Four new metabolites, including three new oblongolides named C1, P1, and X1 (1-3) and 6-hydroxyphomodiol (10), along with eight known compounds--oblongolides B (4), C (5), D (6), O (7), P (8) and U (9), (3R,4aR,5S,6R)-6-hydroxy-5-methylramulosin (11), and (3R)-5-methylmellein (12)--were isolated from the endophytic fungal strain Phomopsis sp. XZ-01 of Camptotheca acuminate. Their structures were elucidated by spectroscopic analyses, including 1H- and 13C-NMR, 2D NMR (HSQC, HMBC, 1H-1H COSY and NOESY) and HR-FT-MS. Cytotoxic activities of these compounds were evaluated. Some of them showed weak selective activities.  相似文献

Thymine glycol (Tg), 5,6-dihydroxy-5,6-dihydrothymine, is formed in DNA by the reaction of thymine with reactive oxygen species. The 5R Tg lesion was incorporated site-specifically into 5'-d(G(1)T(2)G(3)C(4)G(5)Tg(6)G(7)T(8)T(9)T(10)G(11)T(12))-3'; Tg = 5R Tg. The Tg-modified oligodeoxynucleotide was annealed with either 5'-d(A(13)C(14)A(15)A(16)A(17)C(18)A(19)C(20)G(21)C(22)A(23)C(24))-3', forming the Tg(6) x A(19) base pair, corresponding to the oxidative damage of thymine in DNA, or 5'-d(A(13)C(14)A(15)A(16)A(17)C(18)G(19)C(20)G(21)C(22)A(23)C(24))-3', forming the mismatched Tg(6) x G(19) base pair, corresponding to the formation of Tg following oxidative damage and deamination of 5-methylcytosine in DNA. At 30 degrees C, the equilibrium ratio of cis-5R,6S:trans-5R,6R epimers was 7:3 for the duplex containing the Tg(6) x A (19) base pair. In contrast, for the duplex containing the Tg(6) x G(19) base pair, the cis-5R,6S:trans-5R,6R equilibrium favored the cis-5R,6S epimer; the level of the trans-5R,6R epimer remained below the level of detection by NMR. The data suggested that Tg disrupted hydrogen bonding interactions, either when placed opposite to A(19) or G(19). Thermodynamic measurements indicated a 13 degrees C reduction of T(m) regardless of whether Tg was placed opposite dG or dA in the complementary strand. Although both pairings increased the free energy of melting by 3 kcal/mol, the melting of the Tg x G pair was more enthalpically favored than was the melting of the Tg x A pair. The observation that the position of the equilibrium between the cis-5R,6S and trans-5R,6R thymine glycol epimers in duplex DNA was affected by the identity of the complementary base extends upon observations that this equilibrium modulates the base excision repair of Tg [Ocampo-Hafalla, M. The synthesis of (2S,5R)-5-hydroxy-6-oxo-1,2-piperidinedicarboxylates (5) and related (3S,6R)-3-hydroxy-6-alkyl-2-oxo-1-piperidinecarboxylates has been developed. The approach is based on the asymmetric hydroxylation of enolates generated from the corresponding N-protected-6-substituted piperidin-2-ones. The utility of 5a as a precursor in the synthesis of (2S,5R)-5-hydroxylysine (1), an amino acid unique to collagen and collagen-like proteins, has also been demonstrated. (2S)-6-oxo-1,2-piperidinedicarboxylates (6) required for hydroxylation studies were prepared in 38-74% yield, starting from conveniently protected aspartic acid as inexpensive chiral adduct. Hydroxylation of 6 to 5 proceeds in high yield and excellent diastereoselectivity by treatment of their Li-enolate with (+)-camphorsulfonyloxaziridine at -78 degrees C. Ring opening of di-tert-butyl (2S,5R)-6-oxo-1,2-piperidinedicarboxylate ((5R)-5a) under reductive conditions afforded the corresponding 1,2-diol (17) in 91%, which was further transformed to (2S,5R)-5-hydroxylysine in four steps (84%). 17 is also a versatile intermediate in the preparation of tert-butyl (2S,5R)-2-[(tert-butoxycarbonyl)amino]-5-hydroxy-6-iodohexanoate (3) and tert-butyl (2S)-2-[(tert-butoxycarbonyl)amino]-4-[(2R)-oxiranyl]butanoate (4), two amino acid derivatives used in the total synthesis of the bone collagen cross-link (+)-pyridinoline (2a).  相似文献

Two new benzyl gamma-butyrolactone analogues, (R)-5-((S)-hydroxy(phenyl)-methyl)dihydrofuran-2(3H)-one (1) and its 6-acetate (2), and a new naphthalenone derivative (8), together with eight additional known aromatic derivatives, (S)-5-((S)-hydroxy(phenyl)-methyl)dihydrofuran-2(3H)-one (3), (S)-5-benzyl-dihydrofuran-2(3H)-one (4), 5-phenyl-4-oxopentanoic acid (5), gamma-oxo-benzenepentanoic acid methyl ester (6), 3-(2,5-dihydro-4-hydroxy-5-oxo-3-phenyl-2-furyl)propionic acid (7), (3R)-5-methylmellein (9), integracins A (10) and B (11) were isolated from Cytospora sp., an endophytic fungus isolated from Ilex canariensis from Gomera. The structures of these compounds were elucidated by detailed spectroscopic analysis, comparison with reported data, and chemical interconversion. The absolute configurations of the new compounds (1, 2, 8) were established on the basis of optical rotation or CD spectra analysis. Preliminary studies showed antimicrobial activity of these compounds against the fungi Microbotryum violaceum, Botrytis cinerea and Septoria tritici, the alga Chlorella fusca, and the bacterium Bacillus megaterium.  相似文献

A novel angiogenesis inhibitor, 5-demethoxyfumagillol (1), was obtained by isolation, purification and saponification of cultured broth of Aspergillus fumigatus. The structure was assigned as (3R,4R,6R)-4-[(2R,3R)-2-methyl-3-(3-methyl-but-2-enyl)-oxiranyl]-1-oxa-spiro[2,5]octan-6-ol (1) by spectroscopic analysis and confirmed by independent synthesis from fumagillol (3). In addition, 6-O-(chloroacetylcarbamoyl)-5-demethoxyfumagillol (7) showed a potential anti-angiogenic activity in CAPE cells in vitro.  相似文献

The complexes [(C5R5)RuH(dippae)] [R = H (1a), Me (2a); dippae = 1,2-bis(diisopropylphosphinoamino)ethane] and [(C5R5)RuH((R,R)-dippach)] [R = H (1b), Me (2b); (R,R)-dippach = (R,R)-1,2-bis(diisopropylphosphinoamino)cyclohexane] have been prepared and characterized. The cationic ruthenium(IV) dihydride derivatives [(C5R5)RuH2(dippae)][BPh4] [R = H (3a), Me (4a)] and [(C5R5)RuH2((R,R)-dippach)][BPh4] [R = H (3b), Me (4b)] are also reported. No significant intramolecular interaction between the amino protons and the hydrogen atoms bound to the metal has been observed in any of these compounds. The X-ray crystal structure of 4a was determined. The proton-transfer processes over the monohydrides 2a and 2b with HBF4.OEt2 have been studied by NMR spectroscopy. Dicationic dihydride complexes [CpRuH2(LH)]2+ [LH = dippaeH+ (5a), (R,R)-dippachH+ (5b)] and [Cp*RuH2(LH)]2+ [LH = dippaeH+ (6a), (R,R)-dippachH+ (6b)] result respectively from the protonation of either the monohydrides 1a,b or 2a,b or the dihydrides 3a,b or 4a,b at one of the NH groups of the phosphinoamine ligands by an excess of HBF4. These dicationic derivatives exhibit fluxional behavior in solution. In the course of the protonation of 1a with HBF4.OEt2, a cationic dihydrogen complex and a dihydrogen-bonded derivative have been identified as intermediates by NMR spectroscopy. Another dihydrogen species, namely, [CpRu(H...HOOCPh)((R,R)-dippach)], was also identified in the course of the reaction of 1b with benzoic acid in toluene-d8. The reaction of 1a with 0.5 equiv of 1,1,1,3,3,3-hexafluoroisopropanol generates a hydride species having a very short (T1)min of 6.5 ms at 400 MHz, an experimental fact for which no satisfactory explanation has yet been found.  相似文献

Combinatorial solution-phase cycloadditions of (1Z,4R*,5R*)-4-benzoylamino-5-phenylpyrazolidin-3-on-1-azomethine imines 3 to beta-keto esters 4 afforded a library of 26 bicyclic pyrazolidinones 5 in 6-89% yields and in 14-100% de. All products were isolated in >90% purity according to 1H NMR, and 25 of them were analytically pure. The structures of cycloadducts were confirmed by NMR and X-ray diffraction. Most of the products were isolated as mixtures of the major (1S*,2S*,3R*,5R*,6R*)-epimers 5 and the minor (1R*,2S*,3R*,5R*,6R*)-epimers 6. Epimerization of cycloadducts 5/6 at the anomeric position 1 in solution was confirmed by 1H NMR.  相似文献

A convenient approach for the preparation of (2S,3R,4R,5S,6R)-2-(3-(4-ethylbenzyl)-4-chlorophenyl)-6-(hydroxymethyl)- tetrahydro-2H-pyran-3,4,5-triol I is developed. The target compound via four steps is synthesized from 4-bromo-2-(bromomethyl)- 1-chlorobenzene and the isomers of undesired ortho-products were avoided during the preparation.  相似文献

The phosphinoboranes [R(2)PB(C(6)F(5))(2)](2) (R = Et 1, Ph 2) and R(2)PB(C(6)F(5))(2) (R = tBu 3, Cy 4, Mes 5) were synthesized from the reaction of (C(6)F(5))(2)BCl and the corresponding lithium phosphide. The relationships between B-P distance, P pyramidality, and the extent of BP multiple bonding were further explored computationally. Natural Bond Order (NBO) analyses of 3 and 4 showed that the π-bonding highest occupied molecular orbitals (HOMOs) were highly polarized. In addition the Lewis acid-base adducts, R(2)(H)P·B(H)(C(6)F(5))(2) (R = Et 6; Ph 7; tBu 8; Cy 9; Mes 10) were prepared via the reaction of the phosphines R(2)PH with the borane HB(C(6)F(5))(2). Compounds 1 and 2 showed no signs of reaction with H(2); however, reaction of compounds 3 and 4 with H(2) was observed to give 8 and 9. In a related set of reactions compounds 3 and 4 were reacted with H(3)NBH(3) or Me(2)(H)NBH(3) also led to the generation of 8 and 9, respectively. The reaction profile of the reaction of (CF(3))(2)BPR(2) with H(2) was examined computationally and shown to be exothermic. Efforts to effect the reverse reaction, that is, dehydrogenation of adducts 6-10 were unsuccessful. Compound 4 was also shown to react with 4-tert-butylpyridine to give Cy(2)PB(C(6)F(5))(2)(4-tBuC(5)H(4)N) 11 while reactions of 3 and 4 with the Lewis acid BCl(3) gave the dimers (R(2)PBCl(2))(2) (R = tBu 12, Cy 13) and the byproduct ClB(C(6)F(5))(2).  相似文献

The C-O bond cleavage from benzophenone substituted with 4-CH2OR (p-BPCH2OR, 1-3), such as p-phenoxymethylbenzophenone (1, R= C6H5) and p-methoxymethylbenzophenone (2, R= CH3), occurred by a stepwise two-photon excitation during two-color, two-laser flash photolysis. On the other hand, no C-O bond cleavage occurred from p-hydroxymethylbenzophenone (3, R = H). The first 355-nm laser excitation of 1-3 generates p-BPCH2OR in the lowest triplet excited state (T1) which has an absorption at 532 nm. When p-BPCH2OR(T1) is excited with the second 532-nm laser to p-BPCH2OR in the higher triplet excited state (T(n)), the C-O bond cleavage occurred within the laser flash duration of 5 ns. The quantum yields of the C-O bond cleavage during the second 532-nm laser irradiation were found to be 0.015 +/- 0.007 and 0.007 +/- 0.003 for 1 and 2, respectively. Although these values are low, the diminishing 1(T1) or 2(T1) was found to convert, in almost 100% yield, to phenoxyl (C6H5O*) and p-benzoylbenzyl (BPCH2*) radicals or methoxyl (CH3O*) and BPCH2* radicals, respectively. The T(n) excitation energy, the energy barrier along the potential surface between the T(n) states and product radicals, and delocalization of the T(n) state molecular orbital including BP and CH2OR (R = C6H5, CH3, H) moieties are important factors for the occurrence of the C-O bond cleavage. It is found that the C-O bond cleavage and production of free radicals, such as BPCH2*, C6H5O*, and CH3O*, can be performed by a stepwise two-photon excitation. The present study is an example in which the chemical reactions can be selectively initiated from the T(n) state but not from the S1 and T1 states.  相似文献

Four new pyrrolidine alkaloids, broussonetines R, S, T, and V and a new pyrroline alkaloid, broussonetine U were isolated from the branches of Broussonetia kazinoki SIEB. (Moraceae) in low yield. And broussonetines U and V were proposed to be (2S,3S,4S)-2-hydroxymethyl-3, 4-dihydroxy-5-(9-oxo-13-hydroxytridecyl)-5-pyrroline (4), (2R,3S,4R,5R)-2-hydroxymethyl-3,4-dihydroxy-5-[(E)-9-oxo-13-hydroxy-3-tridecenyl] pyrrolidine (5), respectively, by spectroscopic and chemical methods.  相似文献

The title compound, spiro[1-bromo(S)-4-(R)-hydroxy-5-oxa-6-oxo-bicyclo[3.1.0]-hxane-2,2'-(3'-diethyl-α-(S)-4'-Cl-benzyloxyphosphonyl-4'-(1R,2S,5R)-menthyloxybutyrolactone)] has been synthesized via the tandem asymmetric reaction and it crystallizes in a monoclinic system, space group P21 with a = 11.067(3), b = 12.484(2), c = 12.356(2)(A), β = 101.95°,C29H39BrC1O10P, Mr = 693.93, V= 1670.2(6) (A)3, Z= 2, Dc= 1.380 g/cm3, λ(MoKα) = 0.071073nm, μ = 1.410 mm-1, F(000) = 720, the final R = 0.0570 and wR = 0.0758 for 6190 observed reflections with I > 2σ(I). The structure is characterized by the special combination of biologic phosphonyl group and one cyclopropane as well as two butyrolactones. Synthesis and Chirality of (5S,6R)-5,6-Epoxy-5,6-dihydro-β,β-carotene and (5R,6R)-5,6-Dihydro-β,β-carotene-5,6-diol, a Compound with Unexpected Solubility Characteristics Wittig-condensation of azafrinal ( 1e ) with the phosphorane derived from 7 leads to a (1:3)-mixture of (E)-9′- and (Z)-9′-β,β-carotene-diol 3 , from which pure and optically active 3 ((5R,6R)-5,6-dihydro-β,β-carotene-5,6-diol) has been isolated as bright violet leaflets, m.p. 168°. Due to the trans-configuration of the diol moiety and to severe steric hindrance, hydrogen bonding is reduced to such an extent, that 3 behaves much more as a hydrocarbon than as a diol. There is good evidence that the so-called ‘β-oxycarotin’ obtained by Kuhn & Brockmann [15] by chromic acid oxidation of β, β-carotene is the corresponding racemic cis-diol. 3 has been converted into (5S, 6R)-5,6-epoxy-5.6-dihydro-β,β-carotene ( 4 ), m.p. 156°. This transformation establishes for the first time the chirality of a caroteneepoxide (without other O-functions). Full spectral and chiroptical data including a complete assignement of 13C-chemical shifts for azafrin methyl ester and 3 are presented.  相似文献

Two new acaranoic acids, named seimatoporic acid A and B (1, and 2), together with six known compounds, R-(-)-mellein (3), cis-4-hydroxymellein (4), trans-4-hydroxymellein (5), 4R-hydroxy-5-methylmellein (6), (-)-5-hydroxymethylmellein (7), and ergosterol (8) were isolated from an endophytic fungus, Seimatosporium sp, by a bioassay-guided procedure. The structures of the new compounds have been assigned from analysis of the 1H and 13C NMR spectra, DEPT, and by 2D COSY, HMQC, HMBC and NOESY experiments. A mixture of compounds 1 and 2 showed strong antifungal activity against Botrytis cinerea, Septoria tritici, and Pyricularia oryzae.  相似文献

A new steroidal sapogenin named 25-R-spirosta-3,5-dien-12β-ol(1) was isolated from the dried roots of Chlorophytum laxum R. Br. along with five known compounds, namely, diosgenin(2), stigmasterol(3), β-sitosterols(4), estigmasterol-3-O-β-D-glicopyranoside(5) and 3-O-β-authemisol(6). The structure of compound 1 was elucidated by the analysis of IR, HRESI-MS, 1D and 2D NMR spectral data. Compounds 2-5 were isolated from Chlorophytum laxum R. Br. In addition, all the compounds were evaluated for cytotoxicity on the human nasopharyngeal carcinoma cancer cell line 5-8F. Among them, the newly identified 25-R-spirosta-3,5-dien-12β-ol(1) and diosgenin(2) exhibited high cytotoxicity on 5-8F cells, with IC50 values of 24.8 and 41.9 μmol/L, respectively.  相似文献

The reported enzymatic resolution products {acetate of (1S,4aS,8aS)-1,2,3,4,4a,5,6,7,8,8a-decahydro-5,5,8a-trimethyl-2-oxo-trans-naphthalene-1-methanol-2-ethylene acetal} (8aS)-5 (>99% ee)] and [(1R,4aR,8aR)-1,2,3,4,4a,5,6,7,8,8a-decahydro-5,5,8a-trimethyl-2-oxo-trans-naphthalene-1-methanol-2-ethylene acetal (8aR)-4 (98% ee) were converted to (+)-alpha-polypodatetraene (1) and methyl (5R,10R,13R)-labda-8-en-15-oate (2), respectively. For the synthesis of (5R,10R,13R)-2, chiral isoprene congener (3S)-26 corresponding to the right part of 2 was synthesized based on the lipase-assisted resolution of (+/-)-2-methyl-3- (p-methoxyphenyl)propanol (17).  相似文献

2-Aminomethylaniline was converted into the N,N'-bis(pivaloyl) (1) or -bis(trimethylsilyl) (2) derivative, using 2 Bu(t)C(O)Cl or 2 Me(3)SiCl (≡ RCl), respectively, with 2 NEt(3), or for 2 from successively using 2 LiBu(n) and 2 RCl. N,N'-Bis(neopentyl)-2-(aminomethyl)aniline (3) was prepared by LiAlH(4) reduction of 1. From 2 or 3 and 2 LiBu(n), the appropriate dilitiodiamide {2-[{N(Li)R}C(6)H(4){CH(2)N(Li)R}(L)](2) (L absent, 4a; or L = THF, 4b) or the N,N'-bis(neopentyl) analogue (5) of 4a was prepared. Treatment of 4a with 2 Bu(t)NC, 2 (2,6-Me(2)C(6)H(3)NC) or 2 Bu(t)CN (≡ L') furnished the corresponding adduct [2-N{Li(L')R}C(6)H(4){CH(2)N(Li)R}] (4c, 4d or 4e, respectively), whereas 4b with 2 PhCN afforded [2-{N(Li)R}C(6)H(4){CH(2)C(Ph) = NLi(NCPh)}] (6). The dimeric bis(amido)stannylene [Sn{N(R)C(6)H(4)(CH(2)NR)-1,2}](2) (7) was obtained from 4a and [Sn(μ-Cl)NR(2)](2), while the N,N'-bis(neopentyl) analogue 8 of 7 was similarly derived from [Sn(μ-Cl)NR(2)](2) and 5. Reaction of two equivalents of the diamine 2 with Pb(NR(2))(2) yielded 9, the lead homologue of 7. Oxidative addition of sulfur to 7 led to the dimeric bis(diamido)tin sulfide 10. Treatment of 2 successively with 'MgBu(2)' in C(5)H(12) and THF gave [Mg{N(R)C(6)H(4)(CH(2)NR)}(THF)](2) (11a), which by displacement of its THF by an equivalent portion of Bu(t)CN or PhCN produced [Mg{N(R)C(6)H(4)(CH(2)NR)}(CNR')(n)] [R' = Bu(t), n = 1 (11b); R' = Ph, n = 2 (11c)]. The Ca (12), Sr (13) or Ba (14) analogues of the Mg compound 11a were isolated from 2 and either the appropriate compound M(NR(2))(2) (M = Ca, Sr, Ba), or successively 2 LiBu(n) and 2 M(OTos)(2). The new compounds 1-14 were characterized by microanalysis (C, H, N; not for 1, 2, 3, 5), solution NMR spectra, ν(max) (C≡N) (IR for 4c, 4d, 4e, 6, 11b, 11c), selected EI-MS peaks (for 1, 2, 3, 7, 8, 9, 10), and single crystal X-ray diffraction (for 4a, 4b, 11a).  相似文献

A general approach to (5S,6R)-6-alkyl-5-benzyloxy-2-piperidinones based on the regio- and diastereoselective reductive alkylation of (S)-3-benzyloxyglutarimide 7 is described. This method opens an entrance to chiral nonracemic substituted 3-piperidinols. The versatility of the method is illustrated by the asymmetric syntheses of neurokinin substance P receptor antagonist L-733,061 (ent-1), (-)-deoxocassine (4), and an inhibitor of HIV proteases (5a).  相似文献
