Imidazo[1,5-a]- and Thiazolo[3,4-a]quinoxalines Based on 3-(a-Thiocyanobenzyl)quinoxalin-2(1H)-one

When 3-(a-thiocyanobenzyl-2(1H)-one is heated, competing processes of [a]-annelation of the imidazole or thiazole rings occurs with formation of imidazo[1,5-a]- and thiazolo[3,4-a]quinoxalin-4(5H)-ones.     

Synthesis of 5-Phenyl-7-trifluoromethyl-2,3-dihydroimidazo[1,2-a]pyridines

Syntheses are reported for a series of 2-alkylamino-6-phenyl-4-trifluoromethylpyridines. The reaction of 3-cyano-2-(hydroxyalkylamino)-6-phenyl-4-trifluoromethylpyridines with thionyl chloride gave the corresponding 2-(chloroalkylamino)pyridines, 8-cyano-5-phenyl-7-trifluoromethyl-2,3-dihydro-imidazo[1,2-a]pyridines, and 9-cyano-6-phenyl-8-trifluoromethyl-2,3,4-trihydropyrido[1,2-a]-pyrimidines. X-ray diffraction structural analysis was used to study 8-cyano-5-phenyl-7-trifluoromethyl-2,3-dihydroimidazo[1,2-a]pyridine.     

Synthesis of 1- and 2-Carbamoylalkyl-2,3-dihydro-1H-indole and 9-Carbamoylalkyl-2,3,4,4a,9,9a-hexahydro-1H-carbazole Derivatives

Treatment of 1,2,3,9a-tetrahydro-9H-imidazo- and 1,2,3,4,10,10a-hexahydropyrimido[1,2-a]indol-2-one derivatives with formic acid gave 1-carbamoylalkyl-2,3,3-trimethyl-2,3-dihydro-1H-indoles. 9-Carbamoylmethyl- and 9-(2-carbamoylethyl)-4a-methyl-2,3,4,4a,9,9a-hexahydro-1H-carbazoles were prepared from 5,6,7,7a-tetrahydro-1H,4H-imidazo- and 1,2,6,7,8,8a-hexahydro-5H-pyrimido[2,1-k]carbazolones in a similar manner. Synthesis of 2-(2-carbamoylpropyl)-2,3,3-trimethyl-2,3-dihydro-1H-indole was carried out by reduction of 1,3-dihydrospiro[2H-indolo-2,2'-piperidine] derivative with Zn in acetic acid solution.     

Condensation of 1-Substituted 1,2,3,9a-Tetrahydro-9H-imidazo[1,2-a]indol-2-one Derivatives with Aromatic Aldehydes

Condensation of 1-alkyl-, 1-allyl-, and 1-benzyl-1,2,3,9a-tetrahydro-9H-imidazo[1,2-a]indol-2-ones with benzaldehydes in acetic acid and subsequent treatment of the reaction mixture with potassium hydroxide afforded 1-substituted 9a-(2-phenylethenyl)-1,2,3,9a-tetrahydro-9H-imidazo[1,2-a]indol-2-one derivatives. 1-Methyl- and 1-ethyl-9a-[2-(4-dimethylaminophenyl)ethenyl]-1,2,3,9a-tetrahydro-9H-imidazo[1,2-a]indol-2-ones were synthesized by alkylation of 9a-[2-(4-dimethylaminophenyl)ethenyl]-1,2,3,9a-tetrahydro-9H-imidazo[1,2-a]indol-2-one with methyl- and ethyl iodides in DMF in the presence of a strong base.     

Stereoselective synthesis of 4-substituted 1,2,3,4,10,10a-hexahydropyrazino[1,2-<Emphasis Type="Italic">a</Emphasis>]indoles

4-Substituted 1,2,3,4-tetrahydropyrazino[1,2-a]indoles were synthesized from 2-cyanoindole. (R)-4-Methyl-1,2,3,4-tetrahydropyrazino[1,2-a]indole was obtained by the Mitsunobu reaction. Stereoselective reduction of 4-substituted 1,2,3,4-tetrahydropyrazino[1,2-a]indoles gave 4-substituted 1,2,3,4,10,10a-hexahydropyrazino[1,2-a]indoles. (4R, 10aR)-4-Methyl-1,2,3,4,10,10a-hexahydropyrazino[1,2-a]indole was synthesized.__________Published in Russian in Izvestiya Akademii Nauk. Formylation of Furyl-substituted Imidazo[1,2-a]pyridine, Imidazo[1,2-a]pyrimidine and Imidazo[2,1-b]thiazole

Vilsmeier formylation of 2-(2-furyl)-substituted imidazo[1,2-a]pyridine and imidazo[1,2-a]pyrimidine, and also 6-(2-furyl)imidazo[2,1-b]thiazole with 1 mole of reagent occurs at the free position of the imidazole ring, while with an excess of the reagent it also occurs at the position 5 of the furyl group.     

Spatial Configuration of Derivatives of 5,5a,6,7-Tetrahydropyrido[1,2-a]benzimidazole and 6,7-Dihydro-5aH-pyrido[1,2-b]benzoxazole

The spatial configurations of 7,9-diphenyl-5a,6-tetramethylene-5,5a,6,7-tetrahydropyrido[1,2-a]benzimidazole and 7,9-diphenyl-5a,6-tetramethylene-2,5a,6,7-tetrahydropyrido[1,2-a]benzimidazol-2-one have been established by X-ray crystallography. Analogous configurations are proposed for a series of other derivatives of 5,5a,6,7-tetrahydropyrido[1,2-a]benzimidazoles and some derivatives of 6,7-dihydro-5aH-pyrido[1,2-b]benzoxazoles on the basis of ¹H NMR spectroscopic data and the results of quantum chemical calculations using the MNDO, AM1, and PM3 methods.     

Fused polycyclic nitrogen-containing heterocycles 15.* 1,3-bis(4-hydroxy-4-methoxycarbonyl-3,5-diphenylthiazolidin-2-ylideneamino)-benzene in the synthesis of thiazolo[3,4-a]quinoxalines

Reactions of methyl chloro(phenyl)pyruvate with 1,3-bis(phenylthioureido)benzene easily afforded 1,3-bis(4-hydroxy-4-methoxycarbonyl-3,5-diphenylthiazolidin-2-ylideneamino)benzene. Reactions of the latter with 1,2-phenylenediamines gave various derivatives of thiazolo[3,4-a]quinoxalines, including their bisanalogs. Published in Russian in Izvestiya Akademii Nauk. The Use of 4-(Bromomethylene)-5,5-dimethyl-1,3-dioxolan-2-one as “Masked” α-Bromo-α'-Hydroxy Ketone in the Synthesis of Heterocyclic Systems

4-(Bromomethylene)-5,5-dimethyl-1,3-dioxolan-2-one was obtained on the basis of the readily obtainable 4-methylene-5,5-dimethyl-1,3-dioxolan-2-one. It forms 2-imidazo[1,2-a]pyridin-2-yl-2-propanol with 2-aminopyridine, 11a-hydroxy-1,1-dimethyl-3-oxo-1,5,11,11a-tetrahydro[1,3]oxazolo[3,4-a]pyrido[1,2-d]-10-pyrazinium bromide with 2-(aminomethyl)pyridine, and the corresponding derivative of 4-hydroxyoxazolidin-2-one with 2-(3,4-dimethoxyphenyl)ethylamine. The last product was converted by intramolecular amidoalkylation without isolation into 10b-(bromomethyl)-8,9-dimethoxy-1,1-dimethyl-1,5,6,10b-tetrahydro[1,3]oxazolo[3,4-a]isoquinolin-3-one.     

Fused polycyclic nitrogen-containing heterocycles

Upon treatment with acetic acid, 3-(2-phenyl-1-selenocyanatoethyl)-, 3-(3-phenyl-1-selenocyanatopropyl)quinoxalin-2(1H)-ones and their thio analogs undergo intramolecular condensation to form selenazolo-and thiazolo[3,4-a]quinoxalines. 以芳香醛、4-溴茚酮、丙二腈和醋酸铵为原料,分别用微波辐射和传统加热法,一步法合成7种未见文献报道的2-氨基-6-溴基-4-芳基-5H-茚并[1,2-b]吡啶-3-腈衍生物.对比两种方法,在常压下微波具有反应时间短、产率较高等特点.所有化合物均经IR,1HNMR和元素分析确定.     

Cyclization of 1-hydroxy-2-(oxoalkynyl)anthraquinones

Procedures were developed for the heterocyclization of 2-benzoylethynyl-1-hydroxy-and 1-hydroxy-2-(1-oxo-3-phenylpropynyl)anthraquinone. The regioselective preparation of 2-substituted anthra[1,2-b]pyran-4,7,12-triones was demonstrated to be possible under mild conditions.__________Published in Russian in Izvestiya Akademii Nauk. Reactions of 4-morpholino-1,2-naphthoquinone with enamines ando-phenylenediamine

The reactions of 4-morpholino-1,2-naphthoquinone with enamines, cyclohexanone derivatives, afford 7a-amino-5-morpholinohexahydrobenzo[b]naphtho[1,2][1,4]dioxines, while the reaction witho-phenylenediamine yields 5-morpholinobenzo[a]phenazine. Formylation of 3,4-Dihydropyrrolo[1,2-a]pyrazines

Formylation of 3,4-dihydropyrrolo[1,2-a]pyrazines containing alkyl or aryl substituents at position 1 has been studied under the conditions of the Vilsmeier reaction. The direction of the reaction depends on the structure of 3,4-dihydropyrrolo[1,2-a]pyrazine starting materials. Formylation of 1-methyl-substituted 3,4-dihydropyrrolo[1,2-a]pyrazines occurs at the methyl group.     

Acylation of 3,4-Dihydropyrrolo[1,2-a]pyrazines

The direction of trifluoroacetylation with trifluoroacetic anhydride of 3,4-dihydropyrrolo[1,2-a]pyrazines containing an alkyl or aralkyl substituent in position 1 depends on both the structure of the 3,4-dihydropyrrolo[1,2-a]pyrazine starting materials and on the ratio of reagent:substrate. It may lead to both mono- and disubstituted products. Trifluoroacetylation of 1-methyl-3,4-dihydropyrrolo[1,2-a]pyrazines occurs at the methyl group. Acetylation of 3,4-dihydropyrrolo[1,2-a]pyrazines leads only to N-acetyl-substituted reaction products.     

Bromination of Substituted 5H-Imidazo[1,2-b]-1,2,4-triazepin-4-ones and -thiones

The reactions of substituted 5H-imidazo[1,2-b]-1,2,4-triazepin-4-ones and -thiones with bromine and N-bromosuccinimide have been studied. Derivatives of 3- and 8-bromo-, 3,8-dibromoimidazo[1,2-b]-1,2,4-triazepine and 5H-2-bromomethyl-3-methyl-7,8-diphenylimidazo[1,2-b]-1,2,4-triazepin-4-one are formed, depending on the degree of substitution, the nature of the brominating agent, and the reaction conditions.     

Synthesis of polyarene derivatives of fused aziridines by Suzuki-Miyaura cross-coupling

A convenient and effective synthetic approach to potentially radiochromic polyarene derivatives of bi- and tricyclic aziridines utilizing modified Suzuki-Miyaura cross-coupling with catalysis in micellar media has been developed. A series of polyarene substituents were introduced into 1,5-diaryl-3,5a-dihydro-1H-azireno[1,2-c]imidazoles, 1,6-diaryl-1,3,4,6a-tetrahydroazireno[1,2-a]pyrazines, and 1,2-diaryl-1,1a-dihydroazireno[1,2-a]quinoxalines without degradation of the highly reactive aziridine ring system.     

Exotic Amino Acids. 6. Synthesis of Substituted 4-Oxo-4H-pyrido[1,2-a]pyrimidines

2-Pyridylaminomethyleneisopropylidenemalonates, prepared from ethoxymethyleneisopropylidenemalonate and 2-aminopyridines, form 4-oxo-4H-pyrido[1,2-a]pyrimidines at their melting points and are separated from the reaction mixture by sublimation.     

Properties of 8-Cyano-5-phenyl-7-trifluoromethyl-2,3-dihydroimidazo[1,2-a]pyridines

The alkylation, acylation, halogenation, nitration, oxidation, and hydrolysis reactions of 8-cyano-5-phenyl-7-trifluoromethyl-2,3-dihydroimidazo[1,2-a]pyridine have been studied. It was found that the 6-halo derivatives add alcohol to give covalent solvates. X-ray analysis has been carried out on one of the solvates (6-chloro-8-cyano-7-ethoxy-5-phenyl-7-trifluoromethyl-1,2,3,7-tetrahydroimidazo[1,2-a]pyridine) as well as on 8-cyano-5-phenyl-7-trifluoromethyl-2,3-dihydroimidazo[1,2-a]pyridine trifluoroacetate.     

One-Pot Three-Component Synthesis of 3-Aminoimidazo[1,2-<Emphasis Type="Italic">a</Emphasis>]pyridines and -pyrazines in the Presence of Silica Sulfuric Acid

Summary. The synthesis of 3-aminoimidazo[1,2-a]pyridines and 3-aminoimidazo[1,2-a]pyrazines through a condensation reaction of 2-aminopyridine or 2-aminopyrazine, aldehyde, and alkyl or aryl isocyanide in high yields at room temperature in the presence of silica sulfuric acid is described.
