A number of 5-R-substituted 3-nitro-1-trinitromethyl-1H-1,2,4-triazoles (R = Me, Cl, Br, N3, NH2, NO2) were synthesized by nitration of the corresponding ω-(5-R-3-nitro-1H-1,2,4-triazol-1-yl)alkan-2-ones with mixtures of concentrated sulfuric and nitric acids.  相似文献

5-Substituted 2-(1-aryl-5-methyl-1H-1,2,3-triazol-4-yl)-1,3,4-oxadiazoles were synthesized by reaction of 1-aryl-5-methyl-1H-1,2,3-triazole-4-carbonyl chlorides with the corresponding 5-substituted 1H-tetrazoles. 5-Methyl-1-phenyl-1H-1,2,3-triazole-4-carbonyl chloride reacted with N′-hydroxybenzimidamides to give 3-aryl-5-(5-methyl-1-phenyl-1H-1,2,3-triazol-4-yl)-1,2,4-oxadiazoles. Reactions of 4-(5-methyl-1H-1,2,3-triazol-1-yl)benzoic acid with N′-hydroxybenzimidamides resulted in the formation of 3-aryl-5-[4-(5-methyl-1H-1,2,3-triazol-1-yl)phenyl]-1,2,4-oxadiazoles.  相似文献

Herein we describe a novel and efficient method for synthesizing the (R)-atropisomer of 3-[3-ethyl-5-(6-phenylpyridin-3-yl)-4H-1,2,4-triazol-4-yl]-2-methylbenzonitrile 1, a novel GlyT1 inhibitor. The diastereomeric salt formation of 3-[3-ethyl-5-(6-phenylpyridin-3-yl)-4H-1,2,4-triazol-4-yl]-2-methylbenzoic acid 7 with (1R,2S)-(?)-2-amino-1,2-diphenylethanol afforded the desired (R)-atropisomer. We also report the determination of the absolute configuration of (R)-7 by powder X-ray diffraction.  相似文献

Palladium–biscarbene complexes derived from N,N′-bis(1,2,4-triazol-1-yl)methane, which bear an alkyl chain functionalized with a hydroxyl group, have been synthesized ([Pd(L1)Br2] (6) and [Pd(L1)I2] (7) [L1 = 1,1′-(3-hydroxypropylidene)bis(4-butyl-4,5-dihydro-1H-1,2,4-triazol-5-ylidene)]). Each product is obtained as a non-equimolecular mixture of two conformers. The hydroxyl group has been replaced by bromide and methanesulphonate and ( [Pd(L2)Br2] [L2 = 1,1′-(3-bromopropylidene)bis(4-butyl-4,5-dihydro-1H-1,2,4-triazol-5-ylidene)] (9)) and ([Pd(L3)Br2] [L3 = 1,1′-(3-methanesulphonyloxypropylidene)-bis(4-butyl-4,5-dihydro-1H-1,2,4-triazol-5-ylidene)] (10)) were obtained, respectively, as mixtures of conformers. All compounds consist of a six-membered metallacyclic structure in a boat conformation. Major conformers present the functionalized chain in the axial position, while in minor conformers it is located in the equatorial position.  相似文献

Methods were developed for the synthesis of complexes of CoII, NiII, and CuII nitrates and chlorides with N-(1-phenylethylidene)-N-(4H-1,2,4-triazol-4-yl)amine (L1) and N′-(4H-1,2,4-triazol-4-yl)benzamidine (L2). The CoII and NiII complexes have a linear trinuclear structure. The CuII complexes are polynuclear. Both ligands are coordinated to the metal ions in a bidentate-bridging mode through the N(1) and N(2) atoms of the heterocycle. In all compounds, the coordination polyhedron can be described as a distorted octahedron. The molecular and crystal structure of the [Ni3(L1)6(EtOH)2(H2O)4](NO3)6·2EtOH·4H2O complex was established.  相似文献

Treatment of 2-(2-oxo-2-arylethylthio)-N-(4H-1,2,4-triazol-3-yl)acetamides with paraformaldehyde in the presence of a base has led to a tandem chemoselective hydroxymethylation followed by cyclization yielding a set of novel 6-aroyl-4-(4H-triazol-3-yl)thiomorpholin-3-ones. The thiomorpholine ring has been found to have a boat like conformation in these compounds as evidenced by NOESY NMR spectrum.  相似文献

13C NMR spectra (20 and 75 MHz, in DMSO-d6) of a series of 1,3-diaryl-3-(1H-1,2,4-triazol-1-yl)- and 1,3-diaryl-2-(1H-1,2,4-triazol-1-yl)prop-2-en-1-ones were registered. It was shown that the chemical shifts of both the carbon atom of the alkene group and C(3) reflect regio- and stereoisomerism of these compounds. Taking this into account the isomeric structures of several 1,3-diaryl-3-(1H-1,2,4-triazol-1-yl)prop-2-en-1-ones were identified and the configurations relative to the double bond of a number of 1,3-diaryl-2-(1H-1,2,4-triazol-1-yl)prop-2-en-1-ones were determined.  相似文献

The reaction of 3-(2,3-dihydro-4-methyl-3-thioxo-4H-1,2,4-triazol-5-ylmethylene)-2-oxo-1,2,3,4-tetrahydroquinoxaline 4 with o-chlorobenzenediazonium chloride gave 3-[α-(o-chlorophenylhydrazono)-2,3-dihydro-4-methyl-3-thioxo-4H-1,2,4-triazol-5-ylmethyl]-2-oxo-1,2-dihydroquinoxaline 6 , whose refluxing in phosphoryl chloride/pyridine afforded 1-(o-chlorophenyl)-3-(2,3-dihydro-4-methyl-3-thioxo-4H-1,2,4-triazol-5-yl)-1H-pyrazolo[3,4-b]quinoxaline 7. The reactions of 6 and 7 with nitrous acid resulted in sulfur extrusion to provide 1-(o-chlorophenyl)-3-(4-methyl-4H-1,2,4-triazol-5-yl)1H-pyrazolo[3,4-b]quinoxaline 8 and 3-[α-(o-chlorophenylhydrazono)-4-methyl-4H-1,2,4-triazol-5-ylraethyl]-2-oxo-1,2-dihydroquinoxaline 9 , respectively.  相似文献

The title compounds 3al have been synthesized by the reaction of thiocarbohydrazide with substituted phenoxy acetic acid to obtained substituted 1,2,4-triazoles (1). Compound 1 was treated with various substituted aromatic aldehydes which results in 4-(substituted benzylideneamino)-5-(substituted phenoxymethyl)-2H-1,2,4-triazol-3(4H)-thiones (2ag), further 2ag is converted to 2-[4-(substituted benzylideneamino)-5-(substituted phenoxymethyl)-4H-1,2,4-triazol-3-yl thio] acetic acid (3al) derivatives by the reaction with chloroacetic acid. All the newly synthesized compounds were evaluated for in vivo anti-inflammatory and analgesic activities. Among the series 2-[4-(2,4-dichlorobenzylideneamino)-5-(phenoxymethyl)-4H-1,2,4-triazol-3-yl thio] acetic acid (3d), 2-[4-(4-dichlorobenzylideneamino)-5-(phenoxymethyl)-4H-1,2,4-triazol-3-yl thio] acetic acid (3e), 2-[4-(2,4-dichlorobenzylideneamino)-5-[(2,4-dichlorophenoxy)methyl]-4H-1,2,4-triazol-3-yl thio] acetic acid (3j) and 2-[5-[(2,4-dichlorophenoxy)methyl)]-4-(4-chlorobenzylideneamino)-4H-1,2,4-triazol-3-yl thio] acetic acid (3k) showed significant anti-inflammatory activity with P < 0.001 (63.4%, 62.0%, 64.1% and 62.5% edema inhibition, respectively), as compared to the standard drug diclofenac (67.0%) after third hour respectively and also compounds 3j, 3k exhibited significant analgesic activity with P < 0.001 (55.9% and 54.9% protection, respectively) and less ulcerogenic activity as compared with standard drug aspirin (57.8%).  相似文献

Reactions of 3-mono- and 3,5-disubstituted 1,2,4-triazoles with a “model” thiirane, 8-bromo-1,3-dimethyl-7-(thiiran-2-ylmethyl)-3,7-dihydro-1H-purine-2,6-diones proceed at the positions N1 and N2 of the triazole ring and yield 7-(5-R-3-R′-1,2,4-triazol-1-yl)methyl- and/or 7-(5-R′-3-R-1,2,4-triazol-1-yl)methyl-1,3-dimethyl-6,7-dihydro[1,3]thiazolo[2,3-f]-purine-2,4-(1H,3H)-diones. 3-Methylsulfonyl-1,2,4-triazole reacted regiospecifically at the position N1 forming 1,3-dimethyl-7-[(3-methyl-sulfonyl-1,2,4-triazole-1-yl)-methyl]-6,7-dihydro[1,3]thiazolo-[2,3-f]purine-2,4(1H,3H)-dione.  相似文献

Abstract

of 1,4-dihydro-4-oxo-1,8-naphthyridine-3-carbohydrazide (4) with substituted phenyl isothiocyanates (5) in ethanol under reflux for 30 min gave thiosemicarbazide derivatives 6, which on cyclization in 2N NaOH under refluxing conditions for 1 h resulted in 3-(5-mercapto- 4-phenyl-4H-1,2,4-triazol-3-yl)-1,8-naphthyridin-4(1H)-one (7). Alternatively, 7 could also be prepared from following sequence of reactions, i.e., 4 → 8 → 7. In another sequence of reactions, condensation of 7 with chloroacetic acid in dimethylformamide (DMF) and K2CO3 as a mild base at 120 °C for 2 h resulted in 2-((5-(1,4-dihydro-4-oxo-1,8-naphthyridin-3-yl)-4-phenyl-4H-1,2,4-triazol-3-yl)sulfanyl)acetic acid (10). The latter, on reaction with substituted o-phenylenediamine (11) in 6N HCl for 4 h yielded 3-(5-((1H-benzo[d]imidazol-2-yl)methylthio)-4-phenyl-4H-1,2,4-triazol-3-yl)-1,8-naphthyridin-4(1H)-one (12). Alternatively, 12 could also be prepared by reacting 7 with 13 in DMF and K2CO3 as a mild base at 120 °C for 2 h, followed by oxidation with H2O2 resulting in the corresponding sulfonyl derivatives 14.  相似文献

The formation by 1H-1,2,4-triazoline-3-thiones in dilute chloroform solution of n-σ*-complex with molecular iodine of the composition C2H2N3S·I2 was studied by electronic spectroscopy in the UV and visible regions (log β = 2.14±0.05). By the XRD method the crystal and molecular structure of the product of chemical interaction of the thione with molecular iodine in ethanol, 1,2-di(1H-1,2,4-triazol-3-yl) disulfide, was determined.  相似文献

The amination of 5-R- and 6-R-3-X-1,2,4-triazines (R = C6H5, t-C4H9, X = SCH3, SO2CH3, N+ (CH3)3, Cl) by potassium amide in liquid ammonia has been studied. In all reactions the formation of the corresponding 3-amino-1,2,4-triazines takes place; in some reactions by-products were found: from 5-phenyl- and 5-t-butyl-3-(methylthio)-1,2,4-triazine a ring contracted product i.e. 5-phenyl and 5-t-butyl-3-(methylthio)-1,2,4-triazole, from 6-phenyl-3-(methylthio)-1,2,4-triazine the dimer 3,3′-bis-(methylthio)-6,6′-bisphenyl-5,5′-bi-1,2,4-triazine and from 5-t-butyl-3-(trimethylammonio)-1,2,4-triazine chloride compound bis-(5-t-butyl-1,2,4-triazin-3-yl)- amine. Furthermore the conversion of 5-phenyl- and 5-t-butyl-1,2,4-triazin-3-one into the corresponding 3-amino compound by treatment with phenyl phosphorodiamidate (PPDA) was studied. A 15N study of these aminations showed that nearly all compounds undergo substitution according to both SN(AE) and SN(ANRORC) processes. The contribution of each of the competitive mechanisms to the amination is strongly influenced by the character of the leaving group.  相似文献

Ethyl 2-amino-4-methyl-5-(1H-1,2,4-triazol-1-yl)thiophene-3-carboxylate 2, obtained from Gewald reaction of 1-(1H-1,2,4-triazol-1-yl)acetone 1 with ethyl cyanoacetate and sulfur, was transferred into iminophosphorane 3. Further reaction of 3 with aromatic isocyanates gave carbodiimides 4, which were treated subsequently with a secondary amine to give 6-(1H-1,2,4-triazol-1-yl)-thieno[2,3-d]pyrimidin-4(3H)-ones 6 in good yields in the presence of a catalytic amount of sodium ethoxide.  相似文献

A series of novel arylantimony derivatives of analogues of demethylcantharimide with the formulae ArnSbL(5−n) and ArnSbL(5−n)(LH=N-hydroxy-demethyldehydrogencantharimide, LH=N-hydroxy-demethylcantharimide, n=3, 4; ArC6H5, 4-CH3C6H4, 3-CH3C6H4, 2-CH3C6H4, 4-ClC6H4, 4-FC6H4) were synthesized and characterized by elemental analysis, IR, 1H NMR and mass spectroscopy. The crystal structures of (C6H5)4SbL, (4-CH3C6H4)3SbL2 and (3-CH3C6H4)3SbL2 were determined by X-ray diffraction. The in vitro antitumor activities of all compounds against six cancer cells are reported.  相似文献

The preparation of the deep-blue diamagnetic dinitrogen complexes (Cp2TiR)2N2 with R=C6H5, o-, m-, p-CH3C6H4, C6F5, CH2C6H5 is described. Their chemical and physcial properties confirm the formulation in which the R groups are σ-bonded to the Cp2Ti moiety, and the two nitrogen atoms are equivalent. The heats of formation of the complexes from Cp2TiR and N2 in toluene have been determined from spectrophotometric data; for R=C6H5, o-, m-, p-CH3C6H4, C6F5, CH2C6H5, the values are ?18, ?9, ?17, ?20, ?17 and ?14 kcal·mol?1, respectively. The solid complexes vary markedly in thermal stability, and are extremely air sensitive. The complexed nitrogen can be completely reduced with sodium naphthalene; after hydrolysis of the products, NH3 and N2H4 are obtained. In the thermolysis of the solids, some of the nitrogen is reduced.  相似文献

A mild procedure for the preparation of 2-(5-amino-1-aryl-1H-1,2,3-triazol-4-yl)-2-iminoacetonitriles and 2-(5-amino-1-aryl-1H-1,2,3-triazol-4-yl)-2-oxoacetonitriles was achieved by the reaction of diaminomaleonitrile and phenyl/substituted phenyl diazonium chlorides. 4-Nitrophenyl diazonium chloride afforded 2-amino-3-(3-(4-nitrophenyl)triaz-1-en-1-yl)maleonitrile. Triazole iminoacetonitrile and maleonitrile derivatives were reacted further with excess acetone and benzaldehyde with a catalytic amount of 1,8-diazabicyclo[5.4.0]undec-7-ene to yield 5-(5-imino-2,2-dimethyl-2,5-dihydrooxazol-4-yl)-3-aryl-3H-1,2,3-triazol-4-amine and (E)-N-benzylidene-5-(5-imino-2-aryl-2,5-dihydrooxazol-4-yl)-3-aryl-3H-1,2,3-triazol-4-amine, respectively. Two competitive reactions, i.e., nucleophilic substitution and nucleophilic addition, were observed when triazole oxoacetonitrile and maleonitrile derivatives were reacted with hydroxylamine hydrochloride in the presence of sodium acetate.  相似文献

A new modified BINOL, (S)-3-[(1H-1,2,4-triazol-1-yl)methyl]-1,1′-binaphthol, was prepared. In the presence of titanium tetraisopropoxide, this ligand showed moderate catalytic properties for the asymmetric addition of diethylzinc to aldehydes. By treating rac-3-[(1H-1,2,4-triazol-1-yl)methyl]-1,1′-binaphthol with excess titanium tetraisopropoxide, a novel trinuclear titanium(IV) complex was obtained. A C3 axis along Ti1–Ti2–Ti3 is present in the molecule.  相似文献

The reactions of 1,2-bis(tetrazol-5-yl)benzene (1), 1,3-bis(tetrazol-5-yl)benzene (2), 1,4-bis(tetrazol-5-yl)benzene (3), 1,2-(Bu3SnN4C)2C6H4 (4), 1,3-(Bu3SnN4C)2C6H4 (5) and 1,4-(Bu3SnN4C)2C6H4 (6) with 1,2-dibromoethane were carried out by two different methods in order to synthesise pendant alkyl halide derivatives of the parent bis-tetrazoles. This lead to the formation of several alkyl halide derivatives, substituted at either N1 or N2 on the tetrazole ring, as well as the surprising formation of several vinyl derivatives. The crystal structures of both 1,2-[(2-vinyl)tetrazol-5-yl)]benzene (1-N,2-N′) (1b) and 1,3-bis[(2-bromoethyl)tetrazol-5-yl]benzene (2-N,2-N′) (5d) are discussed.  相似文献

Combining different nitrogen-rich heterocycles into a molecule can fine-tune its energetic performance and physical properties as well as its safety for use in energetic materials. Here, 1,2,4-oxadiazole was incorporated into 1,2,4-triazole to construct new energetic backbones. 3-(5-Amino-1H-1,2,4-triazol-3-yl)-1,2,4-oxadiazol-5-amine ( 5 ) was designed and synthesized. Nitramino-functionalized N-(5-(5-amino-1,2,4-oxadiazol-3-yl)-3H-1,2,4-triazol-3-yl)nitramide ( 6 ) and N-(5-(5-(nitramino)-1,2,4-oxadiazol-3-yl)-3H-1,2,4-triazol-3-yl)nitramide ( 7 ) were also obtained, and two series of corresponding nitrogen-rich salts were prepared, leading to the creation of new energetic compounds. All derivatives were fully characterized, and five of them were further confirmed by X-ray diffraction. The theoretical calculations, energetic performance, safety, and the main decomposition gaseous products of 1,2,4-triazole-1,2,4-oxadiazole-derived energetic materials were studied. Compound 7 and its dihydroxylammonium salt ( 7 c ) exhibited prominent detonation performance comparable to that of RDX while possessing satisfying thermal stabilities and mechanical sensitivities.  相似文献
