Topologically mobile difluorinated cyclooctenones undergo rapid, high-yielding, and completely stereoselective epoxidations with methyl(trifluoromethyl)dioxirane. The epoxides resist conventional hydrolysis but react smoothly in basic media under microwave irradiation to afford unique hemiacetals and hemiaminals in good yield. [reaction: see text]  相似文献

2-Functionalised aromatic monoaldehydes were synthesised in good to excellent yields by reacting 4-bromo-2-fluorobenzaldehyde with different secondary amines and phenol. The Suzuki-coupling reaction of the newly functionalised aromatic monoaldehydes with 4-formylphenylboronic acid afforded the corresponding 2-functionalised-4,4'-biphenyldialdehydes in good yields (47-85%). The [3+3]-cyclocondensation reactions of the 2-functionalised-4,4'-biphenyldialdehydes with (1R,2R)-1,2-diaminocyclohexane afforded a mixture of regioisomeric C(3)-symmetrical and non-symmetrical trianglimines. Reduction of the C(3)-symmetrical and the non-symmetrical trianglimines with NaBH(4) in a mixture of THF and MeOH afforded the corresponding trianglamines in high yields.  相似文献

A versatile, one-pot synthesis of 2-arylimidazole-4-carboxylic acids from arylamidines and methyl-2-chloroacetoacetate is described. The transformation is chemoselective, and reaction conditions are mild. Moreover, the flexibility of the strategy offers rapid access to two important classes of biaryl compounds, both 2-arylimidazoles and 2-arylpyrimidines, depending simply upon solvent and base selection. A novel method for peptide bond formation utilizing amino acid 2-formyl-4-nitrophenylthio ester has been developed. The reaction can be performed in water-containing media and is compatible with various types of amino acid side-chain functional groups. Use of N-methylmaleinimide as an additive is essential for the reaction to proceed with high efficiency. It captures liberated formyl-substituted thiophenol through 1,4-addition followed by aldol cyclization.  相似文献

Highly efficient chemoselective 1,2-addition of 2-aminopyrimidines and D,L-amino acid esters to substituted propynals was performed. Acid-catalyzed addition of 2-aminopyrimidine and 2-amino-4-methylpyrimidine to trimethylsilyl- and phenylpropynals gave previously unknown acetylenic aminals. Reactions of α-amino acid methyl esters (alanine and lysine) with trimethylsilylpropynal led to the formation of new aza enynes in high yield. Regardless of the reactant ratio, trimethylsilylpropynal reacted with lysine at both amino groups with formation of bis-aza enyne.  相似文献

An excellent utility of Schmidt reaction of aldehydes to access corresponding nitriles in an instantaneous reaction is demonstrated. The reaction of aldehydes with NaN(3) and TfOH furnishes the corresponding nitriles in near quantitative yields and tolerates a variety of electron-withdrawing and electron-donating substituents on the substrates. Formanilides, a common side product in Schmidt reaction, is not observed in this reaction. Besides these advantages, the salient feature of this reaction is that it exhibits a remarkable chemoselectivity, as acid and ketone functionalities are well tolerated under the reaction conditions. The reaction is easily scalable, high yielding, and nearly instantaneous.  相似文献

A methodology for the successive regiospecific "clicking" together of three components in one pot via two triazole linkages is reported. The protocol utilizes copper(I)-mediated alkyne-azide cycloaddition reactions combined with a silver(I)-catalyzed TMS-alkyne deprotection under mild hydroalcoholic conditions. We exemplify the approach with peptide-based components to illustrate its compatibility with polyfunctionalized biomolecules. The method constitutes a promising tool for peptide ligation. We also provide a procedure for directly using TMS-alkynes as the cycloaddition partner in classical "click" chemistry.  相似文献

A new approach is herein reported to prepare biaryl derivatives via a tandem Pd-catalyzed boron-Heck and Suzuki reactions. This one-pot tandem process avoided purification or addition of extra catalyst between steps. 2-Amino-5-phenyl-1,3,4-oxadiazole ( 1a ) and 2-amino-1,3,4-thiadiazole ( 1b ) reacted with acrylonitrile to yield β-cyanoethylamino derivatives. On the other hand, 2-amino-4-phenylthiazole ( 2 ) reacted with acrylonitrile under the same experimental conditions to yield a di-β-cyanoethylaminothiazole derivative. 3-Phenyl-Δ2-1,2,4-triazoline-5-thione reacted with acrylonitrile to yield the corresponding adduct. Reaction of the fluoride ion with secondary alkyl halides leads to 90% of elimination reaction and only 10% of nucleophilic substitution in dipolar aprotic solvents. Adding water to the organic phase, the SN2 yield increases in the cost of decreased reactivity. Using ab initio calculations, we have shown that it is possible to increase the reaction rate and the selectivity toward the SN2 process through supramolecular organocatalysis. The catalytic concept is based on selective solvation of the transition state through two hydrogen bonds provided by the 1,4-benzenedimethanol. The two hydrogen bonds between the catalyst and the SN2 transition state favor this pathway while just one strong hydrogen bond between the catalyst and the fluoride ion leads to a lower stabilization of the nucleophile, resulting in a higher reaction rate. Our calculations predict that the substitution product increases to 40% yield because of the selective catalysis provided by the 1,4-benzenedimethanol.  相似文献

本文总结了过去几十年特别是近15年来过渡金属催化下各种含磷-氢键的膦氢化合物对炔烃的高选择性膦氢化反应,详尽叙述了其发现、发展和现状.自1996年来,过渡金属催化高选择性膦氢化反应研究工作发展迅速,各种高选择性膦氢化反应不断开发,目前已具有底物适用范围广、过渡金属催化剂活性高、反应选择性高、原子经济性高、以及能满足不同合成需求等优点,并逐步向反应条件温和化、金属催化剂简单化、无配体化、合成步骤简易化以及原料催化剂成本低价化方向发展.虽然如此,至今仍缺乏关于本研究全面的综述和介绍,希望本文可以弥补文献缺陷,对过渡金属催化高选择性膦氢化反应研究有个客观全面的介绍.过渡金属催化烯烃的不对称膦氢化反应合成碳手性或磷手性的光学活性有机磷化合物作为相关研究中的起步最晚的分支,本文也将作阶段小结.  相似文献

Grignard reactants like methylmagnesium chloride are not selective with respect to different carbonyl bonds. We present a theoretical study where shaped laser pulses are utilized to prefer specific bonds in a mixture of more than one carbonyl reactant. A mixture of cyclohexanone and cyclopentanone has been chosen as a representative example. The light pulse is supposed to provide the activation energy and to adopt the function of a protecting group. The control aim is to stretch exclusively the C-O bond of one compound to the length required in the Grignard transition state. To guarantee an experimentally realizable bandwidth for the unshaped pulse, we use our recently developed optimal control theory algorithm, which allows the simultaneous optimization of the light field in the time and frequency domain. Highly selective picosecond control pulses could be optimized in the infrared regime suggesting that laser assisted chemoselectivity is possible to a large extent. To obtain control not only on the final product but also on the excitation mechanism, various initial conditions and frequency restrictions were investigated.  相似文献

Summary The polarographic properties of some aliphatic, arylaliphatic, phenoxyaliphatic and aromatic amidines have been investigated. Aliphatic and arylaliphatic amidines are not reducible in the range 0 to –1.8 volts versus the saturated calomel electrode. Phenoxyacetamidines are reduced to the phenol and acetamidine. Aromatic amidines are reduced to the benzylamine and ammonia. The half-wave potentials of both reducible classes of compound are independent of Ph over the range 7–9 Ph.  相似文献

We recently reported a structure-activity relationship study of 4-epi-jaspine B derivatives toward sphingosine kinase (SphK) and identified selective inhibitors of two SphK isoforms. In this study, we designed and synthesized jaspine B regioisomers on the basis of palladium-catalyzed tetrahydrofuran formation and late-stage cross metathesis reactions to investigate the influence of the substitution position of functional groups on SphK inhibition. Evaluation of the jaspine B regioisomers SphK inhibitory activities revealed that several of these compounds exhibited comparable SphK1/2 inhibitory potency to that of 4-epi-jaspine B.  相似文献

Cyclic and acyclic ether or acetal radicals were generated directly from ethers or acetals by the action of dimethylzinc-air, and their subsequent conjugate addition reaction with alkylidenemalonates afforded the corresponding conjugate adducts in reasonably high yields. The reaction with benzylidenemalonates bearing formyl and imino groups gave chemoselectively the conjugate addition products. The asymmetric reaction of bis(8-phenylmenthyl) benzylidenemalonate proceeded diastereoselectively to provide the adduct with 93:7 dr.  相似文献

[reaction: see text] An efficient one-step synthesis of 2,4-diazabicyclo[4.2.0]octa-1(6),2-dien-5-ones 3 from methyl 2-chloro-2-cyclopropylideneacetate (1) and amidines 2a-c as well as N,N-dimethylguanidine (2d) is described. Similar to the benzocyclobutenes, the cyclobutene-annelated pyrimidones 3 undergo thermal ring opening and the resulting o-quinodimethane analogues readily cycloadd dienophiles to yield tetrahydroquinazolone derivatives.  相似文献
