Polycaprolactone (PCL) is widely used in biomedical applications as electrospun fibers or porous foams. As PCL is synthetic polymer, many researchers have explored blends of PCL–gelatin to combine mechanical and bioactive properties of individual components. High pressure carbon dioxide (CO2) has been studied to foam and impregnate many biocompatible polymers. In case of PCL–gelatin blends, certain compositions can be swelled reversibly under high pressure CO2 without permanent deformation. This allows successful impregnation of PCL–gelatin blends under CO2. This study summarizes effect of different treatments adopted during impregnation process including high pressure CO2 on several blend compositions of PCL–gelatin blends. Stress relaxation, polymer melting and dissolution were observed during several treatments which affects porosity and scaffold structure significantly. Results summarized in this study will aid in optimum selection of PCL–gelatin blend composition for biomedical applications. Furthermore, CO2 solubility in polymers is restricted due to thermodynamic limitations but can be altered in the presence of a co-solvent to produce better foams. PCL can be foamed using supercritical CO2. However, CO2 foaming of PCL–gelatin blend becomes challenging to simultaneous swelling of PCL and compression of gelatin providing blend structural stability. This study has demonstrated ability of supercritical CO2 to foam PCL–gelatin blends in presence of water to create porous structure. These foams were subjected post-fabrication crosslinking and supercritical CO2 without losing porosity of foams. Thus, creating a strategy to use environmentally benign processes to fabricate, crosslink and impregnate porous scaffolds for biomedical applications.  相似文献

Electrospun micro- and nanofiber scaffolds have gained interest in biomedical applications, especially in tissue engineering, because they can be used to reproduce the structure of the extracellular matrix (ECM) of natural tissue. The selection of the solvent is an important factor which affects the diameter, the surface morphology and the crystallinity of the electrospun fibers, and, accordingly, their mechanical properties as well as their degradation kinetics. Furthermore, the surface morphology of the electrospun fibres can be controlled by solvent vapour pressure to produce porous structures which might be helpful for cell adhesion and proliferation. In the present work, poly (L-lactic acid) (PLLA) has been electrospun using solvents with different vapour pressures to investigate the influences of the solvent vapour pressure on morphology, diameter, crystallinity and mechanical properties of the electrospun fiber scaffolds. The results show that the vapour pressure of the solvents (or solvent mixtures) play an important role in the fiber diameter and crystallinity. Furthermore, the crystallinity of the fibers is increased by lowering the vapour pressure of the used solvent. In addition, the mechanical properties (e.g., tensile strength and Young's modulus) are strongly dependent on morphological features such average fibers diameter. The smaller the average diameter, the higher the tensile strength and Young's modulus.  相似文献

Nowadays, hydrogels-based microneedles (MNs) have attracted a great interest owing to their outstanding qualities for biomedical applications. For the fabrication of hydrogels-based microneedles as tissue engineering scaffolds and drug delivery carriers, various biomaterials have been tested. They are required to feature tunable physiochemical properties, biodegradability, biocompatibility, nonimmunogenicity, high drug loading capacity, and sustained drug release. Among biomaterials, human proteins are the most ideal biomaterials for fabrication of hydrogels-based MNs; however, they are mechanically weak and poorly processible. To the best of the knowledge, there are no reports of xeno-free human protein-based MNs so far. Here, human albumin-based hydrogels and microneedles for tissue engineering and drug delivery by using relatively new processible human serum albumin methacryloyl (HSAMA) are engineered. The resultant HSAMA hydrogels display tunable mechanical properties, biodegradability, and good biocompatibility. Moreover, the xeno-free HSAMA microneedles display a sustained drug release profile and significant mechanical strength to penetrate the model skin. In vitro, they also show good biocompatibility and anticancer efficacy. Sustainable processible human albumin-based biomaterials may be employed as a xeno-free platform in vivo for tissue engineering and drug delivery in clinical trials in the future.  相似文献

Recently, tissue engineering and regenerative medicine studies have evaluated smart biomaterials as implantable scaffolds and their interaction with cells for biomedical applications. Porous materials have been used in tissue engineering as synthetic extracellular matrices, promoting the attachment and migration of host cells to induce the in vitro regeneration of different tissues. Biomimetic 3D scaffold systems allow control over biophysical and biochemical cues, modulating the extracellular environment through mechanical, electrical, and biochemical stimulation of cells, driving their molecular reprogramming. In this review, first we outline the main advantages of using polysaccharides as raw materials for porous scaffolds, as well as the most common processing pathways to obtain the adequate textural properties, allowing the integration and attachment of cells. The second approach focuses on the tunable characteristics of the synthetic matrix, emphasizing the effect of their mechanical properties and the modification with conducting polymers in the cell response. The use and influence of polysaccharide-based porous materials as drug delivery systems for biochemical stimulation of cells is also described. Overall, engineered biomaterials are proposed as an effective strategy to improve in vitro tissue regeneration and future research directions of modified polysaccharide-based materials in the biomedical field are suggested.  相似文献

With the ability to form a submicron-sized fibrous structure with interconnected pores mimicking the extracellular matrix (ECM) for tissue formation, electrospinning was used to fabricate ultra-fine fiber mats of hexanoyl chitosan (H-chitosan) for potential use as skin tissue scaffolds. In the present communication, the in vitro biocompatibility of the electrospun fiber mats was evaluated. Indirect cytotoxicity evaluation of the fiber mats with mouse fibroblasts (L929) revealed that the materials were non-toxic and did not release substances harmful to living cells. The potential for use of the fiber mats as skin tissue scaffolds was further assessed in terms of the attachment and the proliferation of human keratinocytes (HaCaT) and human foreskin fibroblasts (HFF) that were seeded or cultured on the scaffolds at different times. The results showed that the electrospun fibrous scaffolds could support the attachment and the proliferation of both types of cells, especially for HaCaT. In addition, the cells cultured on the fibrous scaffolds exhibited normal cell shapes and integrated well with surrounding fibers. The obtained results confirmed the potential for use of the electrospun H-chitosan fiber mats as scaffolds for skin tissue engineering.  相似文献

Bone tissue engineering has become one of the most effective methods for treating bone defects. In this study, an electrospun tissue engineering membrane containing magnesium was successfully fabricated by incorporating magnesium oxide (MgO) nanoparticles into silk fibroin and polycaprolactone (SF/PCL)-blend scaffolds. The release kinetics of Mg2+ and the effects of magnesium on scaffold morphology, and cellular behavior were investigated. The obtained Mg-functionalized nanofibrous scaffolds displayed controlled release of Mg2+, satisfactory biocompatibility and osteogenic capability. The in vivo implantation of magnesium-containing electrospun nanofibrous membrane in a rat calvarial defect resulted in the significant enhancement of bone regeneration twelve weeks post-surgery. This work represents a valuable strategy for fabricating functional magnesium-containing electrospun scaffolds that show potential in craniofacial and orthopedic applications.  相似文献

Electrospinning is a versatile technique providing highly tunable nanofibrous nonwovens. Many biomedical applications have been developed for nanofibers, among which the production of antimicrobial mats stands out. The production of scaffolds for tissue engineering, fibers for controlled drug release, or active wound dressings are active fields of research exploiting the possibilities offered by electrospun materials. The fabrication of materials for active food packaging or membranes for environmental applications is also reviewed. We attempted to give an overview of the most recent literature related with applications in which nanofibers get in contact with living cells and develop a nano-bio interface.  相似文献

Recent advancements in the electrospinning method enable the production of ultrafine solid and continuous fibers with diameters ranging from a few nanometers to a few hundred nanometers with controlled surface and internal molecular structures. A wide range of biodegradable biopolymers can be electrospun into mats with specific fiber arrangement and structural integrity. Through secondary processing, the nanofiber surface can be functionalized to display specific biochemical characteristics. It is hypothesized that the large surface area of nanofibers with specific surface chemistry facilitates attachment of cells and control of their cellular functions. These features of nanofiber mats are morphologically and chemically similar to the extracellular matrix of natural tissue, which is characterized by a wide range of pore diameter distribution, high porosity, effective mechanical properties, and specific biochemical properties. The current emphasis of research is on exploiting such properties and focusing on determining appropriate conditions for electrospinning various polymers and biopolymers for eventual applications including multifunctional membranes, biomedical structural elements (scaffolds used in tissue engineering, wound dressing, drug delivery, artificial organs, vascular grafts), protective shields in specialty fabrics, and filter media for submicron particles in the separation industry. This has resulted in the recent applications for polymer nanofibers in the field of biomedicine and biotechnology.  相似文献

Electrospun polymeric fibers present an emerging alternative for the development of flexible electronics, enabling applications in wearable sensors and biosensors for continuous monitoring, and actuators for tissue engineering. The possibility to prepare sub-micrometric polymeric scaffolds, their processing for increasing the conductivity, their modification with different materials, conductive polymers and biomolecules in order to obtain functional flexible electrodes, allows the development of innovative devices for healthcare, and biomedical applications. In this review, the impact of metallized electrospun polymeric fibers in electrochemical (bio)sensors and actuators is discussed. A relation between their structure and functionality is provided, alongside with an overview of the different methods to obtain functional conductive fibers.  相似文献

Zein, a corn-derived protein, has a variety of applications ranging from drug delivery to tissue engineering and wound healing. This work aims to develop a biocompatible scaffold for dermal applications based on thermally annealed electrospun propolis-loaded zein nanofibers. Pristine fibers' biocompatibility is determined in vitro. Next, propolis from Melipona quadrifasciata is added to the fibers at different concentrations (5% to 25%), and the scaffolds are studied. The physicochemical properties of zein/propolis precursor dispersions are evaluated and the results are correlated to the fibers' properties. Due to zein's and propolis' very favorable interactions, which are responsible for the increase in the dispersions surface tension, nanometric size ribbon-like fibers ranging from 420 to 575 nm are obtained. The fiber's hydrophobicity is not dependent on propolis concentration and increases with the annealing procedure. Propolis inhibitory concentration (IC50) is determined as 61.78 µg mL−1. When loaded into fibers, propolis is gradually delivered to cells as Balb/3T3 fibroblasts and are able to adhere, grow, and interact with pristine and propolis-loaded fibers, and cytotoxicity is not observed. Therefore, the zein–propolis nanofibers are considered biocompatible and safe. The results are promising and provide prospects for the development of wound-healing nanofiber patches—one of propolis' main applications.  相似文献

Polycaprolactone (PCL) is a bioresorbable and biocompatible polymer that has been widely used in long-term implants and controlled drug release applications. However, when it comes to tissue engineering, PCL suffers from some shortcomings such as slow degradation rate, poor mechanical properties, and low cell adhesion. The incorporation of calcium phosphate-based ceramics and bioactive glasses into PCL has yielded a class of hybrid biomaterials with remarkably improved mechanical properties, controllable degradation rates, and enhanced bioactivity that are suitable for bone tissue engineering. This review presents a comprehensive study on recent advances in the fabrication and properties of PCL-based composite scaffolds containing calcium phosphate-based ceramics and bioglasses in terms of porosity, degradation rate, mechanical properties, in vitro and in vivo biocompatibility and bioactivity for bone regeneration applications. The fabrication routes range from traditional methods such as solvent casting and particulate leaching to novel approaches including solid free-form techniques.  相似文献

Designing advanced biomaterials with regenerative and drug delivering functionalities remains a challenge in the field of tissue engineering. In this paper we present the design, development, and a use case of an electrospun nano-biocomposite scaffold composed of silk fibroin (SF), hardystonite (HT), and gentamicin (GEN). The fabricated SF nanofiber scaffolds provide mechanical support while HT acts as a bioactive and drug carrier, on which GEN is loaded as an antibacterial agent. Antibacterial zone of inhibition (ZOI) results indicate that the inclusion of 3–6 wt% GEN significantly improves the antibacterial performance of the scaffolds against Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus) bacteria, with an initial burst release of 10–20% and 72–85% total release over 7 days. The release rate of stimulatory silicon ions from SF-HT scaffolds reached 94.53±5 ppm after 7 days. Cell studies using osteoblasts show that the addition of HT significantly improved the cytocompatibility of the scaffolds. Angiogenesis, in vivo biocompatibility, tissue vascularization, and translatability of the scaffolds were studied via subcutaneous implantation in a rodent model over 4-weeks. When implanted subcutaneously, the GEN-loaded scaffold promoted angiogenesis and collagen formation, which suggests that the scaffold may be highly beneficial for further bone tissue engineering applications.  相似文献

In recent years, the use of organic materials to infer conductivity in biomedical devices has received increasing attention. Typical inorganic semiconductors and conductors are rigid and expensive, usually require multiple processing steps and are unsuitable for biomedical applications. Electrochemically or chemically doped conjugated polymers help to overcome these problems due to their stability, low cost, light weight and excellent electrical and optical properties. The conducting polymer poly(3,4-ethylenedioxythiophene): polystyrene sulfonate (PEDOT:PSS) is the material of choice for biomedical applications as it is water soluble, however, there are growing concerns around its stabilizer, PSS, due to its release of acidic products upon degradation in-vivo. Here, we report the successful synthesis of PEDOT nanoparticles using hyaluronic acid (HA) as a stabilizer via an oxidative miniemulsion polymerisation technique. This improves the bioactivity and hydrophilicity of nanoparticles. The effect of varying amounts of HA and different molar ratios of EDOT:TOS has been studied and their role in the conductive and morphological properties of final nanoparticles has been fully elucidated. Furthermore, bioactivity and biocompatibility of the nanoparticles are demonstrated for customizable in vivo applications. Nanoparticles were found to have a conductivity up to 10 times greater than pristine PEDOT:PSS with increased addition of oxidant. The proposed easy-to-manufacture approach, along with the highlighted superior properties, expands the potential of conductive polymers in future customizable biological applications such as tissue scaffolds, nerve conduits and cardiac patches and represents a real breakthrough from the current state of the art.  相似文献

Nowadays, encapsulated dyes in a polymeric matrix have opened up new perspectives in many applications such as filtration of subatomic particles, composite reinforcement, multifunctional membranes, tissue engineering scaffolds, wound dressing, coatings, medical purposes as well as sensors. In the presented work, we report on electrospinning neat peryelene dianhydride based thermoplastic elastomers. Perylene‐3, 4,9, 10‐tetracarboxylic dianhydride (PDA) is encapsulated into cellulose acetate (CA) electrospun fibers, which was prepared from 12 % cellulose acetate solution, at 20 kV with a distance of 10 cm. The flow rate was 0.2 ml · h–1. These water repellent nanofibrous coatings are anticipated to serve as hydrophobic coatings. Scanning electron microscope is used to study the properties of the electrospun PDA‐CA nanofibers.  相似文献

The characteristics of tissue engineered scaffolds are major concerns in the quest to fabricate ideal scaffolds for tissue engineering applications. The polymer scaffolds employed for tissue engineering applications should possess multifunctional properties such as biocompatibility, biodegradability and favorable mechanical properties as it comes in direct contact with the body fluids in vivo. Additionally, the polymer system should also possess biomimetic architecture and should support stem cell adhesion, proliferation and differentiation. As the progress in polymer technology continues, polymeric biomaterials have taken characteristics more closely related to that desired for tissue engineering and clinical needs. Stimuli responsive polymers also termed as smart biomaterials respond to stimuli such as pH, temperature, enzyme, antigen, glucose and electrical stimuli that are inherently present in living systems. This review highlights the exciting advancements in these polymeric systems that relate to biological and tissue engineering applications. Additionally, several aspects of technology namely scaffold fabrication methods and surface modifications to confer biological functionality to the polymers have also been discussed. The ultimate objective is to emphasize on these underutilized adaptive behaviors of the polymers so that novel applications and new generations of smart polymeric materials can be realized for biomedical and tissue engineering applications.

Electrospinning is a well-known technique since 1544 to fabricate nanofibers using different materials like polymers, metals oxides, proteins, and many more. In recent years, electrospinning has become the most popular technique for manufacturing nanofibers due to its ease of use and economic viability. Nanofibers have remarkable properties like high surface-to-volume ratio, variable pore size distribution (10–100 nm), high porosity, low density, and are suitable for surface functionalization. Therefore, electrospun nanofibers have been utilized for numerous applications in the pharmaceutical and biomedical field like tissue engineering, scaffolds, grafts, drug delivery, and so on. In this review article, we will be focusing on the versatility, current scenario, and future endeavors of electrospun nanofibers for various biomedical applications. This review discusses the properties of nanofibers, the background of the electrospinning technique, and its emergence in chronological order. It also covers the various types of electrospinning methods and their mechanism, further elaborating the factors affecting the properties of nanofibers, and applications in tissue engineering, drug delivery, nanofibers as biosensor, skin cancer treatment, and magnetic nanofibers.  相似文献

The fabrication of three‐dimensional (3D) electrospun composite scaffolds was presented in this study. Layers of electrospun meshes made from composites of poly(lactide‐co‐glycolide acid) (PLGA) and hydroxyapatite (HA) were stacked and sintered using pressurized gas. Three HA concentrations of 5, 10, and 20 wt % were tested, and the addition of the HA nanoparticles decreased the tensile mechanical properties of the meshes with 20 wt % HA. However, after the gas absorption process, the fibers within the mesh sintered, which improved the mechanical properties more than twofold. The fabrication of 3D, porous, electrospun scaffolds was also demonstrated. The resulting 3D scaffolds had open porosity of up to 70% and modulus of ~20 MPa. This technique improves on the current electrospinning technology by overcoming the challenges of depositing a thick, 3D structure. © 2011 Wiley Periodicals, Inc.
