Ion–molecule reactions between the α‐phenylvinyl cation and isomeric naturally occurring phenols were investigated using a quadruple ion trap mass spectrometer. The α‐phenylvinyl cation m/z 103, generated by chemical ionization from phenylacetylene, reacts with neutral aromatic compounds to form the characteristic species: [M + 103]+ adduct ions and the trans‐vinylating product ions [M + 25]+, which correspond to [M + 103]+ adduct after the loss of benzene. Isomeric differentiation of several ring‐substituted phenols was achieved by using collision‐induced dissociation of the [M + 103]+ adduct ions. This method also showed to be effective in the differentiation of 4‐ethylguaiacol from one of its structural isomers that displays identical EI and EI/MS/MS spectra. The effects of gas‐phase alkylation with phenylvinyl cation on the dissociation behavior were examined using mass spectrometryn and labeled derivatives. Matrix‐assisted laser desorption/ionization in‐source decay (MALDI‐ISD) induces N–Cα bond cleavage via hydrogen transfer from the matrix to the peptide backbone, which produces a c′/z? fragment pair. Subsequently, the z? generates z′ and [z + matrix] fragments via further radical reactions because of the low stability of the z?. In the present study, we investigated MALDI‐ISD of a cyclic peptide. The N–Cα bond cleavage in the cyclic peptide by MALDI‐ISD produced the hydrogen‐abundant peptide radical [M + 2H]+? with a radical site on the α‐carbon atom, which then reacted with the matrix to give [M + 3H]+ and [M + H + matrix]+. For 1,5‐diaminonaphthalene (1,5‐DAN) adducts with z fragments, post‐source decay of [M + H + 1,5‐DAN]+ generated from the cyclic peptide showed predominant loss of an amino acid with 1,5‐DAN. Additionally, MALDI‐ISD with Fourier transform‐ion cyclotron resonance mass spectrometry allowed for the detection of both [M + 3H]+ and [M + H]+ with two 13C atoms. These results strongly suggested that [M + 3H]+ and [M + H + 1,5‐DAN]+ were formed by N–Cα bond cleavage with further radical reactions. As a consequence, the cleavage efficiency of the N–Cα bond during MALDI‐ISD could be estimated by the ratio of the intensity of [M + H]+ and [M + 3H]+ in the Fourier transform‐ion cyclotron resonance spectrum. Because the reduction efficiency of a matrix for the cyclic peptide cyclo(Arg‐Gly‐Asp‐D‐Phe‐Val) was correlated to its tendency to cleave the N–Cα bond in linear peptides, the present method could allow the evaluation of the efficiency of N–Cα bond cleavage for MALDI matrix development. Metal complex formation was investigated for di‐exo‐, di‐endo‐ and trans‐2,3‐ and 2,5‐disubstituted trinorbornanediols, and di‐exo‐ and di‐endo‐ 2,3‐disubstituted camphanediols using different divalent transition metals (Co2+, Ni2+, Cu2+) and electrospray ionization quadrupole ion trap mass spectrometry. Many metal‐coordinated complex ions were formed for cobalt and nickel: [2M+Met]2+, [3M+Met]2+, [M–H+Met]+, [2M–H+Met]+, [M+MetX]+, [2M+MetX]+ and [3M–H+Co]+, where M is the diol, Met is the metal used and X is the counter ion (acetate, chloride, nitrate). Copper showed the weakest formation of metal complexes with di‐exo‐2,3‐disubstituted trinorbornanediol yielding only the minor singly charged ions [M–H+Cu]+, [2M–H+Cu]+ and [2M+CuX]+. No clear differences were noted for cobalt complex formation, especially for cis‐2,3‐disubstituted isomers. However, 2,5‐disubstituted trinorbornanediols showed moderate diastereomeric differentiation because of the unidentate nature of the sterically more hindered exo‐isomer. trans‐Isomers gave rise to abundant [3M–H+Co]+ ion products, which may be considered a characteristic ion for bicyclo[221]heptane trans‐2,3‐ and trans‐2,5‐diols. To differentiate cis‐2,3‐isomers, the collision‐induced dissociation (CID) products for [3M+Co]2+, [M+CoOAc]+, [2M–H+Co]+ and [2M+CoOAc]+ cobalt complexes were investigated. The results of the CID of the monomeric and dimeric metal adduct complexes [M+CoOAc]+ and [2M–H+Co]+ were stereochemically controlled and could be used for stereochemical differentiation of the compounds investigated. The relationships between the ionization profile, sensitivity, and structures of 64 exogenous anabolic steroids (groups I–IV) was investigated under electrospray ionization (ESI) conditions. The target analytes were ionized as [M + H]+ or [M + H–nH2O]+ in the positive mode, and these ions were used as precursor ions for selected reaction monitoring analysis. The collision energy and Q3 ions were optimized based on the sensitivity and selectivity. The limits of detection (LODs) were 0.05–20 ng/mL for the 64 steroids. The LODs for 38 compounds, 14 compounds and 12 compounds were in the range of 0.05–1, 2–5 and 10–20 ng/mL, respectively. Steroids including the conjugated keto‐functional group at C3 showed good proton affinity and stability, and generated the [M + H]+ ion as the most abundant precursor ion. In addition, the LODs of steroids using the [M + H]+ ion as the precursor ion were mostly distributed at low concentrations. In contrast, steroids containing conjugated/unconjugated hydroxyl functional groups at C3 generated [M + H ? H2O]+ or [M + H ? 2H2O]+ ions, and these steroids showed relatively high LODs owing to poor stability and multiple ion formation. An LC‐MS/MS method based on the present ionization profile was developed and validated for the determination of 78 steroids (groups I–V) in human urine. Experimental and theoretical studies on the oxidation of saturated hydrocarbons (n‐hexane, cyclohexane, n‐heptane, n‐octane and isooctane) and ethanol in 28 Torr O2 or air plasma generated by a hollow cathode discharge ion source were made. Ions corresponding to [M + 15]+ and [M + 13]+ in addition to [M ? H]+ and [M ? 3H]+ were detected as major ions where M is the sample molecule. The ions [M + 15]+ and [M + 13]+ were assigned as oxidation products, [M ? H + O]+ and [M ? 3H + O]+, respectively. By the tandem mass spectrometry analysis of [M ? H + O]+ and [M ? 3H + O]+, H2O, olefins (and/or cycloalkanes) and oxygen‐containing compounds were eliminated from these ions. Ozone as one of the terminal products in the O2 plasma was postulated as the oxidizing reagent. As an example, the reactions of C6H14+? with O2 and of C6H13+ (CH3CH2CH+CH2CH2CH3) with ozone were examined by density functional theory calculations. Nucleophilic interaction of ozone with C6H13+ leads to the formation of protonated ketone, CH3CH2C(=OH+)CH2CH2CH3. In air plasma, [M ? H + O]+ became predominant over carbocations, [M ? H]+ and [M ? 3H]+. For ethanol, the protonated acetic acid CH3C(OH)2+ (m/z 61.03) was formed as the oxidation product. The peaks at m/z 75.04 and 75.08 are assigned as protonated ethyl formate and protonated diethyl ether, respectively, and that at m/z 89.06 as protonated ethyl acetate. Unusual ionization behavior was observed with novel antineoplastic curcumin analogues during the positive ion mode of matrix‐assisted laser desorption ionization (MALDI) and dopant‐free atmospheric pressure photoionization (APPI). The tested compounds produced an unusual significant peak designated as [M ? H]+ ion along with the expected [M + H]+ species. In contrast, electrospray ionization, atmospheric pressure chemical ionization and the dopant‐mediated APPI (dopant‐APPI) showed only the expected [M + H]+ peak. The [M ? H]+ ion was detected with all evaluated curcumin analogues including phosphoramidates, secondary amines, amides and mixed amines/amides. Our experiments revealed that photon energy triggers the ionization of the curcumin analogues even in the absence of any ionization enhancer such as matrix, solvent or dopant. The possible mechanisms for the formation of both [M ? H]+ and [M + H]+ ions are discussed in this paper. In particular, three proposed mechanisms for the formation of [M ? H]+ were evaluated. The first mechanism involves the loss of H2 from the protonated [M + H]+ species. In the search for novel natural products in plants, particularly those with potential bioactivity, it is important to efficiently distinguish novel compounds from previously isolated, known compounds, a process known as dereplication. In this study, electrospray ionization‐multiple stage tandem mass spectrometry (ESI‐MSn) was used to study the behaviour of 12 pregnane glycosides and genins previously isolated from Marsdenia tenacissima, a traditional Chinese medicinal plant, as a basis for dereplication of compounds in a plant extract. In addition to [M + Na]+ and [M + NH4]+ ions, a characteristic [M‐glycosyl + H]+ ion was observed in full‐scan mode with in‐source fragmentation. Sequential in‐trap collision‐induced dissociation of [M + Na]+ ions from 11,12‐diesters revealed consistent preferred losses of substituents first from C‐12, then from C‐11, followed by losses of monosaccharide fragments from the C‐3 tri‐ and tetrasaccharide substituents. A crude methanol extract of M. tenacissima stems was analysed using high‐performance liquid chromatography coupled to ESI‐MS. Several previously isolated pregnane glycosides were dereplicated, and the presence of an additional nine novel pregnane glycosides is predicted on the basis of the primary and fragment ions observed, including two with a previously unreported C4H7O C‐11/C‐12 substituent of pregnane glycosides. This study is the first report of prediction of the structures of novel pregnane glycosides in a crude plant extract by a combination of in‐source fragmentation and in‐trap collision‐induced dissociation and supports the usefulness of LC‐ESI‐MSn not only for dereplication of active compounds in extracts of medicinal plants but also for detecting the presence of novel related compounds. N‐Boc/Fmoc/Z‐N′‐formyl‐gem‐diaminoalkyl derivatives, intermediates particularly useful in the synthesis of partially modified retro‐inverso peptides, have been characterized by both positive and negative ion electrospray ionization (ESI) ion‐trap multi‐stage mass spectrometry (MSn). The MS2 collision induced dissociation (CID) spectra of the sodium adduct of the formamides derived from the corresponding N‐Fmoc/Z‐amino acids, dipeptide and tripeptide acids show the [M + Na‐NH2CHO]+ ion, arising from the loss of formamide, as the base peak. Differently, the MS2 CID spectra of [M + Na]+ ion of all the N‐Boc derivatives yield the abundant [M + Na‐C4H8]+ and [M + Na‐Boc + H]+ ions because of the loss of isobutylene and CO2 from the Boc protecting function. Useful information on the type of amino acids and their sequence in the N‐protected dipeptidyl and tripeptidyl‐N′‐formamides is provided by MS2 and subsequent MSn experiments on the respective precursor ions. The negative ion ESI mass spectra of these oligomers show, in addition to [M‐H]?, [M + HCOO]? and [M + Cl]? ions, the presence of in‐source CID fragment ions deriving from the involvement of the N‐protecting group. Furthermore, MSn spectra of [M + Cl]? ion of N‐protected dipeptide and tripeptide derivatives show characteristic fragmentations that are useful for determining the nature of the C‐terminal gem‐diamino residue. The present paper represents an initial attempt to study the ESI‐MS behavior of these important intermediates and lays the groundwork for structural‐based studies on more complex partially modified retro‐inverso peptides. The microalga Haematococcus pluvialis produces the pigment astaxanthin mainly in esterified form with a multitude of fatty acids, which results in a complex mixture of carotenol mono‐ and diesters. For rapid fingerprinting of these esters, matrix‐assisted laser desorption ionization time of flight mass spectrometry (MALDI‐TOF/TOF‐MS) might be an alternative to traditional chromatographic separation combined with MS. Investigation of ionization and fragmentation of astaxanthin mono‐ and diester palmitate standards in MALDI‐TOF/TOF‐MS showed that sodium adduct parent masses [M + Na]+ gave much simpler MS2 spectra than radical / protonated [M]+● / [M + H]+ parents. [M + Na]+ fragments yielded diagnostic polyene‐specific eliminations and fatty acid neutral losses, whereas [M]+● / [M + H]+ fragmentation resulted in a multitude of non‐diagnostic daughters. For diesters, a benzonium fragment, formed by polyene elimination, was required for identification of the second fatty acid attached to the astaxanthin backbone. Parents were forced into [M + Na]+ ionization by addition of sodium acetate, and best signal‐to‐noise ratios were obtained in the 0.1 to 1.0 mM range. This method was applied to fingerprinting astaxanthin esters in a crude H. pluvialis extract. Prior to MALDI‐TOF/TOF‐MS, the extract was fractionated by normal phase Flash chromatography to obtain fractions enriched in mono‐ and diesters and to remove pheophytin a, which compromised monoester signals. The detection of layer‐by‐layer self‐assembly multilayer films was carried out using low‐temperature plasma (LTP) mass spectrometry (MS) under ambient conditions. These multilayer films have been prepared on quartz plates through the alternate assembling of oppositely charged 4‐aminothiophenol (4‐ATP) capped Au particles and thioglycolic acid (TGA) capped Ag particles. An LTP probe was used for direct desorption and ionization of chemical components on the films. Without the complicated sample preparation, the structure information of 4‐ATP and TGA on films was studied by LTP‐MS. Characteristic ions of 4‐ATP (M) and TGA (F), including [M]+?, [M‐NH2]+, [M‐HCN‐H]+, and [F + H]+, [F‐H]+, [F‐OH]+, [F‐COOH]+ were recorded by LTP‐MS on the films. However, [M‐CS‐H]+ and [F‐SH]+ could not be observed on the film, which were detected in the neat sample. In addition, the semi‐quantitative analysis of chemical components on monolayer film was carried out, and the amounts of 4‐ATP and TGA on monolayer surface were 45 ng/mm2 and 54 ng/mm2, respectively. This resulted the ionization efficiencies of 72% for 4‐ATP and 54% for TGA. In order to evaluate the reliability of present LTP‐MS, the correlations between this approach and some traditional methods, such as UV–vis spectroscopy, atomic force microscope and X‐ray photoelectron spectroscopy were studied, which resulted the correlation coefficients of higher than 0.9776. The results indicated that this technique can be used for analyzing the films without any pretreatment, which possesses great potential in the studies of self‐assembly multilayer films. Ion/molecule reactions of saturated hydrocarbons (n‐hexane, cyclohexane, n‐heptane, n‐octane and isooctane) in 28‐Torr N2 plasma generated by a hollow cathode discharge ion source were investigated using an Orbitrap mass spectrometer. It was found that the ions with [M+14]+ were observed as the major ions (M: sample molecule). The exact mass analysis revealed that the ions are nitrogenated molecules, [M+N]+ formed by the reactions of N3+ with M. The reaction, N3+ + M → [M+N]+ + N2, were examined by the density functional theory calculations. It was found that N3+ abstracts the H atom from hydrocarbon molecules leading to the formation of protonated imines in the forms of R′R″C?NH2+ (i.e. C–H bond nitrogenation). This result is in accord with the fact that elimination of NH3 is the major channel for MS/MS of [M+N]+. That is, nitrogen is incorporated in the C–H bonds of saturated hydrocarbons. No nitrogenation was observed for benzene and acetone, which was ascribed to the formation of stable charge‐transfer complexes benzene????N3+ and acetone????N3+ revealed by density functional theory calculations. The detection of anabolic androgenic steroids (AAS) is one of the most important topics in doping control analysis. Gas chromatography coupled to (tandem) mass spectrometry (GC–MS(/MS)) with electron ionization and liquid chromatography coupled to tandem mass spectrometry have been traditionally applied for this purpose. However, both approaches still have important limitations, and, therefore, detection of all AAS is currently afforded by the combination of these strategies. Alternative ionization techniques can minimize these drawbacks and help in the implementation of a single method for the detection of AAS. In the present work, a new atmospheric pressure chemical ionization (APCI) source commercialized for gas chromatography coupled to a quadrupole time‐of‐flight analyzer has been tested to evaluate the ionization of 60 model AAS. Underivatized and trimethylsylil (TMS)‐derivatized compounds have been investigated. The use of GC–APCI–MS allowed for the ionization of all AAS assayed irrespective of their structure. The presence of water in the source as modifier promoted the formation of protonated molecules ([M+H]+), becoming the base peak of the spectrum for the majority of studied compounds. Under these conditions, [M+H]+, [M+H‐H2O]+ and [M+H‐2·H2O]+ for underivatized AAS and [M+H]+, [M+H‐TMSOH]+ and [M+H‐2·TMSOH]+ for TMS‐derivatized AAS were observed as main ions in the spectra. The formed ions preserve the intact steroid skeleton, and, therefore, they might be used as specific precursors in MS/MS‐based methods. Additionally, a relationship between the relative abundance of these ions and the AAS structure has been established. The 70 eV electron ionization mass spectra of polycyclic aromatic compounds are characterized by the presence of relatively stable multiply charged molecular ions [M]n+ (n=2–4). When generated from the compounds benzene, napthalene, anthracene, phenanthrene, 2,3-benzanthracene, 1,2-benzanthracene, chrysene, 9,10-benzophenanthrene and pyrene, the relative abundances of the multiply charged ions increase dramatically with the number of rings. These compounds form multiply charged molecular ions (n=2, 3) which undergo unimolecular decompositions indicative of considerable ionic rearrangement. The main charge separation processes observed here [M]2+→m1++m2+, [M]3+˙→m3++m→+m42+) involve, in almost every case, one or more of the products [CH3]+, [C2H3]+˙ and [C3H3]+. This suggests the existence of preferred structures amongst the metastable parent ions. Information on the relative importance of the various fragmentation pathways is presented here along with translational energy release data. Some tentative structural information about the metastable ions has been inferred from the translational energy release on the assumption that the released energy is due primarily to coulombic repulsion within the transition state structure. For the triply charged ions these interpretations have necessitated the use of a coulombic repulsion model which takes account of an extra charge. Vertical ionization energies for the process [M]n++G→[M](n+1)+G+e? (charge stripping) have also been determined where possible for n=1 and 2 and the results from these experiments allow the derivation of simple empirical equations which relate successive ionization energies for the formation of [M]2+ and [M]3+˙ to the appearance energy of [M]+˙.  相似文献

Spin‐labeled nitroxide derivatives of podophyllotoxin had better antitumor activity and less toxicity than that of the parent compounds. However, the 2‐H configurations of these spin‐labeled derivatives cannot be determined by nuclear magnetic resonance (NMR) methods. In the present paper, a high‐performance liquid chromatography‐diode array detection (HPLC‐DAD) and a high‐performance liquid chromatography‐electrospray ionization tandem mass spectrometry (HPLC‐ESI/MS/MS) method were developed and validated for the separation, identification of four pairs of diastereoisomers of spin‐labeled derivatives of podophyllotoxin at C‐2 position. In the HPLC‐ESI/MS spectra, each pair of diastereoisomers of the spin‐labeled derivatives in the mixture was directly confirmed and identified by [M+H]+ ions and ion ratios of relative abundance of [M‐ROH+H]+ (ion 397) to [M+H]+. When the [M‐ROH+H]+ ions (at m/z 397) were selected as the precursor ions to perform the MS/MS product ion scan. The product ions at m/z 313, 282, and 229 were the common diagnostic ions. The ion ratios of relative abundance of the [M‐ROH+H]+ (ion 397) to [M+H]+, [A+H]+ (ion 313) to [M‐ROH+H]+, [A+H‐OCH3]+ (ion 282) to [M‐ROH+H]+ and [M‐ROH‐ArH+H]+ (ion 229) to [M‐ROH+H]+ of each pair of diastereoisomers of the derivatives specifically exhibited a stereochemical effect. Thus, by using identical chromatographic conditions, the combination of DAD and MS/MS data permitted the separation and identification of the four pairs of diastereoisomers of spin‐labeled derivatives of podophyllotoxin at C‐2 in the mixture.  相似文献

Some compounds containing mono- and polynitroxide radicals have been investigated and characterized by electron ionization (EI), chemical ionization (CI), and matrix-assisted laser desorption-ionization (MALDI) mass spectrometry. Some characteristics of the mass spectra of these radicals are demonstrated. It was found that, under CI and MALDI conditions, ions [M+nH]+ are formed along with monoprotonated molecules [M+H]+, depending on the type of the radical n = 2–5.  相似文献

Organophosphate esters (OPEs) are chemical compounds incorporated into materials as flame‐proof and/or plasticizing agents. In this work, 13 non‐halogenated and 5 halogenated OPEs were studied. Their mass spectra were interpreted and compared in terms of fragmentation patterns and dominant ions via various ionization techniques [electron ionization (EI) and chemical ionization (CI) under vacuum and corona discharge atmospheric pressure chemical ionization (APCI)] on gas chromatography coupled to mass spectrometry (GC‐MS). The novelty of this paper relies on the investigation of APCI technique for the analysis of OPEs via favored protonation mechanism, where the mass spectra were mostly dominated by the quasi‐molecular ion [M + H]+. The EI mass spectra were dominated by ions such as [H4PO4]+, [M–R]+, [M–Cl]+, and [M–Br]+, and for some non‐halogenated aryl OPEs, [M]+● was also observed. The CI mass spectra in positive mode were dominated by [M + H]+ and sometimes by [M–R]+, while in negative mode, [M–R] and more particularly [X] and [X2]‐● were mainly observed for the halogenated OPEs. Both EI and APCI techniques showed promising results for further development of instrumental method operating in selective reaction monitoring mode. Instrumental detection limits by using APCI mode were 2.5 to 25 times lower than using EI mode for the non‐brominated OPEs, while they were determined at 50‐100 times lower by the APCI mode than by the EI mode, for the two brominated OPEs. The method was applied to fish samples, and monitored transitions by using APCI mode showed higher specificity but lower stability compared with EI mode. Sesquiterpene lactones (SL) have been reported with various biological effects. Among the described SL skeletons, hirsutinolide and glaucolide have not been extensively studied by mass spectrometry (MS), especially how to distinguish them in organic matrices. Thus, this paper reports (1) a strategy of their differentiation based on MS behavior during the ionization and (2) a proposal of the fragmentation pattern for both SL‐subtypes. ESI(+)‐HRMS data of four isolated SL (hirsutinolides 1 and 3 ; glaucolides 2 and 4 ) were recorded by direct and UPLC water‐sample combined injections. These analyses revealed that hirsutinolides and glaucolides formed [M+Na]+ ion during the operation of the direct MS injection, and ([M+Na]+ and [M+H‐H2O]+) and [M+H]+ ions were respectively observed for hirsutinolides and glaucolides during the operation of combined UPLC water and sample MS injection. Computational simulations showed that the complex hirsutinolide ( 1 )‐Na+ formed with a lower preparation energy compared with the complex glaucolide ( 2 )‐Na+. However, despite their different behavior during the ionization process, ESI(+)‐HRMS/MS analyses of 1 ‐ 4 gave similar fragmentation patterns at m/z 277, 259, 241, and 231 that can be used as diagnostic ions for both skeletons. Moreover, the differentiation strategy based on the nature of the complex SL‐adducts and their MS/MS fragmentation pattern were successfully applied for the chemical characterization of the extract from Vernonanthura tweedieana using UPLC‐ESI‐HRMS/MS. Secondary and tertiary amines have been reported to form [M–H]+ that correspond to dehydrogenation in matrix‐assisted laser desorption ionization time of flight mass spectrometry (MALDI‐TOF MS). In this investigation, we studied the dehydrogenation of amines in MALDI‐TOF MS by isotopic labeling. Aliphatic amines were labeled with deuterium on the methylene of an N‐benzyl group, which resulted in the formation of [M–D]+ and [M–H]+ ions by dedeuteration and dehydrogenation, respectively. This method revealed the proton that was removed. The spectra of most tertiary amines with an N‐benzyl group showed high‐intensity [M–D]+ and [M–H]+ ion peaks, whereas those of secondary amines showed low‐intensity ion peaks. Ratios between the peak intensities of [M–D]+ and [M–H]+ greater than 1 suggested chemoselective dehydrogenation at the N‐benzyl groups. The presence of an electron donor group on the N‐benzyl groups enhanced the selectivity. The dehalogenation of amines with an N‐(4‐halobenzyl) group was also observed alongside dehydrogenation. The amino ions from dehalogenation can undergo second dehydrogenation. These results provide the first direct evidence about the position at which dehydrogenation of an amine occurs and the first example of dehalogenation of haloaromatic compounds in MALDI‐TOF MS. Three liquid chromatography–tandem mass spectrometry (LC‐MS/MS) methods were respectively developed and validated for the simultaneous or independent determination of taurine and edaravone in rat plasma using 3‐methyl‐1‐p‐tolyl‐5‐pyrazolone and sulfanilic acid as the internal standards (IS). Chromatographic separations were achieved on an Agilent Zorbax SB‐Aq (100 × 2.1 mm, 3.5 µm) column. Gradient 0.03% formic acid–methanol, isocratic 0.1% formic acid–methanol (90:10) and 0.02% formic acid–methanol (40:60) were respectively selected as the mobile phase for the simultaneous determination of two analytes, taurine or edaravone alone. The MS acquisition was performed in multiple reaction monitoring mode with a positive and negative electrospray ionization source. The mass transitions monitored were m/z [M + H]+ 175.1 → 133.0 and [M + H]+ 189.2 → 147.0 for edaravone and its IS, m/z [M ? H]? 124.1 → 80.0 and [M ? H]? 172.0 → 80.0 for taurine and its IS, respectively. The validated methods were successfully applied to study the pharmacokinetic interaction of taurine and edaravone in rats after independent intravenous administration and co‐administration with a single dose. Our collective results showed that there were no significant alterations on the main pharmacokinetic parameters (area under concentration–time curve, mean residence time, half‐life and clearance) of taurine and edaravone, implying that the proposed combination therapy was pharmacologically feasible. N‐(3‐Ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 1–4 and N‐(6‐ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 5–8 were prepared by coupling either 3‐ferrocenylnaphthalene‐2‐carboxylic acid or 6‐ferrocenylnaphthalene‐2‐carboxylic acid to the dipeptide ethyl esters GlyGly(OEt) (1, 5), AlaGly(OEt) (2, 6), GlyPhe(OEt) (3, 7) and GlyLeu(OEt) (4, 8), using the standard N‐(3‐dimethylaminopropyl)‐N'‐ethylcarbodiimide hydrochloride, 1‐hydroxybenzotriazole protocol. Electrospray ionization mass spectrometry (ESI‐MS) and laser desorption ionization mass spectrometry (LDI‐MS) were employed in conjunction with tandem mass spectrometry in the analysis of N‐(3‐ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 1–4 and N‐(6‐ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 5–8. Radical cations, [M]+? and [M + H]+ species were both observed in the mass spectra. Intense sodium [M + Na]+ and potassium [M + K]+ adducts were also present. An important diagnostic ion at m/z [M–65]+ was observed in both the MS and MS/MS spectra of the N‐(3‐ferrocenyl‐2‐naphthoyl) dipeptide derivatives. Sequence‐specific ions were generally not observed in the MS/MS spectra of the N‐(3‐ferrocenyl‐2‐naphthoyl) series due to formation of the diagnostic [M–65]+ ion. Sequence‐specific ions were observed in the MS/MS spectra of the N‐(6‐ferrocenyl‐2‐naphthoyl) dipeptide esters with charge retention on the derivatized N‐terminal of the dipeptide.
