A novel approach for analysis of amino acids in individual erythrocytes was established.In this method, the derivatization reagent was in~oduced into the living cells by electroporation.After derivatization, the amino acids in a single cell were determined by capillary electrophoresis with laser-induced fluorescence detection.  相似文献

Chiral separation of dausyl amino acids by capillary electrophoresis using mixed selectors of Mn(ll)-L-alanine complex and β-cyclodextrin (β-CD) was studied. Resolution was considerably superior to that obtained by using either Mn (Ⅱ)-L-alanine complex or β-CD alone. The effects of separation parameters, such as pH value of buffer solution, capillary temperature, the concentration of Mn (Ⅱ)-L-alanine complex, the types of CD and ligand on the migration times and resolutions were investigated. Six different transition metal complexes,Cu(Ⅱ), Zn(Ⅱ), Co(Ⅱ), Ni(Ⅱ), Hg(Ⅱ) and Cd(Ⅱ)-L-alanine complexes have been employed and compared with Mn(Ⅱ)complex. Differences in retention and selectivity were found.The substitution of Cu(Ⅱ), Zn(Ⅱ), Co(Ⅱ) and Ni(Ⅱ) for Mn(Ⅱ) resulted in a better chiral resolution while Hg(Ⅱ) and Cd(Ⅱ) showed poorer resolution abilities. The chiral separation mechanism was also discussed briefly.  相似文献

A new method for the quantitative determination of the enantiomers of salsolinol, an endogenous neurotoxin, was reported. Enantiomeric separation of salsolinol was obtained by capillary zone electrophoresis using hydroxypropyl-β-cyclodextrin (HP-β-CD) as chiral selector. Coupled with UV detection, this separation was applied to the determination of R,S-salsolinol in dried banana chips, and both R- and S-salsolinol were found at 40 ktg/g level.  相似文献

N-Conjugated antigen was synthesized and polyclonal antibody with high specificity was obtained from immunizing animals. With this polyclonal antibody, a rapid and efficient CEIA-LIF method was developed to determine the free morphine in urine of abusers. The detection limit was calculated to be 40 ng/mL. Simulated urine samples were analyzed with good recoveries, which showed the feasibility of its application in specific morphine determination in urine of morphine abusers.  相似文献

In this paper, we first presented a novel method for quantification of circulating DNA in human serum based on capillary zone electrophoresis with laser-induced fluorescence detection (CZE-LIF). The serum was digested by proteinase to release free DNA, and then CZE-LIF system was used for the quantification of total circulating DNA. This method was successfully used to quantify the circulating DNA levels in sera from healthy individuals and certain cancer patients.We found the significantly elevated circulating DNA levels in certain prostate cancer patients. Our results demonstrated that CZE-LIF system has good linearity, excellent sensitivity (0.5 ng/mL DNA),satisfactory reproducibility (RSDs in one day and between days were both less than 5%) and reliability, and is well suitable to the quantification of the circulating DNA in human serum or plasma.  相似文献

The mixtures of two polymers, poly (N,N-dimethylacrylamide) (PDMA) and polyvinylpyrrolidone (PVP) were synthesized and used as the separation medium for double-stranded and single-stranded DNA fragments by capillary electrophoresis with UV detector. On optimal conditions, 2%w/v PDMA 2%w/v PVP can be used to separate the doublet 123/124bp in pBR322/Hae Ⅲ Markers.  相似文献

A capillary electrophoresis (CE) method has been firstly used for the separation of the therapeutically important xanthones from Securidaca inappendiculata. The separation of the nine xanthones was systematically optimized with respect to pH, concentration of running buffers,addition of sulfated β-CD,applied voltage and column temperature.Baseline separation was achieved for the nine xanthones in less than 15 minutes using a background electrolyte consisting of 200 mmol/L borate (pH9.5) and 10 mmol/L sulfated β-CD.  相似文献

制备了一种新的β-环糊精衍生物固定相2,6-二-O-戊基-3-O-[(甲基)5(烯丙基)2]-β-CD,并对其气相色谱分离性能进行研究.实验显示,该固定相具有良好的柱表面性能和较强的色谱分离能力,对一些芳香族位置异构体的分离能力优于2,6-二-O-戊基-3-O-烯丙基-β-CD衍生物,对10余种手性物质显示出较好的选择性能,且对α-位取代的丙酸酯类手性分离效果明显优于杂环类β-CD衍生物.与2,6-二-O-戊基-3-O-烯丙基-β-CD固定相的分离性能比较表明,β-CD 3位羟基部分甲基基团的引入能增强对芳香族位置异构体的选择能力,但不能明显改变烯丙基衍生物的手性分离能力.  相似文献

以环氧氯丙烷(EP)为交联剂,合成水溶性的β-环糊精聚合物(β-CDpoly-met),提出了一种以β-CDpolymer为手性选择剂,在毛细管电泳中分离未衍生芳环氨基酸对映体的方法。考察了手性选择剂浓度、背景电解质溶液pH、柱温及分离电压对分离的影响。在优化的实验条件下,使3种氨基酸对映体得到了基线分离。  相似文献

Quasi-interpenetrating network of polyacrylamide (PAA) and polyvinylpyrrolidone (PVP) had been successfully used for single-base resolution of double-stranded DNA (0.76 for 123 bp/124 bp) and single-stranded DNA fragments (0.97 for 123 b/124 b) with UV detection. This quasi-IPN (interpenetrating network) sieving matrix showed low viscosity (23.5 mPa·s at 25 ℃) and decreased with increasing temperature. This polymer also exhibited dynamically coating capacity and could be used in the uncoated capillary. The effects of temperature and electric field strength on the DNA separation of quasi-IPN matrix were also investigated and found that the temperature and electric field strength could markedly affected the mobility behavior of DNA fragments. This polymer matrix has also applied to separate the bigger DNA fragments by capillary electrophoresis with UV detection. Under the denaturing conditions, this matrix separated the samples with last fragment of 1353 base in 40 rain, in which the doublet of 309/310 base was partial separated and the resolution was 0.88.  相似文献

Enantiomeric separation of epinephrine and salbutamol was investigated by micellar electrokinetic chromatography employing β-cyclodextrin as chiral additive in ammonium chloride-ammonia solution. The analytes were detected by electrochemistry using gold microelectrode at 0.65 V versus SCE reference electrode. The effects of detection potential,concentration of β-cyclodextrin, concentration of sodium dodecyl sulfate, pH value of electrolyte and applied voltage were discussed.  相似文献
