A new, stereoselective formal synthesis of hirsutic acid and medium ring carbocyclic systems from salicyl alcohol is described. Cycloaddition between electron deficient partners such as cyclohexa-2,4-dienone and methylmethacrylate, triplet sensitized 1,2-acyl shift and ring-closing metathesis are the key features of our approach.  相似文献

Synthesis of endotetracyclo[5.5.2.0.2,608,12]tetradeca-3(4),8(12)-dien-13-one from 5-indanol and photoreactions in singlet and triplet excited state leading to complex polycyclic systems is reported. Crystal structure of 14-spiroepoxyendotetracyclo[5.5.2.0.2,608,12] tetradeca-3(4),8(12)-dien-13-one is also reported.  相似文献

A regio- and stereoselective cycloaddition of 6,6-spiroepoxycyclohexa-2,4-dienones with indene leading to benzo-annulated spiroepoxytricyclo[5.2.2.02,6]undec-10-en-9-ones has been described. Manipulation of the oxirane ring in adducts furnished benzotricyclo[5.2.2.02,6]undecanes endowed with a β,γ-enone chromophore. Photochemical reaction of β,γ-enones upon direct irradiation (1S) gave annulated bicyclo[4.2.0]octenones as a consequence of a stereoselective 1,3-acyl shift. Triplet excitation of chromophoric systems led to oxa-di-π-methane reaction and gave pentacyclic products which upon cleavage of cyclopropane ring furnished benzoannulated triquinanes. An interesting effect of substituent on the photoreaction has also been observed.  相似文献

A total synthesis of hirsutene, a triquinane sesquiterpene, from salicyl alcohol is reported. Oxidation of salicyl alcohol in the presence of cyclopentadiene gave 9-spiroepoxy-endo-tricyclo[5.2.2.02,6]undeca-4,10-dien-8-one which was elaborated to the 3-hydroxy-2-methyl-endo-tricyclo[5.2.2.02,6]undeca-10-en-8-one containing major structural and functional features of hirsutene. Photochemical sigmatropic 1,2-acyl shift in 3-hydroxy-2-methyl-endo-tricyclo[5.2.2.02,6]undeca-10-en-8-one followed by radical induced cleavage of peripheral cyclopropane bond, olefination and Simmon-Smith reaction furnished 11-hydroxy-1-methyl-4-spirocyclopropanetricyclo[6.3.0.02,6]undecane that upon treatment with hydrogen on PtO2 and PCC oxidation gave 1,4,4-trimethyltricyclo[6.3.0.02,6]undecan-11-one, a known precursor. Wittig methylenation on this precursor gave hirsutene.  相似文献

Synthesis of azepane annulated bicyclo[2.2.2]octenone via oxidative dearomatization and intramolecular π4s?+?π2s cycloaddition has been described. Manipulation of the adduct led to tricyclic compounds having a β,γ-enone chromophore whose photoreaction upon triplet and singlet excitation furnished polycyclic systems having azepane ring systems.  相似文献

An expedient route to tetracyclo[6.5.2.02,7.09,13]pentadec-2(7),11-dien-14-one and tetracyclic framework of conidiogenol have been reported. Cycloaddition of annulated cyclohexa-2,4-dienone with cyclopentadiene, photochemical oxa-di-π-methane reaction and a highly unusual dehalogenation of bridgehead halogen are the key features of our methodology.  相似文献

A new and efficient synthesis of a variety of highly embellished bicyclooctenones having an endo-vinyl moiety and their sigmatropic shifts in ground and excited states leading to a stereoselective route to substituted cis-decalins and diquinane frameworks have been described. Functionalized bicyclo[2.2.2]octenones having an endo-vinyl group and a beta,gamma-enone chromophore were prepared by in situ generation of 6-chloromethyl-6-hydroxycyclohexadienones and cycloaddition with butadiene (also generated in situ) followed by manipulation of the adducts. The presence of contiguous carbonyl, hydroxyl, and chloromethyl groups in adducts led to the introduction of various alkyl groups alpha to the ketone in a stereoselective fashion. The 3,3-sigmatropic shift in the bridged bicyclic compounds gave the corresponding cis-decalins, whereas the triplet sensitized irradiation led to the formation of diquinanes as a result of a 1,2-acyl shift.  相似文献

Stereoselective route to tricyclo[5.3.1.01,5]undecane and bicyclo[4.2.0]octane ring systems present in the molecular structure of tricycloillicinone, cedrenoids and endiandric acid, elysiapyrones, respectively, is described. A tandem oxidative dearomatization of o-hydroxymethylphenol, π4s+π2s cycloaddition, reactivity modulation in excited state and ring-closing metathesis are the key features of our approach.  相似文献

Total syntheses of title natural products, 1 and 2, have been achieved using the cis-1,2-dihydrocatechol 7 as starting material. Compound 7 is readily obtained in large quantity and enantiomerically pure form through the whole-cell biotransformation of toluene using the genetically engineered micro-organism Escherichia coli JM109 (pDTG601) that over-expresses the enzyme toluene dioxygenase (TDO). Three key chemical steps were employed in these syntheses, the first of which was a high-pressure-promoted Diels-Alder cycloaddition reaction between diene 8 and cyclopentenone to give adduct 9. The second key step was the photochemically promoted oxa-di-π-methane rearrangement of the bicyclo[2.2.2]octenone derivative, 18, of 9 to give 20 while the third key step was the reductive cleavage of the last compound so as to afford the linear triquinane 22. Elaboration of compound 22 to targets 1 and 2 followed conventional and/or established procedures. Single-crystal X-ray analyses were carried out on compounds 10-13, 15, 18, 24, and 34.  相似文献

A novel one step regio- and stereoselective synthesis of functionalised bicyclo[2.2.2]octenones from readily available aromatic precursors is described. The methodology involved in situ generation of reactive spiroepoxycyclohexadienones and π4s2s cycloaddition with methyl vinyl ketone. Study on π-facial alkylation that led to the formation of homobrendane derivatives as a result of stereochemical inversion and aldol condensation in tandem, is also presented. The crystal structure of 6-acetyl-1-methoxy-bicyclo[2.2.2]oct-7-en-2-one-spiro[3,2′]oxirane and 3-methoxy-4,6,9-trimethyltricyclo[4.3.1.03,7]decan-8-en-5-one-spiro[2,2′]oxirane is also reported.  相似文献

An enantiomerically pure cis-1,2-dihydrocatechol, which is readily obtained via a toluene dioxygenase-mediated dihydroxylation of toluene in a whole-cell biotransformation process, has been converted over 17 steps into the linear triquinane (−)-hirsutene. Since the enantiomer of the starting material is also available this work constitutes a formal total synthesis of the naturally occurring (+)-form of hirsutene. Furthermore, minor modifications of the route used here offer the possibility of accessing (+)-hirsutene from the original starting material.  相似文献

Abstract

(1R,2R)-1,2-bis[5-(arylideneamino)-1,3,4-thiadiazol-2-yl]ethane-1,2-diol (2a–d) were synthesized by using appropriate aldehydes and (1R,2R)-1,2-bis(5-amino-1,3,4-thiadiazol-2-yl)ethane-1,2-diol (1) as a starting compound. Then, the phosphinic acid component (3a–d) were obtained from (2a–d) and hypophosporus acid. In addition, the structures of the novel chiral compounds (2a–d) and (3a–d) were confirmed by elemental analyses, IR, 1H-NMR, 13C-NMR, and 31P-NMR spectra.

The reaction of 1,1,1,5,5,5-hexafluoro-2,4-pentanedione with (R)-phenylglycinol was found to proceed via intermediate formation of (R, 4E, 6Z)-5,7-bis-(trifluoromethyl)-2,3-dihydro-3-phenyl-1,4-oxazepine which further underwent a base-catalyzed 1,3-proton shift reaction followed by [1,2] Wittig rearrangement giving rise to 2,4-bis-(trifluoromethyl)-6-phenylpyridine.  相似文献

A practical synthesis of (−)‐englerin A was accomplished in 17 steps and 11 % global yield from commercially available achiral precursors. The key step consists of a platinum‐catalyzed [4C+3C] allenediene cycloaddition that directly delivers the trans‐fused guaiane skeleton with complete diastereoselectivity. The high enantioselectivity (99 % ee) stems from an asymmetric ruthenium‐catalyzed transfer hydrogenation of a readily assembled diene–ynone. The synthesis also features a highly stereoselective oxygenation, and a late‐stage cuprate alkylation that enables the preparation of previously inaccessible structural analogues.  相似文献

1H‐imidazol[1,2‐a]indeno[2,1‐e]pyridine‐6(5H)‐ones derivatives were synthesized in a one‐pot four‐component condensation of corresponding aldehydes, 1,3‐indandione, diamine, and nitro ketene dithioacetal using KAl(SO4)2·12H2O (alum) as nontoxic, reusable, inexpensive and easily available catalyst in good to excellent yields. This green protocol provides a powerful entry into fused polycyclic structures related to bioactive heterocycles.  相似文献

In a search for environmentally friendly metal chelating ligands for industrial applications, the protonation and complex formation equilibria of [S,S,S]- and [R,S,R]-isomers of N-bis[2-(1,2-dicarboxyethoxy)ethyl] aspartic acid (BCA6) with Mg(II), Ca(II), Mn(II), Fe(III), Cu(II) and Zn(II) ions in aqueous 0.1 M NaCl solution were studied at 25°C by potentiometric titration. The model for complexation and the stability constants of the different complexes were determined for each metal ion using the computer program SUPERQUAD. With all metal ions (M n+), stable ML n?6 complexes dominated complex formation for both isomers. Differences in complexation models were found for binuclear species.  相似文献

A metal‐free trifluorosulfonate anhydride (TFAA)‐catalyzed strategy for the synthesis of spiro pyrrolo[1,2‐a]quinoxalines from 1‐(2‐aminophenyl)pyrroles and benzoquinones/ketones has been developed. With this general method, spiro pyrrolo[1,2‐a]quinoxalines have been accessed via nucleophilic addition and cyclization. This reaction exhibits good functional group tolerance, and a wide range of products are obtained in moderate to good yields.  相似文献

Two efficient iodine‐mediated strategies, which are economical and one‐pot, are described to access bis(imidazo[1,2‐a ]pyridin‐3‐yl)sulfanes and bis(imidazo[1,2‐a ]pyridin‐3‐yl)disulfanes in chloroform and acetic acid, respectively, by a direct oxidative homocoupling of imidazo‐heterocycles using inexpensive sodium sulfide as a sulfur source. These strategies are scalable, and an array of substrates delivered their corresponding stable sulfur‐bridged imidazo‐heterocycles in excellent yields.  相似文献
