A NOVEL EFFECT OF UV-B IN A HIGHER PLANT (SORGHUM VULGARE)   总被引:2,自引:0,他引:2  
Abstract— Two non-photosynthetic photoreceptors (phytochrome and the usual blue/UV light photoreceptor) were previously found to be involved in light-mediated anthocyanin synthesis in the mesocotyl of Sorghum seedlings (Drumm and Mohr, 1978). The decisive point is that phytochrome can act only after a blue/UV light effect has occurred. On the other hand, the expression of the blue/UV light effect is controlled by phytochrome ('obligatory sequential action'). A strong positive interaction between the blue/UV-A and the UV-B part of the spectrum was found, in addition to the above sequential action: an inductive effect of blue or UV-A light can only express itself fully if short wavelength UV (approximately 300–320nm. UV-B range) is also given, either after the blue/UV-A light or simultaneously. Since even small amounts of the UV-B are strongly effective it is probable that this effect plays a role under natural conditions and may not be considered as a mere laboratory artifact.  相似文献

Two non-photosynthetic photoreceptors (phytochrome and a blue light photoreceptor) are involved in light-mediated anthocyanin synthesis in the mesocotyl of Sorghum seedlings. The present study was undertaken to investigate the kind of interaction between phytochrome and the blue light photoreceptor. The data show that phytochrome (Pfr) can only act once a blue light effect has occurred. On the other hand, the blue light effect cannot express itself without Pfr. It is concluded that there is an obligatory dependency (or sequential interaction) between the blue light effect and the light effect occurring through phytochrome, although the blue light photoreaction per se is not affected by the presence or absence of phytochrome. The latter statement is based on the results of dichromatic experiments, i.e. simultaneous, high fluence rate irradiation with two kinds of light. Blue light can be replaced by UV light. It is not clarified yet whether the effect of blue and UV light is due to the same photoreceptor.  相似文献

Abstract Anthocyanin synthesis in the hypocotyl of tomato ( Lycopersicon esculentum ) seedlings responds strongly and specifically to blue/UV light while the response to red and far-red light, operating through phytochrome, is weak. The herbicide Norflurazon (SAN 9789) was used to inhibit synthesis of colored carotenoids almost completely without affecting growth and development measurably. Even though carotenoid content was reduced to less than 2% of normal and the fluence rate response function for blue and UV light was linear within the experimental range, Norflurazon treatment did not reduce seedling sensitivity toward blue/UV light. It was concluded that at least'bulk'carotenoids are not the photoreceptor chromophore of the blue/UV photoreceptor pigment.  相似文献

The photocontrol of anthocyanin synthesis in dark-grown seedlings of tomato (Lycopersicon esculentum Mill.) has been studied in an aurea (au) mutant which is deficient in the labile type of phytochrome, a high pigment (hp) mutant which has the wild-type level of phytochrome and the double mutant au/hp , as well as the wild type. The hp mutant demonstrates phytochrome control of anthocyanin synthesis in response to a single red light (RL) pulse, whereas there is no measurable response in the wild type and au mutant. After pretreatment with 12 h blue light (BL) the phytochrome regulation of anthocyanin synthesis is 10-fold higher in the hp mutant than in the wild type, whilst no anthocyanin is detectable in the au mutant, thus suggesting that it is the labile pool of phytochrome which regulates anthocyanin synthesis. The au/hp double mutant exhibits a small (3% of that in the hp mutant) RL/far-red light (FR)-reversible regulation of anthocyanin synthesis following a BL pretreatment. It is proposed that the hp mutant is hypersensitive to the FR-absorbing form of phytochrome (Pfr) and that this (hypersensitivity) establishes response to the low level of Pfl. (below detection limits in phytochrome assays) in the au/hp double mutant.  相似文献

Abstract— The involvement of phytochrome in light-mediated anthocyanin synthesis in the mustard seedling ( Sinapis alba L.) under inductive conditions (law of reciprocity valid) was shown previously (Drumm and Mohr, 1974). In the present paper the hypothesis (Hartmann, 1966) is checked that light-mediated anthocyanin synthesis in continuous high-irradiance far-red light ('high-irradiance response') is also due exclusively to phytochrome. The data indicate that the effectiveness of the far-red light is indeed a function of total phytochrome [ Ptotal ]* and therewith [ Pfr ]*. The data are not consistent with the suggestion (Schneider and Stimson, 1972) that photosynthesis (in particular, photosystem I) is involved in the 'high-irradiance response' of photomorphogenesis.  相似文献

Abstract— In the two fungal species ( Fusarium and Neurospora ) carotenogenesis has been described as a blue light effect. Generally, phytochrome was believed to be absent in fungi. Recently, work was reported suggesting that this might not strictly be the case. Therefore, we very thoroughly tested these two organisms for any red light effect on carotenogenesis. No action of phytochrome was found, either as a 'low irradiance response' or a 'high irradiance response'. Also, no sequential interaction between blue and red light and between UV and red light was observed. Therefore, we conclude that phytochrome is not involved in carotenogenesis in Fusarium and Neurospora.  相似文献

Abstract —The dose response curve for light (phytochrome)-induced anthocyanin synthesis was determined in the mustard seedling. The curve gives the amount of anthocyanin (A) synthesized within 24 h as a function of the amount of Pfr* produced by a brief light pulse. The [Pfr] response curve is composed of two linear parts with very different slopes ( a 1,2) connected by a relatively narrow transient range (curved segment). The [Pfr] response curve extrapolates precisely through zero [Pfr]. The reciprocity law is valid over the whole range investigated (up to 320 s of irradiation). It is concluded that the initial (or primary) reaction of Pfr (Pfr+ X → PfrX) does not involve any significant cooperativity in the case of phytochrome-mediated anthocyanin synthesis. It is speculated that the linear parts of the [Pfr] response curve truly reflect the mode of phytochrome action ( A = a 1,2 [Pfr]; X does not come into play since it is not rate limiting) whereas the curved segment represents a transition of the reaction matrix of Pfr. The large difference between a1 and a2 seems to indicate that the physiological effectiveness of a given amount of Pfr (or PfrX) is determined by [Pfr] through a Pfr-induced change in the reaction matrix.  相似文献

Abstract— Experiments with irradiation sequences where red precedes far-red lead to the conclusion that, in turnip, phytochrome is the only pigment mediating anthocyanin synthesis in red and far-red. Results from experiments where far-red precedes red, however, suggest that more than one reaction is involved. A possible interpretation is that the 'high-energy' reaction in far-red and the low energy red/far-red reversible reaction are mediated by two different forms of phytochrome.

Abstract— Problems concerning the interpretation of interactions of higher plant photomorphogenetic receptors are discussed. The theory that action of a blue light photoreceptor serves only to maintain responsiveness to phytochrome (Responsiveness Theory) is demonstrated to be unable to be properly tested with present techniques. This theory is also unable to explain experimental results any better than an alternative theory that a blue light photoreceptor may require the presence of the active form of phytochrome to express its activity (Presence Theory). This tatter theory is also incapable of being fully tested. There does not appear to be an adequate current theory to explain photoreceptor interactions. Other issues discussed include the use of displacement transducers in growth studies, the induction of phytochrome-type responses by blue light, and the relative importance of the photoreceptors. New data are introduced on the effect of blue light in the end-of-day growth response to phytochrome of the light-grown Cucumis sativus L. hypocotyl, and on the light equivalence principle in the same species.  相似文献

Abstract— Irradiation of the Pr form of phytochrome in the presence of flavin mononucleotide (FMN) which absorbs the actinic blue light yields Pfr at a rate greater than that in the absence of FMN. The actinic blue light absorbed by FMN enhances the phototransformation of Pr via the energy transfer from the former to the latter. On the other hand, the photoreversion of Pfr was inhibited by the presence of FMN when illuminated with blue light. The lack of photo-enhancement of the reversion of Pr, by blue light suggests that the Pfr chromophore (acceptor) transition dipole is virtually perpendicular to the FMN transition dipole, as the result of a chromophore reorientation in the Pr→Pfr phototransformation. The fact that blue light absorbed by flavin preferentially enhances the forward phototransformation of phytochrome while inhibiting the reversion may have an important implication in the high irradiance responses in plants in terms of a preferential accumulation of Pfr by blue light excitation.  相似文献

Abstract— Using carrot cell suspension in 2,4-dichlorophenoxyacetic acid (2,4-D)-depleted culture medium, fluence-response curves for the formation of anthocyanin were determined at various wavelengths from 250 to 800 nm. In the fluence-response curves at wavelengths between 260 and 330 nm, the response showed a sharp fluence-dependent increase after the fluence exceeded threshold level at the respective wavelength. Such a sharp increase in response was not observed by light at 450 nm or longer wavelengths, although the response obtained by higher fluence of such light was always higher than that in the dark control. Action spectra determined at the sharp increasing phase of the response showed the single peak at 280 nm which equals the absorption maximum of UV-B photoreceptor.
Although red (R)-light alone had a minor effect on anthocyanin accumulation, it modulated the action of UV-B light. That is, when carrot cells were irradiated with R-light either before or after UV-B irradiation, anthocyanin formation was greatly enhanced above the level enhanced by UV-B light alone. The most effective wavelength for this enhancement was 660 nm. The effect of R-light on the anthocyanin formation of the UV-B irradiated cells was reversed by immediately following it with far-red light, suggesting the involvement of phytochrome in the R-effect.  相似文献

The UV-mediated induction of anthocyanin and UV-absorbing compounds was characterized in etiolated hypocotyls of wild-type and aurea (au) mutant tomato seedlings. Ultraviolet radiation induced significant increases of anthocyanin and UV-absorbing compounds in hypocotyls of die au mutant and of its isogenic wild-type, but the differences in the time courses of UV-induced pigment accumulation indicate mat different photoregulatory mechanisms are involved for each of these two groups of pigments. It appears mat prolonged presence of adequate levels of UVB (290–320nm) energy and consequently the action of a specific UVB photoreceptor are indispensable for the photoinduction of anthocyanin accumulation in UV-irradiated hypocotyl of the au mutant that is missing the labile phytochrome pool. The large difference found between the wild-type and the au mutant strongly indicate the involvement of labile phytochrome as the primary functional photoreceptor for the photoinduction of anthocyanin accumulation in wild-type tomato hypocotyls. The UVB photoreceptor could at least partly replace the action of labile phytochrome (as far as anthocyanin accumulation is concerned) when the functional phytochrome pool is missing as in the au mutant. The general picture of UV-mediated induction of total UV-absorbing compounds shows only a macroscopic difference between wild-type and die au mutant of tomato: the higher initial level (in darkness) of these compounds in die wild-type in contrast to the au mutant. Although there is UV-induced accumulation of UV-absorbing compounds in bom genotypes, the levels in the au mutant never reach mat of the wild-type under the same UV exposure. A UVB photosensor may play a more important role in the photoinduction of UV-absorbing compounds. Indeed, in the absence of labile phytochrome, i.e. in the au mutant, a UVB-absorbing photoreceptor alone is able to establish high responsiveness for the UV-induced flavonoid accumulation.  相似文献

Abstract— Kinetic studies with the mustard seedling ( Sinapis alba L.) support the hypothesis that the so-called 'high energy reaction' of photomorphogenesis can be understood solely on the basis of phytochrome. Light-induced anthocyanin synthesis (a typical 'positive' photoresponse(1) and light dependent inhibition of hypocotyl lengthening (a typical 'negative' photoresponse(1)) have been investigated. In order to explain the experimental data we have to assume that there are two different types of phytochrome 730 which differ greatly as far as their resistance to irreversible destruction is concerned. The existence of these two different types of phytochrome 730 has already been postulated on the basis of spectrophotometric measurements in vivo .(2)  相似文献

Abstract— The effects of red, blue and green light pulses on total protein accumulation in the green alga Ulva rigida following transfer from a low to high nitrate medium in darkness were examined. Red light pulses prior to transfer to darkness increased protein accumulation by about 55%. Blue and green light pulses also stimulated protein accumulation, but to a lesser extent (40-30% respectively). Stimulation of protein accumulation by red, blue or green light was largely (red light) or partially (blue or green light) reversible by far-red. The role of phytochrome and a red/green photoreversible system in the control of protein synthesis is discussed.  相似文献

PHYSIOLOGICAL CHARACTERIZATION OF A HIGH-PIGMENT MUTANT OF TOMATO   总被引:4,自引:0,他引:4  
Abstract— A high-pigment (hp) mutant, which shows exaggerated phytochrome responses and three other genotypes of Lycopersicon esculenrum Mill. cv. Ailsa Craig: the aurea (au) mutant deficient in the bulk light-labile phytochrome (PI) pool, the au, hp double mutant, and their isogenic wild type, were used in this study. Measurements of phytochrome destruction in red light (R) revealed that the exaggerated responses of the hp mutant are not caused by a higher absolute phytochrome level or a reduced rate of phytochrome destruction. Fluence-response relationships for anthocyanin synthesis after a blue-light pretreatment were studied to test if the hp mutant conveys hypersensitivity to the far-red light (FR)-absorbing form of phytochrome (Pfr), i.e. the threshold of Pfr required to initiate the response is lower. The response range for the hp mutant and wild type was identical, although the former exhibited a 6-fold larger response. Moreover, the kinetics of anthocyanin accumulation in continuous R were similar in the wild-type and hp-mutant seedlings, despite the latter accumulating 9-fold more anthocyanin. Since the properties of phytochrome are the same, the hp mutation appears to affect the state of responsiveness amplification, i.e. the same amount of Pfr leads to a higher response in the hp mutant. We therefore propose that the hp mutation is associated with an amplification step in the phytochrome transduction chain. Escape experiments showed that the anthocyanin synthesis after different light pretreatments terminated with a R pulse was still 50% FR reversible after 4–6 h darkness, indicating that the Pfr pool regulating this response must be relatively stable. However, fluence-rate response relationships for anthocyanin synthesis and hypocotyl growth induced by a 24-h irradiation with 451, 539, 649, 693, 704 and 729 nm light showed no or a severely reduced response in the au and au, hp mutants, suggesting the importance of PI in these responses. We therefore propose that the capacity for anthocyanin synthesis (state of responsiveness amplification) could be established by PI, while the anthocyanin synthesis is actually photoregulated via a stable Pfr pool. The Hp gene product is proposed to be an inhibitor of the state of responsiveness amplification for responses controlled by this relatively stable Pfr species.  相似文献

Four genotypes of tomato (Lycopersicon esculentum Mill.) in the genetic background ‘Aha Craig’ were used: an aurea (au) mutant, deficient in the bulk light-labile phytochrome pool; a high pigment (hp) mutant, showing exaggerated phytochrome responses at the time of de-etiolation; the au,hp double mutant and the isogenic wild type (WT). A dramatic increase in plant height resulting from an increase in the length of all internodes for each of the genotypes studied was observed upon reduction of the red light: far-red light photon ratio (R:FR) from 6.90 to 0.13 by addition of FR for the whole photoperiod. A concomitant increase in leaf length was also observed. Since au and au,hp mutants, deficient in the bulk light-labile phytochrome pool, respond to this reduction in the R:FR these data demonstrate that the phytochrome pool that mediates this response is present and fully functional. Anthocyanin was detectable in the comparably developed young growing leaves of the WT and hp mutant under the high R:FR, but not in the au and au,hp mutants, suggesting that the potential for anthocyanin synthesis is correlated with the presence of the bulk light-labile phytochrome pool. The kinetics of anthocyanin decrease in the young growing leaves were investigated in the hp mutant and the results suggest a very rapid cessation of flavonoid biosynthesis upon reduction of the R:FR. The functions of different phytochrome types are discussed.  相似文献

The intracellular localization and orientation of the receptors for the blue light-induced phototropism in the fern Adianrum protonemata, phytochrome and the blue light-absorbing pigment, were investigated by combining the techniques of cell centrifugation and of microbeam irradiation with linearly polarized light. The phototropic response was induced in the cells even after they had been centrifuged basipetally to spin down the endoplasm from the apical region. When a polarized blue microbeam was given to a flank of the apical region of the protonema, the phototropic response after compensation of phytochrome effect by far-red light was most effectively induced when the polarization plane was parallel to the long axis of the cell. If protonemata were pre-irradiated with blue and far-red light, the phototropic response was mediated through phytochrome alone. If such pre-irradiated protonemata were similarly irradiated with a polarized blue microbeam, polarized light vibrating parallel to the cell axis was again most effective in inducing the response. These results indicate that both the blue light-absorbing pigment and the phytochrome responsible for the blue light-induced phototropism in Adiantum are confined to the plasma membrane and/or the ectoplasm and that the transition moments of their blue-absorption bands are nearly parallel to the cell surface.  相似文献

Abstract —Anthocyanin synthesis in cabbage and mustard seedlings depends upon duration and irradiance of the light treatment. The relative effectiveness of radiation in various spectral regions depends upon the length of the irradiation and decreases with increasing dose. In intermittent light treatments, far-red light can reverse the promoting action of red light if the dark interval between successive irradiations is longer than one hour. If the length of the dark interval is less than one hour, far-red applied immediately after each red irradiation, enhances anthocyanin accumulation. Anthocyanin accumulation under various light treatments seems to correlate, to some extent, with the rate of phytochrome decay, but not with chlorophyll production. Anthocyanin accumulation is inhibited by 2,4-dinitrophenol and by the ammonium ion, but not by DCMU. The ammonium ion inhibits anthocyanin accumulation induced by a single, short red irradiation. This suggest that the ammonium ion may have a wider spectrum of action in vivo than in chloroplast preparations where it acts as a specific uncoupler of photophosphorylation. Streptomycin inhibits chlorophyll synthesis and enhances anthocyanin accumulation. These results suggest that there is very little, if any, interaction between photosynthesis and ‘high-irradiance-reaction’ anthocyanin synthesis in cabbage and mustard seedlings.  相似文献

Abstract— Light-induced anthocyanin formation in Zea mays L. coleoptiles was investigated in seven different varieties of this species. Under the test conditions, four varieties showed practically no response to any waveband used (UV, continuous red and continuous far-red), two responded strongly to both UV and far-red, and one showed a strong response only to far-red. The radiation-sensitive varieties showed, however, only a very weak response to continuous red light. In those varieties sensitive to far-red light, a pretreatment with continuous red light led to a greatly enhanced response to UV or in one case the manifestation of a response to UV that was previously lacking. Further investigations in one radiation-sensitive variety (INRA) showed that the UV response was to UV-B radiation below 350 nm. The UV response, as well as the far-red and blue responses in this variety, showed fluence-rate dependency. Red light was almost ineffective and showed only a very weak fluence-rate dependency.  相似文献

Using 290-nm light, which excites only a UV-B photoreceptor, and 385- and 660-nm light, which activate only phytochrome, the fluence rate-response curves of monochromatic irradiations for anthocyanin synthesis in the first internodes of broom sorghum (Sorghum bicolor Moench, cv. Acme Broomcorn) were analyzed. Although the two photoreceptors absorbed light independently, they multiplicatively increased the action of each other. Accordingly, when the fluence rates of both wavelengths were changed together, the resulting slopes of the fluence rate-response curves of double-log plots were steep compared with the slopes obtained with the respective monochromatic irradiations. The slopes of fluence rate-response curves for monochromatic irradiations at 325 to 345 nm were steeper than those at other wavelengths. This difference was shown to be due to the multiplicative actions of both photoreceptors.  相似文献
