Reaction of 3-methyleneisocamphanone with malononitrile, ethyl cyanoacetate or diethyl malonate in the presence of catalytic quantities of alkali or under catalysis with tetramethylguanidine in ethanol proceeds according to classical scheme of the Michael reaction and gives rise to 3-exo-(2,2-dicyanoethyl)-, 3-exo-(2-cyano-2-ethoxycarbonylethyl)-, or 3-exo-(2,2-diethoxycarbonylethyl)isocamphanone respectively. When the reaction with ethyl cyanoacetate and diethyl malonate is carried out in methanol occurs transalkylation of the ester groups resulting in the corresponding methyl esters, and in THF occurs hydrolysis to form carboxylic acids. In ethanol or methanol in the presence of equimolar or excess amounts of alkali compounds with cyano groups suffer cyclization into the corresponding 2-alkoxy-3-cyano- or 1-alkoxy-3-alkoxycarbonyl-7,7,8-trimethylbicyclo[2.2.1]hepteno[2.3-b]pyridines. In THF partially form analogous tricyclic 2-hydroxypyridines.  相似文献

Zinc enolates formed from 1-aryl-2-bromo-2-phenylethanones and zinc react with dimethyl 2-(1-arylmethylene)malonates to afford dimethyl 2-(1,3-diaryl-3-oxo-2-phenylpropyl)malonates. The latter react with cyclohexylamine, piperidine, or morpholine to give the corresponding monosubstituted amides. The zinc enolate derived from 2-bromoindanone and zinc reacts with dimethyl 2-(4-bromobenzylidene)malonate, yielding dimethyl 2-[(4-bromophenyl)(1-oxoindan-2-yl)methyl]malonate. The final products largely form as a single diastereomer.  相似文献

Palladium(0)-catalyzed deconjugative allylation of alkenylidenemalonates and alkylidenemalonates was achieved for the first time. Reactions of dimethyl 2-((E)-but-2-enylidene)malonate with various allylic acetates using LHMDS as a base in DMF in the presence of Pd(2)dba(3) (2.5 mol %) and PPh(3) (10 mol %) proceeded at room temperature to give the corresponding alpha-allylation products in good yields in a regio- and stereoselective manner. This reaction can also be used for allylation of dimethyl ethylidenemalonate or dimethyl 2-((E)-pent-2-enylidene)malonate and give the desired alpha-allylation products in good yields.  相似文献

Samples of polycrystalline calcite were impregnated with solutions of malonic acid of three concentrations (5 x 10(-2), 5 x 10(-3) , and 5 x 10(-4) M) and different pH values (6.00, 7.00, and 8.00). The impregnation was carried out at room temperature to evaluate the adsorption of malonate ion in the calcite surface to optimize the conditions for possible application on limestone and marble in cultural heritage materials. The affinity of the malonate ion was determined through the potentiometric measurement of the surface charge and the corresponding adsorbed amounts by titration, Raman spectroscopy, and small-angle X-ray scattering (SAXS). The results indicate effective adsorption of the malonate ion on the surface at a pH value close to the point of zero charge (pHpzc approximately 8.20) and changes in some surface morphological properties such as the pore shape and the pore size distribution. The presence of a malonate adsorptive layer on calcite generates an interface interaction potential that may influence the reaction and transport mechanisms within the medium.  相似文献

The condensation of cyclohexylidenemalononitrile or cyclohexylidenecyanoacetic ester with thioamidoethyl(methyl)malonate in the presence of sodium ethylate gave 6-amino-3-carbethoxy-5-cyano-4-spirocyclohexane-1,2,3,4-tetrahydropyridine-2-thione and 3-carbethoxy(methoxy)-5-cyano-6-oxo-4-spirocyclohexanepiperidine-2-thiones which were used in the synthesis of the corresponding substituted 2-alkylthiotetrahydropyridines. 5-Carbethoxy-3-cyano-3-methyl-6-methylthio-4-spiro-cyclohexane-3,4-dihydropyridine-2(1H)-one was studied by X-ray crystallography.  相似文献

The treatment of methyl(E)-4-tosyl-2-butenoate (4) with two equiv. of sodium hydride and different mono and dihalides gives mainly γ,γ- and ,- or ,γ- and ,-dialkylated products (5–7) depending on the electrophile. The corresponding monoanion dimerizes with iodine to afford stereoselectively dimethyl cis-4,5-ditosyl-2,6-cyclohexadiene-1,2-dicarboxylate (11). The tosyl group in compounds 6 and 7 is reduced and in the case of γ,γ-dimethylated 5a substituted by sodium dimethyl malonate under Pd(PPh3)4 catalysis. Michael addition of different nucleophiles provides the corresponding β-substituted methyl γ-tosylbutanoates 16.  相似文献

The reactions of 4-alkyl-4-(phenylsulfonyl)-2-sulfolenes with excess carbanion of dimethyl malonate in refluxing THF have been studied. The substitution reaction takes place at the 3-position via an addition-elimination process so that 3,4-disubstituted 2-sulfolenes are obtained. These products are isomerized to the corresponding3-sulfolenes under basic conditions.  相似文献

2-N-Nitrosomethylaminopyridine but not 2-methylaminopyridine, 3-N-nitrosomethylamino- or 3-methyl-aminopyridine and 4-N-nitrosomethylamino- or 4-methylaminopyridine react with malonate esters to form the corresponding derivatives of carbamates, amides and dihydropyridines respectively. Some derivatives of salicylic acid containing substituents meta to the carboxyl group were used to prepare analogs of the anticoagulant drug warfarin, 3-(1-phenyl-3-oxobutyl)-4-hydroxycoumarin, containing substituents in either the 6-or 8-position of the courmarin ring. When the substituent was the hydroxyl group, the resulting products are previously identified metabolites of warfarin. The substituted salicylic acid is first acetylated with acetic anhydride, then either converted to the acid chloride and condensed with diethyl malonate in the presence of sodium hydroxide or converted to the mixed anhydride with formic acid and condensed with ethoxymagnesium diethyl malonate to yield, in either case, the corresponding 3-carbethoxy-4-hydroxycoumarin substituted in the 6- or 8-position of the coumarin ring. These compounds readily condense with benzalacetone to form the corresponding substituted warfarin in the presence of 5 mole % tertiary amine catalyst. This method offers an improved route for the synthesis of 8-hydroxywarfarin.  相似文献

Products of the reactions of 4- and 2-hydroxy-3,5-di-tert-butyl-benzaldehydes with malonic acid, diethyl malonate, and acetic anhydride in the presence of weak bases were isolated and identified. The reactions of 3,5-di-tert-butyl-4-hydroxybenzaldehyde with malonic acid and acetic anhydride in the presence of sodium acetate and piperidine gave 3,5-di-tert-butyl-4-hydroxycinnamic acid. The reaction of its 2-hydroxy isomer with acetic anhydride stopped at the stage of formation of the corresponding O-acetyl derivative, while in the reaction with malonic acid the corresponding substituted cinnamic acid and its lactone (coumarin derivative) were formed as intermediate products in a transformation sequence finally leading to 3-(3,5-di-tertbutyl-2-hydroxyphenyl)-3-piperidinopropionic acid and 6,8-di-tert-butyl-2-oxo-3,4-dihydro-2H-chromen-4-ylacetic acid. Analogous differences were typical of reactions of isomeric 4- and 2-hydroxy-3,5-di-tert-butylbenzaldehydes with diethyl malonate. The transformations of the 2-hydroxy isomer were accompanied by hydrolysis and formation of an adduct of intermediate coumarin derivative with diethyl malonate and piperidine.  相似文献

The oxidative addition of dimethyl malonate to ring substituted styrenes leads to the formation of substituted dimethyl (2-oxo-2-phenylethyl) malonate and methyl 2-oxo-5-phenyltetrahydrofuran-3-carboxylate along with small amounts of substituted dimethyl [2-(nitrooxy)-2-phenylethyl] malonate and dimethyl 2-methoxy-2-phenylethyl) malonate. A novel cyclization reaction of ethenetricarboxylate derivative aromatic compounds in the presence of various Lewis acids gave benzo-annulated cyclic compounds such as oxindole and benzofuran derivatives via Friedel-Crafts intramolecular Michael addition in high yields. For example, the reaction of diethyl 2-[(N-methyl-N-phenylcarbamoyl)methylene]malonate (1a) in the presence of ZnCl2 at room temperature gave diethyl 2-(1-methyl-2-oxoindolin-3-yl)malonate (2a) in 98% yield. The reactions also proceeded with a catalytic amount of a Lewis acid such as AlCl3, ZnCl2, ZnBr2, Sc(OTf)3, or InBr3.  相似文献

Treatment of dimethyl 4-(phenylsulfonyl)-4,5-hexadiene-1,1-dicarboxylate with potassium tert-butoxide in tert-butanol at room temperature effected successive endo-mode ring closure at the sp-hybridized carbon center and demethoxycarbonylation of the resulting malonate derivative leading to the exclusive formation of methyl 2-methyl-3-(phenylsulfonyl)-1-cyclopentenecarboxylate. An additional seven allenyl sulfones having a 1,3-dicarbonyl functionality gave the corresponding cyclopentene or cyclohexene derivatives in high yields.  相似文献

Reaction of 2-mercapto-4-hydroxy-5H-[1]-benzopyrano-[4,3- d ]-pyrimidin-5-one ( 3 ) with phenyl isothiocyanate and methyl acrylate yielded the corresponding 2-(substituted)thio-4-hydroxy-5H-[1]-benzopyrano-[4,3- d ]-pyrimidin-5-ones ( 4 , 5 ). Hydrolysis and hydrazinolysis of 5 gave acid derivative 6 , and hydrazone 7 . Treatment of hydrazone 7 with ethyl acetoacetate, diethyl malonate, and phenyl isothiocyanate yielded the corresponding 2-(substituted)thio-4-hydroxy-5H-[1]-benzopyrano- [4,3- d ]-pyrimidin-5-ones ( 8 , 9 , and 11 ). Bromination–dehydrobromination of methyl (E)-3-(4-methylbenzenesulfonyl)prop-2-enoate gave methyl (Z)-3-bromo-3-(4-methylbenzenesulfonyl)prop-2-enoate whose structure was determined by X-ray analysis. Methyl (Z)-3-bromo-3-(4-methylbenzenesulfonyl)prop-2-enoate behaved as a synthetic equivalent of methyl 3-(4-methylbenzenesulfonyl)prop-2-ynoate in reactions with dimethyl malonate and methyl acetoacetate, which afforded the corresponding Michael adducts, trimethyl 3-(4-methylbenzenesulfonyl)prop-1-ene-1,1,2-tricarboxylate and dimethyl (Z)-2-acetyl-3-(4-methylbenzenesulfonyl)but-2-enedioate, respectively, via nucleophilic attack on the β-position with respect to the sulfonyl group.  相似文献

A chemo-enzymatic approach to some 6″-O-(3-arylprop-2-enoyl) derivatives of the flavonol glucoside isoquercitrin ( 2a ) was explored to overcome the inability to directly introduce these acyl moieties by an enzyme-catalyzed reaction of 2a with the corresponding activated esters. This new approach was based on the regioselective introduction of a methyl malonate residue at the CH2OH of the sugar moiety by catalysis with the protease subtilisin (→ 22a ). The mixed diester 22a was then subjected to chemoselective hydrolysis of the methoxycarbonyl function by another enzyme, biophine esterase (→ 23 ). Finally, the malonic monoester 23 was reacted in a Knoevenagel-type condensation with benzaldehyde, 4-hydroxybenzaldehyde, or 4-hydroxy-3-methoxybenzaldehyde to afford the target 6″-O-(3-arylprop-2-enoyl) isoquercitrins 2b–d .  相似文献

The synthesis of dimethyl {2-[3-(4-nitrophenyl)-1H-pyrazol-5-yl]ethyl}malonate monohydrate 1, C16H17N3O6·H2O was performed and the molecular structure has been studied by using NMR, single crystal X-ray diffraction and ab initio calculations. The title compound presents a pyrazole ring (N1 to C5), a phenyl ring (C1″ to C6″) attached to C3 and the ethylene dimethyl malonate frame (C1′ to C7′) attached to C5. The torsion angle defined by N2C3C1″C2″ (−12.26°) showed that pyrazole and phenyl rings are not in the same plane. Monohydration in (1) is present in the structure by a NHOH2 hydrogen bonding, with a bond length of 1.782 Å. Experimental and theoretical evidences indicated the preference of the 3-tautomer over the corresponding 5-tautomer in the titled pyrazole.  相似文献

Reactions of 1-adamantyl bromomethyl ketone and 1-(1-adamantyl)-3-bromo-2-propanone with acetylacetone and ethyl acetoacetate in a mixture of dry diethyl ether with anhydrous methanol in the presence of sodium methoxide afforded 3-(1-adamantylcarbonylmethyl)-2,4-pentanedione, ethyl 2-(1-adamantylcarbonylmethyl)-3-oxobutanoate, 4-acetyl-1-(1-adamantyl)-2,5-hexanedione, and ethyl 2-acetyl-5-(1-adamantyl)-4-oxopentanoate. The Knoevenagel-Cope reactions of 1-adamantyl bromomethyl ketone and 1-(1-adamantyl)-3-bromo-2-propanone with diethyl malonate yielded, respectively, diethyl 1-(1-adamantyl)-2-bromoethylidenemalonate and diethyl 1-(1-adamantylmethyl)-2-bromoethylidenemalonate. O-Alkylation of ethyl acetoacetate with 1-adamantyl bromomethyl ketone gave ethyl 3-(1-adamantylcarbonylmethoxy)-2-butenoate. Carboxylic acids reacted with 1-adamantyl bromomethyl ketone to form the corresponding 2-(1-adamantyl)-2-oxoethyl carboxylates.  相似文献

The debenzoylation of 2-[(amino)(benzoylamino)methylidene]cyclohexane-1,3-diones (addition products of cyclohexane-1,3-diones with benzoylcyanamide) affords the corresponding 2-diaminomethylidenecyclohexane-1,3-diones. The latter act as N,N-dinucleophiles in reactions with diethyl malonate in the presence of MeONa to form 6-hydroxy-2-(2,6-dioxocyclohexylidene)-1,2-dihydropyrimidin-4(3H)-ones. This reaction performed at 200 °C in the absence of bases results in the subsequent self-condensation of the dihydropyrimidinones to give 4,5′-bipyrimidine derivatives.  相似文献
