complex mixtures. The technique, termed LC(2)-SPE-NMR, optimally combines multi-dimensional liquid chromatography with SPE technology for isolating, enriching and delivering trace analytes to the NMR probe. The fully automated LC(2)-SPE-NMR system allows for maximal loading capacity (in the first, preparative LC dimension), close to optimal peak resolution (in the second, analytical LC dimension) and enhanced sample concentration (through SPE). Using this system, it is feasible to conveniently conduct a wide range of NMR experiments on, for example, drug impurities at the low microgram per milliliter level, even for components poorly resolved in the first dimension. Such a sensitivity gain significantly elevates the analytical power of online NMR technology in terms of the level at which substances of pharmaceutical significance can be structurally characterized.  相似文献

A series of main‐chain, thermotropic, liquid‐crystalline (LC), hydrogen‐bonded polymers or self‐assembled structures based on 4,4′‐bipyridyl as a hydrogen‐bond acceptor and aliphatic dicarboxylic acids, such as adipic and sebacic acids, as hydrogen‐bond donors were prepared by a slow evaporation technique from a pyridine solution and were characterized for their thermotropic, LC properties with a number of experimental techniques. The homopolymer of 4,4′‐bipyridyl with adipic acid exhibited high‐order and low‐order smectic phases, and that with sebacic acid exhibited only a high‐order smectic phase. Like the homopolymer with adipic acid, the two copolymers of 4,4′‐bipyridyl with adipic and sebacic acids (75/25 and 25/75) also exhibited two types of smectic phases. In contrast, the copolymer of 4,4′‐bipyridyl with adipic and sebacic acids (50/50), like the homopolymer with sebacic acid, exhibited only one high‐order smectic phase. Each of them, including the copolymers, had a broad temperature range of LC phases (36–51 °C). The effect of copolymerization for these hydrogen‐bonded polymers on the thermotropic properties was examined. Generally, copolymerization increased the temperature range of LC phases for these polymers, as expected, with a larger decrease in the crystal‐to‐LC transition than in the LC‐to‐isotropic transition. Additionally, it neither suppressed the formation of smectic phases nor promoted the formation of a nematic phase in these hydrogen‐bonded polymers, as usually observed in many thermotropic LC polymers. The thermal transitions for all of them, measured by differential scanning calorimetry, were well below their decomposition temperatures, as measured by thermogravimetric analysis, which were in the temperature range of 193–210 °C. © 2003 Wiley Periodicals, Inc. The dielectric properties of a polymer‐dispersed liquid crystal (PDLC), a liquid‐crystal (LC) mixture (BL036), and three polymer matrices of PN314 containing different amounts of BLO36 were determined over a range of frequencies and temperatures and, for the LC and PDLC, over a range of voltages leading to homeotropic alignment of the LC. The overall dielectric relaxation process was a weighted sum of contributions from (1) the primary (δ) process in the LC arising from the motions of the dipoles about the short molecular axis and (2) dipole motions in the polymer matrix. The dielectric spectra were determined as a function of frequency, temperature, and, when appropriate, applied voltage. An equivalent electrical circuit was used as a working model to describe the dielectric behavior of the PDLC in the absence and presence of applied voltages. Agreement between the dielectric data and this model was achieved if a portion of the LC phase at the interface was assumed to be immobile. The director order parameter for the LC component in the PDLC was determined from dielectric measurements as the material was aligned homeotropically in an applied electric field. © 2001 John Wiley & Sons, Inc. Three triazine‐based dendrons ( 1 a – c ) were successfully prepared in 70–83 % yields. These newly prepared dendrons are found to be liquid crystalline (LC). Computational investigations on molecular conformations and dipoles of triazine‐based dendrons reveal that the substituent on the central triazine unit interrupts strong dipole or H‐bond interactions to avoid dimeric formation. The obtained dendrons, not favouring self‐assembly into dimers but showing LC behaviours, provides evidence for an approach contrary to the conventional method of inducing LC behaviours of dendrons by dimer or trimer formation, mostly through H‐bond interactions.  相似文献

A selective, sensitive and rapid liquid chromatography-tandem mass spectrometry method has been developed for the determination of caudatin-2,6-dideoxy-3-O-methy-beta-d-cymaropyranoside (CDMC) in rat plasma. This method involves a plasma clean-up step using liquid-liquid extraction, followed by LC separation and positive electrospray ionization mass spectrometry detection (LC/ESI-MS/MS). Chromatographic separation of the analytes was achieved using a C(18) column with a mobile phase of acetonitrile and water (70:30, v/v) at a flow rate of 1.0 mL/min. Low energy collision tandem mass spectrometric analysis (CID-MS/MS) using the multiple reaction monitoring (MRM) mode was used for analyte quantification. For the MRM analysis of CDMC, the following transition at m/z 658.4 --> 529.6 derived from the protonated molecule [M + Na](+). A calibration curve was linear in the 5-500 ng/mL range for CDMC, and the limit of detection was 5 ng/mL. The inter- and intra-day precisions (RSD) were 相似文献

2,3,5,4′‐Tetrahydroxystilbene‐2‐O‐β‐D‐glucoside (THSG) from Polygoni multiflori has been demonstrated to possess a variety of pharmacological activities, including antioxidant, anti‐inflammatory and hepatoprotective activities. Ultra‐performance LC‐quadrupole TOF‐MS with MS Elevated Energy data collection technique and rapid resolution LC with diode array detection and ESI multistage MSn methods were developed for the pharmacokinetics, tissue distribution, metabolism, and excretion studies of THSG in rats following a single intravenous or oral dose. The three metabolites were identified by rapid resolution LC‐MSn. The concentrations of the THSG in rat plasma, bile, urine, feces, or tissue samples were determined by ultra‐performance LC‐MS. The results showed that THSG was rapidly distributed and eliminated from rat plasma. After the intravenous administration, THSG was mainly distributing in the liver, heart, and lung. For the rat, the major distribution tissues after oral administration were heart, kidney, liver, and lung. There was no long‐term storage of THSG in rat tissues. Total recoveries of THSG within 24 h were low (0.1% in bile, 0.007% in urine, and 0.063% in feces) and THSG was excreted mainly in the forms of metabolites, which may resulted from biotransformation in the liver.  相似文献

The synthesis and structural analysis of the twin-dendritic benzamide 10, based on the first-generation, self-assembling, tapered dendrons 3,4,5-tris(4'-dodecyloxybenzyloxy)benzoic acid and 3,4,5-tris(4'-dodecyloxybenzyloxy)-1-aminobenzene, and the polymethacrylate, 20, which contains 10 as side groups, are presented. Benzamide 10 self-assembles into a supramolecular cylindrical dendrimer that self-organizes into a columnar hexagonal (Phi(h)) liquid crystalline (LC) phase. Polymer 20 self-assembles into an imperfect four-cylinder-bundle supramolecular dendrimer, and creates a giant vesicular supercylinder that self-organizes into a columnar nematic (N(c)) LC phase which displays short-range hexagonal order. In mixtures of 20 and 10, 10 acts as a guest and 20 as a host to create a perfect four-cylinder-bundle host-guest supramolecular dendrimer that coorganizes with 10. A diversity of Phi(h), simple rectangular columnar (Phi(r-s)) and centered rectangular columnar (Phi(r-c)), superlattices are produced at different ratios between 20 and 10. This diversity of LC lattices and superlattices is facilitated by the architecture of the twin-dendritic building block, polymethacrylate, the host-guest supramolecular assembly, and by hydrogen bonding along the center of the supramolecular cylinders generated from 10 and 20.  相似文献

Cefuroxime lysine is a new second‐generation cephalosporins, which can penetrate the blood–brain barrier to cure the meningitis. In order to investigate its acute toxicokinetic study after intraperitoneal injection of 675 mg/kg cefuroxime lysine, a sensitive and clean ultra‐fast liquid chromatography–tandem mass spectrometry (UFLC‐MS/MS) method for the determination of cefuroxime lysine in microdialysate samples was developed and validated, which was compared with UFLC‐UV as a reference method. Chromatographic separation was performed on a Shim‐pack XR‐ODS C18 column (75 × 3.0 mm, 2.2 µm), with an isocratic elution of 0.1% formic acid in acetonitrile–0.1% formic acid in water (45:55, v/v) for LC‐MS and acetonitrile–20 mm potassium dihydrogen phosphate (pH 3.0,20:80, v/v) for LC‐UV. The lower limit of detection was 0.01 µg/mL for LC‐MS and 0.1 µg/mL for LC‐UV method, with the same corresponding linearity range of 0.1–50 µg/mL. The intra‐ and inter‐day precisions (relative standard deviation) for both methods were from 1.1 to 8.9%, while the accuracy was all within ±10.9%. The results of both methods were finally compared using paired t‐test; the results indicated that the concentrations measured by the two methods correlated significantly (p < 0.05), which suggested that the two methods based on LC‐MS and LC‐UV were suitable for the acute toxicokinetic study. One of the main objectives of the experiment was to achieve the vertical aligned (VA) effect. To accomplish this, we employed liquid crystal (LC)/photo‐curable acrylic monomers mixture systems to prepare vertical alignment copolymer film (VACOF) for LC molecules with the photo‐polymerization induce phase separation (PIPS) process. From previous experimental results, we successfully fabricated LC devices without the micro‐protrusion structure. After the application of a saturated voltage, the LC molecules actually exhibited such interesting phenomena as uniaxial orientation, uniform single‐domain display state, etc. In this study, to obtain VACOF with smooth surface, we similarly controlled appropriate experimental conditions such as UV light exposure intensity and curing temperature, and altered process parameters such as the cell thickness, chemical structure length of the main chain type biphenol acrylic monomer [to simulate the main chain function of the traditional vertical alignment type polyimide (PI)], etc. During the experiment, we discovered that regardless of the cell thickness, this photo‐alignment system would yield the VACOF instead of the polymer disperse liquid crystal (PDLC) film morphology. Another notable finding was that the contrast ratio was heavily influenced by the length of the main chain type acrylic monomer structure for LC/monomer mixture systems, with enhancement of up to ~56%. Therefore, we further investigated the display effects, electro‐optical properties, etc. for these two main chain type acrylic monomer systems with different lengths and cell thicknesses. An optimized 2-D liquid chromatography (LC×LC) set-up, based on the different selectivities of a silver ion (Ag) and a non-aqueous reversed phase (NARP), employed in the first (D1) and the second dimension (D2), respectively, in combination with evaporative light-scattering detection (ELSD), has been developed for the analysis of the triacylglycerol (TAG) fraction in a Borago officinalis oil. The 2-D set-up, thanks to the complementary separation selectivity provided by the two columns, allowed to distribute 78 TAGs throughout the 2-D LC retention plane otherwise unachievable by 1-D LC.  相似文献

Multidimensional separation techniques play an increasingly important role in separation science, especially for the analysis of complex samples such as proteins. The combination of reversed‐phase liquid chromatography in the nanoscale and CZE is especially beneficial due to their nearly orthogonal separation mechanism and well‐suited geometries/dimensions. Here, a heart‐cut nano‐LC–CZE–MS setup was developed utilizing for the first time a mechanical 4‐port valve as LC–CE interface. A model protein mixture containing four different protein species was first separated by nano LC followed by a heart‐cut transfer of individual LC peaks and subsequent CZE–MS analysis. In the CZE dimension, various glycoforms of one protein species were separated. Improved separation capabilities were achieved compared to the 1D methods, which was exemplarily shown for ribonuclease B and its different glycosylated forms. LODs in the lower μg/mL range were determined, which are considerably lower compared to traditional CZE–MS. In addition, this study represents the first application of an LC–CE–MS system for intact protein analysis. The nano‐LC–CZE–MS system is expected to be applicable to various other analytical challenges.  相似文献

Pharmaceuticals require careful and precise determination of their impurities that might harm the user upon consumption. Although today, the most common technique for impurities identification is liquid chromatography‐mass spectrometry (LC‐MS/MS), it has several downsides due to the nature of the ionization method. Also, the analyses in many cases are targeted thus despite being present, some of the compounds will not be revealed. In this paper, we propose and show a new method for untargeted analysis and identification of impurities in active pharmaceutical ingredients (APIs). The instrument used for these analyses is a novel electron ionization (EI) LC‐MS with supersonic molecular beams (SMB). The EI‐LC‐MS‐SMB was implemented for analyses of several drug samples spiked with an impurity. The instrument provides EI mass spectra with enhanced molecular ions, named Cold EI, which increases the identification probabilities when the compound is identified with the aid of an EI library like National Institute of Standards and Technology (NIST). We analyzed ibuprofen and its impurities, and both the API and the expected impurity were identified with names and structures by the NIST library. Moreover, other unexpected impurities were found and identified proving the ability of the EI‐LC‐MS‐SMB system for truly untargeted analysis. The results show a broad dynamic range of four orders of magnitude at the same run with a signal‐to‐noise ratio of over 10 000 for the API and almost uniform response.  相似文献
