Reaction of 1‐amino‐3‐arylpyrido[1,2‐a]benzimidazole‐2,4‐dicarbonitrile (1) with dimethylformamide‐dimethylacetal (DMF‐DMA) gave 1 ‐[N,N‐(dimethylaminomethylene)amino]‐3‐arylpyrido[1,2‐a]benzimidazole‐2,4‐dicarbonitrile (2). Compounds (1) reacted with triethylorthoformate yielding 1‐[N‐(ethoxymethylene)amino]‐3‐arylpyrido[1,2‐a]benzimidazole‐2,4‐dicarbonitrile (3). 3‐Amino‐4‐imino‐5‐aryl‐6‐cyanopyrimido[5′,4′:5,6]pyrido[1,2‐α] benzimidazole (4) was synthesized via condensation of either (2) or (3) with hydrazine hydrate. Reactions of (4) with acetic anhydride, ethyl chloroformate or aryl isothiocyanate yielded the respective derivative of the new ring system namely 1,2,4‐triazolo[2″,3″:6′,1′]pyrimido[4′,5′:2,3]pyrido[1,2‐a]benzimidazole (5–7).  相似文献

Stable 3-acetyl derivatives of imidazo[1,2-a]benzimidazole were synthesized by the action of acetic anhydride on 2,9-disubstituted imidazo[1,2-a]benzimidazole. The former were also obtained by cyclization of 1-alkyl (aralkyl) -3-acylmethyl-2-iminobenzimidazoline hydrobromides in acetic anhydride in the presence of anhydrous sodium acetate. 3-Benzoyl-substituted imidazo[1,2-a]benzimidazoles, which are unstable in acidic media, were synthesized by the action of benzoyl chloride in the presence of pyridine or excess starting imidazo [1,2-a] benzimidazole.See [1] for communication XI.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 1, pp. 121– 125, January, 1976.  相似文献

3-Alkoxycarbonyl-2-arylimidazo[1,2-a]benzimidazoles were synthesized by alkaline cleavage of 3-trichloromethyl ketones of the imidazo[1,2-a]benzimidazole series, which are formed by acylation of this three-ring system with trichloroacetyl chloride. The same compounds were also obtained by esterification of the corresponding 3-carboxylic acid. The haloform reaction with 3-acetyl-2-phenylimidazo[1,2-a]benzimidazole proceeds anomalously and leads to bis(9-methy1-2-phenylimidazo[1,2-a]benzimidazo1-3-yl)-2-buten-1,4-dione, the structure of which was confirmed by independent synthesis.See [1] for communication XV.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 522–525, April, 1978.  相似文献

3-Ethoxycarbonylmethyl- and 3-cyanomethyl-1-amino-2-methylbenzimidazolium chlorides cyclized at treatment with acetic anhydride in the presence of potassium carbonate to afford a mixture of derivatives of pyrazolo[1,5-a]benzimidazole and 4-aminopyrrolo[1,2-a]benzimidazole. Therewith in the first case prevails the process of pyrazole ring fusion, and in the latter pyrrole ring is fused. 3-Aroyl(thenoyl)methyl-1-amino-2-methylbenzimidazolium bromides under the same conditions are converted into 1-aroyl(thenoyl)-2-methyl- and 2-4-acetamido-3-acetylpyrrolo[1,2-a]benzimidazoles.  相似文献

Photocyclization of 2-(pentafluoroanilino)-, 2-(4-chloro-2-iodoanilino)-, 2-(2-chloro-4-iodo-anilino)-4,6-dimethylpyrimidines, as well as 2-(2-chloro-3-methylanilino)pyridine was used to prepare con-densed azinobenzimidazoles, including previously unknown 8-chloro-1,3-dimethylpyrimido[1,2-a]benzimidazole and 9-methylpyrido[1,2-a]benzimidazole. With isomeric chloroiodoanilinopyrimidines as example it was shown that the iodine atom affects photocyclization direction. Quaternization of 6,7,8,9-tetrafluoro-1,3-dimethylpyrimido[1,2-a]benzimidazole, 8-chloro-1,3-dimethylpyrimido[1,2-a]benzimidazole, and 9-methylpyrido[1,2-a]benzimidazole with alkylating agents and C-H activity of alkyl groups in the quaternary salts in reactions with orthoformic ester were studied.  相似文献

Three types of compounds were synthesized from carbendazim ( 1 ), a benzimidazole derivative (Scheme 1 ). They included a group of esters at N1 prepared by treating carbendazim with isocyanates bearing ester groups ( 2a , 2b , 2c ), carboxyalkyl‐1,2,3,4‐tetrahydro‐s‐triazino[1,2‐a]benzimidazole‐2,4‐dione esters ( 3a and 3b , 3d and 3c derived from 3a . The antitumor potencies of the N1 esters were in the range of 7 to 40 μM, which compares favorably with carbendazim, but their water solubilities were low. The s‐triazine derivatives showed activity against pancreatic tumor cells, and one of them ( 3b ) was active in mice, but they were not effective against other tumor types. Treatment of carbendazim with 3‐bromopropionyl chloride produced 1‐methoxycarbonyl‐4‐oxo‐1,2,3,4‐tetrahydropyrimido[1,2‐a]benzimidazole ( 4 ), which gave 1‐(3‐aminopropionyl)benzimidazole 2‐methylcarbamates, substituted on the amino nitrogen ( 5a , 5b , and 5d ), when treated with amines. These products showed some antitumor activity in cell cultures, and an ethoxy derivative ( 5c ), obtained by treating 1‐methoxycarbonyl‐4‐oxo‐1,2,3,4‐tetrahydropyrimido[1,2‐a]benzimidazole with sodium ethoxide, was active in the 67–150 μM range. Some of the new compounds had good water solubility. Carbendazim kills tumor cells by inhibiting tubulin; however, s‐triazine 3b , which differs from it in size and functional groups, does not act by this mechanism.  相似文献

The synthesis of a novel benzimidazole derivative with a long‐chain‐ester substituent, namely methyl 8‐[4‐(1H‐benzimidazol‐2‐yl)phenoxy]octanoate, (3), is reported. Ester (3) shows evidence of aggregation in solution and weak gelation ability with toluene. The octan‐1‐ol solvate, methyl 8‐[4‐(1H‐benzimidazol‐2‐yl)phenoxy]octanoate octan‐1‐ol monosolvate, C22H26N2O3·C8H18O, (4), exhibits a four‐molecule hydrogen‐bonded motif in the solid state, with N—H…O hydrogen bonds between benzimidazole molecules and O—H…N hydrogen bonds between the octan‐1‐ol solvent molecules and the benzimidazole unit. The alkyl chains of the ester and the octan‐1‐ol molecules are in unfolded conformations. The phenylene ring is canted by 10.27 (6)° from the plane of the benzimidazole ring system. H…C contacts make up 20.7% of the Hirshfeld surface coverage. Weak C—H…π interactions involving the benzimidazole alkyl chain and three aromatic rings are observed.  相似文献

Cyclization of ethyl 2-ethoxymethylidene-3-oxo-3-polyfluoroalkylpropionates with benzimidazol-2-amine in boiling 1,4-dioxane followed two concurrent pathways with participation of fluoroacyl and ethoxycarbonyl fragments and formation of, respectively, ethyl 4-hydroxy-4-polyfluoroalkyl-1,4-dihydropyrimido-[1,2-a]benzimidazole-3-carboxylates and 3-polyfluoroacylpyrimido[1,2-a]benzimidazol-4-ols. Dihydropyrimido[1,2-a]benzimidazole derivatives undergo dehydration to give ethyl 4-(polyfluoroalkyl)pyrimido[1,2-a]benzimidazole-3-carboxylates, whereas the hydroxy group in 3-polyfluoroacylpyrimido[1,2-a]benzimidazol-4-ols is capable of being replaced by the amino group of the second benzimidazole molecule with formation of 4-(1H-benzimidazol-2-ylamino)-3-polyfluoroacylpyrimido[1,2-a]benzimidazoles.  相似文献

以苯并咪唑(HL1)和3,6 二氯哒嗪在钠催化下反应制得配体3-氯-6-(苯并咪唑-1-基)哒嗪(L2);采用溶液法和水热法,L1分别与含有银、铜过渡金属盐反应合成了3种新的有机金属苯并咪唑配合物[AgL1n(1),[Cu4(HL1)4(O)(Cl)6](2)和{[Cu(HL1)2]·SiF6}(3);采用溶液法,L2与Zn(NO3)2·6H2O经配位反应合成了配合物{[Zn(L2)3(NO3)]NO3}(4),其结构经IR,元素分析和X-射线单晶衍射表征。结果表明:1通过苯并咪唑桥连形成一维链状结构,2由苯并咪唑和氯离子桥接形成四核结构,3和4均显示一个单核结构,3的金属中心原子采用二配位构型,4的金属中心原子采用四配位构型。此外,研究了配体HL1与1,及配体L2与4的荧光性能。结果表明:与HL1相比,1的最大发射波长发生了红移;而4相对于配体L2其最大发射峰发生明显蓝移。  相似文献

Isochromylium cations containing easily detached groupings (NHCOR, NH2, OCH3) in the 3 position react readily with o-phenylenediamine. The reaction is accompanied by opening of the benzopyrylium ring and condensation of the resulting compound at one or two amino groups of o-phenylenediamine. Benzimidazo[3,2-b]isoquinolinium salts were synthesized alternatively by acylation of 2-(3,4-dimethoxybenzyl)benzimidazole. The HeI photoelectron (PE) spectra of indole ( 1 ), benzimidazole ( 2 ), indazole ( 3 ), 3-chloro-indazole ( 4 ), imidazo[1,2-b]pyridazine ( 5 ), 6-chloroimidazo[1,2-b]pyridazine ( 6 ), 2-phenyl-imidazo[1,2-b]pyridazine ( 7 ), 2-phenyl-6-chloroimidazo[1,2-b]pyridazine ( 8 ), tetrazolo[1,2-a]-pyridine ( 9 ) and 8-cyanotetrazolo[1,5-a]pyridine ( 10 ) have been recorded. The spectra of 2–10 are of special interest for studying lone pair interactions. The assignment of the PE spectra submitted here, conjointly with the electronic structure of the studied compounds is discussed on the basis of molecular orbital calculations.  相似文献

Cyanoethylhydrazones of 2-aroylmethyl-1H-benzimidazoles undergo recyclization on reaction with trifluoroacetyl anhydride to give 3-aryl-1-cyanoethyl-5-(2-trfluoroacetaminoanilino)pyrazoles. The debenzylation of N-benzylimidazo[1,2-a]benzimidazoles with sodium in liquid ammonia was studied. The debenzylation of the 2-phenyl-substituted derivative is accompanied by hydrogenation of the outer imidazole ring to give 2-phenyl-2,3-dihydro-1H-imidazo[1,2-a]benzimidazole, the alkylation of which in alkaline media and neutral media, respectively, gives the 1-methyl derivative and the 9-methyl derivative. The 1-methyl derivative was subjected to quaternization and bromination; the latter proceeds in the condensed benzene ring. By reaction of 1,2-diaminobenzimidazole with 1,3-diketones, acetoacetic ester and its derivatives, we have synthesized substituted 1,2,4-triazepino[2,3-a]benzimidazole and 5H-1,2,4-triazepino[2,3-a]benzimidazol-4-one. By reaction of 5H-2-methyl-1,2,4-triazepino[2,3-a]benzimidazol-4-one with aromatic aldehydes or phenyldiazonium chloride, we have obtained 3-arylidene(phenylazo) derivatives of this compound, and by reaction with P2S5 we have obtained 5H-2-methyl-1,2,4-triazepino[2,3-a]benzimidazole-4-thione. We have shown that when reacted with ammonia, primary or secondary amines, the latter forms 4-amino-substituted 2-methyl-1,2,4-triazepino[2,3-a]benzimidazole.  相似文献
