Multiple emulsions consisting of water droplets dispersed in an oil phase containing emulsifier which is emulsified in an outer water phase (W/O/W) are of great interest in pharmacology for developing new drugs, in the nutrition sciences for designing functional food, and in biology as model systems for cell organelles such as liposomes. In the food industry multiple emulsions with high sugar content in the aqueous phase can be used for the production of sweets, because the high sugar content prevents deterioration. However, for these emulsions the refractive indexes of oil and aqueous phase are very similar. This seriously impedes the analysis of these emulsions, e.g., for process monitoring, because microscopic techniques based on transmission or reflection do not provide sufficient contrast. We have characterized the inner dispersed phase of concentrated W/O/W emulsions with the same refractive index of the three phases by micro Raman spectroscopy and investigated the composition and molecular distribution in water-oil-water emulsions by means of three-dimensional laser scanning CARS (coherent anti-Stokes Raman scattering) microscopy. CARS microscopy has been used to study water droplets dispersed in oil droplets at different Raman resonances to visualize different molecular species. Water droplets with a diameter of about 700 nm could clearly be visualized. The advantages of CARS microscopy for studying this particular system are emphasized by comparing this microscopic technique with conventional confocal reflection and transmission microscopies.  相似文献

The presented results show the usefulness of Raman spectroscopy in the investigation of polyacetylenes in carrot root. The components are measured directly in the plant tissue without any preliminary sample preparation. Compared with the strong polyacetylene signals the spectral impact of the surrounding biological matrix is weak, except for carotenoids, and therefore it does not contribute significantly to the obtained results. Three different Raman mapping techniques applied here have revealed essential information about the investigated compounds. Using point acquisition several spectra have been measured to demonstrate the complex composition of the polyacetylene fraction in carrot root. The molecular structures of falcarinol, falcarindiol and falcarindiol 3-acetate are similar but their Raman spectra exhibit differences demonstrated by the shift of their -C triple bond C- mode. Line mapping performed along the diameter of transversely cut carrot roots has been used to investigate the relative concentration of polyacetylenes and carotenoids. An area map provides detailed information regarding the distribution of both components. It has been found that high accumulation of polyacetylenes is located in the outer section of the root, namely the pericyclic parenchyma, and in the phloem part close to the secondary cambium. The highest concentration of carotenes is seen in the immediate vicinity to polyacetylene conglomerates.  相似文献

It is the first report of direct, in situ detection of carotenoids at the subcellular level by using Raman microspectroscopy. Single crystals sequestered in a carrot cell were measured using FT-Raman spectrometer equipped with a microscope and 40× objective. The observed characteristic bands centered at 1518 cm−1 and 1156 cm−1 proved the crystals were composed of carotenoids with β-carotene being predominant. The obtained results show the potential of Raman microspectroscopy for identification and analysis of compounds localized in cytoplasm by taking measurements directly from a single plant cell.  相似文献

The formation of the interconnected morphology of open-cell styrene/divinylbenzene (DVB) PolyHIPE copolymers has been studied by scanning electron microscopy on frozen HIPE samples at different stages of polymerisation, a technique known as cryo-SEM. The transition from discrete emulsion droplets to interconnected cells was observed to occur around the gelpoint of the polymerising system. This would suggest that the formation of holes between adjacent cells is due to the contraction of the thin monomeric films on conversion of monomer to polymer, as a result of the higher density of the latter.  相似文献

Digital microfluidics based on electrowetting-on-dielectric (EWOD) has recently emerged as one of the most promising technologies to realize integrated and highly flexible lab-on-a-chip systems. In such EWOD-based digital microfluidic devices, the aqueous droplets have traditionally been manipulated either directly in air or in an immiscible fluid such as silicone oil. However, both transporting mediums have important limitations and neither offers the flexibility required to fulfil the needs of several applications. In this paper, we report on an alternative mode of operation for EWOD-based devices in which droplets enclosed in a thin layer of oil are manipulated in air. We demonstrate the possibility to perform on-chip the fundamental fluidic operations by using such water-oil core-shell droplets and compare systematically the results with the traditional approach where the aqueous droplets are manipulated directly in air or oil. We show that the core-shell configuration combines several advantages of both the air and oil mediums. In particular, this configuration not only reduces the operation voltage of EWOD-based devices but also leads to higher transport velocities when compared with the manipulation of droplets directly in air or oil.  相似文献

Dog rose (Rosa canina L.) is a wild native species in Iran, with a significant genetic diversity. This plant serves as a rich source of vitamin C, anthocyanins, phenolic contents and carotenoids. Rose hips have been used in several food products, as well as perfumery and cosmetics industries. In this research, we investigate biochemical characteristics of five dog rose ecotypes (Kopehjamshid, Zarneh, Miyankish, Aghcheh and Sadeghiyeh), that were collected from the central part of Iran (Isfahan province). Amounts of vitamin C, total carotenoids, total phenolic contents, total anthocyanins, macro and micro minerals were measured. Seed oil are extracted by soxhlet method and analysed by gas chromatography. The macro and micro minerals levels in the fruit vary significantly among these regions. The results of this study demonstrate that dog rose have great diversity and can be used in breeding programmes in order to increase nutrient values as a food resource additive.  相似文献

The mummification ritual in ancient Egypt involved the evisceration of the corpse and its desiccation using natron, a naturally occurring evaporitic mineral deposit from the Wadi Natrun, Egypt. The deposit typically contains sodium carbonate, sodium bicarbonate and impurities of chloride and sulfate as its major elemental components. It is believed that the function of the natron was to rapidly remove the water from the cadaver to prevent microbial attack associated with subsequent biological tissue degradation and putrefaction. Several specimens of natron that were recently collected from the Wadi Natrun contained coloured zones interspersed with the mineral matrix that are superficially reminiscent of extremophilic cyanobacterial colonisation found elsewhere in hot and cold deserts. Raman spectroscopy of these specimens using visible and near-infrared laser excitation has revealed not only the mineral composition of the natron, but also evidence for the presence of cyanobacterial colonies in several coloured zones observed in the mineral matrix. Key Raman biosignatures of carotenoids, scytonemin and chlorophyll have been identified. To understand the mechanism of the photoprotective and antioxidative functions of carotenoids, it is essential to have a profound knowledge of their excited electronic and vibronic states. In the present study we investigate the most powerful antioxidants: β-carotene and lutein by means of resonance Raman spectroscopy. The aim was to study in detail their Raman spectra in solution at room temperature and their changes as a function of temperature. To measure the spectra in their natural environment pyridine has been used as a solvent. It has been chosen because of its polarizability (n=1.5092) which is close to that of membrane lipids and proteins. The temperature dependence of the most intensive ν(1) band in the range from 77 K to 295 K at 514.5 nm excitation has been obtained. It was found that in pyridine the CC stretching frequency, its intensity, line shape, and line width are very sensitive to the temperature (the sensitivity being different for the two studied carotenoids). The observed linear temperature dependence of the CC stretching frequency is explained by a mechanism involving changes of the vibronic coupling and the extent of π-electron delocalization. The different behavior of the temperature-induced broadening of the ν(1) band and its intensity for the two studied carotenoids can be associated with the different nature of their solid matrices: glassy for β-carotene and crystalline-like for lutein, owing to their different chemical structures.  相似文献

Photoprotective nanoemulsions are able to attenuate skin damage from overexposure to the sun, thus avoiding the immediate effects caused by ultraviolet radiation. The global cosmetics market understands that there is a demand and greater acceptance by consumers for formulations containing natural products compatible with the skin. Consequently, there is an increasingly need to develop such products that are safe and effective. Furthermore, there is a growing interest in nanoemulsions (NE) in the pharmaceutical industry, due the versatility of incorporating lipophilic substances into cosmetic formulations. In the present work, oil-in-water photoprotective nanoemulsions containing microbial carotenoids, buriti oil and chemical filters were developed and characterized. The essential physical properties of the droplets, the transmission electronic microscopy (TEM), the sun protection factor (SPF) as well as the stability of the formulations were determined. In vitro phototoxicity was evaluated using Balb 3 T3 with relative cell viability estimated by Neutral Red Uptake, with the Photo Irritation Factor (PIF) and the Medium Photo Effect Factor (MPF) as the measurement parameters. Nanoemulsion 3 (NE3) showed spherical morphology with an average droplet size of 142.11 ± 0.92 nm and polydispersity index (PDI) of 0.198 ± 0.017. This nanoemulsion containing microbial carotenoids and buriti oil exhibited a SPF of 36 ± 1.5. Neutral Red Uptake revealed that the cells kept their viability even after irradiation and those nanoemulsions containing the microbial carotenoids and buriti oil were not phototoxic. The addition of microbial carotenoids and buriti oil in nanoemulsions was positive in increasing the mean SPF values compared to the control formulation.  相似文献

There is growing evidence that a metastable phase of ice, cubic ice, plays an important role in the Earth's troposphere and stratosphere. Cubic ice may also be important in diverse fields such as cryobiology and planetary sciences. Using X-ray diffraction, we studied the formation of cubic ice in pure water droplets suspended in an oil matrix as a function of droplet size. The results show that droplets of volume median diameter 5.6 microm froze dominantly to cubic ice with stacking faults. These results support previous suggestions that cubic ice is the crystalline phase that nucleates when pure water droplets freeze homogeneously at approximately 235 K. It is also shown that as the size of the water droplets increased from 5.6 to 17.0 microm, the formation of the stable phase of ice, hexagonal ice, was favoured. This size dependence can be rationalised with heat transfer calculations. We also investigated the stability of cubic ice that forms in water droplets suspended in an oil matrix. We observe cubic ice up to 243 K, much higher in temperature than observed in many previous studies. This result adds to the existing literature that shows bulk ice I(c) can persist up to approximately 240 K. The transformation of cubic ice to hexagonal ice also showed a complex time and temperature dependence, proceeding rapidly at first and then slowing down and coming to a halt. These combined results help explain why cubic ice forms in some experiments described in the literature and not others.  相似文献

Droplet microfluidics is an enabling platform for high‐throughput screens, single‐cell studies, low‐volume chemical diagnostics, and microscale material syntheses. Analytical methods for real‐time and in situ detection of chemicals in the droplets will benefit these applications, but they remain limited. Reported herein is a novel heterogeneous chemical sensing strategy based on functionalization of the oil phase with rationally combined sensing reagents. Sub‐nanoliter oil segments containing pH‐sensitive fluorophores, ionophores, and ion‐exchangers enable highly selective and rapid fluorescence detection of physiologically important electrolytes (K+, Na+, and Cl?) and polyions (protamine) in sub‐nanoliter aqueous droplets. Electrolyte analysis in whole blood is demonstrated without suffering from optical interference from the sample matrix. Moreover, an oil phase doped with an aza‐BODIPY dye allows indication of H2O2 in the aqueous droplets, exemplifying sensing of targets beyond ionic species.  相似文献

The development of plant protein-based delivery systems to protect and control lipophilic bioactive compound delivery (such as vitamins, polyphenols, carotenoids, polyunsaturated fatty acids) has increased interest in food, nutraceutical, and pharmaceutical fields. The quite significant ascension of plant proteins from legumes, oil/edible seeds, nuts, tuber, and cereals is motivated by their eco-friendly, sustainable, and healthy profile compared with other sources. However, many challenges need to be overcome before their widespread use as raw material for carriers. Thus, modification approaches have been used to improve their techno-functionality and address their limitations, aiming to produce a new generation of plant-based carriers (hydrogels, emulsions, self-assembled structures, films). This paper addresses the advantages and challenges of using plant proteins and the effects of modification methods on their nutritional quality, bioactivity, and techno-functionalities. Furthermore, we review the recent progress in designing plant protein-based delivery systems, their main applications as carriers for lipophilic bioactive compounds, and the contribution of protein-bioactive compound interactions to the dynamics and structure of delivery systems. Expressive advances have been made in the plant protein area; however, new extraction/purification technologies and protein sources need to be found Their functional properties must also be deeply studied for the rational development of effective delivery platforms.  相似文献

Emulsions are widely used to produce sol-gel, drugs, synthetic materials, and food products. Recent advancements in microfluidic droplet emulsion technology has enabled the precise sampling and processing of small volumes of fluids (picoliter to femtoliter) by the controlled viscous shearing in microchannels. However the generation of monodispersed droplets smaller than 1 microm without surfactants has been difficult to achieve. Normally, the generation of satellite droplets along with parent droplets is undesirable and makes it difficult to control volume and purity of samples in droplets. In this paper, however, several methods are presented to passively filter out satellite droplets from the generation of parent droplets and use these satellite droplets as the source for monodispersed production of submicron emulsions. A passive satellite droplet filtration system and a dynamic satellite droplet separation system are demonstrated. Satellite droplets are filtered from parent droplets with a two-layer channel geometry. This design allows the creation and collection of droplets that are less than 100 nm in diameter. In the dynamic separation system, satellite droplets of defined sizes can be selectively separated into different collecting zones. The separation of the satellite droplets into different collecting zones correlates with the cross channel position of the satellite droplets during the breakup of the liquid thread. The delay time for droplets to switch between the different alternating collecting zones is nominally 1 min and is proportional to the ratio of the oil shear flows. With our droplet generation system, monodispersed satellite droplets with an average radius of 2.23 +/- 0.11 microm, and bidispersed secondary and tertiary satellite droplets with radii of 1.55 +/- 0.07 microm and 372 +/- 46 nm respectively, have been dynamically separated and collected.  相似文献

Multiple water-in-oil-in-water (W/O/W) emulsions offer a huge potential as encapsulation systems in different food, cosmetic, and pharmaceutical applications. Because of their complex structure, however, it is difficult to characterize these systems. Typical measurement techniques to determine the size and stability of the inner water droplets encapsulated in the oil droplets show limitations and inaccuracies. Determining the total amount of water in the inner droplets is most often done by indirect methods to date. We describe an analytical method based on differential scanning calorimetry (DSC) for characterizing the total amount of encapsulated water droplets and their stability in W/O/W multiple emulsions. It uses the possibility to directly determine the latent heat of freezing of water droplets of the same size and composition as in the multiple emulsions. The amount of water in the inner droplets of a W/O/W emulsion can thus be calculated very accurately. It is shown that this method enables furthermore detecting multi-modalities in the size distribution of inner water droplets in W/O/W emulsions. Changes in droplet size distribution of the inner droplets occurring during the second emulsification step of processing or during storage can be detected. DSC thus offers a powerful tool to characterize the structure of multiple W/O/W emulsions.  相似文献

Fat perception of food emulsions has been found to relate to in-mouth friction. Previously, we have shown that friction under mouth-like conditions strongly depends on the sensitivity of protein-stabilized emulsion droplets to coalescence. Here, we investigated whether this also implies that oral fat retention depends in a similar manner on the stability of the emulsion droplets against coalescence. We investigate the separate contributions of droplet adhesion and droplet spreading to fat retention at the tongue, as well as the role of saliva. We perform ex vivo (Confocal Raman Spectroscopy; Confocal Scanning Laser Microscopy) experiments using pig's tongue surfaces in combination with human in vivo experiments. These reveal that protein-poor (unstable) emulsions are retained more at the tongue than protein-rich (stable) emulsions. Furthermore, the layer formed by adhering protein-poor droplets is more stable against rinsing. Saliva is found to be very efficient in removing fat and emulsion droplets from the oral surface but its role in fat retention needs further research. We relate our results to the colloidal forces governing droplet adhesion and spreading.  相似文献

Abstract Resonance Raman spectroscopy was used to study the polyene-chain structure of carotenoids in light-harvesting pigment-protein complexes from purple photosynthetic bacteria. When major carotenoids of Rhodobacter sphaeroides were incorporated into the light-harvesting complexes of Chromatium vinosum , they showed a relatively intense Raman band at 965 cm-1 arising from the CH out-of-plane wagging modes of the polyene chain. This result was almost the same as that for the intrinsic carotenoids in Chromatium vinosum , but completely different from that for the intrinsic carotenoids in Rhodobacter sphaeroides . On the other hand, the intrinsic carotenoids of Chromatium vinosum lost the intensity of the 965 cm-1 Raman band upon protein denaturation. These results support the view that the intensity of the 965 cm-1 Raman band reflects distortion of the polyene chain, independent of its chemical structure. The polyene-chain distortion is affected by the apoprotein to which carotenoids are bound. The distortion of the polyene chain is correlated with a decrease in the efficiency of the energy transfer from carotenoids to bacteriochlorophyll.  相似文献

By-products from fruits and are of great interest for their potential use in the food industry due to their high content of bioactive compounds. Herein, we examined the ultrasound-assisted extraction (UAE) of carotenoid and carotenoid esters from papaya pulp and peel using soybean oil and sunflower oil as alternative green solvents. Response surface methodology (RSM) was established to optimize the UAE process. Three independent variables, ultrasonic amplitude (20–60%), time (10–60 min), and co-solvent percentage (ethanol) (5–20%, v/v), were applied. The highest total carotenoid content in the UAE extracts was obtained from papaya pulp extracts (58.7 ± 1.6 and 56.0 ± 1.5 μg carotenoids/g oil) using soybean oil and sunflower oil, respectively (60% amplitude/ 10 min/ 20% ethanol). On the other hand, the highest carotenoid content (52.0 ± 0.9 μg carotenoids/g oil) was obtained from papaya peel using soybean oil applying the UAE process (20% amplitude/ 77 min/ 20% ethanol); a minor content of 39.3 ± 0.5 μg carotenoids/g oil was obtained from papaya peel using sunflower oil at 60% amplitude/ 60 min/ 5% ethanol. Lycopene was the most abundant carotenoid among all individual carotenoids observed in papaya oil extracts, obtaining the highest yields of this carotenoid when papaya pulp and peel were extracted using soybean oil (94% and 81%, respectively) and sunflower oil (95% and 82%, respectively). Great extraction of xanthophyll esters was detected using 20% of ethanol in the vegetable oil extraction solvent (v/v). High correlations (>0.85) was obtained between total carotenoid content and color determination in the UAE oil extracts. UAE vegetable oil extracts enriched with carotenoids from papaya by-products could be useful to formulate new food ingredients based on emulsions with interesting potential health benefits.  相似文献

Light-absorbing pigments from different chemical classes have been identified from the lichens Usnea sp. and Crocodia aurata using Raman spectroscopy supported by quantum mechanical DFT calculations. Raman spectra were obtained directly from the lichen tissues as well as from isolated extracts. Usnic acid, a chemomarker of Usnea spp has been reported together with a minor constituent, namely stictic acid, which has been unambiguously identified by 1H and 13C NMR spectral analysis. The structures of calycin and pulvinic dilactone isolated from Crocodia aurata have been confirmed by single crystal X-ray diffraction. The ubiquitous carotenoids have been characterized by FT-Raman and dispersive Raman microimaging in tissues of Usnea sp. and C. aurata, respectively. The Raman map has revealed the presence of a mixture of carotenoids heterogeneously distributed in the upper layer of C. aurata. In this work we have demonstrated that Raman spectroscopy can be used to monitor aromatic and conjugated polyenic pigments in different layers of lichen tissues.  相似文献

Flax plant of the third generation (F3) overexpressing key genes of flavonoid pathway cultivated in field in 2008 season was used as the plant material throughout this study. The biochemical properties of seed, oil and seedcake extracts from natural and transgenic flax plants were compared. Overproduction of flavonoids (kaempferol), phenolic acids (coumaric, ferulic/synapic) and lignan-secoisolariciresinol diglucoside (SDG) in oil and extracts from transgenic seeds has been revealed providing a valuable source of these compounds for biotechnological application. The changes in fatty acids composition and increase in their stability against oxidation along three plant generations were also detected. The analysis of oil and seedcake extracts was performed using Raman and IR spectroscopy. The wavenumbers and integral intensities of Raman and IR bands were used to identify the components of phenylpropanoid pathway in oil and seedcake extracts from control and transgenic flax seeds. The spectroscopic data were compared to those obtained from biochemical analysis.  相似文献
