A facile method has been developed for the synthesis of 4H-chromene-3-carboxylates 3ad by the nucleophilic substitution reaction of 2-hydroxy-2H-chromene-3-carboxylates 2ad with triethylsilane in the presence of BF3·O(C2H5)2. Cyclocondensation of 4H-chromene-3-carboxylates 3ad with benzylamines 4ad afforded a series of 2,3-dihydrochromenopyrrolones 5ap and with propargylamine afforded 2-propynyl-2,3-dihydrochromenopyrrolones 6ad. Click reaction of 6ad with benzyl azides 7ad provided a series of 1H-1,2,3-triazolylmethyl-2,3-dihydrochromenopyrrolones 8ap. Thus synthesized compounds 3ad, 5ap, 6ad, and 8ap are novel heterocyclic compounds and being reported for the first time.  相似文献

A facile route toward substituted aryl diarylvinyl ethers 4 is developed from CuI-mediated cross-coupling reaction of substituted phenols 2 with diarylvinyl bromides 3 in the presence of various bidentate-based ligands in DMF. Skeleton 3 is prepared by Yan’s bromomethylenation of diarylketones 1 with CHBr3–TiCl4–Mg in the co-solvent of DME and CH2Cl2. The synthetic route obtains moderate yields from the one-step operation and the key structure of 4k is confirmed by X-ray crystallographic analysis. The CADD docking experiments of 4k have been included.  相似文献

Reaction of 2 with bis(tributyltin) in the presence of 3 mol % Pd2(dba)3, 6 mol % XPhos, and 30 equiv of LiBr in wet and air bubbled THF at reflux for 8 h afforded the desired products 3 in 73–74% yields. The cross-coupling reaction of 3a with aryl iodides in the presence of 10 mol % Pd(PPh3)4 and 10 mol % CuI afforded the coupled products 4ap in 47–90% yields. The coupling reaction of 3b with various alkynyl bromides having aryl-, alkyl, or trialkylsilyl group also afforded the corresponding 1,3-enynes 5ag in 61–77% yields.  相似文献

A tandem three stages process to a series of trifluoromethyl and halodifluoromethyl 2,4-unsaturated ketones 4a-c is described. This process started with the preparation of 2-fluoroalkyl substituted propargyl vinyl ether 3a-d by treatment of a mixture of individual ethyl α-per(poly)fluoroalkyl acetates 1a-d and propargyl alcohol 2 in CH2Cl2 with the mixed base (Na2CO3/TEA) at ambient temperature. When heated in toluene at 80°C, these ethers readily underwent a tandem propargyl-allenyl Claisen rearrangement and isomerization of the resultant 3,4-dienone to give 2,4-unsaturated fluoroalkyl ketones 4a-c (Z/E mixture). The reaction of ethyl α-per(poly)fluoroalkyl acetate 1 with 1-phenyl propargyl alcohol 5 in refluxing CH2Cl2 in the presence of the mixed base (Na2CO3/TEA) directly afforded the corresponding unsaturated fluoroalkyl ketone 6a-c in one pot. In the presence of NaH, the reaction of ethyl 3-halo-3-fluoroalkylacrylates 8a-b with 1,1-dimethyl propargyl alcohol 9 at −50°C to 0°C also gave the unsaturated fluoroalkyl ketones 10a-b in one pot. The difluorovinyl propargyl ether 11 produced by reduction of 2-bromodifluoromethyl substituted propargyl vinyl ether 3b rearranged in hot benzene to give the corresponding allene 12 bearing a gem-difluoromethylene group in the middle of the aliphatic chain.  相似文献

Treatment of 1-trimethylsilyloxy-1-cyclohexene (1a) in the presence of a catalytic amount of the acidic dihydrogen complex [RuCl(η2-H2)(dppe)2]OTf (4a) [dppe=1,2-bis(diphenylphosphino)ethane, OTf=OSO2CF3] (10 mol.%) under 1 atm of H2 in anhydrous ClCD2CD2Cl at 50 °C for 8 h afforded cyclohexanone (3a) and Me3SiH in quantitative NMR yields. Silyl enol ethers such as 1-triethylsilyloxy-1-cyclohexene (1b), 1-t-butyldimethylsilyloxy-1-cyclohexene (1c), and other trimethylsilylethers (1d, 1e, and 1f) reacted similarly with H2 to afford the corresponding ketones and trialkylsilanes. The direct proton transfer from H2 to the trimethylsilyl enol ethers (1a and 1d-1f) was confirmed by the experiments employing D2 gas, where α-monodeuterated ketones (3a′ and 3d′-3f′) were obtained in high yields. The enantioselective protonation of prochiral silyl enol ethers with 1 atm of H2 by employing [RuCl(η2-H2)((S)-BINAP)2]OTf (4e) [BINAP=2,2′-bis(diphenylphosphino)-1,1′-binaphthyl] and [RuCl(η2-H2)((R, R)-CHIRAPHOS)2]OTf (4f) [CHIRAPHOS=2,3-bis(diphenylphosphino)butane] showed that no enantioselectivity was observed in either catalytic or stoichiometric protonation reactions under various reaction conditions. The reaction of [RuHCl(dppe)2] (5a) with one equivalent of Me3SiOTf under 1 atm of H2 produced rapidly 4a, concurrent with the formation of Me3SiH. Based on these studies, the mechanism for this novel hydrogenolysis of silyl enol ethers is proposed which involves heterolytic cleavage of the coordinated H2 on the ruthenium atom caused by the nucleophilic attack of the oxygen atom of enol ethers to give ketones and Me3SiOTf, and the subsequent reaction of the resultant complex 5a with Me3SiOTf under 1 atm of H2 to regenerate the original dihydrogen complex 4a. On the other hand, the stoichiometric reaction of a lithium enolate 6e with one equivalent of 4e at −78 °C in CH2Cl2 under 1 atm of H2 afforded 2-methyl-1-tetralone (3e) with 75% ee (S) in >95% yield, together with the formation of [RuHCl((S)-BINAP)2] (5e).  相似文献

The reaction of bromoalkanes (R–Br; (3), R=CnH2n+1, n=4 (a), 8 (b), 12 (c),18 (d)) and bromobenzyl derivatives (R′–Br; (4), R′=CH2C6H2(CH3)3-2,4,6 (a); CH2C6H(CH3)4-2,3,5,6 (b); CH2C6(CH3)5 (c)) with 1H-imidazo[4,5-f][1,10]-phenanthroline (IP)(L2) gave the corresponding 1-R-imidazo[4,5-f][1,10]-phenanthroline (IPR)(L3ad) and 1-R′-imidazo[4,5-f][1,10]-phenanthroline(IPR')(L4ac) ligands, respectively. Treatment of L3ad and L4ad with [Ru(p-cymene)Cl2]2 led to the formation of [Ru(p-cymene)(IPR)Cl]Cl (RuL3ad) and [Ru(p-cymene)(IPR′)Cl]Cl (RuL4ac). New ruthenium(II) complexes RuL3ad and RuL4ac were characterized by elemental analysis, FTIR, UV–visible and NMR spectroscopy. In order to understand effects of these changes on the N-substituent of imidazol on IP and how they translate to catalytic activity, these new RuL2, RuL3ad and RuL4ac were applied in the transfer hydrogenation of ketones by 2-propanol in presence of potassium hydroxide. The activities of the catalysts were monitored by NMR and GC analysis.  相似文献

New acyclovir analogues were obtained by reaction of 1,2,4-triazolo[1,5-a]pyrimidin-7-ones 4ai with (2-acetoxyethoxy)methyl acetate 5 in the presence of trimethylsilyl trifluoromethanesulfonate (TMSOTf) as catalyst (Vorbrüggen procedure). Coupling between compounds 4af and 5 led to a mixture of N3- and N4-isomers 6 and 7, respectively. On the contrary, the reaction of compounds 4gi with 5 proceeded selectively with formation of N3-isomers only. It was found that the ratio of 6af and 7af depends on the presence or the absence of N,O-bis(trimethylsilyl)acetamide (BSA). Glycosylated products 6af and 7af underwent reversible isomerization under TMSOTf treatment. The ratio of glycosylated products of the coupling reaction between 4 and 5 was thermodynamically controlled. A similar reaction occurred if ZnCl2 was chosen as a catalyst, although lower yields of the acyclic analogues of nucleosides were observed. The glycosylation of other purines (adenine and guanine) can be achieved via the non-BSA modification of the Vorbrüggen procedure.  相似文献

β-Alkoxyvinyl bromodifluoromethyl ketones 1a, 1b and 1c were synthesized by the reaction of bromodifluoroacetic anhydride with appropriate vinyl ethers in high yields. The acyclic enone 1a reacted with amines to give the corresponding β-aminovinyl bromodifluoromethyl ketones 2 in good yields. The reaction of 1a with electrophilic reagent ICl yielded α-iodoenone 4. The substitution reaction of the cyclic enones 1b and 1c with thio-nucleophiles gave the corresponding difluoromethylene thioethers 6. The three-component reactions of 2 with primary amines and formaldehyde gave multifunctional 1,2,3,4-tetrahydropyrimidine 3 in moderate yields.  相似文献

An efficient one-pot synthesis of fused pyrazoles has been developed. The procedure involved three components reaction of o-alkynylaldehydes 1as with ketones 2am and hydrazine under mild, metal-free reaction conditions. The desired products were obtained in one-step up to 85% yield. The molecular structure was confirmed by the X-ray crystallographic analysis.  相似文献

The quinolinylcyclopalladated complexes 3ab were synthesised in good yields (81% and 77%) by the insertion reaction of the prepared dinuclear palladium complexes [Pd(C,N-2-C9H4N-CHO-3-R-6)Cl(PPh3)]2 [(R = H (2a), R = OMe (2b)] with isonitrile XyNC (Xy = 2,6-Me2C6H3). The cyclopalladated complexes 3ab were also obtained in low yields (39% and 33.5%) via a one pot oxidative addition reaction of quinoline chloride 1ab with isonitrile XyNC:Pd(dba)2 (4:1). The reactions of 3ab with Tl(TfO) (TfO = triflate, CF3SO3) in the presence of H2O or EtOH causes depalladation reactions of the complexes to provide the corresponding organic compounds 4ab, 5ab and 6ab in yields (41%, 27% and 18–19%). The products were characterized by satisfactory elemental analyses and spectral studies (IR, 1H, 13C and 31P NMR). The crystal structures of 2a, 3a and 3b were determined by X-ray diffraction studies.  相似文献

The reactions of hexachlorocyclotriphosphazatriene, N3P3Cl6, with N-alkyl-N-ferrocenylmethylethylene diamines, FcCH2NH(CH2)2NHR1 [R1 = Me (1) and Et (2)], and sodium [3-(N-ferrocenylmethylamino)-1-propanoxide] (3) produce spirocyclic monoferrocenyl tetrachlorophosphazenes (1a3a). The tetrapyrrolidinophosphazenes (1b3b) are prepared from the reactions of corresponding phosphazenes (1a3a) with excess pyrrolidine. The reaction of 1a with excess morpholine affords geminal-morpholino phosphazene (1c), whilst the reactions of 2a and 3a give diethylaminotrimorpholino (2c) and fully substituted morpholino products (3c), respectively. The structural investigations of the compounds are examined by Fourier transform IR, MS, 1H, 13C, 31P NMR, DEPT, HETCOR, and HMBC techniques. The crystal structures of 3b and 3c are determined using X-ray crystallography. Cyclic voltammetric and chronoamperometric data show that compounds 1a3a, 1b3b, and 1c3c exhibit electrochemically reversible one-electron oxidation of Fc redox centers which are hardly affected by the substituents on the phosphazene ring. The compounds 1b, 2b, 3b, and 3c are screened for antibacterial activities against Gram-positive and Gram-negative bacteria and for antifungal activities against yeast strains. In addition, the antituberculosis activities (in vitro) of these compounds are evaluated against INH-susceptible reference strain M. tuberculosis H37Rv, and six multi-drug resistant clinical M. tuberculosis isolates. Compound 2b is found to be the most active against the susceptible the reference strain. In addition, 1b, 2b, and 3c are active against all the multidrug-resistant clinical isolates at the highest concentrations. Gel electrophoresis data indicate that the compounds promote the formation of strand breaks in plasmid DNA. Almost all the concentrations lost of supercoiled DNA suggests that the compound 3b is very efficient plasmid-modifier. The compounds inhibit BamHI cleavage of pUC18 DNA while restricting HindIII.  相似文献

An efficient tandem approach for the selective synthesis of 4,5-dihydroimidazo[1,5-a]quinoxalines 6ag and imidazo[1,5-a]quinoxalines 7ah by the reaction of 2-imidazolyl anilines 4ac with aryl aldehydes 5ak under mild reaction conditions is described. Conjugated C3 symmetric molecules have been pursued for their fluorescent and electrochemical properties. The synthesis of conjugated C3 symmetric aryl tripyrroles has been performed by a route featuring three steps from trimethyl 1,3,5-benzene tricarboxylate 6: copper-catalyzed cascade addition of vinylmagnesium bromide to convert the carboxylates into γ,δ-unsaturated ketones, Tsuji–Wacker olefin oxidations to form tris(1,4-dione) 9, and Paal–Knorr condensation with ammonia and different amines to furnish the final tripyrroles (30–60% yields). In addition, incomplete reaction of vinylmagnesium bromide to 6 provided benzoate 8 possessing two γ,δ-unsaturated ketones, which were similarly converted to 3,5-dipyrrolylbenzoates 11. The absolute fluorescence quantum yields (Φfl) and electrochemical properties of 5ac and 11ac were investigated. The Φfl are the first reported for such compounds and they ranged between 2 and 40%, contingent on structure and solvent polarity. Cyclic voltammetry revealed that the compounds could be both oxidized and reduced, albeit irreversibly. The oxidation potentials (Eox) varied between 0.73 V and 1.2 V and the reduction potentials (Ered) varied from −0.83 V to −1.36 V. The lowest redox processes were measured for the NH–pyrrole 5a. Moreover, tripyrrole 5a was air stable and on oxidative doping with ferric chloride exhibited a 50 nm bathochromic shift in its absorbance spectrum.  相似文献

A stereoselective 6-step synthesis of the C11H12 hydrocarbon cis,anti,cis-tricyclo[5.4.0.02,5]undeca-3,8,10-triene (2a) is described. The dichloro compound 3c was prepared by the selective reduction of 3a with chromous perchlorate-ethylenediamine complex, a general reagent for this type of transformation. Irradiation of 3c produced 6a and 7a in ∼ 7:1 ratio. Reduction of the mixture with sodium produced ketal mixture 6b and 7b. The ketals were hydrolyzed to ketones 6c and 7c and the ketones decarbonylated to 2a and an isomeric hydrocarbon tentatively assigned as homobasketene 2b′. Hydrocarbon 2a was readily obtained pure by a silver nitrate extraction procedure and 2b′ by preparative VPC. It was shown that pure 6c gives only 2a and therefore, 7c is the source of 2b′. The stereochemical assignments, based on both spectral and chemical evidence, are discussed.  相似文献

Synthesis of novel C2-symmetric chiral crown ethers and their application to enantioselective trifluoromethylation of aldehydes and ketones are discussed. The use of a series of C2-symmetric chiral crown ethers 2 or 3 derived from commercially available (R)-1,1′-bi-2-naphthol for the enantioselective trifluoromethylation of 2-naphthyl aldehyde 1a with (trifluoromethyl)trimethylsilane in the presence of a base was attempted. Iodo-substituted crown ether 2b was found to be the most effective in the model reaction. Moderate enantioselectivities were observed for the trifluoromethylation of both aryl or alkyl aldehydes and alkyl aryl ketones in 21-44% ees. Although the ees are still improvable, this is the first example of a chiral crown ether-catalyzed enantioselective trifluoromethylation reaction.  相似文献

A new series of DTPA-N,N″-bis(amide) ligands functionalized by alkyl carboxylates on the amide side-arms (1a1l) and their Gd(III) complexes of the type [Gd(1)(H2O)] · xH2O (2a2l) were synthesized and characterized by analytical and spectroscopic techniques. Potentiality of 2a2l as contrast agent for magnetic resonance imaging (MRI CA) was investigated by measuring some relevant physicochemical properties such as (i) the protonation constants of 1a1l, (ii) thermodynamic and conditional stability constants of 2a2l, (iii) the selectivity (pGd) of 1a1l for the Gd(III) ion over the endogenous metal ions such as Zn(II), Ca(II), and Cu(II), and (iv) the relaxivities (R1 and R2) of 2a2l in aqueous and aqueous HP-β-CD solutions. Comparative studies reveal that most of new Gd(III) complexes show enhanced thermodynamic stability and selectivity as compared with those of [Gd(DTPA-BMA)(H2O)] (DTPA-BMA = N,N″-di(methylcarbamoylmethyl)diethylenetriamine-N,N′,N″-triacetate). Also enhanced with 1a1l (except 1f and 1h) is affinity for Gd(III) as compared with [DTPA-BMA]3− under physiological conditions. The relaxivities (R1 and R2) of aqueous solutions of 2a2l, on the other hand, drop significantly as compared with [Gd(DTPA-BMA)(H2O)] although they increase dramatically (6–10 fold) in aqueous hydroxypropyl-β-cyclodextrin (HP-β-CD) solutions.  相似文献

The reactions of hexachlorocyclotriphosphazene, N3P3Cl6, with N/O donor type N-alkyl or (aryl)-o-hydroxybenzylamines HO(C6H4)CH2NHR(Ar), [R(Ar) = C(CH3)3 (1), Ph (2)] produce monospirocyclic tetrachlorocyclotriphosphazenes (1a and 2a). The geminal substituted cyclotriphosphazenes (1b, 1d, 2b and 2d) are obtained from the reactions of 1 equiv. of 1a and 2a with 2 equiv. of pyrrolidine or morpholine in THF, while the fully substituted phosphazenes (1c, 1e, 2c and 2e) are formed from the reactions of 1a and 2a with the excess pyrrolidine or morpholine in toluene, between 24 and 48 h. The microwave-assisted reactions of 1a and 2a with excess pyrrolidine or morpholine in toluene afford the fully substituted products with higher yields than those which were obtained by conventional methods. The structural investigations of the compounds have been verified by elemental analyses, ESI-MS, FTIR, 1H, 13C, 31P NMR and HETCOR techniques. The crystal structure of 2a is determined by X-ray crystallography and the phosphazene ring is in the flattened boat form. Compounds 1b, 1d, 2b and 2d in which the spiro aryloxy moiety provides the one centre of chirality exist as racemates and the chirality has been confirmed by 31P NMR spectroscopy on addition of a chiral solvating agent (CSA), (S)-(+)-2,2,2-trifluoro-1-(9′-anthryl)ethanol.  相似文献

l-(N-Cbz)-7-azaisotryptophan, l-(N-Cbz)-1a, a new isostere of tryptophan, was synthesized by reacting Li2-(N-Boc)-2-amino-3-picoline, Li2-(N-Boc)-2a, with appropriately protected l-aspartic acid followed by simple functional group manipulation. This synthetic success led us to access a set of analogs of azaisotryptophan (4ac; 6ac) as well as a new class of chiral amines (7ac; 8ac) for future application in asymmetric synthesis and design of homochiral ligands. Further, we have generalized the method substantiating a variety of new azaindol-2-yl derivatives (10aa10lc) with functionalized substituents. In a preliminary luminescence characterization, l-(N-Cbz)-1a has exhibited about 30 nm bathochromic shifted fluorescence emission compared to tryptophan and (N-Cbz)-tryptophan.  相似文献

The dimeric starting material [Ru(η6-p-cymene)(μ-Cl)Cl]2 reacts with the phosphino-amides o-Ph2P–C6H4CO–NH–R [R = iPr (a), Ph (b), 4-MeC6H4 (c), 4-FC6H4 (d)] to give the mononuclear compounds 1ad [RuCl(η6-p-cymene)(o-Ph2P–C6H4–CO–NH–R)]Cl. The subsequent reaction of these complexes with KPF6 produced the cationic species 2ad [RuCl(η6-p-cymene)(o-Ph2P–C6H4–CO–NH–R)][PF6] in which phosphino-amides also act as rigid P,O-chelating ligands. The molecular structures of 2bd were determined crystallographically. Amide deprotonation is achieved when complexes 2ad were made react with 1 M aqueous solution of KOH, affording the corresponding neutral species 3ad [RuCl(η6-p-cymene)(o-Ph2P–C6H4–CO–N–R)] in which a P,N-coordination mode is suggested.  相似文献
