The enantiomeric separation of d ,l ‐tryptophan (Trp) and d ,l ‐kynurenine (KYN) was investigated by high‐performance liquid chromatography using pre‐column fluorescence derivatization with a chiral fluorescent labeling reagent, R(−)‐4‐(3‐isothiocyanatopyrrolidin‐1‐yl)‐7‐ (N,N‐dimethylaminosulfonyl)‐2,1,3‐benzoxadiazole [R(−)‐DBD‐PyNCS]. Using an octadecylsilica column, namely, an Inertsil ODS‐3 column (250 × 2.0 mm; i.d., 3 µm), four fluorescence peaks of D‐ and l ‐Trp as well as d ‐ and l ‐KYN derivatized with R(−)‐DBD‐PyNCS were clearly observed, and their chemical structures were confirmed by HPLC–time‐of‐flight–mass spectrometry. Simultaneous separation was achieved under the mobile phase condition of 1.5% acetic acid in H2O–CH3CN (60:40), and the separation factors of d ,l ‐Trp and d ,l ‐KYN derivatized with R(−)‐DBD‐PyNCS were 1.22 and 1.19, respectively. Fluorescence detection was carried out by setting the emission wavelength at 565 nm, and the excitation wavelength at 440 nm, and the detection limits were approximately 0.3–0.5 pmol (signal‐to‐noise ratio of 3). The Rose Bengal‐sensitized photooxidations of the dipeptides l ‐tryptophyl‐l ‐phenylalanine (Trp‐Phe), l ‐tryptophyl‐l ‐tyrosine (Trp‐Tyr) and l ‐tryptophyl‐l ‐tryptophan (Trp‐Trp) have been studied in pH 7 water solution using static photolysis and time‐resolved methods. Kinetic results indicate that the tryptophan (Trp) moiety interacts with singlet molecular oxygen (O2(1Δg)) both through chemical reaction and through physical quenching, and that the photooxidations can be compared with those of equimolecular mixtures of the corresponding free amino acids, with minimum, if any, influence of the peptide bond on the chemical reaction. This is not a common behavior in other di‐ and polypeptides of photooxidizable amino acids. The ratio between chemical (kr) and overall (kt) rate constants for the interaction O2(1Δg)‐dipeptide indicates that Trp‐Phe and Trp‐Trp are good candidates to suffer photodynamic action, with krlkt values of 0.72 and 0.60, respectively (0.65 for free Trp). In the case of Trp‐Tyr, a lower krlkt value (0.18) has been found, likely as a result of the high component of physical deactivation of O2(1Δg) by the tyrosine moiety. The analysis of the photooxidation products shows that the main target for O2(1Δg) attack is the Trp group and suggests a much lower accumulation of kynurenine‐type products, as compared with free Trp. This is possibly because of the occurrence of another accepted alternative pathway of oxidation that gives rise to 3a‐oxidized hydrogenated pyrrolo[2,3‐b]indoles.  相似文献

A method for the detection of unlabeled and 15N2‐labeled l ‐tryptophan (l ‐Trp), l ‐kynurenine (l ‐Kyn), serotonin (5‐HT) and quinolinic acid (QA) in human and rat plasma by GC/MS is described. Labeled and unlabeled versions of these four products were analyzed as their acyl substitution derivatives using pentafluoropropionic anhydride and 2,2,3,3,3‐pentafluoro‐1‐propanol. Products were then separated by GC and analyzed by selected ion monitoring using negative ion chemical ionization mass spectrometry. l ‐[13C11, 15N2]‐Trp, methyl‐serotonin and 3,5‐pyridinedicarboxylic acid were used as internal standards for this method. The coefficients of variation for inter‐assay repeatability were found to be approximately 5.2% for l ‐Trp and 15N2‐Trp, 17.1% for l ‐Kyn, 16.9% for 5‐HT and 5.8% for QA (n = 2). We used this method to determine isotope enrichments in plasma l ‐Trp over the course of a continuous, intravenous infusion of l ‐[15N2]Trp in pregnant rat in the fasting state. Plasma 15N2‐Trp enrichment reached a plateau at 120 min. The free Trp appearance rate (Ra) into plasma was 49.5 ± 3.35 µmol/kg/h. The GC/MS method was applied to determine the enrichment of 15N‐labeled l ‐Trp, l ‐Kyn, 5‐HT and QA concurrently with the concentration of non‐labeled l ‐Trp, l ‐Kyn, 5‐HT and QA in plasma. This method may help improve our understanding on l ‐Trp metabolism in vivo in animals and humans and potentially reveal the relative contribution of the four pathways of l ‐Trp metabolism. The crystal structure of N‐(l ‐2‐amino­butyryl)‐l ‐alanine, C7H14N2O3, is closely related to the structure of l ‐alanyl‐l ‐alanine, both being tetragonal, while the retro‐analogue 2‐(l ‐alanyl­amino)‐l ‐butyric acid 0.33‐hydrate, C7H14N2O3·­0.33H2O, forms a new type of molecular columnar structure with three peptide mol­ecules in the asymmetric unit.  相似文献

Tryptophan (TRP) is an essential amino acid catabolized mainly through the kynurenine pathway, and part of it is catabolized in the brain. The abnormal depletion of TRP and production of kynurenine (KYN) by two enzymes, tryptophan 2,3-dioxygenase (TDO) and indoleamine 2,3-dioxygenase (IDO), have been linked to various neurological diseases. The ratio of TRP/KYN in plasma is a valuable measure for IDO/TDO activity and the prognosis of disease conditions. The 4-vinylphenylboronic acid (4-VPBA) was evaluated as a novel stationary phase for OT–CEC–MS/MS. TRP, KYN, and 3-hydroxykynurenine were separated using optimum conditions of 15 mM (NH4)2CO3 at pH 8 as a background electrolyte and 25 kV separation voltage on a 90 cm column. The usefulness of the 4-VPBA column for simple, fast, repeatable, and sensitive CEC–ESI–MS/MS application was demonstrated for the quantitation of TRP and KYN in the plasma of healthy human subjects and neuroinflammation subjects. The plasma sample was extracted on a zirconia-based ion-exchange cartridge for simultaneous protein precipitation and phospholipid removal. The method of standard addition, in combination with the internal standards approach, was used to prepare the calibration curve to overcome matrix matching and eliminate procedural errors. The developed quantitation method was validated according to FDA guidelines for sensitivity, accuracy, precision, and extraction recovery. The measured plasma level of TRP and KYN in healthy humans is aligned with the human metabolome database for the same two metabolites.  相似文献

An ultra‐high‐performance liquid chromatography–mass spectrometry (UPLC/MS/MS) method was developed and validated for the quantification of trimethylamine‐N‐oxide (TMAO) simultaneously with TMAO‐related molecules l ‐carnitine and γ‐butyrobetaine (GBB) in human blood plasma. The separation of analytes was achieved using a Hydrophilic interaction liquid chromatography (HILIC)‐type column with ammonium acetate–acetonitrile as the mobile phase. TMAO determination was validated according to valid US Food and Drug Administration guidelines. The developed method was successfully applied to plasma samples from healthy volunteers. When PEG (M.W.~5000 Daltons) is conjugated to poly(l ‐alanine), the polymer aqueous solutions (<10.0 wt.%) undergo sol‐to‐gel (thermal gelation), whereas it is conjugated to poly(l ‐lactic acid), the polymer aqueous solutions (>30.0 wt.%) undergo gel‐to‐sol (gel melting) as the temperature increases. In the search for molecular origins of such a quite different phase behavior, poly(ethylene glycol)‐poly(l ‐alanine) (PEG‐PA; EG113‐A12) and poly(ethylene glycol)‐poly(l ‐lactic acid) (PEG‐PLA; EG113‐LA12) are synthesized and their aqueous solution behavior is investigated. PEG‐PAs with an α‐helical core assemble into micelles with a broad size distribution, and the dehydration of PEG drives the aggregation of the micelles, leading to thermal gelation, whereas increased molecular motion of the PLA core overwhelms the partial dehydration of PEG, thus gel melting of the PEG‐PLA aqueous solutions occurs. The core‐rigidity of micelles must be one of the key factors in determining whether a polymer aqueous solution undergoes sol‐to‐gel or gel‐to‐sol transition, as the temperature increases. © 2014 Wiley Periodicals, Inc. We have developed and validated a high‐performance liquid chromatography method that uses monolithic silica disk‐packed spin columns and a monolithic silica column for the simultaneous determination of NG‐monomethyl‐l ‐arginine, NG,NG‐dimethyl‐l ‐arginine, and NG,NG′‐dimethyl‐l ‐arginine in human plasma. For solid‐phase extraction, our method employs a centrifugal spin column packed with monolithic silica bonded to propyl benzenesulfonic acid as a cation exchanger. After pretreatment, the methylated arginines are converted to fluorescent derivatives with 4‐fluoro‐7‐nitro‐2,1,3‐benzoxadiazole, and then the derivatives are separated on a monolithic silica column. l ‐Arginine concentration was also determined in diluted samples. Standard calibration curves revealed that the assay was linear in the concentration range 0.2–1.0 μM for methylated arginines and 40–200 μM for l ‐arginine. Linear regression of the calibration curve yielded equations with correlation coefficients of 0.999 (r2). The sensitivity was satisfactory, with a limit of detection ranging from 3.75 to 9.0 fmol for all four compounds. The RSDs were 4.3–4.8% (intraday) and 3.0–6.8% (interday). When this method was applied to samples from six healthy donors, the detected concentrations of NG‐monomethyl‐l ‐arginine, NG,NG‐dimethyl‐l ‐arginine, NG,NG′‐dimethyl‐l ‐arginine and l ‐arginine were 0.05 ± 0.01, 0.41 ± 0.07, 0.59 ± 0.11, and 83.8 ± 30.43 μM (n = 6), respectively.  相似文献

A rapid and sensitive LC–MS/MS method with good accuracy and precision was developed and validated for the pharmacokinetic study of quercetin‐3‐O‐β‐d ‐glucopyranosyl‐7‐O‐β‐d ‐gentiobioside (QGG) in Sprague–Dawley rats. Plasma samples were simply precipitated by methanol and then analyzed by LC–MS/MS. A Venusil® ASB C18 column (2.1 × 50 mm, i.d. 5 μm) was used for separation, with methanol–water (50:50, v/v) as the mobile phase at a flow rate of 300 μL/min. The optimized mass transition ion‐pairs (m/z) for quantitation were 787.3/301.3 for QGG, and 725.3/293.3 for internal standard. The linear range was 7.32–1830 ng/mL with an average correlation coefficient of 0.9992, and the limit of quantification was 7.32 ng/mL. The intra‐ and inter‐day precision and accuracy were less than ±15%. At low, medium and high quality control concentrations, the recovery and matrix effect of the analyte and IS were in the range of 89.06–92.43 and 88.58–97.62%, respectively. The crystal structures of the four dipeptides l ‐seryl‐l ‐asparagine monohydrate, C7H13N3O5·H2O, l ‐seryl‐l ‐tyrosine monohydrate, C12H16N2O5·H2O, l ‐tryptophanyl‐l ‐serine monohydrate, C14H17N3O4·H2O, and l ‐tyrosyl‐l ‐tryptophan monohydrate, C20H21N3O4·H2O, are dominated by extensive hydrogen‐bonding networks that include cocrystallized solvent water molecules. Side‐chain conformations are discussed on the basis of previous observations in dipeptides. These four dipeptide structures greatly expand our knowledge on dipeptides incorporating polar residues such as serine, asparagine, threonine, tyrosine and tryptophan.  相似文献

l ‐4‐Chlorokynurenine (l ‐4‐Cl‐Kyn) is a neuropharmaceutical drug candidate that is in development for the treatment of major depressive disorder. Recently, this amino acid was naturally found as a residue in the lipopeptide antibiotic taromycin. Herein, we report the unprecedented conversion of l ‐tryptophan into l ‐4‐Cl‐Kyn catalyzed by four enzymes in the taromycin biosynthetic pathway from the marine bacterium Saccharomonospora sp. CNQ‐490. We used genetic, biochemical, structural, and analytical techniques to establish l ‐4‐Cl‐Kyn biosynthesis, which is initiated by the flavin‐dependent tryptophan chlorinase Tar14 and its flavin reductase partner Tar15. This work revealed the first tryptophan 2,3‐dioxygenase (Tar13) and kynurenine formamidase (Tar16) enzymes that are selective for chlorinated substrates. The substrate scope of Tar13, Tar14, and Tar16 was examined and revealed intriguing promiscuity, thereby opening doors for the targeted engineering of these enzymes as useful biocatalysts.  相似文献

A sensitive and reliable ultra‐high‐performance liquid chromatography with tandem mass spectrometry (UHPLC–MS/MS) method was developed and validated for simultaneous determination of l ‐tetrahydropalmatine (l ‐THP) and its active metabolites l ‐isocorypalmine (l ‐ICP) and L ‐corydalmine (l ‐CD) in rat plasma. The analytes were extracted by liquid–liquid extraction and separated on a Bonshell ASB C18 column (2.1 × 100 mm; 2.7 μm; Agela) using acetonitrile–formic acid aqueous as mobile phase at a flow rate of 0.2 mL/min in gradient mode. The method was validated over the concentration range of 4.00–2500 ng/mL for l ‐THP, 0.400–250 ng/mL for l ‐ICP and 1.00–625 ng/mL for l ‐CD. Intra‐ and inter‐day accuracy and precision were within the acceptable limits of <15% at all concentrations. Correlation coefficients (r ) for the calibration curves were >0.99 for all analytes. The quantitative method was successfully applied for simultaneous determination of l ‐THP and its active metabolites in a pharmacokinetic study after oral administration with l ‐THP at a dose of 15 mg/kg to rats.  相似文献

Thin silica gel layers impregnated with optically pure l ‐glutamic acid were used for direct resolution of enantiomers of (±)‐isoxsuprine in their native form. Three chiral derivatizing reagents, based on DFDNB moiety, were synthesized having l ‐alanine, l ‐valine and S‐benzyl‐l ‐cysteine as chiral auxiliaries. These were used to prepare diastereomers under microwave irradiation and conventional heating. The diastereomers were separated by reversed‐phase high‐performance liquid chromatography on a C18 column with detection at 340 nm using gradient elution with mobile phase containing aqueous trifluoroacetic acid and acetonitrile in different compositions and by thin‐layer chromatography (TLC) on reversed phase (RP) C18 plates. Diastereomers prepared with enantiomerically pure (+)‐isoxsuprine were used as standards for the determination of the elution order of diastereomers of (±)‐isoxsuprine. The elution order in the experimental study of RP‐TLC and RP‐HPLC supported the developed optimized structures of diastereomers based on density functional theory. The limit of detection was 0.1–0.09 µg/mL in TLC while it was in the range of 22–23 pg/mL in HPLC and 11–13 ng/mL in RP‐TLC for each enantiomer. The conditions of derivatization and chromatographic separation were optimized. The method was validated for accuracy, precision, limit of detection and limit of quantification. Starting from methyl 2,3‐O‐isopropylidene‐α‐D ‐mannofuranoside ( 5 ), methyl 6‐O‐benzyl‐2,3‐O‐isopropylidene‐α‐D ‐lyxo‐hexofuranosid‐5‐ulose ( 12 ) was prepared in three steps. The addition reaction of dimethyl phosphonate to 12 , followed by deoxygenation of 5‐OH group, provided the 5‐deoxy‐5‐dimethoxyphosphinyl‐α‐D ‐mannofuranoside derivative 15a and the β‐L ‐gulofuranoside isomer 15b . Reduction of 15a and 15b with sodium dihydrobis(2‐methoxyethoxy)aluminate, followed by the action of HCl and then H2O2, afforded the D ‐mannopyranose ( 17 ) and L ‐gulopyranose analog 21 , each having a phosphinyl group in the hemiacetal ring. These were converted to the corresponding 1,2,3,4,6‐penta‐O‐acetyl‐5‐methoxyphosphinyl derivatives 19 and 23 , respectively, structures and conformations (4C1 or 1C4, resp.) of which were established by 1H‐NMR spectroscopy.  相似文献

Elution profiles of kynurenic acid (KYNA) and 7‐chlorokynurenic acid (Cl‐KYNA) were examined by high‐performance liquid chromatography (HPLC) using a triazole‐bonded stationary phase column (Cosmosil® HILIC) under isocratic elution of a mobile phase consisting of CH3CN–aqueous 10 mm ammonium formate between pH 3.0 and 6.0. The capacity factors of KYNA and Cl‐KYNA varied with both the CH3CN content and the pH of the mobile phase. The elution order of KYNA and Cl‐KYNA was reversed between the CH3CN‐ and H2O‐rich mobile phases, suggesting that hydrophilic interactions and anion‐exchange interactions caused retention of KYNA and Cl‐KYNA in the CH3CN‐ and H2O‐rich mobile phases, respectively. The present HPLC method using a triazole‐bonded column and fluorescence detection (excitation 250 nm, emission 398 nm) was applied to monitor in vitro production of KYNA from d ‐kynurenine (d ‐KYN) by d ‐amino acid oxidase (DAO) using Cl‐KYNA as an internal standard. A single KYNA peak was clearly observed after enzymatic reaction of d ‐KYN with DAO. Production of KYNA from d ‐KYN was suppressed by the addition of commercial DAO inhibitors. The title peptide N‐benzyl­oxy­carbonyl–ΔLeu–l ‐Ala–l ‐Leu–OCH3 [methyl N‐(benzyl­oxy­carbonyl)‐α,β‐de­hydro­leucyl‐l ‐alanyl‐l ‐leucinate], C24H35N3O6, was synthesized in the solution phase. The peptide adopts a type II′β‐turn conformation which is stabilized by an intramolecular 4 1 N—H?O hydrogen bond. In the chiral polymeric title compound, poly[aqua(4,4′‐bipyridine)[μ3S‐carboxylatomethyl‐N‐(p‐tosyl)‐l ‐cysteinato]manganese(II)], [Mn(C12H13NO6S2)(C10H8N2)(H2O)]n, the MnII ion is coordinated in a distorted octahedral geometry by one water molecule, three carboxylate O atoms from three S‐carboxyatomethyl‐N‐(p‐tosyl)‐l ‐cysteinate (Ts‐cmc) ligands and two N atoms from two 4,4′‐bipyridine molecules. Each Ts‐cmc ligand behaves as a chiral μ3‐linker connecting three MnII ions. The two‐dimensional frameworks thus formed are further connected by 4,4′‐bipyridine ligands into a three‐dimensional homochiral metal–organic framework. This is a rare case of a homochiral metal–organic framework with a flexible chiral ligand as linker, and this result demonstrates the important role of noncovalent interactions in stabilizing such assemblies.  相似文献

Reliable methods for the determination of tryptophan and its metabolites are vital to the monitoring of biochemical states during the initiation and progression of cardiovascular disease. In the present study, a single‐run liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method was developed for the simultaneous determination of tryptophan (Trp) and its metabolites, including kynurenine (Kyn), kynurenic acid (KA), xanthurenic acid (XA) and 5‐hydroxytryptamine (5‐HT), in human plasma. The plasma samples were prepared using a protein precipitation approach, and the chromatographic separation was performed by gradient elution on a C18 column within a total analysis time of 3.5 min. The calibration ranges were 40–20,000 ng/mL for Trp, 4–2000 ng/mL for Kyn, 0.2–100 ng/mL for KA, 0.4–200 ng/mL for XA and 1–500 ng/mL for 5‐HT, and the precision and accuracy were acceptable. The evaluation of recovery and internal standard‐normalized matrix effect proved that the sample preparation approach was effective and the matrix effect could be negligible. The newly developed method was successfully applied to the analysis of plasma samples from healthy individuals and myocardial infarction patients. The findings suggested that the plasma concentrations of Trp, Kyn, 5‐HT as well as the concentration ratios of Kyn/Trp and Trp/5‐HT might serve as biomarkers for the monitoring of acute myocardial infarction.  相似文献

The title compound (with the systematic name 2‐{[(1S)‐1‐(methoxy­carbonyl)‐3‐methyl­butyl]amino­carbonyl}benzoic acid), C15H19NO5, crystallizes in the monoclinic space group P21, with two independent mol­ecules per asymmetric unit. The most notable difference between the two mol­ecules is in the dihedral angles between the planes of the carboxyl group and the benzene ring, which are 3.5 (3) and 25.7 (1)°. This difference may account for the fact that two competing reactions are observed in aqueous solution, namely cyclization to form the imide N‐phthaloyl­leucine and hydrolysis of N‐(2‐carboxy­benzoyl)‐l ‐leucine methyl ester to phthalic acid and leucine.  相似文献

In the crystal structure of the l ‐His–cIMP complex, i.e.l ‐histidinium inosine 3′:5′‐cyclic phosphate [systematic name: 5‐(2‐amino‐2‐carboxyethyl)‐1H‐imidazol‐3‐ium 7‐hydroxy‐2‐oxo‐6‐(6‐oxo‐6,9‐dihydro‐1H‐purin‐9‐yl)‐4a,6,7,7a‐tetrahydro‐4H‐1,3,5,2λ5‐furo[3,2‐d][1,3,2λ5]dioxaphosphinin‐2‐olate], C6H10N3O2+·C10H10N4O7P, the Hoogsteen edge of the hypoxanthine (Hyp) base of cIMP and the Hyp face are engaged in specific amino acid–nucleotide (His...cIMP) recognition, i.e. by abutting edge‐to‐edge and by π–π stacking, respectively. The Watson–Crick edge of Hyp and the cIMP phosphate group play a role in nonspecific His...cIMP contacts. The interactions between the cIMP anions (anti/C3′–endo/transgauche/chair conformers) are realized mainly between riboses and phosphate groups. The results for this l ‐His–cIMP complex, compared with those for the previously reported solvated l ‐His–IMP crystal structure, indicate a different nature of amino acid–nucleotide recognition and interactions upon the 3′:5′‐cyclization of the nucleotide phosphate group.  相似文献
