GC-MS analyses of the essential oils of leaves, stems and flower of Nasturtium officinale resulted in the identification of 9, 8 and 15 compounds, representing 97%, 100% and 94.7% of the oils, respectively. The main compounds of the oil of leaves were myristicin (57.6%), α-terpinolene (8.9%) and limonene (6.7%). Caryophyllene oxide (37.2%), p-cymene-8-ol (17.6%), α-terpinolene (15.2%) and limonene (11.8%) were the main components in stems, whereas limonene (43.6%), α-terpinolene (19.7%), p-cymene-8-ol (7.6%) and caryophyllene oxide (6.7%) were the major constituents in the oil of flowers. All the samples were subjected to a screening for their possible antioxidant activities using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene-linoleic acid assays. In the above tests, methanol extracts of leaves showed higher antioxidant activity than the oils and methanol extracts of stems and flowers.  相似文献

The chemical composition of the essential oils, antioxidant activity (DPPH and beta-carotene/linoleic acid assays) and total phenolic content (Foline-Ciocalteu) of the flowers and leaves of Salvia reuterana were determined. Essential oils extracted from the flowers and leaves by hydrodistillation were analyzed by GC and GC/MS. Forty-four constituents, representing 99.7-99.9% of the oils, were identified. The major components were germacrene D, benzoic acid hexyl ester, bicyclogermacrene, beta-gurjunene and ishwarene, constituting 33.7-31.9% of the oils. The highest radical-scavenging activity (DPPH test) was shown by the methanol extract of the flowers (IC50 = 77.6 microg/mL). In the beta-carotene/linoleic acid assay, the methanol extract of the leaves showed the highest inhibition (40.3%) which was only slightly lower than that shown by BHT (82.9%). The total phenolic contents of the methanol extracts of the flowers and leaves as gallic acid equivalents were 81.4 and 88.3 microg/mg, respectively. The plant also showed good antimicrobial activity against three strains of tested microorganisms.  相似文献

Essential oils of the leaves and flowers of Anthemis cotula L. (family Asteraceae) grown in Jordan were extracted by hydro-distillation and then analyzed by GC–MS. Sesquiterpenes hydrocarbons (SH) were the dominant components in the oils extracted from leaves and flowers of A. cotula. γ-Muurolene and aromadendrene, were the major compounds that were obtained from the flowers oil, while γ-muurolene and trans-cadinene ether were detected as major ingredients in the leaves extract. LC-MS analysis was carried out to identify the significant compounds from each extract. Additionally, butanol (B), aqueous methanol (M) and water (W) extracts prepared from the flowers and the leaves of A. cotula were analysed by LC-MS/MS. Apigenin and chlorogenic acid were the main constituents detected in the flowers’ alcoholic extracts and leaves’ aqueous extract. Moreover, the essential oils and all prepared extracts were assayed for their total antioxidant activity using the DPPH, ABTS, and ferrous ion chelating effect (FIC) assay methods. All investigated oils and extracts showed interesting activity as compared to the positive controls employed (α-tocopherol and ascorbic acid).  相似文献

The leaves and twigs of Piper krukoffii, collected in the Carajás National Forest, north Brazil, yielded essential oils (2.0% and 0.8%), the main constituents of which were myristicin (40.3% and 26.7%), apiole (25.4% and 34.1%) and elemicin (2.8% and 3.0%). The antioxidant activities of the oils, methanol extract and its sub-fractions were evaluated. The DPPH EC50 values varied from the ethyl acetate sub-fraction (73.4 +/- 3.7 microg/mL) to the methanol extract (24.9 +/- 0.8 microg/mL), and the ABTS TEAC values ranged in the same order from 265.7 to 349.2 microMol TE/g. These results indicated a significant antioxidant activity for the plant. The lignans (-)-kusunokin, yatein, (-)-hinokin and cubebin were identified in the methanol extract. The hydro-methanolic sub-fraction showed a high value for total phenol content (106.5 +/- 0.7 mg GAE/g), as well as 1H NMR signals for sugar moieties. Crude extracts and sub-fractions were also able to inhibit beta-carotene bleaching, varying from 22.4 to 47.1%. The oils from the leaves and twigs showed strong larvicidal (21.4 and 3.6 microg/mL) and fungicide (0.5 and 0.1 microg/mL) activities.  相似文献

This study was designed to evaluate the in vitro antioxidant activities of n-hexane (Hex), dichloromethane (DCM), methanol (MeOH) and essential oils (EO) extracts obtained from Salvia euphratica var. euphratica and Salvia euphratica var. leiocalycina and to determine their essential oil and phenolic acid compositions. The samples were screened for their antioxidant activity by using DPPH and β-carotene/linoleic acid assays. Methanol extracts of both varieties exhibited strong antioxidant activities. Our results showed that rosmarinic acid was dominant phenolic acid of MeOH extracts (39.4 and 55.8?μg?mg?1, respectively). The chemical compositions of essential oils of two varieties were analysed and their main components were determined as eucalyptol (18.4%) and trans-pinocarvyl acetate (24.9%), respectively. It can be said that these varieties could be used as natural antioxidant.  相似文献

Aerial parts essential oil of Nepeta betonicifolia and N. saccharata were obtained by hydrodistillation and analysed by GC-FID and GC-MS. Thirty-three and eighteen components represented 97.9% and 98.2% of the total oils identified, respectively. Main compounds of the oil of N. betonicifolia were 4aα,7β,7aα-nepetalactone (42.0%), germacrene D (6.0%), triplal (5.2%), 1-nor-bourbonanone (4.0%) and 1,8-cineole (3.2%). The principal constituents of the essential oil of N. saccharata were found to be 4aβ,7α,7aβ-nepetalactone (66.9%), germacrene D (12.9%), sabinene (6.5%) and trans-caryophyllene (3.3%). The radical scavenging capacity (RSC) of methanol extracts and chloroform, butanol and water subfractions of aerial parts of N. betonicifolia and N. saccharata were evaluated by using DPPH, FRAP and ABTS assays. TPC of each extract was measured using Folin-Ciocalteau. The antioxidant activity of the butanolic subfractions of both plants was higher than other extracts examined.  相似文献

Extra-virgin olive oils contain many bioactive substances that are phenolic compounds. The survival of Arcobacter-like strains in non-buffered (WEOO) and buffered (BEOO) extracts of olive oils were studied. Time kill curves of different strains were measured in the environment of olive oil extracts of different grades. The activity of the extracts was also monitored for biofilm formation using the Christensen method. In vitro results revealed that extra-virgin olive oil extracts exhibited the strongest antimicrobial effects, especially non-buffered extracts, which exhibited strain inhibition after only 5 min of exposure. The weakest inhibitory effects were observed for olive oil extracts. A decrease in biofilm formation was observed in the environment of higher WEOO concentrations, although at lower concentrations of extracts, increased biofilm formation occurred due to stress conditions. The dialdehydic forms of oleuropein derivatives, hydroxytyrosol, and tyrosol were the main compounds detected by HPLC-CoulArray. The results indicate that not all olive oils had a similar bactericidal effect, and that bioactivity primarily depended on the content of certain phenolic compounds.  相似文献

The objective of this study was to develop a consecutive preparation method for the isolation and purification of hainanmurpanin, meranzin, and phebalosin from leaves of Murraya exotica L. The process involved supercritical fluid extraction with CO2, solvent extraction, and two‐step high‐speed countercurrent chromatography. Pressure, temperature, and the volume of entrainer were optimized as 27 MPa, 52°C, and 60 mL by response surface methodology in supercritical fluid extraction with CO2, and the yield of the crude extracts was 7.91 g from 100 g of leaves. Subsequently, 80% methanol/water was used to extract and condense the three compounds from the crude extracts, and 4.23 g of methanol/water extracts was obtained. Then, a two‐step high‐speed countercurrent chromatography procedure was developed for the isolation of the three target compounds from methanol/water extracts, including conventional high‐speed countercurrent chromatography for further enrichment and consecutive high‐speed countercurrent chromatography for purification. The yield of concentrates from high‐speed countercurrent chromatography was 2.50 g from 4.23 g of methanol/water extracts. Finally, the consecutive high‐speed countercurrent chromatography produced 103.2 mg of hainanmurpanin, 244.7 mg of meranzin, and 255.4 mg of phebalosin with purities up to 97.66, 99.36, and 98.64%, respectively, from 900 mg of high‐speed countercurrent chromatography concentrates in one run of three consecutive sample loadings without exchanging a solvent system.  相似文献

依次采用石油醚、环己烷、二硫化碳、丙酮、甲醇和等体积的丙酮/二硫化碳混合溶剂对白音华褐煤(BL)进行分级萃取得到萃取物(E_1-E_6),利用甲醇、甲苯和等体积的甲醇/甲苯混合溶剂对萃取残渣进行连续热溶得到热溶物(SP_1-SP_3)。利用傅里叶变换红外光谱(FT-IR)、气相色谱质谱(GC/MS)和实时直接分析-质谱(DART-MS)对萃取物和热溶物的组成和结构特征进行了分析。BL总的萃取物产率和热溶物产率分别为9.37%和21.84%。E_5和E_6的FT-IR谱图中有较强的羟基吸收峰,而E_1的FT-IR谱图中脂肪C-H的伸缩振动峰强度明显高于其他萃取物;三种溶剂热溶物的FT-IR谱图相似,但SP_1和SP_3的FT-IR谱图中羟基吸收峰较强。E_1和E_5中GC/MS可检测化合物分别以烷烃和芳烃为主,E_5中还含有较多的含氧化合物;三种热溶物中化合物均以烷烃和芳烃为主,SP_3中酚类、酮类和酯类等含氧化合物的含量较高。DART-MS可检测出萃取物和热溶物中GC/MS无法检测出的较强极性和难挥发化合物;热溶物中含量较高的化合物的碳数和等价双键数(Double bond equivalent,DBE)分别集中分布在10-25和2-16,并且DBE随碳数增加而增加。  相似文献

建立了超声提取-液相色谱-电喷雾三重串联四极杆质谱测定玉米、大米、大豆等粮油固体样品中黄曲霉毒素B1、B2、G1和G2(AFB1、AFB2、AFG1和AFG2)的方法。分析前对样品进行超声提取,优化得到最佳超声提取条件: 溶剂为甲醇-水(含40 g/L NaCl) (80:20, v/v)溶液、料液比为1:3(g:mL)、温度为50 ℃、时间为3 min。然后对提取的样品进行免疫亲和特异性净化。最后与液相色谱-电喷雾三重串联四极杆质谱联用,使用C18反相色谱柱,流动相为甲醇-10 mmol/L乙酸铵水溶液梯度洗脱,以黄曲霉毒素M1(AFM1)作为内标进行定量测定。结果表明,AFB1、AFB2、AFG1和AFG2的检出限分别为0.002、0.004、0.004和0.012 μg/kg。方法的加标回收率为87%~111%,日内相对标准偏差(RSD)和日间RSD分别不大于6.7%和5.6%。实验结果表明该方法可以有效地降低基质效应的影响,相比于外标法能极大地提高方法的准确度。  相似文献

Extracts from acacia, chestnut, cherry, mulberry, and oak wood, used in making barrels for aging wine and spirits were studied by GC/MS positive ion chemical ionization (PICI). Wood chips were extracted by a 50% water/ethanol solution and a tartrate buffer pH 3.2-12% ethanol (model wine) solution. The principal compounds identified in extracts were guaiacol-containing aldehydes and alcohols, such as benzaldehyde and derivatives, vanillin and syringaldehyde, cinnamaldehyde and coniferaldehyde, eugenol and methoxyeugenol, guaiacol and methoxyguaiacol derivatives. PICI using methane as reagent gas produced a high yield of the protonated molecular ion of volatile phenols, compound identification was confirmed by collision-induced-dissociation (CID) experiments on [M + H](+) species. MS/MS fragmentation patterns were studied with standard compounds: guaiacol-containing molecules were characterized by neutral methyl and methanol losses, benzaldehyde derivatives by CO loss. Acacia wood extracts contained significant syringaldehyde and anisaldehyde, but no eugenol and methoxyeugenol. Significant syringaldehyde, eugenol and methoxyeugenol, and high vanillin were found in chestnut and oak wood extracts; low presence of volatile benzene compounds was found in mulberry wood extracts. Cherry wood extracts were characterized by the presence of several benzaldehyde derivatives and high trimethoxyphenol.  相似文献

Commercial solid-phase extraction (SPE) cartridges with C18 bonded silica packings effectively cleaned up acetonitrile extracts of 3-g samples of fats and oils for determination of organophosphorus pesticide residues by gas chromatography with flame photometric detection. Cartridges from three different sources were tested and found to differ in lipid capacity and inertness (free silanol activity). Consequently, the amount of packing (i.e., number of cartridges) and/or the choice of eluent used were adjusted for each brand of cartridge to achieve optimum clean-up and analyte recovery. Seven pesticides with a wide range of polarity (acephate, azodrin, chlorpyrifos, diazinon, malathion, methamidophos and methyl parathion) were separated from coextracted lipids by elution with either acetonitrile or methanol, depending on the brand of cartridge used. Cartridges were regenerated by purging lipids with dichloromethane and were reused numerous times without apparent loss of effectiveness. Recoveries from vegetable oils and butterfat fortified with the seven compounds at levels of 0.05–0.87 μg g?1 ranged from 80 to 103%. Practical limits of determination range from 0.01 to 0.08 μg g?1, depending on analyte response.  相似文献

The volatile components of essential oil (EO), SPME, and SPME of solvent extracts ( n -hexane, methanol, and water) obtained from fresh Serapias orientalis subsp. orientalis ( Soo ) were analyzed by GC-FID/MS. EO of Soo gave 11 compounds in the percentage of 99.97%; capronaldehyde (37.01%), 2-( E )-hexenal (23.19%), and n -nonanal (19.05%) were found to be major constituents. SPME GC-FID/MS analyses of fresh plant and solvent extracts of Soo revealed 7, 12, 7, and 4 compounds within the range of 99.7% to 99.9%. Limonene (76.5%, 41.7%, and 61.3%) was the major compound in SPMEs of the n -hexane and methanol extracts. α -Methoxy- p -cresol (52.9%) was the main component in its water extract. The antimicrobial activity of EO and the solvent extracts of Soo were screened against 9microorganisms. EO showed the best activity against Mycobacterium smegmatis , with 79.5 µg/mL MIC value. The n -hexane, methanol, and water extracts were the most active against the Staphylococcus aureus within the range of 81.25–125.0 µg/mL (MIC). IC 50 values for the lipase enzyme inhibitory activity of EO and solvent extracts ( n -hexane, methanol, and water) were determined to be 59.87 µg/mL, 64.03 µg/mL, 101.91 µg/mL, and 121.24 µg/mL, respectively.  相似文献

建立了气相色谱-质谱(GC-MS)同时测定植物油中α-、β-、γ-、δ-生育酚和α-、β-、γ-、δ-生育三烯酚等8种维生素E的分析方法。植物油样品经甲醇超声提取、浓缩、定容,在分时段选择离子监测(SIM)模式下分离分析,采用外标法进行定量。结果表明,8种维生素E可实现基线分离;在0.01~1 mg/L范围内,所有目标物均呈良好线性关系,相关系数均大于0.99;检出限和定量限分别为0.03~0.25 mg/kg和0.10~0.83 mg/kg;在芝麻油中分别添加10、50和250 mg/kg 3个水平的8种维生素E进行加标试验,平均回收率为87.5%~107.4%,相对标准偏差(RSD)≤ 7.5%。所建立的方法简单、准确、可靠,且灵敏度高,可用于测定植物油中8种维生素E的含量。采用上述方法对芝麻油、大豆油、菜籽油、葵花籽油、花生油、玉米油和棕榈油等7种共75个植物油样品中维生素E的含量进行测定。结果显示,芝麻油与其他6种植物油中的8种维生素E的组成和含量均有显著差异性,因此该方法可作为芝麻油掺入其他植物油的特征鉴定指标。  相似文献

Summary RPHPLC methods for analysis of photosynthetic pigments (chlorophylls and carotenoids) usually require addition of water to methanol or acetone extracts to prevent distortion of early-eluting peaks corresponding to the more polar compounds. In this work we have investigated the short-(<2 min) and long-term (up to 48 h) effect of adding water to acetone and methanol extracts from two marine phytoplankton species,Emiliania hyxleyi andDunaliella tertiolecta. Solvent extracts were prepared and separated into fractions that were subsequently diluted with water to 90%, 80%, 70%, 60%, 50%, and 40% for methanol, and the same range extended to 30% and 20% for acetone. Changes in pigment concentration with time were followed spectrophotometrically and chromatographically. Losses of pigments as a result of precipitation were clearly observed immediately after dilution of acetone extracts to 60% or less and methanol extracts to 80% or less. For chlorophyll a the most substantial losses were recorded for 50% acetone (up to 27% decrease) and for 70% methanol (31% decrease). This effect increased considerably with time. Only for 90% and 80% acetone were the initial concentrations of all the pigments unchanged after 24h, and even up to 48 h. In contrast, more than 60% and 57% of the initial amounts of chlorophyll a were lost after 24 h in 50% acetone and 70% methanol extracts, respectively. These losses increased to 83% and 60% after 48 h. There was a clear correlation between the polarity of a pigment and the polarity of the solvent at which maximum precipitation occurred. Losses of pigment from pure acetone and methanol extracts with time were also observed, although we attribute these to pigment degradation rather than precipitation. Some of the losses occurring with time can be avoided by use of autosamplers in which the sample can be mixed with water immediately before injection.  相似文献

Production of biodiesel from pure oils through chemical conversion may not be applicable to waste oils/fats. Therefore, enzymatic conversion using immobilized lipase based on Rhizopus orzyae is considered in this article. This article studies this technological process, focusing on optimization of several process parameters, including the molar ratio of methanol to waste oils, biocatalyst load, and adding method, reaction temperature, and water content. The results indicate that methanol/oils ratio of 4, immobilized lipase/oils of 30 wt% and 40°C are suitable for waste oils under 1 atm. The irreversible inactivation of the lipase is presumed, and a stepwise addition of methanol to reduce inactivation of immobilized lipases is proposed. Under the optimum conditions the yield of methyl esters is around 88–90%.  相似文献

The methanolic and aqueous extracts of Garcinia kola seeds were screened for their anti-Vibrio activities against 50 Vibrio isolates obtained from wastewater final effluents in the Eastern Cape Province, South Africa. The crude extracts at 10 mg/mL exhibited appreciable inhibitory activities against most of the test Vibrio isolates, with zones of inhibition ranging from 10-19 mm for methanol extract and 8-15 mm for the aqueous extracts. The minimum inhibitory concentrations (MIC) of the methanol extract varied from 0.313 to 2.5 mg/mL while that for the aqueous extract was 10 mg/mL for all the susceptible Vibrio isolates. Rate of kill assay of the methanolic extracts against three selected Vibrio species showed bacteriostatic activities against all of them achieving 58% and 60% (Vibrio vulnificus AL042); 68% and 69% (Vibrio parahaemolyticus AL049); and 70% and 78% (Vibrio fluvialis AL040) killing of the test bacteria at 3× and 4 ×MIC values, respectively, after 2 h exposure time. We conclude that Garcinia kola seeds hold promise as a potential source of therapeutic compounds of relevance in Vibrio infections management.  相似文献

In 1981 Spain went through a unique epidemic associated with a food-borne vector, affecting more than 20,000 people with over 800 deaths, which came to be known as the Toxic Oil Syndrome (TOS). Early epidemiological studies showed a link between this illness and the ingestion of rapeseed oil denatured with 2% aniline. This oil, originally aniline-denatured for industrial use, was fraudulently processed in an attempt to remove free aniline, and marketed as edible oil. Fatty acid anilides (FAA), monoesters and diesters of 3-(N-phenylamino)-1,2-propanediol (PAP) are present in oil samples as they arise in the refining process from reactions of aniline with constituent fatty acids and triglycerides of the oil matrix and are the only extraneous compounds found in these samples. To expand the search for the causative agents in TOS-associated oils and to look for new aniline-related compounds, an exhaustive characterization of laboratory-processed oils was undertaken. These oils, in the presence of aniline doped with 14C labelled aniline, were submitted to the laboratory conditions required for the generation of PAPs and FAAs. Laboratory-generated oil samples were submitted to a liquid-liquid extraction procedure to remove the unreacted aniline. The extract was processed by double solid-phase extraction to improve detection limits for minor amine-containing compounds in oils. The extracts enriched in aniline derivatives were submitted to on-line HPLC-UV-APCI-MS. Using two-dimensional ion maps, the components of several families of derivatives were readily identified. Additionally, the extracts were also fractionated by HPLC-UV and the fractions were analyzed by HPLC-APCI-MS/MS to obtain structural information. Standards of some of these compounds were synthesized and analyzed to confirm the results. A total of 115 aniline derivatives from 9 aniline-related families were identified in these oil samples. These included fatty acid anilides and an extensive array of phenylaminopropanediol esters distributed in eight major compound classes.  相似文献

Phenolic compounds in Sicilian olive oils were analyzed by GC-MS and GC-MS/MS after extraction with methanol:water 80:20 and derivatization with bis(trimethylsilyl)trifluoracetamide and trimethylchlorosilane (BSTFA:TMCS 99:1). Numerous compounds were detected and 23 were identified. Tyrosol, hydroxytyrosol and the decarbomethoxy ligstroside and oleuropein aglycons in the dialdehydic forms were the most abundant compounds. 4-(Acetoxyethyl)-1-hydroxybenzene, 3,4-dihydroxyphenylacetaldehyde, syringaldehyde and the cis form of ferulic acid were identified: these compounds were not found in olive oils before. The presence of the dialdehydic form of elenolic acid (without carbomethoxy group) linked to 3-methoxy-4-hydroxyphenylethanol was hypothesized. There were quantitative differences in oils from the varieties Nocellara del Belice, Santagatese and Cerasuola; these differences could depend on the olive varieties and ripeness.  相似文献

This paper analyzed thiophenic compounds in Sunniland oils and source rock extractsfrom the South Florida Basin, USA using GC-MS, and compared them with those of oilsfrom shales. We found that oils derived from carbonates versus shales have different distribu-tions of thiophenic compounds. Used in conjunction with such other geochemical parametersas n--alkane, sterane and terpane compositions, thiophenic compounds will be a useful toolfor oil--oil and oil--source correlations.  相似文献
