采用超负荷电动供给(electrokinetic supercharging, EKS)预浓缩技术,在微芯片电泳(MCE)上对脱氧核糖核酸(DNA)片段进行浓缩和分离。EKS是集合样品电动进样(EKI)和过渡等速电泳(tITP)的一种在线浓缩方法。研究表明:采用该方法后,在40.5 mm长的单通道芯片上能够实现对低浓度样品的大量进样、浓缩和基线分离。在普通的紫外检测条件(检测波长为260 nm)下,对DNA片段的平均检出限(S/N=3)约为0.07 mg/L,仅为十字芯片上的微芯片电泳检出限的1/40。本文还对浓缩过程中的一些关键因素和定性分析进行了探讨。  相似文献

A flow injection analysis (FIA) procedure for the determination of free chlorine in industrial formulations and water samples is proposed. The manifold is provided with a gas-diffusion unit which permits the removal of interfering species and also the preconcentration of chlorine. The determination of chlorine is performed on the basis of the oxidation by o-dianisidine as a chromogenic reagent to a coloured product which can be monitored at 445 nm. The method (for a preconcentration step of 60 s) is linear over the range 0.04-1.00 mg l(-1) of chlorine, the limit of detection is 0.04 mg l(-1), the reproducibility of the procedure (as RSD of the slope) is 3.7% for a series of four independent calibrations, the precision (as RSD of a series of 30 continuous FIA peaks of 0.56 mg l(-1) of chlorine) is 1.4% and the sample throughput is 40 h(-1). A detailed comparative study of the analytical characteristics of a single mono-channel reverse FIA assembly and the same system but provided with a Fluoropore membrane filter of 0.5 microm pore size was performed to check the advantages of the new approach in terms of sensitivity, selectivity and limit of detection.  相似文献

The concentrations of Cd, Co, Cu, Mn, Ni, Pb, and Zn in natural and sea waters are too low to be directly determined with by flame atomic absorption spectrometry (FAAS) or graphite furnace atomic absorption spectrometry (GFAAS). Specific sample preparations are requested that make possible the determination of these analytes by preconcentration or extraction. These techniques are affected by severe problems of sample contamination. In this work Cd, Co, Cu, Mn, Ni, Pb, and Zn were determined by inductively coupled plasma mass spectroscopy (ICP-MS) or by atomic absorption spectrometry, in fresh and seawater samples, after on-line preconcentration and following solvent elution with a flow injection system. Bonded silica with octadecyl functional group C18, packed in a microcolumn of 100-μl capacity, was used to collect diethyldithiocarbamate complexes of the heavy metals in aqueous solutions. The metals are complexed with a chelating agent, adsorbed on the C18column, and eluted with methanol directly in the flow injection system. The methanolic stream can be addressed to FAAS for direct determination of Cu, Ni, and Zn, or collected in a vial for successive analysis by GFAAS. The eluted samples can be also dried in a vacuum container and restored to a little volume with concentrated HNO3and Milli-Q water for analysis by ICP-MS or GFAAS.  相似文献

An automated system with a C(18) bonded silica gel packed minicolumn is proposed for spectrophotometric detection of arsenic using flow-injection hydride generation following sorbent extraction preconcentration. Complexes formed between arsenic(III) and ammonium diethyl dithiophosphate (ADDP) are retained on a C(18) sorbent. The eluted As-DDP complexes are merged with a 1.5% (w/v) NaBH(4) and the resulting solution is thereafter injected into the hydride generator/gas-liquid separator. The arsine generated is carried out by a stream of N(2) and trapped in an alkaline iodine solution in which the analyte is determined by the arsenomolybdenum blue method. With preconcentration time of 120 s, calibration in the 5.00-50.0 mug As l(-1) range and sampling rate of about 20 samples h(-1) are achieved, corresponding to 36 mg ADDP plus 36 mg ammonium heptamolybdate plus 7 mg hydrazine sulfate plus 0.7 mg stannous chloride and about 7 ml sample consumed per determination. The detection limit is 0.06 mug l(-1) and the relative standard deviation (n=12) for a typical 17.0 mug As l(-1) sample is ca. 6%. The accuracy was checked for arsenic determination in plant materials from the NIST (1572 citrus leaves; 1573 tomato leaves) and the results were in agreement with the certified values at 95% confidence level. Good recoveries (94-104%) of spiked tap waters, sugars and synthetic mixtures of trivalent and pentavalent arsenic were also found.  相似文献

A preconcentration and determination method for humic and fulvic acids at trace levels in natural water samples was developed. Cloud point extraction was successfully employed for the preconcentration of humic acid (HA) and fulvic acid (FA) prior to the determination by using a flow injection (FI) system coupled to a spectrophotometric UV-Vis detector. The quantitative extraction of HA and FA within the pH range 1-12 was obtained by neutralization of the anionic charge on the humic substances with a cationic surfactant, hexadecyltrimethylammonium bromide (CTAB). This generated a hydrophobic species that was subsequently incorporated (solubilized) into the micelles of a non-ionic surfactant polyethylene glycol, tert-octylphenyl ether (Triton X-114). The FI method for HA and FA determination was developed by injection of 100 microl of the extracted surfactant-rich phase using an HPLC pump with spectrophotometric detection at 350 nm. A 50 ml sample solution preconcentration allowed an enrichment factor of 167. The limit of detection (LOD) obtained under the optimal conditions was 5 microg l(-1). The precision for ten replicate determinations at 0.2 mg l(-1) HA was 3.1% relative standard deviation (RSD), calculated from the peak heights. The calibration using the preconcentration system for HA and FA was linear with a correlation coefficient (r2) of 0.9997 at levels near the detection limits up to at least 1 mg l(-1). The method was successfully applied to the determination of HA and FA in natural water samples (river water).  相似文献

The research adopted a single-channel microchip as the probe, and focused electrokinetic injection combined with transient isotachophoresis preconcentration technique on capillary electrophoresis microchip to improve the analytical sensitivity of DNA fragments. The channel length, channel width and channel depth of the used microchip were 40.5 mm, and 110 and 50 μm, respectively. The separation was detected by CCD (charge-coupled device) (effective LENGTH=25 mm, 260 nm). A 1/100 diluted sample (0.2 mg/l of each DNA fragment) of commercially available stepladder DNA sample could be baseline separated in 120 s with S/N=2–5. Compared with conventional chip gel electrophoresis, the proposed method is ideally suited to improve the sensitivity of DNA analysis by chip electrophoresis.  相似文献

Ion exchange is incorporated in flow injection analysis by using a resin column in the sample loop of an electronically operated proportional injector. Effects of sample aspiration rate, sampling time, eluting agent concentration, pumping rate of the sample carrier stream, resin column size and sample acidity, were investigated to develop a preconcentration procedure for ammonium ion determination in natural waters at the μg 1-1 level with pulsed Nessler reagent. The possibility of sample buffering before the adsorption step was studied. The proposed method is characterized by a precision of about 2%, a sampling rate of about 40 samples per hour. and a reagent consumption of 40 μl per sample, and is almost free of interferences. Recoveries from 95 to 105% were achieved in analysing rain-water samples with ammonium contents of less than 200 μg 1-1. Alternative flow diagrams and the injector command unit are discussed.  相似文献

Chip gel electrophoresis was explored for high-sensitivity detection of DNA by combining electrokinetic injection with transient isotachophoresis preconcentration (here named electrokinetic supercharging (EKS)). Low concentrations (0.2 mg/L) of DNA sample could be detected without fluorescence labeling using a conventional UV detector (at 260 nm). On a single-channel microchip, identification of PCR product was performed by exploiting both external and internal calibration methods. The deviation between the two calibration methods was about 3.6%, and the identified DNA fragment size matched with the predicted size of the template DNA. On the cross microchip the EKS preconcentration has also been achieved when changing the injection reservoir differing from the one used previously. The procedure was computer-simulated and the influence of the voltage applied to two-side reservoirs on sample preconcentration and dilution was also discussed.  相似文献

A new flow injection on-line adsorption preconcentration system adapted to flame atomic absorption spectrometry (FAAS) for copper determination at the mug l(-1) level was developed. Polytetrafluoroethylene (PTFE) turnings packed in a mini-column were used as sorbent material. The copper ammonium pyrrolidine dithiocarbamate (APDC) complex was sorbed on the PTFE turnings, from which it could be eluted on-line instantly by isobutyl methyl ketone (IBMK) into the flame at a flow rate of 2.3 ml min(-1). The system was optimized and offered good performance characteristics with practically unlimited life time, greater flow rates and improved flexibility, as compared with other sorbent materials and the knotted reactor preconcentration systems. With 1 min preconcentration time, and a sample frequency of 40 h(-1), the enhancement factor was 340, which could be further improved by increasing the preconcentration time. The detection limit was c(L)=0.05 mug l(-1), and the precision was 1.5%, at the 2.0 mug l(-1) Cu level. The method has been applied successfully to the analysis of potable, river and seawater, and its accuracy was tested by the analysis of certified reference materials and by recovery measurements on spiked samples. No significant interferences exist from other substances usually occurring in natural water.  相似文献

A flow-injection configuration based on the repeated change of the flow direction is proposed for the direct analysis of gaseous samples. The sample is injected into a liquid carrier-reagent stream and the repeat passage of the liquid zone close to the gas-liquid interface nearest to the detector is monitored to obtain an absorbance-time multi-peak recording. The system has been applied to the determination of nitrogen dioxide in gaseous samples for the range 0.5–100 μl ml?1, with a relative standard deviation of 4–5% and a sampling frequency of 60 h?1. A modification of the injection system allows preconcentration steps to be implemented.  相似文献

Acidic aluminium solutions (120 μl) are injected into a buffered (pH 5.7) carrier stream and merge with a chrome azurol S/cetylpyridinium chloride stream; a 2.25-m reaction coil is used with a total flow rate of 4 ml min ?1. Ethanol (30% v/v) in the reagent stream enhances the absorbance of the ternary complex; the molar absorptivity is then 1.34×10 5 l mol ?1 cm ?1 at 625 nm. Calibration is linear over the range 0–400 ng ml?1 aluminium; the limit of determination is 5 ng ml ?1. Iron is masked in the usual way; fluoride is tolerated at ˇ1 mg l ?1. The enzyme catalase, which catalyses the decomposition of hydrogen peroxide to oxygen and water, was immobilized in a membrane by entrapping it in polyacryl amide and contacted to a Clark-type oxygen electrode. With the resulting catalase biosensor it was possible to detect the substrate hydrogen peroxide and the inhibitors fluoride and cyanide in phosphate buffer.The sensor was integrated into a flow system. In the concentration range from 5–200 mg/l a linear dependence of the peak height on the hydrogen peroxide concentration was obtained. The average decrease in activity during 30 days of storage at 6 °C was 17%. Fluoride and cyanide could be determined by measuring the inhibition of the enzymatic reaction in the same flow system. The analysis was executed in three steps; namely determination of the original activity by pumping substrate solution, inhibition of the enzyme by pumping inhibitor solution, and determination of the activity after the inhibition.The decrease in activity correlated with the inhibitor concentration of the sample, but a linear dependence was not found. The inhibition of fluoride and cyanide was both reversible, the enzyme membrane could be reactivated completely by pumping substrate solution. The detection limit was 1 mg/l for fluoride and 1.5 mg/l for cyanide.  相似文献

We developed a novel on-line preconcentration procedure for microchip gel electrophoresis (MCGE), which enables application of electrokinetic supercharging (EKS) for highly sensitive detection of DNA fragments on a cross-geometry microchip. In comparison with conventional pinched injection using the cross microchip, the present approach allows loading a much larger amount of the sample by taking advantage of a newly developed operational mode. In order to obtain high preconcentration effect and prevent splitting of an enriched sample into subchannels, i.e., off the detector range, effects of the voltage applied on the reservoirs and the time of isotachophoretic preconcentration were examined. The optimal balance between the voltage and time was found for a high-sensitivity analysis of DNA fragments. After experimental optimization the detection limit of a 150 bp fragment was as low as 0.22 mg/L (S/N = 3) that is 10 times better than using the conventional pinched injection.  相似文献

A sequential injection analysis (SIA) assembly for the atomic absorption determination of Fe(III) in natural waters is proposed. Iron is preconcentrated on a microcolumn packed with a chelating resin (Chelex 100) that is inserted in the manifold. The sample is passed through the column and the iron retained by the resin is subsequently eluted with 2 M HNO(3). The proposed SIA system affords automatic preconcentration, elution, detection of Fe(III), data acquisition and treatment. When 9 ml of iron solution containing 0.4 or 1 mg l(-1) was passed through the resin, the retention efficiency was 93.1 +/- 0.6 and 7.4 +/- 3.0% respectively, and when 27 ml of iron solution of 0.2 mg l(-1) was preconcentrated, the retention was 8.4 +/- 2.9%. The detection limits thus achieved is 12 mug l(-1) when 9 ml of sample are preconcentrated and 6 mug l(-1) for 27 ml.  相似文献

A simple, sensitive and low cost, flow injection time-based method was developed for on-line preconcentration and determination of copper, lead and chromium(VI) at sub mug l(-1) levels in natural waters and biological samples. At the optimum pH, the on-line formed metal-ammonium pyrrolidine dithiocarbamate (APDC) complexes were sorbed on the unloaded commercial polyurethane foam (PUF), and subsequent eluted quantitatively by isobutylmethylketone and determined by flame atomic absorption spectrometry (FAAS). All chemical, and flow injection variables were optimized for the quantitative preconcentration of each metal and a study of interference level of various ions was also carried out. The system offered improved flexibility, low backpressure and applicability to all the studied metals. At a sample frequency of 36 h(-1) and a 60 s preconcentration time, the enhancement factor was 170, 131 and 28, the detection limit was 0.2, 1.8 and 2.0 mug l(-1), and the precision, expressed as relative standard deviation (s(r)), was 2.8 (at 10 mug l(-1)), 3.4 (at 50 mug l(-1)) and 3.6% (at 50 mug l(-1)) for Cu(II), Pb(II) and Cr(VI), respectively. The accuracy of the developed method was sufficient and evaluated by the analysis of certified reference materials and spiked water samples. Finally, the method was applied to the analysis of environmental samples.  相似文献

Summary A flow injection method for the determination of formaldehyde was developed. It is based on its reaction with 3-methyl-2-benzthiazolinone hydrazone (MBTH) to yield a blue formazan anion. Typically 180 l samples are injected into a buffered carrier stream with a frequency of 30 per hour. This sample line merges with the reagent stream containing MBTH, whereupon an azine is formed. After merging with a second stream containing an acidic solution of FeCl3 as reagent, MBTH is oxidized to an intermediate that attacks the azine yielding the formazan dye with an max at 635 nm. The method was applied to formaldehyde concentrations in the range from 0.15 mg/l to 15 mg/l. The relative standard deviation was 2.3% for 3 mg/l formaldehyde, and 5.1% for 0.3 mg/l, respectively. Strong reducing agents like nitrite interfere seriously. Out of the family of carbonyl compounds only acetaldehyde and propionaldehyde cause strong positive interference.  相似文献

Glycerol of different quality classifications served as a model for a neutral excess component in the isotachophoretic determination of low-molecular-mass anionic trace impurities. Potential anionic contaminants such as nitrate, sulphate, chlorate, nitrite, oxalate, fluoride formate and phosphate were analysed up to an analyte-to-excess ratio of 1:4·107, thus providing the possibility of checking the sample for the mentioned analytes in the order of 2.5·10−6–9.5·10−6%. Because we used a column-coupling isotachophoretic instrument the electrolyte system consisted of two different leading electrolytes, one for the pre-separation (10 mmol/l HCl, β-alanine, pH 3.2) in the first capillary and one for the final separation (5 mmol/l HCl, 1,3-bis[tris(hydroxymethyl)methylamino]propane, β-alanine, pH 3.6) in the second capillary. The terminating electrolyte was citric acid. Due to an increased injection volume of 300 μl, limits of detection (LODs) in the nanomolar range were realised by conductivity detection. The developed method allows simultaneous analysis without sample preparation and/or preconcentration within 25 min and is for that reason suitable for in-place process control.  相似文献

建立了一种测定水中痕量及超痕量溴酸盐的在线富集大体积进样离子色谱法。采用实验室常备的柱容量较高的Dionex IonPac AG23保护柱作为浓缩柱,连接在定量环处富集溴酸盐。淋洗液自动发生装置在线生成高纯度氢氧化钾淋洗液进行梯度洗脱,抑制电导检测。实验结果表明:溴酸盐质量浓度范围在0.05~51.2 μg/L之间时线性关系良好,相关系数r≥0.9995,方法检出限为0.01 μg/L。与常规进样相比,进样量可高达5 mL,浓缩因子约为240倍。本方法成功应用于市售纯净水中溴酸盐的测定,2个加标水平的回收率在90%~100%之间,RSD(n=6)为2.1%~6.4%。该方法无需前处理,操作简单,准确度和精密度良好。通过大体积进样实现高倍富集,适用于痕量及超痕量溴酸盐的分析。  相似文献

A procedure for the preconcentration and determination of lead in vegetable and biological samples was developed in the continuous mode coupled to a flame atomic absorption spectrometer. Lead is quantitatively preconcentrated in acetic buffer as its diethyldithiocarbamate chelate onto a C-18 minicolumn, placed in the loop of a proportional injector, eluted by a stream of methyl isobutyl ketone and introduced directly into the nebuliser. A detection limit of 3 microg 1(-1) is obtained using a time-based technique for 2 min preconcentration and an RSD of 3.8% was readily achieved for three measurements of 25 microg Pb 1(-1). The sample throughput is 24 h(-1). Using preconcentration times of 10 min an enrichment factor of 189 can be obtained. The continuous flow system was used for some reference sample analysis and the obtained results reveal that the methodology can be easily applied for vegetable and biological sample analysis.  相似文献
