The presence of human and veterinary pharmaceuticals in the environment has caused increasing concern due their effects on ecological receptors. Improving the risk assessment of these compounds necessitates a quantitative understanding of their metabolism and elimination in the target organism (toxicokinetics), particularly via the ubiquitous cytochrome P-450 (CYP) system and their mechanisms of toxicity (toxicodynamics). This review focuses on a number of pharmaceuticals and veterinary medicines of environmental concern, and the differences and similarities between ecological and human risk assessment. CYP metabolism is discussed with particular reference to its ubiquity in species of ecological relevance. The important issue of pharmaceutical mixtures is discussed to assess how emerging technologies such as ecotoxicogenomics may assist in moving towards a more mechanism-based environmental risk assessment of pharmaceuticals.  相似文献

The recycling and recovery of organic matter and nutrients from sewage sludge for application in agricultural soils is gaining interest, while the presence of pharmaceutically active compounds (PhACs) in this matrix may have a great impact on the environment and human health. The aim of this review paper is to outline recent research on the occurrence of PhACs and their metabolites in sewage treatment lines. A total of 32 classes of therapeutic groups including 180 PhACs and 45 metabolites have been included. In a first part, a summary of the analytical methods with a critical overview of the extraction and determination techniques, quality control issues and methodological challenges for their determination is included. Subsequently, the study gives a snapshot of the concentration levels and distribution patterns found in primary, secondary, digested (aerobically and anaerobically), dehydrated and composted sludge. Data have been systematically summarized and categorized according to matrix type, treatment processes available for PhAC degradation in sludge, and geographical areas. Our literature review showed that antimicrobials, antibiotics, non-steroidal anti-inflammatory drugs (NSAIDs), antidepressants and antidiabetics were the most abundant PhACs found in sludge matrices.Overall, attenuation of PhACs concentrations occurs during sludge stabilization, in particular during anaerobic digestion and composting. PhAC sorption onto sludge is strongly affected by the physicochemical properties, the sludge matrix and the operating and environmental conditions. Lastly, the paper discusses the impact of PhACs on sludge-amended agricultural land. The potential ecotoxicological risk associated with the presence of PhACs in amended soil is medium-low for most PhACs. The most critical compounds found in sludge-amended soil are ciprofloxacin, 17α-ethinylestradiol, 17β-estradiol, and triclocarban and triclosan.  相似文献

Field-effect transistors (FETs) are one of the most widely-used electronic sensors for continuous monitoring and detection of contaminants such as pharmaceuticals and endocrine-disrupting compounds at low concentrations. FETs have been successfully utilized for the rapid analysis of these environmental pollutants due to their advantageous material properties like the disposability, rapid responses and simplicity. This paper presented an up-to-date overview of applied strategies with different bio-based materials in order to enhance the analytical performances of the designed sensors. Comparison and discussion were made between characteristics of recently engineered FET bio-sensors used for the detection of famous and selected pharmaceutical compounds in the literature. The recent progress in environmental research applications, comments on interesting trends, current challenge for future research in endocrine-disrupting chemicals’ (EDCs) detection using FETs biosensors were highlighted.  相似文献

A spectrofluorimetric method to determine acrivastine is proposed and applied to its determination in human urine and pharmaceuticals. The fluorimetric method allows the determination of 58-2000 ng ml(-1) of acrivastine in aqueous solutions containing acetic acid-sodium acetate buffer (pH 6.5) with lambda(exc)=230 nm and lambda(em)=380 nm.  相似文献

Human health risk assessment is a site-based approach used to identify the potential health hazards which are induced by an old site contamination. For a proper evaluation of the daily doses of contaminants to which people will be exposed given the future occupation of the site, both a characterization and a quantification of soil pollution are needed. Such information can be provided by soil sampling. Thus the choice of the location, the number, depth and type of soil samples is very important and ought to follow a well-defined strategy. A review of contaminated site sampling practices in Europe and North America could not identify any completely formalized sampling strategy for human health risk assessment. On the contrary there are several approaches which can be roughly classified into two categories: a systematic sampling scheme over the whole site, on the one hand, and a sampling design driven by an initial knowledge of the contamination sources and fitted to the suspected pollution pattern, on the other. The first approach provides a complete coverage of the site but it may be rather expensive and entail useless sampling. The performance of the second depends on the quality of prior information. Actually both methods can be combined as explained hereafter. In view of the specificity of each site, the requirements of health risk assessment and the time and cost constraints, it seems difficult to work out a typical soil sampling strategy suitable for all sites. However, some recommendations can be made according to the site dimensions, the nature, degree and heterogeneity of contamination, and the (future) use of the site. The scientist should thus rely on a thorough examination of all available information (site history, geology and hydrogeology, soil properties, contaminants behaviour , etc.) to delimit contaminated areas as homogeneous as possible and then distribute the sampling points (e.g.using a sampling grid). They should also take the potential exposure paths into account in order to define the areas and soil strata to be sampled as a priority. Hydrophilic interaction capillary electrochromatography (HI-CEC) for the determination of basic pharmaceuticals spiked in human serum is described. The organic modifier content, ionic strength, and pH value of the mobile phase as well as the applied voltage are optimized for separation and elution of these drug analytes. Excellent separation was achieved for drugs using a mobile phase composition of 80% v/v acetonitrile in 100 mM triethylamine phosphate (TEAP) buffer at pH 2.8 with column efficiencies for analytes more than 200,000 plates/m. The samples of human serum spiked with basic drugs were directly injected after a simple acetonitrile treatment. The linear range and reproducibility of these basic drugs using an external and internal standard method were compared. As a result, the reproducibility could be greatly improved by using the internal standard method. Good calibration curves with regression coefficients more than 0.998 in the range of 5-160 microg/mL were observed with the internal standard method. The limits of quantitation, based on standards with acceptable relative standard deviations (RSDs), were below 5 microg/mL. The intra- and inter-day precisions, determined as RSDs, were less than 4.57%.  相似文献

Spectrofluorimetric methods to determine cisatracurium and mivacurium are proposed and applied to the determination of both substances in human serum and to the determination of mivacurium in pharmaceuticals. The fluorimetric methods allow the determination of 5-500 ng ml(-1) of mivacurium in aqueous solutions and 5-500 ng ml(-1) of cisatracurium in water-acetonitrile solutions, both containing acetic acid-sodium acetate buffer (pH 5.5) with lambda(exc)=230 nm and lambda(em)=324 nm.  相似文献

Micellar electrokinetic capillary chromatography (MEKC) was performed at 25 °C and 30 kV (under a pressure of 15 mbar), using 30 mM borate buffer containing 60 mM sodium dodecysulfate (SDS) and 5% (v/v) methanol as background electrolyte (pH 9.0) to determine doxycycline. UV detection was at 350 nm. The method was shown to be specific, accurate (recovery was 100.3 ± 1.0%), linear over the tested range (correlation coefficient 0.9995) and precise (RSD <1.9%). The method was used to determine doxycycline in tablets, capsules and human urine after oral application.  相似文献

A simple and selective method for penicillin V potassium (PVK) determination by chemiluminescence (CL) was developed. Oxidation of PVK by alkaline hydrogen peroxide produces CL, which is greatly enhanced by N, N-dimethyl formamide (DMF) and N-cetyl-N,N,N-trimethylammonium bromide (CTMAB). Optimum conditions were established using luminometry. There is a linear relationship between the chemiluminescent peak height and the amount of PVK within the range 0.5–129.5 mg L−1, with a detection limit of 0.2 mg L−1. The coefficient of variation was 1.2% for 40 mg L−1 PVK solution (n = 7). The method is very simple, has high sensitivity and good selectivity, and is usable for process control. It was successfully utilized for the determination of PVK in pharmaceuticals and spiked human urine.   相似文献

A sensitive, reliable and reproducible HPLC method with electrochemical detection (HPLC-ECD) has been developed for the separation and quantification of levodopa methyl ester (LDME) and its impurities such as levodopa (l-DOPA), 3-methoxytyrosine (MTS) and l-tyrosine (TS) in bulk drug and pharmaceutical dosage form. The separation was performed on an LC18 column by isocratic elution with methanol-acetonitrile-50 mm potassium dihydrogen phosphate (8:2:90, v/v/v) containing 5 mm sodium 1-hexanesulfonate, 5 mm EDTA and 5 mm sodium chloride, adjusted with phosphoric acid to a pH of 3.2 as mobile phase. The correlation coefficients of linear regression for LDME, L-DOPA, MTS and TS were more than 0.999. The detection limits for L-DOPA, MTS and TS were 3.15, 2.04 and 2.88 ng/mL, respectively. The precision was checked in terms of F-test variance ratio using potentiometric titration as reference. The separation of dopa methyl ester enantiomers by chiral chromatography is also described. This method is capable of separating the two enantiomers with a selection of 1.4 and a resolution of 8.4. Both methods were found to be stable and useful in the quality control of the bulk material and formulations.  相似文献

This paper describes an environmentally friendly method for quantitative determination of ranitidine using diffuse reflectance spectroscopy. This method is based on the reflectance measurements of the colored product produced from the spot test reaction between ranitidine and p-dimethylaminocinnamaldehyde (p-DAC), in acid medium, using filter paper as solid support. Experimental design methodologies were used to optimize the optimal conditions. All reflectance measurements were carried out at 590 nm and the linear range was from 1.42x10(-3) to 3.42x10(-2) mol L(-1), with a correlation coefficient of 0.997. The limit of detection was estimated to be 1.09x10(-3) mol L(-1) (R.S.D.=1.9%). The proposed method was successfully applied to the determination of ranitidine in commercial brands of pharmaceuticals and no interferences were observed from the common excipients in formulations. The results obtained by the proposed method were favorably compared with those obtained by an official procedure at 95% confidence level. Additionally, the method was also applied to the determination of ranitidine in human urine showing excellent recoveries (99.6-100.3%).  相似文献

An improved analytical method for determination of human pharmaceuticals in natural and wastewaters with ng L−1 sensitivity is presented. The method is applicable to pharmaceuticals from a wide range of therapeutic classes including antibiotics, analgesics, anti-inflammatories and anti-cancer compounds. Pharmaceuticals were extracted from waters using solid-phase extraction, and after concentration, analysed by high performance liquid chromatography with tandem mass spectrometric detection (HPLC-MS/MS). Identification of each compound was secured using retention time and by the selected reaction monitoring of two transitions, one of which was additionally used for quantification. Limits of detection ranged from 0.03 to 0.96 ng L−1 and were up to two orders of magnitude lower than those of previously published methods. The method was validated using spiked samples prepared from tap, river and sea water as well as wastewater effluents, collected from the North of Scotland. Analysis of wastewater effluents revealed the presence of mefenamic acid, ibuprofen, erythromycin, diclofenac and trimethoprim. None of the selected pharmaceuticals were detected in river, tap or sea water samples.  相似文献

Proteomic analysis of human vitreous humor (VH) may elucidate the pathogenesis of retinal ocular diseases and may provide information for the development of potential therapeutic targets due to its pivotal location near lens and retina. The discovery of whole VH proteome involves a complex analysis of thousands of proteins simultaneously. Therefore, in proteomic studies the protein fractionation is important for reducing sample complexity, facilitating the access to the low‐abundant proteins, and recognizing them as biotargets for clinical research. Although several separation methods have been used, gel‐based proteomics are the most popular and versatile ones applied for global protein separation. However, chromatographic methods and its combination with other separation techniques are now beginning to be used as promising set‐ups for VH protein identification. This review attempts to offer an overview of the techniques currently used with VH, exploring its methodological demands, exposing its advantages, and helping the reader to plan future experiences. Moreover, this review shows the relevance of VH proteomic analysis as a tool for the study of the mechanisms underlying some ocular diseases and for the development of new therapeutic approaches.  相似文献

A selective molecularly imprinted polymer (MIP) has been synthesized for isoxicam pre-concentration, followed by its spectrophotometric determination based on hydrogen bonding interactions between examined drug and alizarin yellow GG. This method is able to evaluate isoxicam in range of 1.0 × 10−3 to 20.0 μg mL−1, with a limit of determination of 1.0 ng mL−1. The retention capacity and pre-concentration factor of prepared sorbent are 18.5 mg g−1 and 200, respectively; and the prepared MIPs can be reused at least for five times. The MIP capability for isoxicam selection and extraction from the solution is higher than non-imprinted polymer (NIP). Under optimum conditions, this procedure can be successfully applied to assay trace amounts of isoxicam in pharmaceutical and biological samples.  相似文献

The quinolone antibacterial agent nalidixic acid (NAL) was studied by cyclic voltammetry (CV) and cathodic adsorptive stripping voltammetry (CASV). A sensitive method is described for the determination of NAL in its pure form, dosage forms and biological fluids. Controlled adsorptive accumulation of NAL on a hanging mercury drop electrode provides the basis for the direct stripping measurement of that compound in the nanomolar concentration level. Different variables were studied and optimized. The proposed method depends upon the voltammetric activity of NAL in Britton-Robinson buffer, whereby a well-defined cathodic peak is produced at pH 5.0 in presence of NO(3)(-). The calibration graph to determine NAL was linear in the range 7.4x10(-8)-2.5x10(-5) M by CASV. CAS voltammetry has been proved to be advantageous over a liquid chromatographic (LC) technique, allowing to detection limit signal to noise ratio, (s/n=3) of 0.766 ng ml(-1) (3.3x10(-9) M) NAL to be reached. The relative standard deviation (n=5) was 5.2% at concentration level of 1.0x10(-7) M NAL. The degree of interference from coexisting metal ions on the CASV signal for NAL was evaluated. The method was applied to two different commercial pharmaceutical products (Negram tablets and suspension) with very good recoveries. It was also shown that the method was successfully applied to the determination of NAL in human urine and blood serum. Mean recoveries were 98.8+/-0.3 and 98.9+/-0.41%, respectively.  相似文献

An automated procedure for the assay of procaine hydrochloride in human blood and pharmaceuticals was developed using a sequential injection (SI) technique with fluorometric detection and fluorescamine as the fluorescence probe. A few microliters of fluorescamine and procaine hydrochloride solutions were used in the SI system leading to the formation of a derivative, which was then excited by a 400-nm LED and whose emitted fluorescence was monitored at a wavelength of 494 nm. A linear calibration graph was obtained with 10–200 ng mL−1 (procaine) by loading 10.0 μL of sample solution and 5.0 μL of fluorescamine solution (both 0.125 % m/v). A detection limit of 2.6 ng mL−1, defined as 3 times the blank standard deviation (3σ), was achieved along with a sampling frequency of 25 h−1 and a precision of 2.1 % RSD at the 50.0 ng mL−1 level. Procaine contents in injection solutions from various pharmaceutical manufactures were analyzed and reasonable agreement was achieved between the values obtained by using the present procedure and the documented spectrophotometry, and both were coincident with the nominal concentrations. In addition, the degradation of procaine in human blood was investigated. A fast degradation of procaine in human blood was observed for the first 30 min, while afterwards the degradation was retarded.  相似文献

A method has been developed for the determination of Ibuprofen (2-[4-isobutylphenyl]-propionic acid) in pharmaceuticals by FT-IR, using the carbonyl band which this compound presents at 1710 cm(-1) in carbon tetrachloride solutions. Samples are dissolved in carbon tetrachloride. In this solvent the excipients are not soluble and so the drug can be directly determined without any additional treatment. The use of a simple FIA manifold permits one to carry out this analysis with a low consumption of reagent and the FT-IR provides a continuous monitoring of the spectral base-line which permits an accurate determination of the maximum in the absorbance band. Also, the FIA system permits easy and fast sampling and cleaning of the measurement cell. The method has a dynamic range between 0.5 and 20 mg/ml with a sensitivity of 0.366 +/- 0.004 au . mg(-1) . ml . mm(-1) and a variation coefficient of 0.8% for 5 independent measurements of a real sample containing 200 mg of Ibuprofen per capsule. The developed procedure provides concentration values comparable with those found by UV spectrophotometry in the analysis of real samples but is free from matrix interferences.  相似文献

New Spectrophotometric methods for the assay of clozapine (CZP) in pure and dosage forms are described. Method A is based on the oxidative coupling reaction of CZP with 3-methyl-2-benzothiazolinone hydrazone hydrochloride (MBTH) in the presence of ceric ammonium sulphate to form a coloured species (max 570 nm). Method B is also based on the oxidative coupling reaction of CZP withP-,N,N-dimethylpheny-lenediamine dihydrochloride (DMPD) in the presence of sodium hypochlorite to form a coloured species (max 690 nm). Method C is based on the formation of coloured charge-transfer complex between CZP and chloranilic acid (max 540 nm). Regression analysis of Beer-Lambert plots showed good correlation in the concentration ranges 2–25, 10–120 and 15–300 g/ml for methods A, B and C, respectively. The validity of the proposed methods was tested by analysing pharmaceutical dosage forms containing CZP and the relative standard deviation values were within 1.0%.  相似文献
