Methods for the regioselective synthesis of Mannich bases starting from unsymmetrical ketones are described. The Mannich base on the more substituted carbon is obtained by reaction with (CH3)2N+CH2,CF3CO2- in CF3CO2H and its isomer on the less substituted carbon with (iPr)2N+CH2,ClO4- in CH3CN. In the first case, the orientation corresponds to a non-selective attack of the immonium salt on a mixture of enols in which the more-substituted isomer is predominant. In the second ease, use of a bulky immonium salt induces a selectivity favoring the less-substituted enol.  相似文献

The asymmetric Diels–Alder reaction of α‐substituted acrolein proceeds in the presence of the trifluoroacetic acid salt of trifluoromethyl‐substituted diarylprolinol silyl ether to afford the exo‐isomer with both excellent diastereoselectivity and high enantioselectivity. In the Diels–Alder reaction of a β,β‐disubstituted α,β‐unsaturated aldehyde, good exo‐selectivity and excellent enantioselectivity was obtained when the perchloric acid salt of the bulky triisopropyl silyl ether of trifluoromethyl substituted diarylprolinol was employed as an organocatalyst in the presence of water. In both cases, all‐carbon quaternary stereocenters are constructed enantioselectively.  相似文献

A straightforward synthesis of substituted 1,2-dihydroquinazolinium triflates (3) is reported by reaction of 2-imino-substituted anilines with a range of carbonyl compounds in the presence of triflic acid via intermediate iminium salts. Similar reactions with di- or trialdehydes and triflic acid give bis- or tris-(1,2-dihydroquinazolinium) salts. Some 4-methyl substituted 1,2-dihydroquinazolinium salts rearrange, under various conditions, to their corresponding 4-iminium-1,2,3,4-tetrahydroquinolinium isomers (7). Most of salts 3 derived from ketones are rather unstable, which prevents their isolation or full characterization. The crystal structures of various 3 and 7 salts have been determined.  相似文献

(E )‐δ‐Boryl‐substituted anti ‐homoallylic alcohols are synthesized in a highly diastereo‐ and enantioselective manner from 1,1‐di(boryl)alk‐3‐enes and aldehydes. Mechanistically, the reaction consists of 1) palladium‐catalyzed double‐bond transposition of the 1,1‐di(boryl)alk‐3‐enes to 1,1‐di(boryl)alk‐2‐enes, 2) chiral phosphoric acid catalyzed allylation of aldehydes, and 3) palladium‐catalyzed geometrical isomerization from the Z to E isomer. As a result, the configurations of two chiral centers and one double bond are all controlled with high selectivity in a single reaction vessel.  相似文献

A new polymeric laponite nanoclay heterogeneous catalytic system based on HPMC (hydroxypropyl methyl cellulose) was developed for direct Mannich-type reaction of ketones with substituted benzaldehydes and anilines to afford corresponding β-amino ketones in good to high yields. Interestingly, cyclic ketones exhibited different chemoselectivity. Cyclopentanone underwent aldol condensation to give crossed-aldol product, while cyclohexanone and cyclopentanone afforded corresponding Mannich adducts. In the case of cyclohexanone, stereoselectivity was changed depending on the nature of the substitution on benzaldehydes, in which, moderate electron-donating and electron-withdrawing groups afforded the anti isomer as major products, but strongly electron-donating substituted benzaldehydes led to syn isomer as the major Mannich adducts. Mannich reaction with cycloheptanone led to Mannich adducts with excellent syn selectivity.  相似文献

The conversion of 4-vinyl substituted β-lactams into 4-vinyl substituted 1-azetines and their subsequent reaction with diphenylcyclopropenone (DPP) results in the formation of a highly functionalised 7-azabicyclo[4.2.1]nonene. This heterocyclic system is an isomer of the homotropane (9-azabicyclo[4.2.1]nonene) nucleus, which is the core structure of a class of alkaloid natural products that includes anatoxin-a. The key processes involved are two ring expansions, one of which constitutes a formal [3+2] cycloaddition and the other of which is an unusual aza-Cope (amino-Claisen) [3,3]-sigmatropic rearrangement.  相似文献

Fused bicyclic pyran-2-ones with pendant furan side chains and an oxygenated stereogenic center adjacent to the pyranone ring oxygen were prepared via FeCl(3)-catalyzed Michael addition. Irradiation furnished the corresponding lactone-bridged tricyclic [4 + 4]-cycloadducts with good facial selectivity. Results of the catalytic hydrogenation of Baylis-Hillman adducts obtained from substituted 3-, 4- and 5-isoxazolecarbox-aldehydes and their corresponding acetates in the presence of Raney-Ni and Pd-C are presented. The hydrogenation of Baylis-Hillman adducts of substituted 5-isoxazolecarbaldehydes and 3-isoxazolecarbaldehydes in the presence of Raney-Ni furnishes diastereoselectively syn enaminones over anti and in the presence of boric acid as an additive further enhancement of diastereoselectivity in favor of syn isomer is observed. The Pd-C-promoted hydrogenation of these substrates is also diastereoselective in favor of syn isomer but occurs without the hydrogenolysis of isoxazole-ring. The presence of boric acid as an additive in this hydrogenation exhibits no pronounced effect on diastereoselectivity. The Raney-Ni-mediated hydrogenation of Baylis-Hillman adducts of substituted 4-isoxazolecarbaldehydes yield pyridone derivatives and Pd-C-promoted hydrogenation of the same substrate is diastereoselective to afford the anti isomer of the resulting products. The enaminones derived from Baylis-Hillman adducts of 3- and 5-isoxazolecarbaldehydes serve as versatile precursors for α′-hydroxy-1,3-diketones, which undergo acid-catalyzed ring-closure reaction to afford the furanone derivatives in excellent yields.  相似文献

The palladium-catalyzed cross-coupling reaction of 2-bromonitrobenzenes or 2-bromoacetanilides with ethylene has been used to produce a variety of substituted indoles. The steric selectivity of the Horner-Emmons reaction between isovaleraldehyde and 3-carboethoxy-2-methyl-2-propenylphosphonates depends on the structure of the alkoxy substituants at the phosphorus atom. Side chains of docetaxel and its isomer were obtained through Staudinger cycloaddition and catalytic hydrogenation of chlorophenyl intermediates, using chlorobenzaldehyde as starting material. Syntheses of three novel chiral azetidinone derivatives through the Staudinger cycloaddition reaction of chlorophenyl chi‐ral amine Schiff base with different substituted positions were described and their ring‐opening reaction under the catalysis of Pd/MgCO3 or Pd/C to afford side chains of docetaxel and its isomer in high yields was investigated. Finally, docetaxel and its isomer were obtained. Single crystal of (3S,4R)‐3‐hydroxy‐N‐[(S)‐(l‐phenyl)ethyl]‐4 ‐(2′‐chlorophenyl) ‐2‐azetidinone ( 4c ) was obtained, the configuration of which was determined by X‐ray diffraction. Because of the mild cyclization reaction condition and convenient asymmetric resolution operation when p‐chlorobenzaldehyde was employed instead of benzaldehyde, the yield of cyclization and hydrogenation increased dramatically and the total yield of docetaxel was higher than the result in literature. When o‐chlorobenzaldehyde was employed instead of benzaldehyde an isomer of docetaxel was obtained by the same way.  相似文献

A number of substituted indenols have been synthesized using ruthenium-mediated ring-closing metathesis (RCM) with Grubbs second generation catalyst as the key step. The required dienes were synthesized by two strategies. The first entailed the isomerization of 2-allyl-3-isopropoxy-4-methoxybenzaldehyde to its styrene derivative, isopropoxy-4-methoxy-2-propenylbenzaldehyde using [RuClH(CO)(PPh3)3]. This compound and 3-isopropoxy-4-methoxy-2-(1-phenyl-propenyl)-benzaldehyde were then treated with vinyl- or isopropenyl-magnesium bromide to afford four of the scaffolds required for the metathesis. As the compound 3-isopropoxy-4-methoxy-2-(1-methyl-2-propenyl)benzaldehyde proved to be difficult to isomerize, the diene substrates 1-[3-isopropoxy-4-methoxy-2-(1-methylpropenyl)-phenyl]-prop-2-en-1-ol and 1-[3-isopropoxy-4-methoxy-2-(1-methylpropenyl)-phenyl]-2-methylprop-2-en-1-ol were synthesized by the addition of the Grignard reagents to 3-isopropoxy-4-methoxy-2-(1-methyl-2-propenyl)benzaldehyde, followed by isomerization of the arylallyl group to the thermodynamically favoured isomer with potassium t-butoxide. The use of harsher conditions (higher temperature and catalyst loadings) for the metathesis reactions resulted in the formation of substituted indenones, formed by a tandem RCM-dehydrogenative oxidation in the absence of a hydrogen acceptor. Further manipulations of the reaction conditions generated two substituted indanones by way of a tandem RCM-formal redox isomerization sequence. Finally the synthesis of some substituted indenes was accomplished from their corresponding dienes by the use of RCM.  相似文献

5,10,15-Triphenylcorrole (1) reacts with the Vilsmeier reagent (POCl(3)/DMF) to give the corresponding 3-formyl derivative 3 as the major product. The regioselectivity of the reaction was proven by X-ray crystallography and only traces of the 2-formyl isomer were observed. A more polar product is also observed and this compound becomes the major product when an excess of DMF is used for the preparation of the Vilsmeier reagent, while the formation of the 3-formyl isomer is almost completely suppressed. X-ray crystallography allowed us to identify this compound as the fully substituted N-ethane bridged derivative 4, formed from the attack of the Vilsmeier reagent at the inner core of the macrocycle. This compound is unique among porphyrinoid macrocycles, and further confirms the peculiarity of corrole chemistry.  相似文献

An efficient Cu(I)-catalyzed asymmetric [3 + 2] cycloaddition of N-(2-pyridylmethyl) imines has been developed. In the presence of a Cu(CH(3)CN)(4)PF(6)/bisoxazoline catalyst system, high levels of enantioselectivity (up to 97% ee) and moderate to high exo selectivity were achieved with a wide variety of substituted dipolarophiles, including maleimides, fumarates, fumarodinitrile, enones, and nitroalkenes. Diels–Alder cycloaddition between cyclopentadiene and p-benzoquinone has been studied using amorphous silica and different ITQ-2 and MCM-41 pure silica and metal containing materials as catalysts. The reaction can afford different products depending on the molecular reacting site, and the possibility of consecutive additions. Structured solid catalysts increase the selectivity to the endo–endo isomer. Silanol groups have not enough Brönsted acidity to interact with the carbonyl groups present in the dienophile, to reduce LUMO's energy and provide a better overlap between HOMO and LUMO, according to the frontier molecular orbital theory.

The introduction of transition metal atoms in the framework increases the reaction rate for the Diels–Alder reaction while preserving the selectivity to the endo–endo isomer. The presence of more acidic OH groups enhance the retro-Diels–Alder reaction increasing the selectivity to the endo–exo isomer.  相似文献

对[Co(2,3-tri)(amp)Cl]^2^+四个几何经式异构体在不同条件下的取代及重排反应进行了详细的考察。因二元胺中吡啶环造成的空间拥塞,使cis异构体碱水解速度比trans异构体约快100倍,控制碱水解实验结果表明,每一异构体的水解产物均含四个经式异构体,且具有相同分布。实验过程中未观察到面式异构体。在二甲亚砜中加热的重排反应中,异构体m1表现出最高的反应性,cis异构体均首先转化为trans异构体m3,而后可观察到trans异构体m3与m4的平衡。利用时间分辨核磁共振仪测定了氘代水里各异构体中各活性氢的氘代化速度。反应活性最低的异构体m4具有氘代速度最快的活性氢,当这些活性氢完全氘代化时仍未见其水解或重排产物;而相同实验条件下异构体m1或m2中相应活性氢的氘代化速度则要缓慢得多,但同时可观察到水解和重排产物。这说明在碱催化水解过程中氘代速度快的活性氢与异构体的反应性并非正比关系,然而活性氢的氘代化是观察到异构体水解重排的必要条件。利用量子化学从头计算法在赝势基组RHF/LANL2DZ的水平上对该体系各异构体进行了结构优化计算,与对应异构体的晶体结构参数比较,一般相对误差不超过3%,从能量角度来看,cis异构体比trans异构体高出约4kJ/mol,而面式异构体则至少比经式异构体高约17kJ/mol;考虑溶剂化影响,一般约低5kJ/mol。考察结构参数结果显示,结构变形性参数能较好地解释异构体反应活性。  相似文献

A series of 3(5),4-trimethylene and 3(5),4-tetramethylenepyrazoles (tetrahydroindazoles) have been prepared from 2-acylcyclanones using two methods: direct reaction with a substituted hydrazine and through an NH-pyrazole followed by nucleophilic substitution. The results mainly concern the 2-benzothiazolyl substituent, but 2,4-dinitrophenyl derivatives were also studied for comparison. The orientation of the reactions (isomer ratio), the deshielding in 1H nmr of the 5-methyl and 5-methylene signals when a benzothiazolyl residue is at position 1, and the ring strain effect on heterocyclic carbons chemical shifts, are discussed.  相似文献

To understand the selectivity of the crown ether type chiral stationary phase (CSP), the retention selectivity for aniline and the positional isomers of substituted anilines were studied. In various substituted isomers, except nitroaniline, a remarkable decrease of retention due to steric hindrance was observed for the 2-substituted isomer. To determine the detailed molecular recognition mechanism, quantum chemical calculations were performed for the aggregates between the crown ether and the anilines. The results suggested that the 20-Crown-6, which includes a phenyl-substituted 1,1′-binaphthyl moiety, interacts with alkyl and aryl amines in an unconventional form different from the proposed one for 18-Crown-6.  相似文献
