4-hydroxy-2-quinolones. 109. Alkylation of 4-substituted ethyl 2-oxo-1,2-dihydro-quinoline-3-carboxylates

The alkylation of the ethyl esters of 4-methyl, 4-chloro-, and 4-amino substituted 2-oxo-1,2-dihydroquinoline-3-carboxylic acid by ethyl iodide in the system DMF/K₂CO₃ has been studied. A convenient synthesis of 3-alkyl-2,3-dihydro-2,4-dioxo-6-phenyl-, 3-alkyl-2,3-dihydro-4-oxo-6-phenyl-2-thioxo-, and 3-alkyl-2-arylimino-2,3-dihydro-4-oxo-6-phenyl-4H-1,3-thiazines

The reactions of 1-alkylamino-1-alkylthio-3-phenylpropene-3-thiones 3 with thiophosgene and phosgene in toluene, followed by treatment of the reaction mixture with triethylamine gave 3-alkyl-2,3-dihydro-4-oxo-6-phenyl-2-thioxo- 4 , 3-alkyl-2,3-dihydro-2,4-dioxo-6-phenyl-4H-1,3-thiazines 5 , respectively in good to excellent yields. Similarly treatment of compounds 3 with N-arylimidoyl dichloride in benzene at room temperature gave 3-alkyl-2-arylimino-2,3-dihydro-4-oxo-6-phenyl-4H-1,3-thiazines 6 in excellent yields. The reactions of compounds 3 with oxalyl chloride in toluene gave also 5 in good yields.     

Reaction of 2-substituted 4-oxo-5, 6-benzo-1, 3, 2-dioxaphosphorinanes with ethyl pyruvate

The reaction of 2-substituted 4-oxo-5, 6-benzo-1, 3, 2-dioxaphosphorinanes with ethyl pyruvate affords ring-expansion products, i.e., diastereomeric 2-substituted 2, 5-dioxo-6, 7benzo-1, 3, 2-dioxaphosphepanes.Translated fromIzvestiya Akademii Nauk. 4-hydroxy-2-quinolones. 110. Bromination of 1-r-4-hydroxy-2-oxo-1,2-dihydroquinoline-3-carboxylic acid anilides

1-R-4-Hydroxy-2-oxo-1,2-dihydroquinoline-3-carboxylic acid anilides have been prepared. It has been shown experimentally that these compounds are brominated by molecular bromine in glacial acetic acid at position 4 of the anilide fragment. Synthesis of alkyl 4-alkyl-2-hydroxy-3-oxo-3,4-dihydro-2H-1,4-benzoxazine-2-carboxylates as peptidomimetic building blocks

A general synthesis of new alkyl 4-alkyl-2-hydroxy-3-oxo-3,4-dihydro-2H-1,4-benzoxazine-2-carboxylate peptidomimetic building blocks from the corresponding alkyl 3-oxo-3,4-dihydro-2H-1,4-benzoxazine-2-carboxylates through carbanion oxidation is described.     

Synthesis of (S,S)-1,5-bis(4-alkyl-5-hydroxy-2-oxo-3-azapentyl)-1,5-diazacyclooctane

Crown ether derivatives have been extensively developed to provide enhanced structural recognition of substrate alkyl-ammonium cations. In particular a number of investigations of complexation of chiral alkylammonium salts by chiral crown ethers have been reported. The enantioselectivity has been applied in the chromatographic resolution of amino acid perchlorates. Synthetic approaches to 2-substituted 1-oxo- and 3-oxotetrahydroisoquinolines

2-Substituted homophthalimides 2a-c were reduced regioselectively with sodium borobydride to carbinol-lactam intermediates 3a-c , which were dehydrated, followed by hydrogenation, to give 1-oxo-tetrabydroisoquinolines or 3,4-dihydroisoquinolin-1(2H)ones 5a-c . The isomeric 3-oxo-tetrahydro-isoquinolines or 1,4-dihydroisoquinolin-3(2H)-ones 8a-i were obtained in satisfactory yields via heating 3-isochromanone ( 6 ) with the corresponding amines 7a-i in the presence of aluminum chloride.     

Self-Sensitized Photo-Oxygenation of 1-Benzyl-3, 4-dihydroisoquinolines

The air oxidation of 1-benzyl-3, 4-dihydroisoquinolines is shown to be a self-sensitized photo-oxidation proceeding via singlet oxygen.     

4-Hydroxy-2-quinolones. 26. Bromination of 3-substituted 2-oxo-4-hydroxyquinolones

The bromination of 3-alkyl- and 3-ethoxycarbonyl-2-oxo-4-hydroxyquinolones by molecular bromine gave 3-bromo-3-R-2,4-dioxoquinolones. Under analogues conditions, 1-R-2,4-dioxo-3H-quinolone-3-carboxylic acids form 1-R-3-bromo-2-oxo-4-hydroxyquinolones. 4-hydroxy-2-quinolones. 18. Synthesis and antithyroid activity of 1-R-2-oxo-3-(4-oxo-3H-quinazolin-2-yl)-4-hydroxyquinolines

Preparative methods for the synthesis of 1-R-2-oxo-3-(4-oxo-3H-quinazolin-2-yl)-4-hydroxyquinolines based on ethyl esters or nitrites of 1-R-2-oxo-4-hydroxyqidnoline-3-carboxylic acids and anthranilamide or ethyl anthranilate correspondingly were proposed. Synthesis of 2-oxo-3-aryl-4-(1′-hydroxy-2′,2′,2′-trichloro-ethyl) imino-5,5-dimethyloxazolidines and some of their transformations

The condensation of chloral with iminooxazolidines was investigated, and some of the transformations of the products were studied.     

Dioxomolybdenum(VI) complexes containing 1-alkyl-2-ethyl-3-hydroxy-4-pyridin-4(1H)-ones

Dioxomolybdenum(VI) complexes derived from ethyl maltol (2-ethyl-3-hydroxy-4-pyrone) and 1-alkyl-2-ethyl-3-hydroxy-4-pyridin-4(1H)-ones have been prepared and characterized using physical methods including ¹H- and ¹³C-n.m.r. spectroscopy, i.r., elemental analysis, and X-ray diffraction for the pyrone and 3-methylpyridine pyridinone derivatives. The octahedral complexes have the general formula cis-MoO₂L₂ where L = the deprotonated pyrone or pyridinone ligand.     

Synthesis of 3-alkyl-8-substituted- and 4-hydroxy-8-substituted-2,3,4,5-tetrahydro-1H-2-benzazepines

Based on the Schmidt reaction and an iodolactone ring expansion reaction, two different synthetic routes to substituted 2,3,4,5-tetrahydro-1H-2-benzazepines were developed. The Schmidt reaction on 2,3-dihydro-2H-1-naphthalenone ( 1 ) gave 3 , the product resulting from the alkyl group migration, as the major product instead of the tetrazole 2. This prompted the investigation of the Schmidt reaction on aromatic ketones 8 and 12. The product 9 due to alkyl group migration was the major product of the Schmidt reaction on 2-methyl-3,4-dihydro-2H-1-naphthalenone ( 8 ). The β-keto diester 12 gave a mixture of decarb-oxylated lactams after the Schmidt reaction. In this case, the lactam 13 resulting from the migration of the aromatic ring dominated over the other lactam 14. When lactam 14 was subjected to nitration, a single regioisomer was produced and transformed to the bromo alcohol 19. The other approach was based on the single pot ring expansion of the iodolactone 22 to the lactam 23 in the presence of methanolic ammonia. The iodolactone 22 was readily prepared from 2-allylbenzoic acid.     

4-hydroxy-2-quinolones. 179*. Synthesis,structure, and anti-inflammatory activity of 4-hydroxy-1-methyl-2-oxo-1,2-dihydroquinolin-3-ylacetic acid and its derivatives

The synthesis of 4-hydroxy-1-methyl-2-oxo-1,2-dihydroquinolin-3-ylacetic acid has been carried out and the structure and acid properties studied. Preparative methods for its esterification and amidation are proposed. The anti-inflammatory properties of the synthesized compounds have been evaluated.
